ABSTRACT

This study aimed to investigate outbreak with high mortality in cultured juvenile cobiaoccurred in Southeast Brazil in 2011. Fish displayed retarded growth rates, lethargy, fin ulceration, skin depigmentation, corneal opacity, and physical deformities. Internally, livers were increased in volume and pale in different degrees. Firm whitish nodules were disseminated in the liver, kidney and spleen. A moderate number of parasites identified as Neobenedenia melleni were recovered from the body surface. Microscopically, severe hepatic steatosis and extensive granulomatous lesion were identified in all fish sampled. Microbiological analysis of moribund fish revealed the presence in pure culture of a Gram-negative bacterium identified as Photobacterium damselae subsp. piscicida using biochemical and molecular characteristics. Analysis of the partial 16S rRNA sequences confirmed the results demonstrating high identity (98%). The isolates were sensitive to chloramphenicol and enrofloxacin and resistant to ciprofloxacin, florfenicol, doxycycline hydrochloride, norfloxacin, oxytetracycline, and tetracycline. Chronic pasteurellosis was considered as the main problem in the farm, while hepatic steatosis and parasitic infestation may have contributed to the development of the process.


Este estudo objetivou investigar um surto com alta mortalidade em cobia juvenis cultivadas na região Sudeste do Brasil em 2011. Os peixes apresentavam baixa taxa de crescimento, letargia, ulceração nas nadadeiras, despigmentação da pele, opacidade da córnea e deformidades físicas. Internamente o fígado apresentava aumentado e pálido em diferentes graus, com nódulos esbranquiçados e firmes disseminados no fígado, rins e baço. Na superfície corporal dos peixes foram observados moderado número de parasitas identificados como Neobenedenia melleni. Microscopicamente verificou-se esteatose hepática grave e extensa lesão granulomatosa em todos os peixes amostrados. A análise microbiológica dos peixes moribundos revelou a presença, em cultura pura de uma bactéria Gram-negativa identificada como Photobacterium damselae subsp. piscicida usando características bioquímicas e moleculares. A análise das sequências parciais de 16S rRNA confirmou os resultados demonstrando alta identidade (98%). Os isolados foram sensíveis a cloranfenicol e enrofloxacina e resistente a ciprofloxacina, florfenicol, cloridrato de doxiciclina, norfloxacina, oxitetraciclina e tetraciclina. A pasteurelose crônica foi considerada como o principal problema na maricultura, enquanto a esteatose hepática e a infestação parasitária podem ter contribuído para o desenvolvimento do processo.

ABSTRACT

The present study aimed to estimate the prevalence of antibodies against Brucella ovis-epididymitis, smooth-Brucella, leptospirosis, toxoplasmosis and Maedi-visna in sheep slaughtered in Minas Gerais, Brazil and to study their simultaneous occurrence, including caseous lymphadenitis, at sheep and flock levels. The study was conducted at a sheep slaughterhouse with Federal Inspection Service. Sera from 594 animals from 21 flocks were collected, in 2007. The agar gel immunodiffusion (AGID) was employed to detect anti-B. ovis and anti-Maedi Visna antibodies, whereas Rose Bengal (RB) and the 2-mercaptoethanol test (2ME) were used to test anti-smooth Brucella antibodies. For the detection of anti-Leptospiraantibodies, sera were examined by microscopic agglutination test (MAT), while for the detection of IgG antibodies to Toxoplasma gondii ELISA was used. Prevalence of antibodies against smooth Brucella, B. ovis-epididimitis, Leptospiraspp., toxoplasmosis and Maedi-Visna found in sheep from Minas Gerais was 0.00%, 24.04%, 25.96%, 10.46% and 3.08%, respectively; whereas the seroprevalence in flocks was 0.00%, 80.95%, 90.48%, 71.43% and 23.81%, respectively. Moreover, when data on antibodies anti-Corynebacterium pseudotuberculosis, previously obtained, were included, about 60% of the flocks showed animals that were exposed to four or more of the studied agents. However, only 25.47% of the sheep exhibited simultaneously antibodies against more than one pathogen. Thus, data from the present study on sheep slaughtered in Minas Gerais, Brazil, showed no antibodies to smooth-Brucella and a low frequency of antibodies anti-Maedi Visna lentivirus, and a high and widespread seroprevalence of B. ovis, Leptospira spp., and T. gondii among animals and flocks.(AU)


O presente estudo teve como objetivo estimar a prevalência de anticorpos contra Brucella ovis (epididimite ovina), Brucellalisa, leptospirose, toxoplasmose e Maedi-visna em ovinos abatidos em Minas Gerais, Brasil, e estudar sua ocorrência simultânea, incluindo linfadenite caseosa, nos ovinos e nos rebanhos. O estudo foi realizado em um abatedouro de ovinos com Serviço de Inspeção Federal. Soros de 594 animais de 21 rebanhos foram coletados, em 2007. A imunodifusão em gel de ágar (IDGA) foi empregada para detectar anticorpos anti-B. ovis e anticorpos anti-Maedi Visna, enquanto o teste do antígeno acidificado tamponado (AAT) e o teste de 2-mercaptoetanol (2ME) foram utilizados para testar anticorpos anti-Brucella lisa. Para a detecção de anticorpos anti-Leptospira, os soros foram examinados pelo teste de aglutinação microscópica (MAT), enquanto que para a detecção de anticorpos IgG para Toxoplasma gondii, foi usado o ELISA. A prevalência de anticorpos anti-Brucella lisa, B. ovis, Leptospira spp., toxoplasmose e Maedi-Visna encontrados em ovinos de Minas Gerais foi de 0,00%, 24,04%, 25,96%, 10,46% e 3,08%, respectivamente; enquanto a soroprevalência em rebanhos foi de 0,00%, 80,95%, 90,48%, 71,43% e 23,81%, respectivamente. Além disso, quando dados de anticorpos anti-Corynebacterium pseudotuberculosis, previamente obtidos, foram incluídos, cerca de 60% dos rebanhos apresentaram animais expostos a quatro ou mais dos agentes estudados. No entanto, apenas 25,47% dos ovinos exibiram simultaneamente anticorpos contra mais de um patógeno. Assim, os dados do presente estudo sobre ovinos abatidos em Minas Gerais, Brasil, mostram que ausência de anticorpos anti-Brucella lisa e baixa frequência de anticorpos anti-Maedi Visna, e uma soroprevalência alta e generalizada de B. ovis, Leptospira spp. e T. gondii entre animais e rebanhos.(AU)

ABSTRACT: The present study was conducted to investigate in 20 extensive sheep farms for the seroprevalence of Corynebacterium pseudotuberculosis (n=402) and Toxoplasma gondii (n=228). Enzyme-linked immunosorbent assay (ELISA) was used for the detection of antibodies to C. pseudotuberculosis/T. gondii. It was observed that C. pseudotuberculosis showed the highest prevalence in the region (34.07%) with statistically significant presence (p<0.05) in ewes. Antibodies to T. gondii was reported in 14.91% of the animals studied. About C. pseudotuberculosis/T. gondii coinfection the categories of rams showed significant (p<0.05) differences, suggesting that this gender could perpetuate the diseases in the flocks. It was concluded that the knowledge about the diseases in the region under study would facilitate the execution of prophylactic measures, especially against the diseases that pose risks to the public health and cause damages to the producer.


RESUMO: O presente estudo foi conduzido para investigar a soroprevalência em 20 fazendas de criação extensiva de ovinos quanto à presença de anticorpos para Corynebacterium pseudotuberculosis (n=402) e Toxoplasma gondii (n=228). Ensaio de imunoabsorção enzimático (ELISA) foi utilizado para a detecção de anticorpos contra C. pseudotuberculosis/T. gondii. Observou-se que C. pseudotuberculosis apresentou a maior prevalência na região (34,07%), com presença estatisticamente significante (p <0,05) nas categorias de ovelha. Anticorpos contra T. gondii foram encontrados em 14,91% dos animais estudados. Sobre a coinfecção de C. pseudotuberculosis/T. gondii, as categorias carneiro apresentaram diferenças significativas (p <0,05), sugerindo que esse gênero poderia perpetuar as doenças nos rebanhos. Concluiu-se que o conhecimento sobre as doenças na região em estudo facilitaria a execução de medidas profiláticas, principalmente contra as doenças que apresentam riscos à saúde pública e causam danos ao produtor.

BACKGROUND: Yersinia pseudotuberculosis is known to cause fever, gastroenteritis, or acute kidney injury (AKI). There have been several Y. pseudotuberculosis infection outbreaks to date associated with ingestion of contaminated food or unsterile water. While this disease was considered to have practically been eradicated with the improvement in public health, we encountered several cases of AKI associated with Yersinia infection. METHODS: We retrospectively collected data from medical records of patients with suspected Y. pseudotuberculosis infection who visited Seoul National University Children’s Hospital in 2017. RESULTS: There were nine suspected cases of Yersinia infection (six males and three females; age range 2.99–12.18 years). Among them, five cases occurred in May, and seven patients were residing in the metropolitan Seoul area. Three patients had history of drinking mountain water. Every patient first presented with fever for a median of 13 days, followed by gastrointestinal symptoms and oliguria. Imaging studies revealed mesenteric lymphadenitis, terminal ileum wall thickening, and increased renal parenchymal echogenicity. Creatinine levels increased to 5.72 ± 2.18 mg/dL. Urinalysis revealed sterile pyuria, proteinuria, and glycosuria. Oliguria continued for 4 to 17 days, and two patients required dialysis; however, all of them recovered from AKI. Mucocutaneous manifestations developed later. In the diagnostic work-up, Yersinia was isolated from the stool culture in one patient. Anti-Yersinia immunoglobulin (Ig) A and IgG were positive in 6 patients. CONCLUSION: Y. pseudotuberculosis infection is an infrequent cause of interstitial nephritis presenting with AKI. When a patient presents with fever, gastroenteritis, and AKI not resolving despite hydration, the clinician should suspect Y. pseudotuberculosis infection.

ABSTRACT

O presente estudo objetivou determinar a prevalência e distribuição de lesões abscedativas, identificação do agente etiológico e avaliação das lesões histológicas em caprinos e ovinos abatidos em um matadouro-frigorífico com Serviço de Inspeção Federal do estado da Bahia. Foram coletadas 153 amostras de vísceras e linfonodos com abscessos de 1.148 animais abatidos. A maior prevalência na espécie ovina foi em macho, com faixa etária de 12 meses, sendo os principais órgãos acometidos fígado (21,2%) e linfonodo pré-escapular (20,3%). Na espécie caprina, a prevalência maior foi em macho, com faixa etária de 30 meses, sendo os linfonodos retro faríngeo (25%) e pré-escapular os mais acometidos (25%). Isolou-se os seguintes micro-organismos das amostras: Corynebacterium pseudotuberculosis em 33,33%, Escherichia coli (19,61%), Proteus mirabilis (9,80%), Pseudomonas aeruginosa (7,19%), Trueperella pyogenes (5,22%), Streptococcusspp. (5,22%) e Staphylococcus aureus (4,57%). As lesões macroscópicas e histológicas dos abscessos coletados não apresentaram diferenças entre micro-organismos isolados.(AU)


The study aimed to determine the prevalence and distribution of abscessed lesions, etiologic agent identification and assessment of histological lesions in sheep and goats slaughtered in a slaughter plant refrigerator with Federal Inspection Service in the State of Bahia. The amount of 153 samples of viscera and lymph nodes with abscesses of 1.148 slaughtered animals were collected. The highest prevalence in sheep was in males, aged 12 months, as in liver (21.2%) and prescapular lymph nodes (20.3%) the main affected organs. The prevalence in goats in male, aged 30 months and in retropharyngeal (25%) and prescapular lymph nodes (25%). The following microorganisms were isolated from the samples: Corynebacterium pseudotuberculosis 33.33%, Escherichia coli 19.61%, Proteus mirabilis 9.80%, Pseudomonas aeruginosa 7.19%, Trueperella pyogenes 5.22%, Streptococcus spp. 5.22% and Staphylococcus aureus 4.57%. The macroscopic and histological lesions of abscesses collected presented no difference between isolated microorganisms.(AU)

ABSTRACT

A caprinocultura na região Nordeste do Brasil desempenha importante função socioeconômica e estratégica. A rusticidade desta atividade nesta região, aliada a sua rápida expansão em outros estados, tem gerado perdas na sua cadeia produtiva a nível regional e nacional, causadas por doenças infecciosas, destacando-se a Linfadenite Caseosa (LC), afecção amplamente difundida nos rebanhos caprinos, acarretando sérios prejuízos econômicos à atividade. Embora LC tenha sido detectada como um problema endêmico no nordeste brasileiro, uma análise abrangente e mais recente desta prevalência nos rebanhos caprinos brasileiros se faz necessária. O objetivo desse estudo foi determinar a frequência de anticorpos anti-Corynebacterium pseudotuberculosis em cinco dos nove estados que compõem a região Nordeste do Brasil. Foram processadas amostras de soro de 2571 caprinos provenientes de 218 propriedades rurais oriundas de cinco estados da região Nordeste do Brasil, coletadas entre os anos de 2010 a 2012. O diagnóstico da infecção por C. pseudotuberculosis foi realizado pela técnica de ELISA-indireto. Em 88,5% (193/218) das propriedades investigadas, pelo menos um caprino foi soropositivo para C. pseudotuberculosis, sugerindo que o agente se encontra disseminado nos rebanhos do Nordeste, com a maior prevalência encontrada no Rio Grande do Norte (94,5%) e a menor no estado de Sergipe (70,3%). Foram identificados 783 (30,4%; IC 95%=28,7-32,2%) caprinos soropositivos, com a maior prevalência entre animais encontrada no Piauí (41,4%) e a menor no estado de Sergipe (22,5%). De um total de 279 reprodutores avaliados, 106 (37,9%) resultaram soropositivos, destacando-se o Rio Grande do Norte, com 45,30%, e Piauí com 46,8% dos reprodutores positivos para LC. Das 1420 matrizes testadas, 599(42,1%) apresentaram sorologia positiva para C. pseudotuberculosis. Entre os estados esta distribuição também se manteve semelhante, destacando-se o Rio Grande do Norte, com 47,5% e Piauí com 59,5% das matrizes positivos para LC. Entre os 872 caprinos jovens avaliados, 78 (8,9%) foram soropositivos para LC, observando-se uma diferença estatística na frequência de sopositividade obtidas entre os adultos e os jovens (P<0,001). Reforça-se a necessidade do diagnóstico da enfermidade em rebanhos caprinos da região Nordeste para possível implementação de programas de controle da doença e medidas mais precisas no manejo da LC junto aos criadores de caprinos.(AU)


Goat farming in the Northeast region of Brazil plays an important socioeconomic and strategic role. The rusticity of this activity in this region, allied with its rapid expansion in other states, has caused losses in its production chain in regional and national levels, caused by infectious diseases, especially caseous lymphadenitis (CL), a widespread goat herds disease which has been causing serious economic loss to this activity. Although CL has been detected as an endemic problem in the Brazilian Northeast, a comprehensive and more recent analysis of this prevalence in Brazilian goats is necessary. The objective of this study was to determine the frequency of anti-Corynebacterium pseudotuberculosis antibodies in five of nine states of the northeast region of Brazil. Serum samples were collected from 2571 goats from 218 farms in five states in the Northeast region of Brazil, collected between 2010 and 2012. The diagnosis of C. pseudotuberculosis infection was made using the ELISA-indirect technique. In 88.5% (193/218) of the investigated properties, at least one goat was seropositive for C. pseudotuberculosis, suggesting that the agent is widespread in Northeast herds, with the highest prevalence found in Rio Grande do Norte (94.5%) and the lowest one in the state of Sergipe (70.3%). A total of 783 (30.45%; 95% CI = 28.71-32.26%) seropositive goats were found, and the highest prevalence among animals was found in Piauí (41.4%) and the lowest in the state of Sergipe (22.5%). Out of a total of 279 breeding herds, 106 (37.99%) were seropositive, standing out the Rio Grande do Norte, with a total of 45.30%, and Piauí 46.8% of positive breeding for CL. A total of the 1420 tested matrices, 599 (42.1%) presented positive serology for C. pseudotuberculosis. Among the States, this distribution also remained similar, standing out Rio Grande do Norte with 47.5%, and Piauí with 59.5% of positive matrices for CL. Among 872 young goats evaluated, 78 (8.9%) were seropositive for CL, observing a statistical difference in the frequency of soropositivity obtained between adults and young adults (P<0.001). The necessity to diagnose the disease in goat herds in the Northeast region is reinforced for the possible implementation of disease control programs and more precise measures to manage CL along with goat farmers.(AU)

ABSTRACT

The semiarid northeast of Brazil contains a unique biome known as caatinga, with a maximum temperature of 40 ºC and a relativity humidity of 56%. The caatinga is characterized by a variety of plants, including Cereus jamacaru Dc (mandacaru), Poincianella microphylla Mart. ex G. Don (catingueira), Pilosocereus gounellei FAC Weber (xique-xique) and Mimosa tenuiflora (Willd.) Poir (jurema preta). Sheep and goat industries are economically strong in that region, despite the fact that caseous lymphadenitis is highly prevalent. The aim of the present study was to assess the survival and biofilm production of Corynebacterium pseudotuberculosis isolates in the environment and under controlled temperatures (28°C, 37°C and 42°C) under different surfaces (plants, soil, wood, wire and thorns). In addition, we investigated the effects of applying the disinfectants chlorhexidine, hypochlorite and quaternary ammonia in soil, tiles, wood and vegetation cover. Four strains of C. pseudotuberculosis were selected (two from goats and two from sheep) for inoculation according to their in vitro biofilm production. Adherence to microplates was used to assess the biofilm-forming ability of the bacteria. Lower survival rates were observed when isolates of C. pseudotuberculosis were subjected to a temperature of 42°C. In terms of caatinga biome plants, contamination of jurema-preta plants resulted in the lowest survival rates. The disinfectant quaternary ammonia promoted a lower inoculum survival in all surfaces. The disinfectants and the higher temperature contributed to the reduction of biofilm production in isolates of C. pseudotuberculosis. knowledge of these patterns is important for the establishment of disease control measures, given the questionable efficacy of the treatment and the immuno-prophylaxis of caseous lymphadenitis.(AU)


O semiárido nordestino do Brasil possui um bioma exclusivo, a caatinga, que apresenta temperatura máxima de 40oC e uma umidade relativa do ar de 56%. A caatinga é caracterizada por uma diversidade de plantas, entre elas Cereus jamacaru DC. (mandacaru), Poincianella microphylla Mart ex G. Don (catingueira), Pilosocereus gounellei F.A.C. Weber (xique-xique) e Mimosa tenuiflora (Willd.) Poir (jurema preta). A produção de ovinos e caprinos está em franca expansão, porém a linfadenite caseosa é uma enfermidade de alta prevalência na região. Objetiva-se com o presente estudo, verificar a sobrevivência e produção de biofilme em isolados de Corynebacterium pseudotuberculosis em temperaturas de 28oC, 37oC e 42oC, quando inoculado em superfícies de solo, madeira, arame e espinho e em plantas nas condições ambientais da caatinga. Além disto, foi verificado o efeito da aplicação dos desinfetantes clorexidine, hipoclorito e amônia quaternária sobre o solo, piso (lajota), madeira e vegetação de cobertura do solo. Foram selecionadas quatro amostras de C. pseudotuberculosis (dois caprinos e dois ovinos) para inoculação de acordo com a sua produção in vitro de biofilme. A adesão a microplacas foi utilizado para avaliar a capacidade de formação de biofilme das bactérias. As menores taxas de sobrevivência foram observadas quando isolados de C. pseudotuberculosis foram submetidos a uma temperatura de 42°C. Com relação as plantas do bioma caatinga, a contaminação na planta jurema-preta apresentou menores índices de sobrevivência. O desinfetante amônia quartenária promoveu uma menor sobrevivência do inóculo em todas as superfícies. Os desinfetantes e temperatura contribuíram para a redução na produção de biofilme nos isolados de Corynebacterium pseudotuberculosis. O conhecimento destes padrões é importante para o estabelecimento de medidas de controle da enfermidade, dada a eficiência questionável do tratamento e imunoprofilaxia da linfadenite caseosa.(AU)

This study aimed to evaluate aspects of host immune response using an experimental infection model of Corynebacterium pseudotuberculosis (CP) in C57/Black6 wild-type and knockout for nitric oxide (KO-NO) mice. 28 mice were evaluated: 4 wild-type controls; 10 wild-type infected with CP; 4 KO-NO controls; 10 KO-NO infected with CP. Infection procedures were carried out by intraperitoneal inoculation using 107. Infected C57/Black6 KO-NO mice began to die after the 5° day post-inoculation, up until the 14º day. Neutrophils were found in increased numbers in the infiltrate of KO-NO murine peritoneal cavities. Examination of splenic tissue revealed an accumulation of lymphocytes, predominantly CD8 T-cells, in experimental animal groups. KO-NO animals were found to have a predominance of granulomas 7 days post-inoculation, primarily in the lymph nodes. In addition, greater amounts of bacteria were recovered from the mesenteric lymph nodes of KO-NO mice. There was no statistically significant difference in the levels of total IgG and its subclasses 14 days post-inoculation between KO-NO and wild groups. The results suggest the importance of nitric oxide in the process of controlling CP infection, as KO-NO animals were observed to be markedly more affected by infection with this bacterium.(AU)


O objetivo deste estudo foi avaliar os aspectos da resposta imune do hospedeiro, mediante o uso de um modelo experimental de infecção de Corynebacterium pseudotuberculosis (CP) em camundongos C57/Black6 do tipo selvagem e em C57/Black6 knockout para o óxido nítrico (KO-NO). Foram avaliados 28 camundongos: quatro controles de tipo selvagem; 10 do tipo selvagem infectados com CP; quatro controles KO-NO; e 10 KO-NO infectados com CP. A infecção foi realizada via intraperitoneal, usando-se 107. Os animais C57/Black6 KO-NO infectados começaram a vir a óbito no quinto dia pós-inoculação, o que aconteceu até o 14º dia. Um número maior de neutrófilos foi encontrado na sua cavidade peritoneal. O exame do baço revelou um acúmulo de linfócitos, predominantemente células T CD8, nos grupos de animais experimentais. Nos animais KO-NO, foi observada a presença de granulomas, sete dias pós-inoculação, principalmente nos gânglios linfáticos. Além disso, uma maior quantidade de bactérias foi detectada dos linfonodos mesentéricos desses animais. Não houve diferença estatisticamente significante nos níveis séricos IgG total e em suas subclasses aos 14 dias pós-inoculação nos grupos KO-NO e selvagem. Os resultados obtidos sugerem a importância do óxido nítrico no processo de controle da infecção por CP.(AU)

ABSTRACT

Corynebacterium pseudotuberculosis se transmite entre ovinos de la Patagonia argentina y ocasiona pseudotuberculosis. Esta bacteria, que infecta ganglios superficiales y vísceras, ingresa al animal a través de heridas en la piel o membranas mucosas. Durante la esquila, se cortan abscesos superficiales que drenan el contenido purulento y contaminan las herramientas y el suelo. El objetivo de este trabajo fue evaluar la capacidad de C. pseudotuberculosis de sobrevivir a lo largo del tiempo en suelos de la Patagonia extraandina. Se recolectaron 5 muestras de suelos de distintas zonas de la provincia del Chubut (Patagonia extraandina) con características fisicoquímicas diferenciales en lo que respecta a contenido de materia orgánica, pH, conductividad eléctrica y textura. Diferentes alícuotas de estas muestras se inocularon con la cepa de C. pseudotuberculosis PAT10, aislada de un ovino patagónico, y se conservaron a temperatura ambiente. A distintos tiempos se determinó el número de bacterias supervivientes. El 60% de la población de C. pseudotuberculosis inoculada sobrevivió durante 80 y 210 días en suelos con moderadas y altas concentraciones de materia orgánica, respectivamente. Los suelos de características diversas de la Patagonia contaminados con poblaciones de C. pseudotuberculosis cumplirían un papel importante en el ciclo de infección en la naturaleza. La supervivencia de la bacteria por largos períodos se vio favorecida por el mayor contenido de materia orgánica sumado a la textura franco-limosa y no se observó la influencia de variables como el pH y la salinidad.


Corynebacterium pseudotuberculosis is transmitted among sheep in Argentine Patagonia causing pseudotuberculosis. The bacterium penetrates the skin or mucous membrane wounds, infecting the superficial lymph nodes and viscera. When surface abscesses are cut during shearing, they drain their purulent contents and contaminate tools and the soil. The objective of this work was to evaluate the survival capacity of C. pseudotuberculosis over time, in soils from the extra-Andean Patagonia region. Five types of superficial soils were collected from different areas in Chubut province (extra-Andean Patagonia), having distinctive physicochemical properties including organic matter content (very high to nonexistent), pH (neutral to strongly alkaline), electrical conductivity (saline to non-saline) and texture (sandy, clayey, silty loam). Different aliquots of each type of soil were inoculated with C. pseudotuberculosis PAT10 strain isolated from a Patagonian sheep, and were stored at room temperature. The number of surviving bacteria was determined at various times. Sixty percent (60%) of the inoculated C. pseudotuberculosis population survived for 80 to 210 days in soils with moderate to high organic matter content respectively. Silty soils favored bacterial survival, whereas the variables pH and salinity had no effect on survival.

Moreover, the pathogenicity of two Y. pseudotuberculosis strains was investigated in order to detect potential differences. Methods Twelve European brown hares (Lepus europaeus, Pallas) were experimentally infected per os and via conjunctival mucosae with Y. pseudotuberculosis: six subjects were infected with a strain isolated from a naturally infected hare (YpH) and six subjects with a strain isolated from a naturally infected rabbit (YpR). Two hares were used as negative controls. All animals were subjected to clinical, bacteriological and serological examinations during 9 weeks following the infection and, at the end of the control period, subjects still alive were euthanized and submitted to a complete post mortem examination. Results All faecal samples collected during the control period were positive for bacteriological examinations and to a PCR for the inv gene of Y. pseudotuberculosis, while only one YpH-infected hare showed a positive haemocultures. From the 2nd to the 9th week post infection (pi), serological analysis revealed specific antibodies with titers ranging from 1:10 to 1:160 in all YpH-infected and two YpR-infected subjects. All the YpH-infected and two YpR-infected hares scored positive for Y. pseudotuberculosis by means of bacteriological investigations. Grossly, suppurative multifocal lesions were detected in liver, spleen, kidney and sub-mandibular lymph nodes in both YpH- and YpR-infected hares and confirmed with histopathology. Pulmonary lesions were observed only in YpH-infected subjects. Immunohistochemistry confirmed the presence of bacterial antigen in all infected animals. Conclusion Results of this study revealed that YpH strain is more pathogenic for hares than the YpR strain; moreover the serological test performed in this study could be used for the diagnosis of pseudotuberculosis in hares, whereas post mortem diagnosis should be confirmed by means of bacteriological examination, PCR, histopathology and immunohistochemistry.

La linfadenitis caseosa (LC) es una enfermedad infecciosa de carácter crónico causada por Corynebacterium pseudotuberculosis; afecta a pequeños rumiantes y genera pérdidas económicas por reducción del peso, eficiencia reproductiva, producción de leche y lana, decomiso de canales y depreciación de pieles. Dada la importancia socioeconómica de la producción caprina en el estado Falcón (Venezuela), el objetivo de esta investigación fue caracterizar epidemiológicamente la enfermedad en rebaños de la península de Paraguaná. La investigación es de carácter descriptivo. La fase de campo se efectuó durante seis semanas, tiempo en que se inspeccionaron los ganglios linfáticos superficiales, de los que se obtuvieron 71 muestras de secreción purulenta en aquellos animales en los que se presumía padecían de LC, según las manifestaciones clínicas. Una vez en el laboratorio, los especímenes se analizaron bacteriológicamente; se compararon los aislamientos de C. pseudotuberculosis con la cepa de referencia ATCC 19410. Solo se detectó como factor de riesgo de la LC (p < 0,05) la procedencia de los caprinos según la unidad de producción, de las que se observaron con las mayores prevalencias en aquellas ubicadas en el municipio Falcón, entre las cuales las lesiones en los ganglios preescapulares fueron las más frecuentes (p < 0,05) entre todos los animales diagnosticados. Se identificaron cepas con resistencia a penicilina, trimetoprima-sulfametoxazol y novobiocina. Estos resultados son de importancia para la concientización de los productores, dado que esta actividad es de vital trascendencia en la región y en muchos casos se pudo constatar el desconocimiento del tema.


Caseous lymphadenitis (CL) is a chronic infectious disease caused by Corynebacterium pseudotuberculosis; it affects small ruminants and generates economic loss due to a reduction in weight, reproductive performance, milk and wool production forfeiture, and depreciation of skins. Given the socio-economic importance of goat production in the Falcón state (Venezuela), this research aimed to epidemiological^ characterize the disease in herds in the Paraguaná peninsula. The research is descriptive. Field work lasted six weeks, during which superficial lymph nodes were inspected, and 71 samples of purulent discharge were obtained from animals suspected to suffer from CL, according to their clinical manifestations. Back in the laboratory, specimens were bacteriologically analyzed; C. pseudotuberculosis isolates were compared with the reference strain ATCC 19410. The only risk factor detected for CL (p < 0.05) was the origin of goats by production units; those with the highest prevalence were located in the municipality of Falcón. Injuries in subscapular lymph nodes were the most frequent (p < 0.05) among the diagnosed animals. Penicillin, trimethoprim-sulfamethoxazole, and novobiocin-resistant strains were identified. These results are important to raise awareness among producers, given that this activity is of vital importance for the region and in many cases ignorance on the subject was evidenced.

Devido à importância socioeconómica da produção caprina no estado Falcón (Venezuela), o objetivo desta pesquisa foi caracterizar a epidemiologia da doença em rebanhos da península de Paraguaná. A pesquisa é de caráter descritivo. A fase de campo foi realizada durante seis semanas, tempo em que se inspecionaram os gânglios linfáticos superficiais, do que se obtiveram 71 amostras de secreção purulenta em aqueles animais nos que se presumia padeciam de LC, segundo as manifestações clínicas. Já no laboratório, os espécimenes se analisaram bacteriologicamente; se compararam os isolamentos de C. pseudotuberculose com a cepa de referência ATCC 19410. Identificaram-se cepas com resistência a penicilina, trimetoprima-sulfametoxazol e novobiocina. Estes resultados são importantes para a conscientização dos produtores, sendo que esta atividade tem uma vital transcendência na região e em muitos casos pôde-se constatar o desconhecimento do tema.

Caseous lymphadenitis is a chronic infectious disease caused by Corynebacterium pseudotuberculosis , which is a bacterium responsible for a great number of economic losses on goat and sheep production. It is characterized by the formation of abscesses in superficial lymph nodes and in internal organs and lymph nodes. This study aimed at determining the agreement between microbiological culture and PCR in the identification of C. pseudotuberculosis , in samples collected from animals in slaughterhouses and in animals that presented lymph node enlargement in field conditions. From the 202 samples analyzed through microbiological culture, 113 (56%) were positive for Corynebacterium sp.; from these positive samples, 38 (34%) were identified as C. pseudotuberculosis by microbiological culture. From the amount of samples, 110 (54%) were positive and 92 (46%) were negative in the PCR. Kappa index (0.193) presented a weak agreement between PCR and microbiological culture. We concluded that molecular diagnosis (PCR) in clinical samples proved to be more efficient, reproducible, and faster than the microbiological culture, both on clinical samples analyses and in the confirmation of C. pseudotuberculosis in colonies that were classified by Corynebacterium genus. Thus, the present study demonstrated the importance of PCR to confirm C. pseudotuberculosis diagnosis, and the best contribution for the epidemiological surveillance of the disease in sheep.(AU)


A linfadenite caseosa é uma doença infectocontagiosa crônica, causada pelo agente etiológico Corynebacterium pseudotuberculosis , que é uma bactéria responsável por grandes perdas econômicas na produção de ovinos e caprinos. Caracteriza-se pela formação de abscessos em nódulos linfáticos superficiais e em órgãos internos e linfonodos. O presente trabalho teve por objetivo verificar a concordância entre as metodologias de isolamento microbiológico com a PCR na identificação do C. pseudotuberculosis , em amostras clínicas colhidas em abatedouros e em animais que apresentavam aumento de linfonodo em condições de campo. Das 202 amostras analisadas no cultivo microbiológico, 113 (56%) foram positivas para o gênero Corynebacterium sp., e 38 (34%) colônias foram identificadas como C. pseudotuberculosis por meio de cultura microbiológica. Das amostras clínicas extraídas, 110 (54%) foram positivas e 92 (46%) foram negativas na PCR. A concordância estimada entre as técnicas de PCR e o cultivo microbiológico pelo indicador Kappa foi considerada fraca (0,193). Concluímos que o diagnóstico molecular (PCR) provou ser mais eficiente, rápido e com reprodutibilidade quando comparado ao cultivo microbiológico das amostras clínicas bem como da confirmação do C. pseudotuberculosis de colônias pertencentes ao gênero Corynebacterium . Dessa forma, o presente trabalho demonstrou a importância do uso da PCR na confirmação diagnóstica do C. pseudotuberculosis , visando contribuir com a melhoria da vigilância epidemiológica da doença em ovinos.(AU)

ABSTRACT

Corynebacterium pseudotuberculosis is the etiologic agent of caseous lymphadenitis (CLA), a chronic disease that affects goats and sheep, characterized by granuloma formation in subcutaneous and internal lymph nodes. CLA causes significant economic losses to commercial goat herds. In this study, we aimed to test secreted antigens secreted from T1 strain bacteria grown in brain heart infusion (BHI) broth in an indirect ELISA system to determine the presence of specific immunoglobulins against C. pseudotuberculosis. We analyzed the BHI antigen electrophoretic profile and the recognition pattern by infected sheep sera samples. The ELISA results were compared with multiplex PCR assay and IFN-gamma production. The ELISA was able to discriminate between negative and positive animals, with a sensitivity of 89% and a specificity of 99%, using microbiological isolation as gold standard. When this assay was compared with multiplex PCR and specific IFN-gamma quantification, six discrepant results were found among thirty-two samples. We concluded that the ELISA using antigens secreted from C. pseudotuberculosis T1 strain growth in BHI broth culture can be used for the serodiagnosis of CLA in sheep.


Corynebacterium pseudotuberculosis é o agente etiológico da linfadenite caseosa (LC) uma doença crônica que afeta ovinos e caprinos caracterizada pela formação de granulomas em linfonodos. A LC causa perdas econômicas significativas em criações de pequenos ruminantes. Este trabalho teve como objetivo testar antígenos secretados da cepa T1 da bactéria em um sistema de ELISA indireto para detecção de anticorpos específicos contra C. pseudotuberculosis. O perfil eletroforético do antígeno foi analisado, bem como o padrão de reconhecimento por soros de animais infectados. Os resultados do ELISA foram comparados com ensaio de multiplex PCR e com teste de indução de produção específica de IFN-gama. O ELISA foi capaz de discriminar animais positivos de animais negativos, com sensibilidade de 89% e especificidade de 99%, usando o isolamento microbiológico como padrão ouro. Quando o ensaio foi comparado com o multiplex PCR e a produção específica de IFN-gama, somente seis resultados discrepantes foram encontrados em trinta e duas amostras. Pode-se concluir que o ensaio de ELISA desenvolvido pode ser utilizado com alto grau de confiança para o diagnóstico da LC em ovinos.

ABSTRACT

To verify the occurrence of caseous lymphadenitis in sheep and goats on farms of Pernambuco, Brazil, and in animals slaughtered in two Brazilian cities (Petrolina/PE and Juazeiro/BA), and to characterize the susceptibility profile of Corynebacterium pseudotuberculosis to disinfectants and antimicrobials, and its relationship with biofilm production were the objectives of this study. 398 samples were tested for sensitivity to antimicrobial drugs, disinfectants, and biofilm production. Among the 108 samples collected on the properties, 75% were positive for C. pseudotuberculosis. Slaughterhouse samples indicated an occurrence of caseous lymphadenitis in 15.66% and 6.31% for animals slaughtered in Petrolina and Juazeiro respectively. With respect to antimicrobials, the sensitivity obtained was 100% for florfenicol and tetracycline; 99.25% for enrofloxacin, ciprofloxacin and lincomycin; 98.99% for cephalothin; 98.74% for norfloxacin and sulfazotrim; 97.74% for gentamicin; 94.22% for ampicillin; 91.71% for amoxicillin; 91.21% for penicillin G; 89.19% for neomycin and 0% for novobiocin. In analyzes with disinfectants, the efficiency for chlorhexidine was 100%, 97.20% for quaternary ammonium, 87.40% for chlorine and 84.40% for iodine. 75% of the isolates were weak or non-biofilm producers. For the consolidated biofilm, found that iodine decreased biofilm formation in 13 isolates and quaternary ammonia in 11 isolates. The reduction of the biofilm formation was observed for iodine and quaternary ammonium in consolidated biofilm formation in 33% and 28% of the isolates, respectively. The results of this study highlight the importance of establishing measures to prevent and control the disease.

Para o biofilme consolidado, observou-se que o iodo diminuiu a formação de biofilme em 13 isolados e a amônia quaternária em 11 isolados. A redução da formação de biofilme foi observada no biofilme consolidado, para iodo e amônia quaternária, em 33% e 28% das amostras, respectivamente. Os resultados deste estudo destacam a importância de se estabelecer medidas para prevenir e controlar a doença.

ABSTRACT

This paper reports the clinical, bacteriological and pathological findings of a thoracic aortic aneurysm in a four-year-old Anglo-Nubian goat buck, related to a framework of visceral caseous lymphadenitis. General clinical examination showed heart rate of 75 beats per minute, respiratory rate of 20 movements per minute and ruminal movements of four movements per minute. Superficial lymph nodes were normal upon palpation. Rectal temperature was slightly high (40.5°C). Blood test showed an intense leukocytosis (54,000/µL), characterized by strong neutrophil shift to the left. At necropsy, a large blood clot was detected in the thoracic cavity. The thickening of the myocardium and dilatation of the aorta in the thoracic portion, presenting a saculiform format was also observed. A large number of abscesses were disseminated in the media and intima layers of aorta. The aorta lumen obstruction by arterial plaques consisting of inflammatory infiltrate, predominantly neutrophilic was also detected. Abscesses were found spread in turbinate, rumen, reticulum, kidneys, liver, spleen, testicles and aorta wall. The microbiological exam of exudate confirmed Corynebacterium pseudotuberculosis as the causal agent.


Este trabalho descreve os achados clínicos, bacteriológicos e patológicos de um aneurisma da aorta torácica em um reprodutor caprino da raça Anglo-Nubiana, de quatro anos de idade, relacionado a um caso de linfadenite caseosa visceral. Ao exame clínico geral observaram-se: frequência cardíaca de 75 batimentos por minuto, frequência respiratória de 20 movimentos por minuto e movimentos ruminais de quatro movimentos por minuto. Os linfonodos superficiais encontravam-se normais à palpação. A temperatura retal estava ligeiramente aumentada (40,5°C). No hemograma completo, observou-se leucocitose intensa, 54.000/µL, caracterizada por um forte desvio neutrofílico à direita. Nos achados de necropsia, observou-se, na cavidade torácica, presença de um grande coágulo de sangue. No coração, foi identificado espessamento do miocárdio. Uma dilatação na porção torácica da artéria aorta foi detectada, apresentando um aspecto saculiforme. Um grande número de abscessos estava presente nas camadas média e íntima da aorta. Observou-se, também, obstrução do lúmen da aorta por placas de infiltrado inflamatório, predominantemente neutrofílico. A disseminação de abscessos nos cornetos, rúmen, retículo, fígado, baço, rins, testículos e parede da aorta foi detectada. O exame microbiológico do exsudato confirmou o Corynebacterium pseudotuberculosis como o agente causal.

ABSTRACT

Corynebacterium diphtheriae, Corynebacterium ulcerans and Corynebacterium pseudotuberculosis constitute a group of potentially toxigenic microorganisms that are related to different infectious processes in animal and human hosts. Currently, there is a lack of information on the prevalence of disease caused by these pathogens, which is partially due to a reduction in the frequency of routine laboratory testing. In this study, a multiplex polymerase chain reaction (mPCR) assay that can simultaneously identify and determine the toxigenicity of these corynebacterial species with zoonotic potential was developed. This assay uses five primer pairs targeting the following genes: rpoB (Corynebacterium spp), 16S rRNA (C. ulcerans and C. pseudotuberculosis), pld (C. pseudotuberculosis), dtxR (C. diphtheriae) and tox [diphtheria toxin (DT) ]. In addition to describing this assay, we review the literature regarding the diseases caused by these pathogens. Of the 213 coryneform strains tested, the mPCR results for all toxigenic and non-toxigenic strains of C . diphtheriae, C. ulcerans and C. pseudotuberculosis were in 100% agreement with the results of standard biochemical tests and PCR-DT. As an alternative to conventional methods, due to its advantages of specificity and speed, the mPCR assay used in this study may successfully be applied for the diagnosis of human and/or animal diseases caused by potentially toxigenic corynebacterial species.

ABSTRACT

The objective of this experiment was to evaluate the efficacy of iodine tincture at 10% and sodium hypochlorite at 2.5% applied into the abscess of animals affected by Caseous Lymphadenitis (CL). Eighteen ewes were used, assorted into three groups: one treated with iodine tincture at 10% (IT), another one with sodium hypochlorite at 2.5% (SH) and the last group underwent the conventional treatment (CT). Conventional treatment was based on surgical drainage and chemical cauterization of the lesion with iodine tincture at 10%. Natural rupture of six abscesses from IT group was observed and in five of them the viability of Corynebacterium pseudotuberculosis was confirmed on the lesion place, after rupture of lymph node. As for the SH group, spontaneous rupture was observed in five out of six abscesses treated, and the microorganism was identified on the lesion of five animals, after rupture. In the sixth animal of this group, abscess involution was noticed. A severe swelling was identified in the region of lymph node treated, resulting in wide lesion in animals from groups IT and SH. No difference (p > 0,05) was found in blood parameters due to treatments. As for the serological monitoring of animals, comparative analysis between months within each group showed that months 1, 2, 3 and 4 were different (p 0,05) from month 0, for groups IT and SH. For CT group, there was no difference (p > 0,05) between months 1 to 5 and 0. Then, the application of iodine tincture at 10% or sodium hypochlorite at 2.5% into the abscess of animals affected by CL, at stage in which lesions are detected through inspection, is not effective for its control.


Objetivou-se com este experimento avaliar a eficácia da tintura de iodo a 10% e do hipoclorito de sódio a 2,5% aplicados no interior do abscesso de animais acometidos pela Linfadenite Caseosa (LC). Foram utilizadas 18 fêmeas ovinas, divididas em 3 grupos: o primeiro foi tratado com tintura de iodo a 10% (TI), o segundo com hipoclorito de sódio a 2,5% (HS) e o último grupo foi submetido ao tratamento convencional (CT). O tratamento convencional da doença consistia na drenagem e cauterização química dos abscessos, utilizando-se solução de iodo a 10%. Foi observada a ruptura natural dos seis abscessos do grupo TI, sendo que em cinco deles a viabilidade do Corynebacterium pseudotuberculosis foi confirmada no local da lesão, após a ruptura do linfonodo. Quanto ao grupo HS, a ruptura espontânea foi observada em cinco dos seis abscessos tratados, sendo o microrganismo identificado na lesão dos cinco animais após o rompimento. No sexto animal desse grupo, foi constatada a involução do abscesso. Foi verificado um edema intenso na região do linfonodo tratado com formação de uma lesão de grande extensão nos animais dos grupos TI e HS. Não foi detectada diferença (p > 0,05) nos parâmetros hematológicos em decorrência dos tratamentos avaliados. Quanto ao acompanhamento sorológico dos animais, a análise estatística comparativa entre os meses, dentro de cada grupo, mostrou que os meses 1, 2, 3 e 4 foram diferentes (p 0,05) do mês 0 para os grupos TI e HS. No grupo CT, não houve diferença (p > 0,05) entre os meses de 1 a 5 e o mês 0. Conclui-se que a aplicação de tintura de iodo a 10% ou hipoclorito de sódio a 2,5% no interior do abscesso de animais acometidos pela LC, em estágio no qual as lesões são detectadas por meio da inspeção, não é eficaz para o seu controle.
