ABSTRACT

ABSTRACT: Aster (Aster ericoides L.) is a flower crop of North American origin, which belongs to the family Asteraceae. This plant presents capitulum-type inflorescences, widely used as cut flowers. In Brazil, it has been grown only recently, but still has a high expansion potential. In floriculture, pulsing and conditioning solutions are often used to extend the vase life of cut flowers. The present study aimed to evaluate the influence of silver thiosulphate (STS) pulsing and sucrose solutions on the postharvest life of A. ericoides stems. The experiment was carried out under a complete randomized design with treatments arranged in a 2x5 factorial scheme. Treatments consisted of flower stems treated with STS pulsing for 30 minutes, combined with four sucrose concentrations (5, 10, 15, and 20%) for six hours, plus a control (without treatment). Each plot contained four replicates of 10 stems, totalizing 400 stems. The following variables were daily: vase life, fresh mass, and relative water content (RWC). For pulsing data, means were compared by the Tukey's test (p≥5%). Yet for sucrose concentrations, means were submitted to a polynomial regression to verify the behavior of variables as a function of concentrations increases. Likewise, each treatment was evaluated for fresh mass and RWC variations over time by a polynomial regression. Results indicated no effect of STS pulsing (2mM) on the vase-life length of aster stems. In contrast, the sucrose solution at 10% extended the vase life of stems by two days, decreased fresh mass loss, and helped maintain water balance in stems.

ABSTRACT

Objective To evaluate the value of reducing radiation dose with decreased size of ECG-pulsing windows and influence on image quality in dual-source CT coronary angiography. Methods 120 patients with stable heart rate(HR) were divided into four groups according to HR and the rang of ECG-pulsing windows in dual-source CT coronary angiography: HR < 70 bpm and 61% ~ 77% R-R interval of ECG-pulsing windows, HR < 70 bpm and 25% ~ 80% R-R interval of ECG-pulsing windows,HR > 80 bpm and 31% ~47% R-R interval of ECG-pulsing windows, and HR > 80 bpm and 25% ~ 80%R-R interval of ECG-pulsing windows was employed, respectively. The radiation dose parameters were recorded and image quality scores were performed. The image quality and radiation dose between two slow HR groups and between two fast HR groups were compared respectively. Result The effective doses were (7.06 ± 2. 13 ), ( 11.34 ± 3.65 ), ( 6. 67 ± 1.97 ) and ( 9. 92 ± 3. 15 ) mSy for four groups, respectively. The effective dose was decreased by 37.74% for slow HR and by 32. 76% for fast HR using narrow ECG-pulsing windows. There was no difference on image quality between two slow HR groups and two fast HR grouvs. Concluslons The proper application of narrow ECG-pulsing windows can reduce radiation exposure significantly to stable slow or fast HR patients in dual-source CT coronary angiography withont sacrificing the image quality.

ABSTRACT

Oncidium varicosum from the Orchidaceae family has been widely used as pot flower, and, most recently,as cut flower. The objective of this work was to evaluate the longevity of the Oncidium varicosum, dueto its low post-harvest resistance, after being conditioned in preservation solutions with differentconcentrations of sucrose. Oncidium varicosum stems grown in Holambra (SP) were transported toLondrina (PR) in climatized trucks at 5-7oC. Pre-treatment with pulsing (15% sucrose for 24 hours)followed by conditioning in maintenance solution (1, 2, 3, and 4% of sucrose) as well as conditioning inmaintenance solution without pre-treatment were evaluated. Flower stems were distributed in an entirelycasualized design, with ten treatments and five replications. Flowers were evaluated daily, using a scoresystem developed to compare and verify the conservation of visual aspects. No treatment was superiorto the control in maintaining the longevity of the flowers

ABSTRACT

BACKGROUND: Tumor cell lysate has been considered as a preferential antigen source for the therapeutic dendritic cell pulsing. Our experiences with in vivo study with animal tumor model indicate the tumor cell lysate dependent differential effect of DC therapy. Our previous data show that MC38 lysate pulsed-DC induced stronger ag-specific immunity than CT26 lysate pulsed-DC in vitro. In this study we tried to reveal the mechanism for differential induction of ag-specific immunity of different colon cancer cell lysate pulsed-DCs. METHODS: MC38 and CT26 cell lines were prepared as lysate by freezing-thawing procedure. Tumor cell antigenicity was confirmed by detecting the surface expression of MHC I/II & B7.1/2 molecules. IL-10, IL-12 and TGF-beta in the tumor cell lysate were detected by ELISA and the presence of heat shock proteins were analysed by western blotting. RESULTS: The secretion of IL-10, a immune-inhibitory cytokine was about 470% higher in CT26 lysate than in MC38. Hsp 70 was detected only in the MC38 lysate but not in the CT26. On the other hand, Hsp 60 and 90 expression were not different in two colon cancer cell lysates. CONCLUSION: In two different colon caner cell lysate, immune inhibitory IL-10 (higher in CT26) and Hsp70 (MC38 superiority) were differentially expressed. These data indicate that higher ag- specific immunity induction by MC38 lysate pulsed-DC may due to the expression of hsp70 and lower secretion of IL-10, a immune-inhibitory cytokine than CT26 lysate. The significance of other cytokine and the surface marker expression will be discussed.
