ABSTRACT

Objective To explore the mechanisms of synergic action of commonly-used herb pair of Radix Sophorae Flavescentis (Kushen, KS) and Fructus Cnidii (Shechuangzi, SCZ) for the treatment of pruritus. Methods We predicted and analyzed the potential targets of KS and SCZ based on network pharmacology method, and then established Chinese herbs-compound skeletons-targets networks to reveal anti-pruritus targets, predicting targets, and the interaction of KS and SCZ, as well as the components of the herb pair. The RAW264.7 inflammatory cell model was established to observe the synergistic anti-inflammatory activity of KS and SCZ. Results KS and SCZ had possible synergic action on the pruritus-related targets such as histamine receptors, cannabinoid receptors and proteinase-activated receptor 2. Additionally, KS and SCZ probably had synergic regulation of inflammation-related pathway (Toll -like receptor signaling pathway , chemokine signaling and Fc epsilon RI signaling pathway) and nerve-related pathway(neurotrophin signaling pathway) during the treatment of pruritus. Flavones from KS and coumarin of SCZ had various synergistic anti-inflammatory activities(P<0.05), indicating that they played an important role in inhibiting pruritus induced by inflammation. Conclusion The method may reveal the molecular mechanism of KS and SCZ in inhibiting pruritus, which is important for the modernization of Chinese medicine and new drug development.

ABSTRACT

Objective To investigate the regulative effect of the extract of Radix Ranunculi Ternati,Radix Sophorae Flavescenti,Prunella vulgaris L.and Stellera chamaejasme L.on cellular immunity induced by multiple drugs resistant bacillus tuberculosis(MDR-TB) from pneumoconiosis patients complicated with tuberculosis in rats.Methods MDR-TB model in rats was induced by MDR-TB.Normal control group were feed by standard feed,model group were irrigated by normal saline,and the other groups were respectively feed by the extract of Chinese herbal medicines.The content of IFN-γ,IL-4,IL-10 and IL-12 were examined by ELISA.RT-PCR was used to detect the mRNA levels of them.Results The content of IFN-γof the four extracts of Chinese herbal medicines were (2.01 ±0.73 ),( 1.92±0.56),( 1.98 ±0.67 ) and (1.94±0.59) pg/ml,IL-4 were (6.01±1.46),(6.12±1.35),(6.47±1.46) and (6.15±1.44) pg/ml,IL-10 were (12.09±3.07),( 12.45±4.01 ),( 12.13±3.43) and (12.54±3.78) pg/ml,IL-12 were (2.99±0.89),(2.75±0.84),(3.02±0.86) and (2.89±0.75) pg/ml.Compared to the model group,they resulted in significant in serum IFN-γ,IL-12,IL-4 and IL-10 (P<0.05 or P<0.01 ),the mRNA levels of them were significantly (P<0.05 or P<0.01 ).Conclusion The four extracts of Chinese herbal medicines can enhance the cellar immunological function in rats from up-regulation of the level of genetic transcription,accordingly provide the theory base of healing of pneumoconiosis patients complicated with tuberculosis with them.

ABSTRACT

Objective To study the refining of Radix Sophorae Flavescentis (RSF) decoction by microfiltration combined with ultrafiltration method,and to compare the refining effect with that refined by alcohol sedimentation method.Methods HPLC was applied to analyze the content of oxymatrine,ultroviolet (UV) spectrophotometry was used to determine the content of total flavonoids in RSF decoction,and suitable concentration of NaOH solution for the detergent were chosen in order to wash the membrane.Results The refining effect is better and the recovery ratio of efficient components is higher in RSF decoction refined by microfiltration combined with ultrafiltration method than in those refined by alcohol sedimentation method.Microfiltration combined with ultrafiltration method is of simple operation and short period,and ultrafiltration membranes can be easily washed.Conclusion The microfiltration combined with ultrafiltration method can be used as the alternative for the traditional alcohol sedimentation method in refining of RSF decoction.

ABSTRACT

Objective To establish the quality standard for Xuanshi Texiao Yaoshui(mainly composed of Radix Sophorae Flavescentis,Cortex Pseudolaricis,etc).Methods Radix Sophorae Flavescentis and Cortex Pseudolaricis were identified by TLC.The content of oxymatrine was determined by HPLC.Symmetry C18 column(4.6 mm? 250 mm,5 ? m)with mobile phase of methanol-water-triethylamine(40:60:0.04)was used.The detective wavelength was 220 nm.Results TLC for Radix Sophorae Flavescentis and Cortex Pseudolaricis was specific .HPLC for oxymatrine had a good separation with other components,and the linear range of oxymatrine was 0.60~ 1.80 ? g and recovery rate was 99.34 %(RSD =0.71 %).Conclusion This method can be used to control the quality of Xuanshi Texiao Yaoshui.

ABSTRACT

AIM: To establish the quality standard for Lixieling Pill(Herba Andrographis; Radix Sophorae flavescentis etc.). METHODS: Herba Andrographis and Radix Sophorae flavescentis in Lixieling Pill were identified by TLC. Andrographolide, dehydroandrographolide, matrine and oxgmatrine of pill were determined by HPLC together. RESULTS: The characteristic identification by TLC was distinct and highly specific. The quantitative evaluation of andrographolide had the linear range of 0.969 -2.422 ?g, The average recovery was 98.45% and RSD was 0.88. The quantitative evaluation of dehydroandrographolide had the linear range of 0.481 -1.204 ?g , The average recovery was 100.53% and RSD was 0.63. The quantitative evaluation of matrine had the linear range of 1.032 -2.064 ?g , The average recovery was 99.06% and RSD was 1.04. The quantitative evaluation of oxgmatrine had the linear range of 1.016 -2.032 ?g , The average recovery was 99.67% and RSD was 1.20. CONCLUSION: The method is reliable, accurate and specific. It can be used for quality control of Lixieling Pill.

ABSTRACT

AIM: To establish the quality standard for Jieeryin Effervescent Tablets (Fructus Cnidii, Folium Artemisiae Argyi, Radix Sophorae Flavescentis, Cortex Phellodendri, etc.). METHODS: Radix Sophorae Flavescentis、Cortex Phellodendri、Radix Scutellariae and Fructus Gardeniae in Jieeryin Effervescent Tablets were identified by TLC. The content of geniposide was determined by HPLC. RESULTS: The average recovery was 99% and relative standard deviation was 1.6%. CONCLUSION: The precision and reproducibility of the method are good. The method can be used for quality control of Jieeryin Effervescent Tablets.

ABSTRACT

AIM:To establish an assay of oxymatrine and matrine in Compound Shiwei Tablets(Folium Pyrrosiae, Radix Sophorae Flavescentis, Radix Astragali, Herba Polygoni Avicularis, etc.) and Radix Sophorae Flavescentis by HPLC. METHODS: Diamonsil C 18 (10 ?m,250 mm?4.6 mm) analytical column was used.The mobile phase consisted of methanol-0.2% phosphoric acid solution (45∶55). The detection wavelength was 220nm. RESULTS : The average recoveries of oxymatrine and matrine in Compound Shiwei Tablets were 100.3% and 100.6% , respectively and in Radix Sophorae Flavescentis were 101.0% and 101.2%, respectlively. CONCLUSION: This method could be used for the quality evaluation of Compound Shiwei Tablets and Radix Sophorae Flavescentis.

ABSTRACT

Objective: To intvestigate the effects of processing on contents of matrine and oxymatrine of Radix Sophorae Flavescentis. Methods: The determinations of matrine and oxymatrine were carried out by dual wavelength TLC scanning. Results: There were obvious differences between the crude and the different processd products of Radix Sophorae Flavescentis in matrine and oxymatrineConclusion: The processing temperature, material, wine and vinegar can make the contents of matrine and oxymatrine of Radix Sophorae Flarescentis change.
