ABSTRACT

Diversos compuestos bioactivos de los alimentos se han empleado en el tratamiento alterativo de la diabetes mellitus. El jugo de granada posee un alto contenido en compuestos fenólicos a los cuales se les atribuyen propiedades biológicas como hipolipemiante, hipoglucemiante y protector del tejido pancreático. El objetivo de este estudio fue evaluar el efecto del libre acceso de jugo de granada sobre los niveles de glucosa, colesterol, triglicéridos e integridad del tejido pancreático in vivo. Se emplearon 18 ratas macho Wistar inducidas a hiperglucemia con 60 mg estreptozotocina/kg de peso corporal intraperitoneal (IP). Se formaron tres grupos experimentales. El grupo HS expuesto a solución de sacarosa, el grupo HJG expuesto a jugo de granada y el grupo HSI expuesto a solución de sacarosa y tratamiento con insulina. Los resultados mostraron que, las ratas del grupo HJG consumieron jugo de granada durante 21 días, lo que resultó en la reducción de los niveles de glucosa con respecto a su valor inicial de 417 a 356 mg/dL, no de manera significativa (p>0.05). Los niveles de lípidos mostraron una reducción no significativa al finalizar la intervención (p>0.05). El estudio histológico del páncreas en el grupo HJG mostró conservación de la arquitectura pancreática y presencia de islotes de Langerhans; mientras que el grupo HS mostró extensa necrosis pancreática y el grupo HSI mostró daño intermedio con escasos islotes de Langerhans. Se sugiere que el jugo de granada posee efectos hipoglucemiantes y protege el tejido pancreático en ratas hiperglucémicas inducidas(AU)


Diverse bioactive compounds of foods have been used in the alternative treatment of diabetes mellitus. Pomegranate juice has a high content of phenolic compounds to which biological properties are attributed as lipid-lowering, hypoglycemic and protective of pancreatic tissue. The objective of this study was to evaluate the effect of free access of pomegranate juice on the levels of glucose, cholesterol, triglycerides and the integrity of pancreatic tissue in vivo. Eighteen male Wistar rats were induced to hyperglycemia with 60 mg streptozotocin/kg of body weight intraperitoneal (IP). Three experimental groups were formed. The HS group exposed to sucrose solution, the HJG group exposed to pomegranate juice and the HSI group exposed to sucrose solution and insulin treatment. The results showed that the rats of the HJG group consumed pomegranate juice for 21 days, which resulted in the reduction of glucose levels from its initial value of 417 to 356 mg/dL, not significantly (p>0.05). Lipid levels showed a non-significant reduction at the end of the intervention (p>0.05). The histological study of the pancreas in the HJG group showed conservation of the pancreatic architecture and the presence of islets of Langerhans; while the HS group showed extensive pancreatic necrosis and the HSI group showed intermediate damage with few islets of Langerhans. It is suggested that pomegranate juice has hypoglycaemic effects and protects pancreatic tissue in induced hyperglycemic rats(AU)

ABSTRACT

Aims: The consumption of foods and beverages containing probiotic microorganisms is a growing, global consumer trend. In this research, production of probiotic pomegranate juice containing Lactobacillus plantarum and Lactobacillus delbrueckii was studied. Study Design: Plackett-Burman statistical design was used to evaluate the impact of eleven process variables on the viability of both probiotics. Impact of incorporation of grape juice, tomato juice and pomegranate peel extract as well as phenolic compounds and vitamins have been investigated. Place and Duration of Study: Department of Food Science and Technology, Varamin Branch, Islamic Azad University, Varamin, Tehran, Iran, between Sep 2012 and July 2013. Methodology: Pomegranate juices were inoculated with probiotic bacteria and their survival was evaluated every week by pure plate method. The effect of 11 variables (in two levels) on survival of bacteria by the statistical design of Plackett-Burman was evaluated. For this purpose, 12 treatments in triplicate by the Minitab (version = 11.0) software at significant levels α= .01 were analyzed. Results: The highest survival rate of L. plantarum (4.74 ×106 CFU/mL) and L. delbrueckii (4 ×106 CFU/mL) was obtained by 10% v/v inoculation of a 48 h inoculum culture in MRS broth medium to enriched pomegranate juice (10% v/v Grape juice, 5% v/v tomato juice, 0.1% v/v pomegranate peel extract and 2.0 g.L glucose) which was inoculated in anaerobic condition for 72 h at 37ºC and kept for 2 weeks at environment temperature. Sensory evaluation shows the probiotic juice was accepted by consumers with no significant difference in comparison to control in terms of taste, odour and overall acceptability (P>.05). Conclusion: The results of this study suggest that grape, tomato juices and pomegranate peel extract exert a protective effect on L. plantarum and L. delbrueckii viability under acidic condition of pomegranate juice and storage time, which was associated with the chemical composition of them. This study indicates that develop of probiotic pomegranate juices with acceptable viability and stability of the probiotic is possible.

ABSTRACT

Objective To explore the influence of pomegranate juice on cognitive behavior of chroniccerebral hypoperfusion in rats and its mechanism.Methods 48 healthy male Wistar rats were selected and randomly divided into 3 groups, sham operation group (SO), permanent bilateral persistence commancarotid artery occlusion or two-vesselocclusion (2VO) group, pomegranate juice group.Chronic cerebral hypoperfusion model was prepared in rats by permanent bilateral common carotid artery ligation.Given the intervention after operation 1 weeks, before the 2 groups were given by sugar water intragastric ( every 100 mL syrup containing 25 g,sucrose 2 g, fructose glucose 2 g) with pomegranate juice, pomegranate juice group ( polyphenol content of 2.4-4.8 mg/mL) were given by gavage,2 mL/d, a total of 7 weeks.8 th weeks after surgery, cognitive behavior ability of 3 groups was tested by space exploration target quadrant swimming time after an interval of 1day;CA1 region of 8-hydroxy deoxyguanosine (8-OHdG), 4-hydroxyl nonenal ( 4-HNE ) expression of 3 groups in cortex and hippocampus detected by immunohistochemical method; observed whether it could improve the chronic cerebral hypoperfusion in rats learning, memory ability, or had antioxidative stress injury.Results On the 4th day and 5th day, the time of rats in SO to find the platform and the shortest time, group 2VO for the longest time, compared with statistical significance between the three groups (P<0.01); the 3 groups of rats in the target quadrant emergence time in sham operation group, the longest (10.05+1.688)s, 2VO group has the shortest time (6.343 ±1.220) s, and there was statistical significance (P<0.05).The integral optical density of positive cells in 3 groups of rats cortex and hippocampal CA1 region of 4HNE, the value of the 8-OHdG IOD is the lowest in the sham operation group, 2VO group was the highest, with statistical significance.Pomegranate juice intervention, cortex of rats and hippocampal CA1 region of 8-OHdG, 4-HNE decreased significantly ( P <0.05).Conclusion Chronic cerebral hypoperfusion may cause oxidative stress injury, pomegranate juice can improve the low irrigation learning and memory ability of rats with chronic cerebral, decrease 8-OHdG and 4-HNE expression may play a protective function through antioxidative stress pathway.
