ABSTRACT

OBJECTIVE Insomnia is the most fre-quent sleep disorder worldwide and the clinical applica-tion of therapeutic drugs has various adverse effects. In recent years, drugs developed from natural herbs have become potential alternative therapies for insomnia.Nuciferine, one of the main bioactive components obtained from the lotus leaves, has been reported to possess extensive pharmacological activities. However, its hypnotic and sleep regulatory effects have rarely been reported.Hence, this study was intended to investigate the pharma-cological effects of nuciferine and its mechanisms of action in insomnia.METHODS The hypnotic and seda-tive effects of nuciferine were investigated using the eval-uation of locomotor activity test and pentobarbital-induced sleep test in normal and serotonin(5-HT)depletion-induced insomniac mice.Furthermore, the sleep regulatory effects, including sleep time, sleep architecture, and δ-wave power spectral density, were explored using elec-troencephalography/electromyogram(EEG/EMG)-based sleep profiling in normal rats. Finally, the mechanisms of the hypnotic and sedative effects of nuciferine were explored usingin vivoand in silico experiments.RESULTS Nuciferine reduced locomotor activity and prolonged pen-tobarbital-induced sleep time in a dose-dependent man-ner in normal and insomniac model mice.Nuciferine sig-nificantly increased the total sleep time and non-rapid eye movement(NREM)sleep time, inhibited NREM sleep fragmentation, and improved delta power between 0.5 Hz and 1 Hz in normal rats. The results of molecular experiments showed that nuciferine could increase the 5-HT content and 5-HT1A receptor level in the hypothala-mus of insomnia model mice.CONCLUSION This study combined network pharmacological prediction and experi-mental pharmacological techniques to discover the seda-tive-hypnotic effect of nuciferine for the first time Nucif-erine can ameliorate sleep disorder in mice with insom-nia,possibly via serotonergic system.Nuciferine may rep-resent a novel treatment that alleviate the insomnia-like symptoms by modulating 5-HT system.

ABSTRACT

Objective To explore the role of functional connectivity( FC) between the subgenual anterior cingulate cortes(sgACC) and limbic cortices in predicting the efficacy of early antidepressant treat-ment. Methods Resting state magnetoencephalography( MEG) data were collected from 32 patients with major depressive disorder(MDD) and 20 matched healthy control subjects at baseline.According to whether the HAMD17total score reduction rate reached 50% after 2 weeks, the patients were divided into responder group and non-responder group. The difference of the FC,the relationship between the FC and the change rate of HAMD17were explored. Results Compared with controls,MDD patients showed decreased connectivity of the sgACC with the right hippocampus(MNI coordinate:x=30,y=-3,z=-24; F=5.11,P=0.028),the right amygdala(MNI coordinate:x=22,y=-2,z=-15; F=6.90,P=0.012) and the left amygdala(MNI coordi- nate:x=-24,y=-6,z=-16; F=5.75,P=0.020).Compared with non-responders,the connectivity of the sgACC with the right hippocampus(MNI coordinate:x=30,y=-3,z=-24;F=8.74,P=0.0065),the right a-mygdala(MNI coordinate:x=22,y=-2,z=-15; F=12.417,P=0.0016) were higher in responders.Pretreat-ment connectivity of the sgACC with the right hippocampus(r=-0.50,P=0.0066),the right amygdala(r=-0.56,P=0.0018) were negatively correlated with clinical improvement.ROC analyses confirmed the value of the connectivity of sgACC with the right hippocampus(area under curve(AUC)=0.71,P=0.042),the right amygdala(AUC=0.73,P=0.029) in classifying responders versus non-responders in 2 weeks of treatment. Conclusion The decreased FC of the sgACC with the right hippocampus and the right amygdala at baseline is associated with a better improvement, which can potentially serve as a treatment outcome biomarker in ear-ly curative effect of depression.

ABSTRACT

ABSTRACT Purpose: To evaluate intraocular pressure (IOP) and extraocular orbital vessels with color Doppler ultrasound (CDU) and investigate the effects of obesity on retrobulbar blood flow. Methods: Fifty-nine patients were included in this prospective study. Patients were divided into two groups according to body mass index: Group 1 (31 obese patients) and Group 2 (28 non-obese patients). IOP was measured with a Goldmann applanation tonometer, and CDU was used to evaluate the retrobulbar vessels. Results: The mean IOP was 18 ± 6.68 mmHg in the obese group and 13.71 ± 1.60 mmHg in the control group (p<0.001). When the CDU values for the central retinal artery were compared between the groups, the pulsatility index was found to be significantly lower in the obese group than in the control group (p<0.001). When the CDU values for the ophthalmic artery (OA) were compared between the groups, the peak systolic velocity (p<0.001) and end-diastolic velocity (p=0.002) values were found to be significantly lower in the obese group than in the control group. Conclusions: Obese patients have a higher mean IOP and lower flow velocity than non-obese patients. Increased IOP together with decreased retrobulbar blood flow, particularly in obese individuals, may increase the risk of glaucoma development.

ABSTRACT

The fatty acid (FA) profiles of fish species with a similar genetic or geographic origin are important quality parameters that attract bioecological interest because they are influenced by the nutritional habits of the species. The aim of this study was to determine the proximate compositions and FA profiles of heart, brain and muscle tissues from three fish species (Lutjanus purpureus, Lutjanus synagris and Ocyurus chrysurus) captured from the Amazonian Atlantic Coast. In addition to performing nutritional analysis of the FA profiles, principal components analysis (PCA) was performed as a chemometric tool to discriminate among the species. Lutjanus synagris had the highest protein content (18.23%), while O. chrysurus possessed the highest lipid content (4.25%). All of the nutritional quality parameters (the n-6/n-3 and Σ polyunsaturated FA/Σ saturated FA ratios and dietary indices of atherogenicity and thrombogenicity) of the FA profiles of the three species are of interest to consumers. In general, the predominant FA in the tissues of all species analyzed was palmitic acid, followed by stearic acid. Oleic acid was predominant among the monounsaturated FAs, while docosahexaenoic acid was the most prevalent polyunsaturated FA. Chemometric analysis was an effective tool for fish identification. PCA showed that the FA profiles of the brain tissues were optimal for discriminating among the species, with O. chrysurus showing the most different FA profile from the others. The muscle FA profile was the most useful for identifying L. purpureus.

ABSTRACT

Objective:This study aimed to establish a mouse model of breast cancer by inoculating human breast cancer cells into mice with normal immune function. Methods:Forty female BALB/C mice were randomized into four groups, with 10 mice in each group. The four groups were established according to the dosage of cyclophosphamide and prednisone, namely, the control group, low dose group, medium dose group, and high dose group. The mouse models of breast cancer were established by injecting human breast cancer cells into the fat pad of the right second breast of mice in the groups. Mice in the four groups were observed based on the time of tumorigenesis, rate of tumor formation, tumor imaging and pathological features, and metastasis of vital internal organs. Results:In the high dose group, the time of tumor formation was lower than that of the other groups, but the rate of tumor formation was high. Some visceral metastases occurred in the mice. By contrast, the medium dose group revealed completely opposite results. No death and tumor formation in both the control and low dose groups were reported. Conclusion:A human breast carcinoma model in mice was successfully established. Using this model, the onset and development of breast cancer could be much better imitated in the normal immune system of mice.

ABSTRACT

The innovation of Chinese materia medica (CMM) is the central driver of CMM modernization. The application of modern theories and techniques in life sciences to innovating CMM research remains to be an urgent issue to be solved. Currently, the international R&D for drug is stepping into a new trend, in which these transformations are going through a signal target to multiple drug combination and active constituents selection by pharmacodynamics to optimize the design with pharmacokinetics. In the face of changes, the pace of CMM R&D is slow. Based on the R&D content of system biology and Huangqin Decoction extract (PHY906), the "preceded pharmacokinetics" concept is introduced. Taking pharmacokinetics as the tool to rapidly screen the potential constituents with the aims at CMM R&D. Furthermore, why the "preceded pharmacokinetics" is needed and how to start the innovation model are preliminarily discussed in this paper.

ABSTRACT

Objective To explore the the analytical method for the quality difference of Radix Salviae Mihiorrhizae (RSM) injection. Methods Parallel intercomparison experiment was carried out in different batches of RSM injection samples which had different degrees of adverse drug reaction. The samples were analyzed by gel size exclusion chromatog-zraphy and C18 HPLC fingerprint. Subtractive analysis was used to reveal the information of the non-low molecular weight phenolic acids. Results There were obvious differences between the results of size exclusion chromatography. The amount of the non-low molecular weight phenolic acids from different batches of the samples was also different, which was correlated with the degrees of acute toxicity in guinea pigs. Conclusion The amount of the non-low molecular weight phenolic acids varies with different batches of Radix Salviae Mihiorrhizae injection. It is suggested that non-low molecular weight phenolic acids can be used for the quality control of Radix Salviae Mihiorrhizae injection. For the first time, the perspective that the corresponding relationship between the fingerprint and their toxicities of Traditional Chinese medicine injection may be one of the most important fields is put forward.

ABSTRACT

The method for analysis and determination the cleavage of soybean sterol, in which the soybean sterol was degraded and the products androst-1,4-diene-,17-dione (ADD) and androst-4-ene-3,17-dion (AD) were developed by Liquid Chromatography-mass Spectrometry. The HPLC conditions adopted were: a All- tima ODS-2 column (250 mm?4.6 mm, 5 ?m), a mobile phase consisted of menthanol-water (70:30), a flow rate of 1.0 mL/min, a room column temperature. and the detective wavelength was 244 nm.The ZMD Micromass electrospray ionization (ESI)-mass spectrometer was employed. In such conditions the corre- sponding HPLC chromatogram and MS spectrum were obtained. The method has a linear ranger of 0.01 mg/mL ~ 0.09 mg/mL, R2 =0.9999, the recoveries of ADD and AD were 102.6% and 105.90%, the RSD of ADD and AD were 3.02%, 3.5% and 3.08%, 3.24%. This method showed high sensitivity, accuracyand easy to perform. It is suitable to analysis the process cleavage of soybean sterol as well as quality control of product.
