ABSTRACT

Objective@#To investigate the clinicopathological features of mammary analogue secretory carcinoma (MASC) of salivary glands, and its diagnosis, differential diagnosis, immunohistochemistry and molecular pathology.@*Methods@#Seventeen cases of MASC were enrolled, with 9 cases of salivary acinar cell carcinoma and 18 cases of adenoid cystic carcinoma as control groups from Nanjing General Hospital from 1997 to 2014 were included in this retrospective study, combined with immunohistochemistry and molecular detection of ETV6-NTRK3 gene fusion. All cases were histologically reviewed with immunohistochemical staining (EnVision) for S-100 protein, SOX10, GATA3, CD117 expression in each group. Fluorescence in situ hybridization (FISH) was used to detect the ETV6-NTRK3 gene fusion.@*Results@#The age of MASC patients ranged from 27 to 74 years with mean age of 47 and ratio of male and female was 4∶3. All cases showed infiltrative growth and diverse cytology and histology, including lobular (8 cases), cystic papillary (3 cases), cribriform mixed with papillary and glandular structures (6 cases) at various proportions. Some tumors of MASC also exhibited solid growth areas with occasional microcystic honeycombed pattern composed of small cysts merged into larger cysts resembling thyroid follicles. S-100 protein and SOX10 were strongly positive in all MASC cases (17/17). In addition, there was insignificant positivity for GATA3 (3/17) and CD117 (4/17). ETV6 gene fusion detection was informative in 12 MASC cases by FISH with 10 positive cases and 2 negative cases.@*Conclusions@#Combined immunohistochemical positivity of S-100 protein, CD117 and SOX10 are useful in the diagnosis and differential diagnosis of MASC. FISH detection of ETV6-NTRK3 fusion offers an additional molecular diagnostic marker for the diagnosis.

ABSTRACT

Objective To investigate the expression of Pim‐1 in non small cell lung cancer and adjacent normal tissues ,and study the relationship between c‐Myc and Pim‐1 in the corresponding tissue gene expression .Methods Totally 30 cases of non small cell lung cancer tissue and adjacent normal lung tissues were collected by surgical operation in department of thoracic surgery . Clinical data were statisticed and tracking late pathologic results ,using RT‐PCR ,qRT‐PCR and immunohistochemical method to de‐tect Pim‐1 mRNA ,c‐Myc and Pim‐1 protein expression in lung cancer and adjacent normal tissue ,and to analyze the relationship be‐tween the expression of Pim‐1 and c‐Myc .Results The positive rate of Pim‐1 mRNA and protein expression in non small cell lung cancer was obviously higher than that in adjacent normal tissue ,the mRNA expression levels were 0 .798 ± 0 .083 and 0 .394 ± 0 .107 (P0 .05) ,bue had related to ymph node metastasis and TNM stages of tumor (P<0 .05) ,with lymph node metastasis and TNM sta‐ges increases ,its expression quantity also rise .There was a positive correlation between Pim‐1 and c‐Myc protein expression ,corre‐lation coefficient (r) was 0 .433 (P=0 .017) .Conclusion High expression of Pim‐1 in non small cell lung cancer gene and is also increased with lymph node metastasis and TNM stages ,Pim‐1 and c‐Myc expression has positive correlation ,this could provide clues to the early diagnosis and prognosis evaluation of non small cell lung cancer ,and also provides a new train of thought and to find a new target for gene therapy of lung cancer .

ABSTRACT

Objective To investigate the effect of different doses valsartan on atherosclerosis in rabbits. Methods 53 white rabbits were randomly divided into control group, cholesterol group, and valsartan group of high-,middle-,and low-dose.The positive expression of pro-oncogene c-myc and c-fos mRNA was studied with in situ reverse transcription-PCR)ISRT-PCR).The histoppathological changes of aorta were observed.Results)1) Positive expression rate of pro-oncogene c-myc and c-fos mRNA was significantly higher in cholesterol than that in the qontrol group. The rate of positive expression was remarkably lower in the high-dose valsartan group than that in the cholesterol group)P
