ABSTRACT

A resistência bacteriana tem aumentado progressivamente no mundo, assim, há necessidade de novas opções de tratamentos. A fitoterapia tem ganhado notoriedade para combater infecções, principalmente as causadas por bactérias resistentes aos antibacterianos disponíveis. Diante do exposto, o presente estudo teve como objetivo preparar e analisar a composição fitoquímica e a ação antibacteriana dos extratos hidroetanólicos de canela (EHC) e romã (EHR) isolados e associados frente culturas planctônicas e biofilmes de cepas padrão e clínicas de Acinetobacter baumannii e Pseudomonas aeruginosa, além disso, analisar a ação citotóxica dos extratos em queratinócitos humanos (HaCat). Para isso, os EHC e EHR foram preparados e quantificado o teor de sólidos solúveis. Posteriormente, foi quantificado o teor de flavonoides e fenóis totais, análise antioxidante por meio da redução do radical 2,2'-difenil-1-picrilhidrazila (DPPH), e a fitoquímica por cromatografia líquida (HPLC). Em relação a ação antibacteriana dos extratos, foi aplicado o teste de microdiluição em caldo (CLSI ­ M7-A9) e a ação sinérgica realizada por meio do ensaio de checkerboard. As concentrações mais efetivas foram analisadas sobre biofilmes em formação (prevenção) e biofilmes formados (tratamento de 24 h), e quantificada a viabilidade por meio do teste colorimétrico MTT. Para avaliar a citotoxidade, os tratamentos foram aplicados sobre cultura celular de HaCat por 24 h e analisados por meio do teste colorimétrico MTT. A análise estatística foi realizada com 5% de significância (p<0.05), analisados pelo método ANOVA complementado pelo Teste de Tukey. Os resultados demonstraram que os EHC e EHR possuem ação antioxidante e presença de fitocompostos. Os extratos apresentaram ação antibacteriana para todas as cepas avaliadas, quando os mesmos foram associados, obteve-se concentrações sinérgicas para as cepas clínicas de A. baumannii. Em relação a ação antibiofilme, o EHC inibiu a formação em 95% e EHR em 96% do biofilme de #Ab 1, enquanto a cepa #Pa 2 teve 92% e 93% de inibição quando em contato com EHC e EHR, respectivamente. Após tratamento de 24 h em biofilmes formados, as reduções da viabilidade foram de 72% para as cepas #Ab 2 e #Ab 3 quando em contato com o EHC, já EHR inibiu em 83% a viabilidade da cepa #Ab ATCC. Para P. aeruginosa (#Pa 2), as reduções da viabilidade foram de 84% e 88,5% quando tratados com EHC e EHR, respectivamente. A avaliação da citotoxicidade em HaCat demonstrou que após tratamentos com diferentes concentrações dos extratos a viabilidade celular se manteve acima de 70% em todos os grupos. Diante disso, conclui-se que os EHC e EHR apresentam importante ação antioxidante e antibacteriana, tanto em culturas planctônicas quanto em biofilmes, e não apresentaram efeitos citotóxicos na faixa de concentração testada. Bacterial resistance has progressively increased in the world, thus, there is a need for new treatment options. Phytotherapy has gained notoriety for fighting infections, mainly those caused by bacteria resistant to available antibacterials. In view of the above, the present study aimed to prepare and analyze the phytochemical composition and antimicrobial action of hydroethanolic extracts of cinnamon (EHC) and pomegranate (EHR) isolated and associated against planktonic cultures and biofilms of standard and clinical strains of Acinetobacter baumannii and Pseudomonas aeruginosa, in addition, analyze the cytotoxic action of the extracts on human keratinocytes (HaCat). For this, the EHC and EHR were prepared and the soluble solids content was quantified. Subsequently, the content of flavonoids and total phenols, antioxidant analysis through the reduction of the radical 2,2'-diphenyl1-picrylhydrazyl (DPPH), and phytochemistry by liquid chromatography (HPLC) were quantified. Regarding the antimicrobial action of the extracts, the broth microdilution test (CLSI ­ M7-A9) was applied and the synergistic action was performed through the checkerboard test. The most effective concentrations were analyzed on forming biofilms (prevention) and formed biofilms (24 h treatment), and viability was quantified using the MTT colorimetric test. To evaluate the cytotoxicity, the treatments were applied on HaCat cell culture for 24 h and analyzed using the MTT colorimetric test. Statistical analysis was performed with 5% significance (p<0.05), analyzed by the ANOVA method complemented by the Tukey test. The results showed that the EHC and EHR have antioxidant action and presence of phytocompounds. The extracts showed antibacterial action for all evaluated strains, when they were associated, synergistic concentrations were obtained for the clinical strains of A. baumannii. Regarding the antibiofilm action, EHC inhibited formation by 95% and EHR by 96% of the #Ab 1 biofilm, while the #Pa 2 strain had 92% and 93% inhibition when in contact with EHC and EHR, respectively. After 24 h treatment in formed biofilms, viability reductions were 72% for strains #Ab 2 and #Ab 3 when in contact with EHC, whereas EHR inhibited the viability of strain #Ab ATCC by 83%. For P. aeruginosa (#Pa 2), viability reductions were 84% and 88.5% when treated with EHC and EHR, respectively. The evaluation of cytotoxicity in HaCat showed that after treatments with different concentrations of extracts, cell viability remained above 70% in all groups. Therefore, it is concluded that EHC and EHR have important antioxidant and antibacterial action, both in planktonic cultures and in biofilms, and did not show cytotoxic effects in the tested concentration range. @#Punica granatum L.(pomegranate) is a medicinal plant belonging to the genus Punica Linn..The peel, seed, flower, leaf and root of P.granatum is widely used as traditional medicine in China.Phytochemical studies showed that the major chemical constituents of P.granatum were tannins, flavonoids, terpenes, alkaloids, phenolic acids, anthocyanins, fatty acids, etc.Biological studies on extracts and active ingredients of P.granatum show some bioactivities, such as antioxidant, hypoglycemic, anti-inflammatory, anti-tumor, antibacterial activities.Herein, the chemical constituents and pharmacological effects of different parts of pomegranate were reviewed, providing a theoretical basis for the further research and utilization of pomegranate.

ABSTRACT: The use of machine vision to recognize mature pomegranates in natural environments is of major significance in improving the applicability and work efficiency of picking robots. By analyzing the color characteristics of color images of mature pomegranates under different illumination conditions, the feasibility of the YCbCr color model for pomegranate image recognition under different illumination conditions was proven. First, the Cr component map of pomegranate image is selected and then the pomegranate fruit is segmented by the kernel fuzzy C-means clustering algorithm to obtain the pomegranate image. Contrast experiments of pomegranate image segmentation under different illumination conditions were then performed using the proposed kernel fuzzy C-means clustering algorithm, the fuzzy C-means clustering algorithm, the Otsu algorithm and the threshold segmentation algorithm. Results of the experiments verified the effectiveness and superiority of the proposed algorithm.


RESUMO: O uso de máquina para reconhecer romãs maduras em ambientes naturais é de grande importância para melhorar a aplicabilidade e a eficiência do trabalho de robôs de colheita. Ao analisar as características de cor das imagens coloridas de romãs maduras sob diferentes condições de iluminação, a viabilidade do modelo de cores YCbCr para o reconhecimento de imagens de romãs sob diferentes condições de iluminação foi comprovada. Primeiro, o mapa do componente Cr da imagem da romã é selecionado e, em seguida, o fruto da romãzeira é segmentado pelo algoritmo de agrupamento C-means fuzzy do kernel para obter a imagem da romã. Experimentos contrastados de segmentação de imagens de romã sob diferentes condições de iluminação foram então realizados usando o algoritmo proposto de agrupamento C-means fuzzy, o algoritmo fuzzy de agrupamento C-means, o algoritmo Otsu e o algoritmo de segmentação de limiares. Os resultados dos experimentos verificaram a efetividade e superioridade do algoritmo proposto.

The natural products derived from medicinal plants are proven to be the abundant source of biologically active compounds, which have been the basis of development of new pharmaceuticals. The present study was carried out to understand the antibacterial activity of methanolic extract of Punica granatum L. seed (MPG) against the clinically isolated urinary tract infections (UTI) causing bacteria i.e. Escherichia coli and Klebsiella pneumoniae using different methods i.e. disc diffusion, agar well diffusion, modified agar well diffusion. Colony forming units per ml (CFU/ml) were also determined for wild and MPG treated bacteria by spread plate and absorbance method. Phytochemical analysis of MPG and biochemical characterization of K. pneumoniae and E. coli was carried out using a standard protocol. MPG was a dark red colour sticky mass with a yield of 26.87% (w/w). Phytochemical analysis revealed the presence of steroids, tannins, coumarins, glycosides, flavonoids and phenolic compounds. The present investigation showed that MPG was effective against K. pneumoniae and E. coli. K. pneumoniae was a multi-drug resistant strain, found to be more sensitive to MPG in comparison to E. coli. The CFU/ml was found to be reduced in a dose-dependent manner in MPG treated urobacteria, which suggests that P. granatum seeds can be used as a remedy for UTI and also can be taken regularly to prevent the recurrence of UTI.

AIM To optimize the subcritical aqueous extraction for polysaccharides from the leaves of Punica granatum L.and to evaluate the in vitro antioxidant activity.METHODS With reaction pressure,solid-liquid ratio,extraction time and extraction temperature as influencing factors,yield of polysaccharides as an evalution index,the extraction was optimized by Box-Behnken method on the basis of single factor test.Then the scavenging effects of polysaccharides on hydroxyl free radical,superoxide anion and DPPH free radical were detected.RESULTS The optimal conditions were determined to be 5 MPa for reaction pressure,1 ∶ 27 for solid-liquid ratio,11 min for extraction time,and 155 ℃ for extraction temperature,the yield of polysaccharides was 1.809%.There was a dose-effect relationship between scavenging rate and polysaccharides' concentration.0.1 mg/mL Polysaccharides displayed the strongest scavenging effects on hydroxyl free radical,superoxide anion and DPPH free radical with the clearance rates of 57.36%,70.51% and 58.02%,respectively.CONCLUSION This stable and reliable method can be used for the subcritical aqueous extraction for polysaccharides from P.granatum leaves with obvious in vitro antioxidant activity.

ABSTRACT

Inflammatory bowel disease (IBD) is a common gastrointestinal disease in which the etiology and pathogenesis are still unclear. Generally speaking, it is caused by environment, inheritance, and immune. The related factors include immune system, oxidative stress, microflora in gastrointestinal tract, NF-κB, NO, COX-2, LB4, and the correlation factors. There are many plants that are effective to inflammatory bowel disease in traditional Uygur medicine, such as Solanum nigrum, Quercus infectoria, and Punica granatum. For some of these plants, only one or two mechanisms of action have been found. Besides, some constituents of them are effective to anti-inflammatory bowel disease. This article reviewed the role of factors in IBD and the mechanism of anti-inflammatory bowel disease of traditional Uygur medicine.

ABSTRACT

RESUMO O objetivo desse estudo foi observar a ação da romã (Punica granatum L.) em estomatites induzidas por queimaduras no dorso da língua de ratos Wistar. Foram utilizados 24 ratos Wistar machos adultos, provenientes do Biotério da UNIVASF. Foram formados quatro grupos (G1: Polpa da romã por gavagem; G2: Polpa da romã por gavagem + aplicação local do chá da casca do fruto; G3: aplicação local do chá da casca do fruto e G4: Controle negativo). As queimaduras foram confeccionadas com instrumental odontológico padrão. Os tratamentos foram realizados duas vezes ao dia, durante 14 dias. Metade dos animais de cada grupo (n=3) foi eutanasiada no sétimo dia de experimentação, enquanto o restante foi eutanasiado no 14º dia. As línguas foram removidas e fixadas com formaldeído a 10% tamponado, processadas com cortes de 5 µm e coradas em HE. Clinicamente, os animais do grupo G2 tiveram melhores resultados. Na análise histológica qualitativa foi avaliada a reepitelização e os graus de inflamação numa escala de 0 a 4. Na análise estatística, utilizou-se o teste Qui-quadrado de Pearson. Houve significância estatística (p=0,026 e p=0,023) quando se comparou o tratamento com os graus de reepitelização e inflamação nos quatro grupos estudados. O grupo G2 apresentou cicatrização completa com 14 dias. Os piores escores obtidos foram atribuídos ao Grupo G4 nos dois parâmetros de avaliação qualitativa. Diante dos resultados obtidos, observa-se que a romã (Punica granatum L.) possui ação cicatrizante na mucosa lingual de ratos Wistar.


ABSTRACT The objective of this study was to observe the action of pomegranate (Punica granatum L.) on stomatitis induced burns on the dorso-lingual musosa in Wistar rats. Twenty-four male, adult Wister albino rats were used, from the bioterium of UNIVASF. There were four groups (G1: Pomegranate juice by gavage; G2: Pomegranate juice by gavage + local application of fruit peel tea; G3: Local application of fruit peel tea only and G4: a negative control). The burns were made with standard dental instruments. The treatments were performed twice a day for 14 days. Half the animals in each group (n = 3) were euthanized on the seventh day of experimentation, while the remainder were euthanized on day 14. The tongues were removed and fixed with a 10% formaldehyde buffer, processed as 5µm sections and stained with HE. Clinically the animals treated with tea showed better healing. For statistical analysis the Pearson chi-squared test was used. There was a statistical significance (p = 0.026 and p = 0.023) when compared to treatment with the degree of re-epithelialization and inflammation of the four groups studied. The G2 group showed complete healing within 14 days. The worst scores were found in the G4 group in both qualitative assessment parameters. Based on these results, it was observed that pomegranate (Punica granatum L.) has a healing action on the lingual mucosa of Wistar rats.

INTRODUCCIÓN: la especie vegetal Punica granatum L. se conoce en Cuba como granada. A esta planta se le atribuye acción anticatarral en la medicina tradicional, entre otras propiedades. En estudios previos realizados al extracto hidroalcohólico liofilizado preparado con el fruto de esta planta, se informaron la presencia de numerosos componentes químicos, la mayoría flavonoides. Estos compuestos, unidos a la variedad de metabolitos secundarios detectados, le confieren a este liofilizado la posibilidad de poseer acción antiinfluenza, tal como ha sido demostrado en estudios previos del mismo. OBJETIVO: evaluar la acción virucida directa del extracto hidroalcohólico liofilizado de P. granatum frente a la cepa Influenza A/Mississippi/1/85 (H3N2), con diferentes variantes de tratamiento. MÉTODOS: las variantes de tratamiento directo se realizaron en 27 combinaciones de las variables; tiempo de contacto de: 60, 30 y 15 minutos; concentración del extracto de: 1000, 200 y 125 µg/mL y temperatura de incubación de la mezcla extracto-virus: 4, 25 y 37 0C. La presencia o no de la hemaglutinina del virus influenza en cada una de las muestras tratadas, se midió por la titulación de la Inhibición de la Hemaglutinación (IHA) y el título infectivo viral (DIE50). RESULTADOS: la concentración mínima efectiva para ejercer el extracto liofilizado, la acción virucida directa, in vitro, fue 125 µg/mL, a los 15 minutos. La acción virucida directa de éste extracto estuvo siempre presente en las diferentes temperaturas y tiempos de exposición ensayados. CONCLUSIONES: la variante de ensayo que utilizó 125 µg/mL, con independencia del tiempo y la temperatura de tratamiento, resultó suficiente efectiva para reducir la presencia de hemaglutinina del virus influenza.


INTRODUCTION: Punica granatum L. is known in Cuba as grenade. In traditional medicine, among other properties, antiflu action is attributed to this plant. In previous studies with the hydroalcoholic lyophilized extract, prepared with the fruit of this specie, several chemical compounds were reported, specially the presence of flavonoids. The variety of secondary metabolites detected for this lyophilized extract, added to a flavonoid fraction confers a high possibility of anti-influenza activity, as has been demonstrated in previous studies. OBJECTIVE: to evaluate the virucidal activity of the hydroalcoholic lyophilized extract of P. granatum against Influenza A/Mississippi/1/85 (H3N2), with different schemes of treatments. METHODS: the different schemes of treatments included 27 variants of the variables such as contact time of 60, 30 and 15 minutes; extract concentration of 1000, 200 and 125 µg/mL; an incubation temperature of the mix extract-virus of 4, 25 and 37 0C; and the corresponding replicas. The presence or not of hemaglutinin of influenza virus in each one of the treated samples, was measured by titration of Hemaglutination Inhibition (HAI) and the Embryo Infection Doses (EID50). RESULTS: the minimal effective concentration to show in the hydroalcoholic lyophilized extract the direct virucidal action in vitro was 125 µg/mL, in 15 minutes of contact. This virucidal action was always present in the different assessed temperature and time exposures. CONCLUSIONS: the treatment of 125 µg/mL, regardless of time and temperature of treatment, was effective enough to reduce the presence of hemaglutinin of the influenza virus.

ABSTRACT

Punica granatum L., locally known as romanzeira, is native to Asia but found throughout Brazil. P. granatum is used for treating inflammatory, infectious and respiratory diseases. The aim of this study was to evaluate the chromatography and genotoxicity of an aqueous extract of P. granatum (pomegranate) fruit peel using the Allium cepa L. test. The experiment set-up entailed 7 treatments: T1-distilled water, T2-tea 5 g.L-1, T3-tea 10 g.L-1, T4-glyphosate at 9.6%, T5-glyphosate with subsequent recovery in distilled water, T6-glyphosate with subsequent recovery in tea 5 g.L-1 and T7-glyphosate with subsequent recovery in tea 10 g.L-1. The rootlets were collected and fixed in ethanol:acetic acid (3:1) for 24 hours, then stored in 70% ethanol under refrigeration. Analysis was performed using high performance liquid chromatography for the quantification of the extracted phenolic compounds. Gallic acid, catechin, caffeic acid, and rutin were abundant in the extracts of P. granatum. The extracts were found to exhibit antiproliferative potential but not antimutagenic or genotoxic activity.


Punica granatum L., conhecida como romanzeira, é originária da Ásia e encontra-se distribuída por todo Brasil. É usada para o tratamento de doenças inflamatórias, infecciosas e respiratórias. Em decorrência da grande utilização de recursos fitoterápicos, é necessário esclarecer à população sobre a grande quantidade de substâncias existentes nas plantas e sobre os benefícios e prejuízos de tais substâncias à saúde. O presente trabalho objetivou realizar a análise cromatográfica e o estudo da genotoxicidade dos extratos aquosos das cascas dos frutos de P. granatum através do teste de Allium cepa L. Para a montagem do experimento, foram utilizados 7 tratamentos: T1-água destilada, T2-chá 5 g.L-1, T3-chá 10 g.L-1, T4-glifosato a 9,6%, T5-glifosato para recuperação em água destilada, T6-glifosato para recuperação em chá 5 g.L-1 e T7-glifosato para recuperação em chá 10 g.L-1. As radículas foram coletadas e fixadas em etanol:ácido acético (3:1) por 24 horas, e armazenadas em álcool 70%, sob refrigeração. Realizou-se análise por cromatografia líquida de alta eficiência para quantificação dos compostos fenólicos. Nos extratos de P. granatum foram observados em maior quantidade: ácido gálico, catequina, ácido cafeico e rutina. Além disso, os extratos demonstraram potencial antiproliferativo, sem apresentar atividade antimutagênica e genotóxica.

ABSTRACT

This article reviewed pharmacological study development of Punica granatum L. both at home and abroad in recent years. P. granatum has a long medical usage history. Different parts of P. granatum have wide applications in both traditional Chinese medicine (TCM) and other ethnic medicine. Its main chemical compositions were tannins, flavonoids, alkaloids and organic acids. It has obvious pharmacological activities. In recent years, the pharmacological research progress on P. granatum in terms of antibacterial, antiviral, hypoglycemic, antitumor, adjusting blood lipids, antioxidant, and immunoregulation has rapid development. The scientific collection and analysis on effects and new pharmacological research results of P. granatum are hoped to provide references for the study, clinical application, development and utilization of P. granatum and its products.

ABSTRACT

O uso de plantas medicinais no tratamento de doenças (fitoterapia) é uma prática de diversos povos, sendo realizada desde a Antiguidade. Diversos estudos são realizados para determinar a eficácia dessas plantas, sendo desconhecida a existência de interações e interferências na combinação das mesmas, quer seja benéfica, quer cause algum dano para o organismo. A fim de descobrir a existência dessas interações ou a interferência na ação do antibiótico amoxicilina testou-se Punica granatum e Plantago major contra as bactérias Staphylococcus aureus e Escherichia coli. A matéria prima foi obtida nas regiões de Montes Claros e Curvelo e seus extratos aquosos foi obtido através da decocção. A Concentração Inibitória Mínima (CIM) dos extratos foi determinada pela técnica de microdiluição em caldo. Para o teste de interferência realizou-se o teste de disco-difusão. No teste de microdiluição em caldo, Punica granatum apresentou CIM de 6,25% contra o S. aureus e de 25% contra E. coli. O extrato de Plantago major não apresentou atividade contra as bactérias. O teste de disco-difusão confirmou a existência de interações, sendo apenas quatro resultados significativos (Amoxicilina + Plantago major Pura (PgP), Amoxicilina + Plantago major Diluição 1 (PgD1) e Amoxicilia + Plantago major Diluição 2 (PgD2) contra E. coli e Amoxicilina + PgP contra S. aureus). Conclui-se que existe interação dos extratos entre si e dos extratos com a amoxicilina, o que também pode acontecer com outros antibióticos e gerar riscos para a população. Portanto é indispensável à realização de testes que complementem essa pesquisa oferecendo mais informação à população para segurança na utilização de plantas medicinais. informações à população assegurando sua segurança na utilização de plantas medicinais.


The use of medicinal plants in treating diseases (herbal medicine) is a practice of many peoples, being done since ancient times. Several studies are conducted to determine the effectiveness of these plants, but the existence of interactions and interference with their combined use, whether beneficial or detrimental to the body, are unknown. In order to discover the existence of these interactions or interferences in the activity of the antibiotic amoxicillin, we tested in this research Punica granatum and Plantago major against Staphylococcus aureus and Escherichia coli. The raw material was obtained in the regions of Montes Claros and Curvelo, and their aqueous extracts were obtained from decoction. The minimum inhibitory concentration (MIC) of the extracts was determined by the broth microdilution technique. For interference, we performed the disk diffusion test. In the broth microdilution test, Punica granatum had MICs of 6.25% against S.aureus and 25% against E. coli. The Plantago major extract showed no activity against the bacteria. The disk diffusion test confirmed the existence of interactions and only four significant results (amoxicillin + pure Punica granatum (PgP), amoxicillin + Punica granatum dilution 1 (PgD1) and amoxicillin + Punica granatum dilution 2 (PgD2) against E. coli and amoxicillin + PgP against S. aureus). We conclude, as a result, that there really is interaction between the extracts themselves and between the extracts and amoxicillin, which can also occur with other antibiotics and cause some risk to the population. Therefore, it is essential to carry out tests to complement this research, providing more information to the public and ensuring the safety in the use of medicinal plants.

ABSTRACT

OBJECTIVE: To study the in vivo metabolism of granatum tannins by liquid chromatography-mass spectrometry and clarify their metabolic pathways. METHODS: The rat urine samples were collected after intragastric administration of tannins preparation of Punica granatum L. husk. The analysis was performed on a Venusil MP-C18 column with gradient elution system of acetonitrile and water containing 0.1% formic acid. Using electrospray ionization (ESI) in negative ion mode with scanning range from m/z 100 to 1000, the metabolites were identified by comparing the MS spectra and MS data with the literatures. RESULTS: Seven metabolites were detected in the rat urine, among which six were identified according to their second mass spectra to be U-1 (urolithin B), U-2 (urolithin A methyl ether), U-3 (urolithin A), U-4 (urolithin A glucuronide), U-5 (ellagic acid) and U-6 (urolithin C). CONCLUSION: Tannins in Punica granatum L. husk are metabolized after oral administration mainly through hydrolysis, glucuronidation gly-cosidation, decomposition and methylation reaction to generate a series of derivatives of urolithin. These results preliminarily indicate the metabolic pathway and material basis of the pharmacological activity of tannins in Punica granatum L. husk in rats.

Introducción: Punica granatum L., (Punicaceae) es una especie conocida en Cuba como granada. Algunos fitocomponentes presentes en la fruta muestran una fuerte actividad antioxidante. En la medicina tradicional esta planta es empleada en el tratamiento de infecciones parasitarias, diarreas y disentería, así como en el tratamiento de asma, catarro, fiebre, inflamaciones y otras. En años recientes, la actividad biológica de los polifenoles presentes en la fruta de esta especie, ha despertado el interés de los investigadores y la industria farmacéutica.Objetivo: establecer un proceso a escala de laboratorio que permita extraer los polifenoles totales de la fruta.Métodos: se aplicó un proceso de maceración para la extracción de los polifenoles de la fruta y se realizó la optimización de los parámetros mediante diseños experimentales. Se evaluaron las variables independientes: tiempo de extracción, relación droga-menstruo y concentración alcohólica.Resultados: las tres variables presentaron una influencia significativa sobre el proceso de extracción de polifenoles.Conclusiones: en las condiciones de trabajo establecidas, la extracción óptima es la siguiente: tiempo de extracción de 72 h, relación droga-menstruo de 1:20 y concentración alcohólica del 50 por ciento (v/v)


Introduction: Punica granatum L., (Punicaceae) is one species known in Cuba as granate. Some phytocomponents of the fruit show strong antioxidant activity. This plant is used in the traditional herb medicine to treat parasitic infections, diarrheas and dysentery, asthma, flu, fever, inflammation and other illnesses. In recent years, the biological activity of phenols present in the fruit has aroused the interest of both researchers and the drug industry. Objective: to set a lab-scale process that makes it possible to extract total phenols from the fruit. Methods: a maceration process was used to extract phenols from the fruit and the parameters were optimized through experimental designs. The independent variables were time of extraction, drug/menstruum ratio and alcohol concentration. Results: the three variables had significant influences over the process of extraction of polyphenols. Conclusions: under the set working conditions, the optimal extraction is as follows: time of extraction of 72 hours, drug/menstruum ratio of 1:20 and alcohol concentration of 50 percent(v/v)

ABSTRACT

For thousands of years, Punica granatum L. has been used in traditional medicine all over the world and predate the introduction of antibacterial drugs. The aim of the present study was to investigate the antibacterial activity of aqueous and ethanolic extracts of Punica granatum L. bark obtained by decoction and maceration. The different extracts of Punica granatum L. (Lythraceae) bark have been tested for antibacterial activity against Gram-positive bacteria (Staphylococcus aureus, Bacillus stearothermophilus) and Gram-negative bacteria (Escherichia coli, Pseudomonas aeruginosa) by disc diffusion method. The ethanolic macerate extract showed the strong in vitro antibacterial activity against Pseudomonas aeruginosa with zone inhibition of 24.4 mm. However, the results tests by disc diffusion method revealed the effectiveness of ethanolic decoctate against Gram-positive bacteria (Staphylococcus aureus and Bacillus stearothermophilus) with diameter zone of inhibition varying with 21.1mm and 23.75 mm respectively.

ABSTRACT

Punica granatum L., Punicaceae, amplamente usada no Brasil, foi avaliada quanto ao seu potencial antitumoral in vitroe in vivo. Investigou-se in vitro a citotoxicidade do extrato etanólico do fruto e folha da P. granatumutilizando células K-562 e células do Tumor Ascítico de Ehrlich (TAE), pelos métodos de redução do MTT e exclusão do azul de tripano. Nos estudos in vivoavaliou-se o aumento da sobrevida de animais portadores do TAE e tratados, por via oral, com diferentes doses dos extratos etanólicos da P. granatum(12,5; 25; 50 e 100 mg/kg) por dez dias consecutivos. Além disso, nestes animais analisou-se o potencial de inibição tumoral e a atividade antiangiogênica da P. granatum. Os resultados dos estudos in vitrodemonstraram uma redução na viabilidade das células K-562 e do TAE, concentração-dependente, nos métodos investigados. Os resultados in vivo demonstraram aumento da sobrevida dos animais portadores do TAE tratados, de forma dose-dependente. Em paralelo, observou-se diminuição do número de células tumorais na cavidade peritoneal dos animais portadores e tratados. Além disto, os tratamentos empregados reduziram o padrão de vascularização da parede abdominal. Dessa forma, os dados apresentados revelaram que o extrato de P. granatumpossui atividade antitumoral in vitroe in vivo em paralelo a redução da angiogênese peritoneal.


Punica granatum L., a plant widely used in Brazil, was tested for its antitumor and antiangiogenic activities in vitroand in vivo. In this work, the in vitrocytotoxicity was evaluated using the K-562 cell line and Ehrlich ascites tumour cells, by MTT tetrazolium reduction test and the trypan blue exclusion test. In vivostudies investigated the increase in the survival time of Ehrlich tumour-bearing mice after treatment with different doses of Punica granatumL. ethanol extract (12.5; 25; 50 e and 100 mg/kg), by gavages, for ten consecutive days. In addition, we also investigated the tumour inhibition potential and antiangiogenic activity of this plant. In vitroresults demonstrated a decrease of K-562 and Ehrlich ascites tumour cells viability, with both methods used, in a dependent-manner concentration. In vivoresults showed a significant antitumor activity against Ehrlich ascites tumour growth, increasing survival time. In parallel, we detected a significant inhibition of the tumour growth, along with a decrease in the vascular pattern of the peritoneal wall. Thus, the data presented herein clearly showed that Punica granatum L. has antitumor and antiangiogenic activities.
