ABSTRACT

Herein, a strategy is proposed for the simultaneous determination of primary coumarins in Peucedani Radix (Chinese name: Qianhu). The methodology consists of three consecutive steps: 1) Semi-preparative LC in combination with a home-made automated fraction collection module to fragment the universal metabolome standard into ten fractions (Frs. I-X); 2) LC-accurate MS/MS and quantitative H NMR spectroscopy conducted in parallel to acquire the qualitative and quantitative data of each fraction; 3) Robust identification and quantification of components by use of LC coupled to multiple reaction monitoring. In this final step, the most significant fractions (Frs. III-X) were pooled to serve as the pseudo-mixed standard solution. Meticulous online parameter optimization was performed to obtain the optimal parameters, including ion transitions and collision energies. Concerns were particularly paid onto pursuing the parameters being capable of monitoring -specific isomers, notably praeruptorin E 3'-isovaleryl-4'-angeloylkhellactone. The quantitative performance of the method was validated according to diverse assays. Eleven primary coumarins (-) were unambiguously identified and absolutely quantified, even though no external reference compound was used. Above all, the integrated strategy not only provides a feasible pipeline for the quality assessment of Peucedani Radix, but more importantly, shows the potential for authentic compound-free quantitative evaluation of traditional Chinese medicines.

ABSTRACT

Peucedanum praeruptorum is a traditional Chinese medicine, which has been recorded in Chinese Pharmacopoeia. Phytochemical studies have shown that the active ingredients of P. praeruptorum can treat anemopyretic cold and cough with abundance of phlegm. Furthermore, the herb is well-known for the treatment of respiratory diseases as well as pulmonary hypertension, and shows great medical values involved in cardiovascular diseases, especially. This paper shows several research achievements involved in plant resources and its adulterants and substitutes identification based on pertinent literatures. DNA barcoding technology shows strong discriminability for medicinal plant and medicinal materials identification of Peucedani Radix. In this paper, recent research advances for identificating Peucedani Radix adulterants and substitutes were elaborated based on modern molecular biotechnology and traditional identification method, merits and drawbacks and practicability of each method are summarized at the same time. The main purpose is to provide new research ideas and solutions for rapid, reliable, and accurate identification of Peucedani Radix.

ABSTRACT

Peucedani Radix is the root of Angelica decursiva Franchet et Savatier (=Peucedanum decursivum Maximowicz) or Peucedanum praeruptorum Dunn in several Asian countries. The coumarins contained in Peucedani Radix were quantitatively analyzed using HPLC-DAD to develop a simultaneous determination for the quality control of A. decursiva and P. praeruptorum. For quantitative analysis, four major coumarins contained in these medicinal plants were assessed. Nodakenin (1), nodakenetin (2), praeruptorin A (3), and praeruptorin B (4) were separated with a Phenomenex Luna C18 column (5 µm, 4.6 × 250 mm) under the gradient conditions using distilled water with 0.1% phosphoric acid and acetonitrile with 0.1% phosphoric acid as the mobile phase, at a flow rate of 1.0 ml/min and a detection wavelength of 330 nm. This method was fully validated for linearity, accuracy, precision, recovery, and limit of detection and quantification. As a result, A. decursiva and P. praeruptorum were clearly classified by the quantification of four major coumarins in extracts. Also, the pattern recognition analysis based on HPLC indicates that all of the samples were largely clustered into two groups. Therefore, it is possible to distinguish between A. decursiva and P. praeruptorum and contribute to quality control.

ABSTRACT

Objective: To establish a UPLC/Q-TOF MS method for analyszing the chemical constituents of total coumarins extract from Peucedani Radix (the rhizome of Peucedanum praeruptorum) and to study its preliminary pharmacodynamics. Methods: The chromatographic column was Acquity UPLC BEH C18, the mobile phase was composed of 0.1% formic acid (A) and acetonitrile (B) with gradient elution; The detection wavelength was set at 321 nm; The ionization mode was ESI+; And the mass scan range was m/z 100-1 000. Using the rat model of type 2 diabetes mellitus (T2DM) induced by streptozocin (STZ), the effect of total coumarins on reducing blood-lipid and blood-sugar was investigated in the experiment. Results: Qianhucoumarin D, peucedanocoumarin II, praeruptorin A, peucedanocoumarin I, praeruptorin B, and praeuptorin E in Peucedani Radix coumarin could be well separated and identified in one run. The preliminary pharmacodynamic results show that the total coumarins had an effect of reducing the level of fasting blood glucose, cholesterol, triglycerides, LDL, and liver index (P < 0.05); On the other hand, it could improve the level of HDL (P < 0.05). The above results prompt that coumarin extract of Peucedani Radix could improve the carbohydrate and lipid metabolism of T2DM, then play a role in the treatment of T2DM. Conclusion: A UPLC/Q-TOF MS method has been established to separate and identify the total coumarin extract of Peucedani Radix, the preliminary pharmacodynamic study shows that the total coumarin extract of Peucedani Radix had the function of reducing blood sugar and blood lipid significantly.

ABSTRACT

Objective To establilsh the quality standard for Qingfei Yihuo tablet. Methods TLC was performed to identify Sophora flavescens Ait, Peucedani and Cortex Phellodendri. HPLC was used to determine Baicalin. ODS column was used. The mobile phase consisted of methanol-water-phosphoric acid (47∶53∶0.2). The flow rate was 1.0 mL/min and column temperature was at 25 ℃. The detecting wave length was at 280 nm. Results The characteristic for identification by TLC was distinct and hightly specific. Baicalin could be determined by HPLC. The linearity of Baicalin was good in the range of 0.163~0.975 ?g (r=0.999 9). The average recovery of Baicalin was 97.43% and RSD was 1.97% (n=6). Conclusion The methods of identification and quantification is simple and reproducible. It can be used effectively for the quality control of Qingfei Yihuo table.
