ABSTRACT

OBJECTIVE@#To study the expression of E-cadherin, N-cadherin, TGF-β1 and Twist protein and investigate its significance in the occurrence and development of prostate cancer.@*METHODS@#The expression of E-cadherin, N-cadherin, TGF-β1 and Twist protein in 59 prostate cancer tissues and 21 adjacent tissues were detected by immunohistochemical SABC staining, and the correlation with clinicopathological features was analyzed.@*RESULTS@#Positive rates of E-cadherin, N-cadherin, TGF-β1 and Twist were 32.2%, 54.2%, 71.2% and 74.6%, respectively, in prostate cancer tissues and 85.7%, 9.52%, 19.0% and 9.52%, respectively, in cancer-adjacent tissues, with significant differences between the two groups (P20μg/L group, but the positive expression of Twist was not significant between groups. The expression of E-cadherin was highly negatively correlated with that of N-cadherin and also highly negatively correlated with that of Twist. The expression of TGF-β1 was correlated with those of E-cadherin, N-cadherin and Twist.@*CONCLUSIONS@#The reduced expression of E-cadherin, abnormal expression of N-cadherin, transformation form E-cadherin to N-cadherin and the increased expression of TGF-β1 and Twist play an important role in the occurrence and development of prostate cancer.

ABSTRACT

Objective To investigate the permeability changes of prostate cancer cells membrane under low intensity ultrasound in vitro.Methods The culture of monolayer adherent LNCaP prostate cancer cells in six-well plate was exposed to continuous ultrasound at frequency of 1 MHz.The cells membrane permeability (stained with Calcein)and cells viability(stained with PI)were evaluated by fluorescent microscope (FM) and cells morphological changes were evaluated by scanning electron microscope (SEM) under ultrasound with acoustic intensity of 160 mW/cm2 for 5 s.The rate of cells with increased cells membrane permeability as function of acoustic intensity (80 mW/cm2,120 mW/cm2 and 160 mW/cm2,for 5 s) and exposure duration (5 s,10 s and 15 s,acoustic intensity of 120 mW/cm2) was evaluated by flow cytometry.ResultsAfter low intensity of ultrasound,the cells with increased cell membrane permeability could be clearly shown with Calcein uptake under FM while no cell showed Calcein uptake in the control group.The SEM showed less microvilli on the cells after low intensity of ultrasound exposure and few cells showed holes on the cell membrane.The rate of cells with increased membrane permeability increased with acoustic intensity and exposure duration.Conclusion Low intensity ultrasound alone could increase membrane permeability of prostate cancer cells and cells with increased membrane permeability showed surface plane,uncommon holes on the cells membrane.The rate of cells with increased membrane permeability positively correlated with acoustic intensity and exposure duration.

ABSTRACT

Purpose: The authors assess the morbidity, functional results and oncologic follow-up of a series of laparoscopic radical prostatectomies performed in over a 10-year period. Material and Methods: The data on 780 laparoscopic radical prostatectomies performed between September 1997 and December 2007 were stored in a personal database. The following parameters are described and critically analyzed: operative time, blood transfusions, conversions, length of hospital stay, complications, functional results of sexual potency and urinary continence, surgical margins and oncologic follow-up. Results: Operative time averaged 125 minutes, with a mean bleeding volume of 335 mL and mean hospital stay of 4.3 days. The rate of conversion to open surgery was 1.36 percent and the overall complication rate was 14.24 percent. The pathology analysis showed pT2 tumors in 82.60 percent and pT3 tumors in 17.39 percent of cases. The overall positive margin rate was 19.58 percent, with a biochemical recurrence of 10.27 percent at a mean follow-up of 62.5 months. Urinary continence and sexual potency yielded rates of 88 percent and 61 percent, respectively, 12 months after surgery. Conclusions: Laparoscopic radical prostatectomy is a technically well-defined procedure that provides good oncologic and functional results after proper training.
