ABSTRACT

OBJECTIVE: To establish a determination method for content of polysaccharide in Acanthopanax senticosus, and to conduct cluster analysis and ultrasonic extraction technology optimization. METHODS: The content of polysaccharide in A. senticosus was determined by phenol-sulfuric acid method. Cluster analysis was conducted by using SPSS 23.0 software of the polysaccharide in A. senticosus from 17 habitats. The ultrasonic extraction technology was optimized with L9(34)orthogonal test using extraction temperature, the ratio of material to liquid, extraction time as factors, the content of polysaccharides as index, and then validated. RESULTS: The linear range of glucose ranged from 0.007 75 to 0.151 mg/mL (r=0.999 1); the limits of quantification and detection were 2.854, 0.856 μg/mL; RSDs of precision, stability, and repeatability tests were less than 2%; recovery rates of the sample were 98.41%-101.58%(RSD=1.23%,n=6). Cluster analysis results showed that 17 batches of samples could be clustered into three classes; S1, S6, S10, S12 and S13 were clustered into one class; S3-S5 and S7 were clustered into one class; and the rest samples were clustered into one class. The optimal ultrasonic extraction technology was extraction temperature 55 ℃, ratio of material to liquid 1 ∶ 10 (g/mL), extraction time 35 min. Results of validation tests showed the content of the polysaccharide under the best process condition was 4.36%(RSD=0.920%,n=3). CONCLUSIONS: Established method is simple,reproducible and suitable for determination of polysaccharide in A. senticosus; the optimized ultrasonic extraction technology is stable and feasible.

ABSTRACT

Objective :To optimize the extraction of polysaccharide from the residue of Periplaneta americana by response surface method and study the effect of P. americana polysaccharide (PAP) on wound healing, so as to provide ideas for the comprehensive utilization of P. americana residues. Methods On the basis of single factor experiment, Box-Behnken response surface method was used to design the best extraction process. And the monosaccharide composition of polysaccharide was analyzed. Polysaccharide concentrations of 0.1, 0.05, and 0.025 g/mL were applied to the wound injury model mice. The wound healing rate was calculated, and hematoxylin-eosin (HE) and Sirius Red staining were used to evaluate the wound healing degree. Results :The optimum extraction technology of PAP were as follows: The concentration of NaOH was 0.02 mol/L; And the extraction time was 1.65 h; The extraction temperature was 62 ℃. The transfer rate of polysaccharide extracted by this process was 13.68%. The polysaccharide of the P. americana consisted of seven monosaccharides of mannose, rhamnose, glucuronic acid, galacturonic acid, glucose, galactose, and fucose. Wound healing experiment showed that wound healing rate of the polysaccharide group on day 3, 6, 9, and 14 was higher than that of the model group, and HE and Sirius Red staining showed that the polysaccharide could promote the formation of collagen and granulation tissue, thus promoting the wound healing. Conclusion: The optimized extraction process of PAP from P. americana residues by response surface method was reliable, with high extraction rate. And PAP could promote wound healing in mice, suggesting that the polysaccharide from P. americana residues could be used as wound related products.

ABSTRACT

Objective To establish the assay method for the total polysaccharide in Pudi Enema.Methods Phenol-sulfuric acid method was used for chromogenic reaction.The content of total polysaccharide was measured by UV spectrophotometry at 488.8 nm.Results The total polysaccharides calibration curve was at the range of 0~22.635 mg/L, with regression function being Y=0.062 06 X-0.003 34(r=0.999 8).The recovery of calycosin was 98.36%(RSD=2.34%).Conclusion This method is sensitive,rapid,accurate and reliable.It can be used to assay the content of total polysaccharide in Pudi Enema.

ABSTRACT

OBJECTIVE:To establish a method for the content determination of saccharide in polysaccharides containing galact-uronic acid based on phenol-sulfuric acid method combined with correction factor method. METHODS:The determination condi-tion of phenol-sulfuric acid was optimized. A series of standard curves were drawn with glacturonic aid-glucose mixed control with different mass ratio. The content of saccharide in Codonopsis pilosula polysaccharides CPP1b samples containing galacturonic acid was determined. According to regression equation of galacturonic acid-glucose ratio of 0-100%,the correction factor was calculated by using C. pilosula polysaccharides CPP1b as the reference polysaccharide,and the results of content determination of saccharide were corrected. The rationality of this method was verified by using mixed monosaccharide control with same composition as C. pi-losula polysaccharides CPP1b. RESULTS:The correction factor of C. pilosula polysaccharide CPP1b to glucose was 3.33;in vali-dation test,the content of saccharide in mixed monosaccharide control with same composition as C. pilosula polysaccharides CPP1b was 103.47%. RSDs of precision and stability tests was <1%. The recoveries ranged 93.52%-107.35%(RSD=5.09%,n=6). CONCLUSIONS:The established method can accurately determine the content of saccharide in C. pilosula polysaccharides CPP1b containing galacturonic acid.

ABSTRACT

Objective:To study the boiling water brewing extraction of astragalus polysaccharides in order to solve the problems of long cycle , high energy consumption and poor clarity in the traditional extraction , and lay foundation for the industrial production . Methods:Using the extraction yield of astragalus polysaccharide as the evaluation index and phenol -sulfuric acid method applied to de-termine the content of astragalus polysaccharide , the boiling water brewing extraction was used for astragalus polysaccharides .The effects of brewing time , boiling water amount and brewing times on the polysaccharide content were studied by single factor experi -ments, and then an orthogonal design method was used to screen the optimum technology parameters .The traditional water decoction extraction process was employed as the control .Results:The optimum conditions of boiling water brewing were as follows:adding 9-fold amount of water and soaking 60 min for three times.The astragalus polysaccharides extraction yield (4.81) of the optimal technology was higher than that of the traditional water decoction extraction (4.06%).Conclusion:Boiling water brewing method used to extract astragalus is with high extraction yield , simple operation , short cycle and low energy consumption , the color of astragalus polysaccha-ride is light, and it is clear after dissovled in water , which is superior to the traditional water decoction extraction method , and provide a new method with broad application prospect for the preparation of astragalus related preparations .

ABSTRACT

OBJECTIVE:To compare the content of polysaccharide from Dendrobium nobile cultivated in Guizhou and Sichuan,and to establish content determination method of polysaccharide from D.nobile.METHODS:Polysaccharide was extracted from D.nobile using water extraction alcohol precipitation.The content of polysaccharide from D.nobile cultivated in different areas was determined by phenol-sulfuric acid method with glucose as standard substance at UV wavelength of 490 nm.RESULTS:The linear range of polysaccharide from D.nobile was 1.25~8.75 ?g?mL-1(r=0.999 4).The average recovery of D.nobile cultivated in Chishui of Guizhou province was 101.6%(RSD=1.9%,n=6),and the content of polysaccharide was 11.456%;the content of polysaccharide from D.nobile cultivated in Hejiang of Sichuan province was 9.226%.CONCLUSIONS:Phenol-sulfuric acid method is simple,reliable,accurate,sensitive and stable for the content determination of polysaccharide from D.nobile.D.nobile cultivated in Chishui of Guizhou province is the best choice for the extraction of polysaccharide from D.nobile.

ABSTRACT

Objective To compare the polysaccharide content of pseudostellaria heterophylla which produced in various areas and in different parts of them. Methods The polysaccharide content of pseudostellaria heterophylla was determined by phenol-sulfuric acid method. Results The polysaccharide content of pseudostellaria heterophylla produced in Fujian is the highest, Jiangsu and Guizhou are second-class. The polysaccharide content has some disparity in the bodies and in the roots of pseudostellaria heterophylla. Conclusions The polysaccharide content in the bodies differs 1.96% with that in the roots. This experiment provides theory foundation for scientific preparation of pseudostellaria heterophylla.

ABSTRACT

OBJECTIVE:To compare the differences of two methods in the determination of Ganoderma or Ganoderma extract and the related products.METHODS:The content of Ganoderma in samples was determined by phenol-sulfuric acid method vs.anthrone-sulfuric acid colorimetric method,and the differences of the two methods were compared.RESULTS:The two methods showed similar determination results.The linear range of the standard Polysaccharide solution was 0.005~0.1 mg?mL-1(rphenol=0.999 4,ranthrone=0.999 7).CONCLUSION:Both of the two methods can be used to determinate the content of Ganoderma.

ABSTRACT

OBJECTIVE: To study on the effect of different extracting methods on the content determination of polysaccharide from Caulis Dendrobii.METHODS: Polysaccharide was extracted from Caulis Dendrobii respectively by reflux extraction,improved reflux extraction,ultrasound wave extraction and ultrasonic-assisted thermal reflux extraction.The content of polysaccharide was determined by phenol-sulfuric acid method.RESULTS: The determined results using these methods were as follows:(13.473?0.634)%,(15.692?0.743)%,(11.87?0.536)% and (16.29?0.407)%.CONCLUSION: The content determination of polysaccharide from Caulis Dendrobii was associated with the extraction method.The polysaccharide from Caulis Dendrobii could be extracted more thoroughly and rapidly by ultrasonic-assisted extraction technology.Therefore this method could be used to pretreat the sample in content determination of Caulis Dendrobili polysaccharide.

ABSTRACT

OBJECTIVE:To establish phenol-sulfuric acid method for the determination of the content of total polysaccharides in Weile granules.METHODS:The samples were deproteinized by Sevag method,colored by phenol-sulfuric acid method(water bath at 60 ℃ for 30 min),and the content of total polysaccharides in Weile granules was determined with Glucose as control.RESULTS:The linear range of Glucose was 20.47~102.36 ?g?mL-1(r=0.999 4),with an average recovery of 99.09%(RSD=0.92%,n=6).The content of total polysaccharides in Weile granules was 41.63%.CONCLUSION:This method was simple,accurate and reproducible,and suitable for quality control of Weile granules.

ABSTRACT

Objective To investigate the change of the content of polysaccharides in crude and processed Fructus Corni,thus to approach the process mechanism of Fructus Corni.Methods The content of polysaccharides in the crude and processed Fructus Corni was determined by phenol-sulfuric acid method.Results After processing with wine,the content of polysaccharides of Fructus Corni decreased from 10.12 %(content of total polysaccharidesbeing 51.41 %) to 5.91 %(content of total polysaccharidesbeing 53.10 %),decreased by 41.60 %as compared with that in the crude Fructus Corni.Conclusion After processing with wine,the content of polysaccharides decreases markedly.The results provide certain evidence for approaching the process mechanism of Fructus Corni.

ABSTRACT

AIM: To measure and compare the content of glycyrrhizic polysaccharides in Glycyrrhiza in three different growth time. METHODS: The contents of polysaccharides were determined by phenol-sulfuric acid method and by reference to glucose, and wavelenth in spectrophotometer was set at 490 nm. RESULTS: There was difference of the content of the extracted polysaccharides among Glycyrrhiza for 1, 2 and 3 year, amounted to 11.75%, 11.07%, 7.88%, respectively.
