ABSTRACT

Objective To study the early prediction of infection in acute pancreatitis in rats by plasma procalcitonin (PCT) and c-reactive (CRP) detection. Methods Eighty SD rats were randomly assigned into acute infected pancreatitis group (I, n=20), pancreatitis control group (C, n=40) and sham-operated group (S, n=20). Blood samples were collected pre- (0h) and post-operatively (12h, 24h and 48h). Plasma CRP was analyzed by ELISA. Plasma and liver PCT was detected by Western blot. Results (1). Ascitic infection occurred in all the group B rats and 16 of 40 rats of group C (analyzed as group C1), and did not occur in the other 20 of 40 rats of group C (analyzed as group C2) and group S. (2). The plasma CRP concentrations elevated gradually after the model setup in group B and C1, which were significantly higher at 48h than those in group C2 and group S. (3). PCT was detected in high levels in plasma and liver tissues in group B and C1 at 48h post-operatively, and they were sighificantly higher than those in group C2 and group S.Conclusions PCT can predict early infection of acute pancreatitis, and detection of PCT combined with plasma CRP may help in the differentiation of acute infected pancreatitis. The liver may be an important organ for synthesis of PCT.

ABSTRACT

0.05).(3)Pathological score of C group at 1h and 3h was significantly higher than the A group(P0.05).Conclusions (Urokinase) LAI can improve the microcirculation of rats with SAP,but can not completely prevent the(pathological) changes of SAP.

ABSTRACT

Objective To study the protective mechanism of adenosine on intestinal barrier function in acute hemorragic necrotizing pancreatitis (AHNP) pigs; Methods Twelve small pigs were equally devided into two groups randomly after the AHNP model sitted up: (1) AHNP controll group (group A) and adenosine treated group (group B). The intestinal blood flow, intestinal permeability, bacterial culture and endotoxin in portal blood were compared between the two groups in pre and post AHNP. Results (1)The intestinal blood flow was dramatically decreased in group A, and much higher in group B than that in group A at the 8 h, 24 hour and day 7 after ANHP model was setted up (P0.001,P

ABSTRACT

Objective To study the expression of cyclooxygenase-2(COX-2) in pancreatic tissues of severe acute pancreatitis(SAP) in rats, and explore the possible role of COX-2 in the pathogenesis of the SAP. Methods Forty adult male Sprague-Dawley rats were randomly divided into SAP group(n=20) and control group(n=20). At 12h after operation, acites, serum levels of amylase activity, tumor necrosis factor-?(TNF-?),TXB 2 and 6-keto-PGF 1? and the expression of COX-2 in pancreatic tissue were detected.the pathological changes of pancreatic tissues were observed and scored under microscope;the survival rate in 72h and mortality in 24h in SAP and NC group were recorded. Results At 12h after the induction of SAP,the serum amylase activity,TNF-?,TXB 2 and 6-keto-PGF 1? were significantly higher in SAP group than those in NC group(P

ABSTRACT

Objective To investigate the expressions of IL-8,MCP-1 and MIP-1? and their biological significance in pancreatic carcinoma and chronic pancreatitis, and investigate the relationships of their expressions in pancreatic cancer tissues. Methods Immunohistochemical method of avidin-biotin complex was used to examine surgically removed specimens from 51 cases of pancreatic carcinoma and 10 cases of chronic pancreatitis. Results The positive rates of expression and the scorings of IL-8,MCP-1,MIP-1? were significantly higher in pancreatic carcinoma than that in chronic pancreatitis(P

ABSTRACT

Objective To study the clinicopathological features of chronic inflammatory mass lesion of the pancreas. Methods The clinical data of 37 patients with focal chronic inflammatory mass lesion of the pancreas were retrospectively studied.Seventeen cases congruent with the standard clinical diagnostic of chronic pancreatitis were separated into group A;and Whipple procedure was carried out in two cases, resection of the body and tail of the pancreas in 2 cases, local resection in one case, and choledochojejunostomy in 12 cases. Those without the stander clnical features of Group A but with the features of pancreatic tumor were separated into Group B;and Whipple procedure was carried out in 4 cases, choledochojejunostomy in 16 cases. Results In group A,except for the local mass lesion, sclerosis of the whole pancreas was found in 88.2% of cases.Pathological examination showed proliferation of fibrous tissue with associated inflammation,as well as acinar atrophy, remnant islet cells, and ductular dilatation and focal calcification. While in Group B,only a local mass lesion of the pancreas was found in 19 cases. The pathological features were characterized histologically by proliferation of fibrous tissue with associated moderate or marked inflammation. No pancreatic carcinoma was found during 1 to 12 years follow-up of 33 cases. Conclusions Chronic inflammatory mass lesion of the pancreas showed the clinicopathological features of pseudotumoral pancreatitis.Internal drainage by choledochojejunostomy is suggested as its effective management.

ABSTRACT

Objective To investigate the role of the spleen in the pathogenesis of acute pancreatitis(AP). Methods The studied rats were divided randomly into four groups:Sham operation group, splenectomy group, AP group and AP with splenectomy group. The rats were sacrificed at 3h,6h and 12h after operation, and spleen and pancreas were removed for examination. Light microscopy was used to examine pancreatic tissues for pathologic change and to assess pathology score. Immunohistochemical method was used to observe the activity of splenic NF-?B(p65) in the rats with AP.Results The pancreas pathology score in the AP with splenectomy group was significantly lower than that in AP group at 6h(7.83?0.753vs 9.67?1.211) and 12 h (9.67?0.816 vs13?0.894) after operation (P

ABSTRACT

Objective To study the damaging effect of tumor necrosis factor-alpha (TNF-?) and Interleukin-1?(IL-1?) on brain of rats in acute pancreatitis(AP). Mothods AP pancreatitis model in rats was induced by retrograde injection of 5% sodium taurocholate into the pancreatobiliary duct(1ml/kg body weight), then pretreated with normal saline (NS) or TNF-??IL-1?, TNF-? plus IL-1? respectively. Animals were killed at 5 h after drugs administration. The changes of brain water contents, leukocyte accumulation and adhesion were measured, and pathological studies of pancreas and brain were also performed. Results In groups with inflammatory cytokines pretreated, brain water contents, leukocyte accumulation and adhesion increased more significantly than those in control group and in NS pretreated group(P0.05). Conclusions Inflammatory cytokines such as TNF-? and IL-1? play important roles in the formation and acceleration of the brain damage in acute pancreatitis.
