ABSTRACT

OBJECTIVETo investigate the protective effects and mechanisms of Radix Astragali Injection on multiple organs of rats with obstructive jaundice (OJ).

METHODSA total of 180 rats were randomly divided into the sham-operated, model control and treated groups (60 in each group). On 7, 14, 21 and 28 days after operation, the serum contents of alanine aminotransferase (ALT), aspartate aminotransferase (AST), r-glutamyl transpeptidase (r-GT), total bilirubin (TBil), direct bilirubin (DBil), blood urine nitrogen (BUN), and creatinine (CREA) were determined. And the pathological changes of livers, kidneys and lungs, and protein expressions of toll-like receptor-4 (TLR-4) of livers, intercellular adhesion molecule-1 (ICAM-1) of lungs, Bax and nuclear factor-kappa B (NF-κB), as well as apoptotic indexes of multiple organs were observed, respectively.

RESULTSThe pathological severity scores of multiple organs (including livers on 7, 14, 21 and 28 days, kidneys on 14 and 28 days, and lungs on 14 days), serum contents of ALT (14 and 21 days), AST (14 days), TBil (7, 14, 21 and 28 days), DBil (14 and 21 days), BUN (28 days), protein expressions of TLR-4 (in livers, 28 days), Bax (in livers and kidneys, 21 days), and apoptotic indexes in livers (7 and 21 days) in the treated group were significantly lower than those in the model control group (P<0.05 or P<0.01).

CONCLUSIONRadix Astragali Injection exerts protective effects on multiple organs of OJ rats by improving the pathological changes of lung, liver and kidney, decreasing the serum index of hepatic and renal function as well as inhibiting the protein expression of TLR-4 and Bax in the livers and Bax in the kidneys.

ABSTRACT

OBJECTIVE:To study the inhibitory action of Radix Astragali injection on human nasopharyngeal carcinoma CNE-2 cell line in vitro.METHODS:Human nasopharyngeal carcinoma CNE-2 cell were treated with different concentrations of Radix Astragali injection(trial group),the cell proliferation state was determined by adenosine triphospate -bioluminescence tumor chemosensitivity assay, cell apoptosis was observed by cell morphology and flow cytometry, and which were compared with the control group.RESULTS:Compared with the control group, the trial group showed significant inhibitory action on the proliferation of the trial cell line(P

ABSTRACT

Objective To observe the therapeutic effect of Chinese medicine combined with ion-introduction of Radix Astragali Injection (RAI)for herpes simplex keratitis (HSK).Methods Ninety HSK patients (104 affected eyes)were randomized into two groups. Both of the two groups received anti-virus drugs, and the treatment group (45 patients with 53 affected eyes)received oral use of decoction of Chinese medicines (mainly composed of Semen Cassiae,Fructus Viticis,Periostracum Cidadae,Ramulus Uncariae cum Uncis,Fructus Gardeniae,Fructus Forsythiae,Flos Chrysanthemi,Rhizoma Chuanxiong, etc.),and ion-introduction of RAI.The changes of vision and clinical symptoms before and after treatment were observed, and a 2-year follow-up was carried out. Results In the treatment group,19 eyes were cured,27 effective,7 ineffective, and the total therapeutic effective rate being 86.79%;in the control group,10 eyes were cured,21 effective,20 ineffective, and the total therapeutic effective rate being 60.78%.The therapeutic effect in the treatment group was superior to that in the control group (P

ABSTRACT

AIM: To examine the quality of Radix Astragali Injection from different manufacturers. METHODS: The HPLC-ELSD method for the determination of astragaloside Ⅳ in Radix Astragali Injection was established with hypersil ODS 2 C 18 column(250mm?4.6mm,5?m), column temperature at 40℃. The mobile phase was acetonitrile-water (36∶64) and flow rate was at 1.0mL?min -1. The evaporation light-scattering detector was used with its drift tube temperature at 100℃ and the gas flow rate: 3.0L?min -1. RESULTS: Within 2.4~12?g astragaioside Ⅳ had a good linearity. The average recovery was 101.6%, RSD=2.9%,n=5. By analyzing the samples of 10 batch of Radix Astragali Injection from 5 manufactuers, it showed there were significant difference in the contents of astragaloside Ⅳ. CONCLUSION: The method is simple and fast. The results are accurate and reproducible and can be used in the quality control of Radix Astragali Injection. The significant differences in the astragaloside content may influence clinical treatmem.
