ABSTRACT

ObjectiveTo observe the therapeutic effect of Radix Paeoniae Rubra-Radix Aconiti Lateralis on acute-on-chronic liver failure (ACLF) rats and its effect on M1/M2 macrophage polarization. MethodMale SD rats were randomly divided into normal group, model group, positive group (lactulose, 1.8 g·kg-1) and traditional Chinese medicine (TCM) group (Radix Paeoniae Rubra-Radix Aconiti Lateralis, 5.85 g·kg-1), six in each group. The ACLF rat model was established by subcutaneous and tail vein injection of bovine serum albumin combined with intraperitoneal injection of D-galactosamine+lipopolysaccharide. Then the modeled rats were intervened with corresponding drugs for one week. The normal group and model group were given the same dose of distilled water. The histopathological changes of liver tissue were observed by hematoxylin-eosin (HE) staining. The mRNA and protein expression levels of CD86, inducible nitric oxide synthase (iNOS), CD206 and arginase 1 (Arg1) were detected by real-time polymerase chain reaction (Real-time PCR), Western blot and immunohistochemistry. ResultCompared with the conditions in the normal group, pseudolobule formation in liver tissue and morphological changes and necrosis of hepatocytes were observed in ACLF rats, accompanied by a large number of inflammatory cell infiltration. Moreover, the mRNA and protein expression levels of CD86, iNOS were up-regulated(P<0.01). Compared with the model group, the treatment groups had improved necrosis and inflammatory infiltration of hepatocytes, down-regulated mRNA and protein expression of CD86 and iNOS (P<0.01) and up-regulated mRNA and protein expression of CD206 and Arg1 (P<0.05, P<0.01), with the up regulation in the TCM group better than that in the positive group. ConclusionACLF rats had unbalanced M1/M2 macrophage polarization, and the imbalance shifted towards M1. Radix Paeoniae Rubra-Radix Aconiti Lateralis inhibited the activation of M1 macrophages and reduced the inflammatory response of liver failure by promoting the polarization of liver macrophages towards M2.

ABSTRACT

<b>Objective::To investigate the effect of Aconiti Lateralis Praeparata Radix combined with Glycyrrhizae Radix et Rhizoma on mRNA expression of cytochrome P450 (CYP450) enzymes in rat' s heart. <b>Method::Male Sprague Dawley rats were randomly divided into the control group, the inducer group (0.08 g·kg<sup>-1</sup>·d<sup>-1</sup>), the aconite group (0.5 g·kg<sup>-1</sup>·d<sup>-1</sup>), the licorice group (0.5 g·kg<sup>-1</sup>·d<sup>-1</sup>) and the compatibility group (aconite and licorice group 0.5 g·kg<sup>-1</sup>·d<sup>-1</sup>), with 6 rats in each group. In each group, the serum concentrations of aspartate transaminase (AST), creatine kinase (CK) andlactate dehydrogenase (LDH) were respectively measured. Hematoxylin-eosin(HE) staining was used to detect the pathologic changes in the heart tissue of the rats. The expression of several CYP genes were measured. <b>Result::After combination, the contents of AST, CK and LDH were lower than those of the aconite group. At the same time, the pathological results showed cardiac injury in rats in the aconite group, and licorice could alleviate the cardiac injury caused by aconite. At the level of gene transcription, the glycyrrhizae group could up-regulate the expressions of CYP2C11 and CYP2J3 to different degrees (<italic>P</italic><0.05), and the combination of licorice and aconite could up-regulate the expression of CYP2B1, CYP2C11 and CYP2J3 family (<italic>P</italic><0.05). It suggested that the combination could promote the formation of EETs in arachidonic acid(AA) metabolism, and the aconite could up-regulate the expressions of CYP4A1, CYP4A3, CYP4F5 and CYP4F6 in CYP4 family (<italic>P</italic><0.05), promote the formation of 20-hydroxyeicosatetraenoic acid(20-HETE). The combination of aconite and licorice can weaken the ability of aconite, down-regulate the expression of CYP4A3, CYP4F1, CYP4F5 and CYP4F6 family mRNA (<italic>P</italic><0.05), inhibit the formation of 20-HETE and reduce the cardiotoxicity caused by aconite. <b>Conclusion::The combination of licorice and aconite can regulate the expression of CYP450 enzyme in the heart, increase the production of EETs and the content of 20-HETE, and finally play a role in reducing toxicity.

ABSTRACT

OBJECTIVE: To explore the optimum harvest time of the Mongolian medicine, Radix Aconiti kusnezoffii herbs, by analysis of the changes of the main components in different developmental stages. METHODS: HPLC was used to determine the contents of the main alkaloids in 6 kinds of Radix Aconiti Kusnezoffii which were harvested in sprouting stage, early flowering stage, flowering stage and withering period, respectively. Taking the content of total alkaloids as index, the appropriate harvest time of Radix Aconiti kusnezoffii was determined. RESULTS: From germination to withering period, the total alkaloids in the 6 kinds of Radix Aconiti Kusnezoffii displayed increasing and then decreasing trend. The highest contents existed in the early flowering stage and the lowest existed in the withering period. The contents of the main components were sorted from high to low in the early flowering stage, germination stage, flowering stage and withering period. CONCLUSION: Based on the unique processing METHODS in Mongolian medicine and clinical medication safety, the withering period of Radix Aconiti Kusnezoffii is the best harvest time and this is consistent with the traditional Mongolian harvest period.

The dried root of Ranunculaceae Aconitum perennial herb of North Aconitum, is commonly used. It has the function of cardiac stimulation, anti-inflammatory and analgesic, anti-tumor and immune regulation. However, its toxicity lower the safety and applicaiton. The pharmacological and toxicological effects were discussed in this paper, in order to provide reference for the intoxication diagnosis and clinical safety.

ABSTRACT

Objective: Comparison of alkaloid and polysaccharides content in different parts of whole plant of wild aconitum in Nanyang, and to explore the medicinal value of aboveground part and fibrous root of wild aconitum in Nanyang. Methods: The contents of three monoester alkaloids, three diester alkaloids in wild aconitum, radix aconiti carmichaeli, fibrous root and aboveground parts in Nanyang were determined by HPLC method; The content of crude polysaccharides was determined by sulfuric-phenol method, crude polysaccharides content from different tissues of wild aconitum in Nanyang in different harvest time was compared, including wild aconitum in Nanyang, wild radix aconiti carmichaeli in Nanyang, fibrous root, aboveground part. Results: Among the 4 parts of the wild Aconitum in Nanyang, the contents of 3 mono ester alkaloids rang from high to low: wild aconitum in Nanyang> wild radix aconiti carmichaeli in Nanyang> fibrous root> aboveground part (P < 0.05), the contents of three diester alkaloids rang from high to low: wild radix aconiti carmichaeli in Nanyang> fibrous root> wild aconitum in Nanyang> aboveground part (P < 0.05), the order of total alkaloid content from high to low is: wild radix aconiti carmichaeli in Nanyang> fibrous root> wild aconitum in Nanyang> aboveground part (P < 0.05), The content of crudepolysaccharides in different tissues of wild aconitum in Nanyang were in the sequence of wild radix aconiti carmichaeli in Nanyang> wild aconitum in Nanyang> aboveground part> fibrous root (P < 0.05) . Conclusion: In this study, an attempt was made to explore the differences of 6 kind salkaloid content in different parts of whole plant of wild aconitum in Nanyang, which provided scientific basis for the medicinal value of the fibrous roots and the aboveground parts.

It includes five parts:modern basic study of Radix Aconiti Lateralis Praeparata; research on toxicity-reducing and efficacy-enhancing of complete or separate prescription of Sini Tang; research of the herb-pair in compatible prescription; studying the efficacy of herbal prescription of Radix Aconiti Lateralis Praeparata by utilizing complex biological networks; further research of Radix Aconiti Lateralis Praeparata and its prospect.

ABSTRACT

Currently, rapid detection of effective components in synthetic drugs and herbal medicine remains an important and difficult issue of medicine research. A novel ionization technique, called dielectric barrier discharge ionization ( DBDI ) , has strong ionization ability, and is suitable for weak polar substances. Besides, this technique possesses many intrinsic advantages, such as simplicity, rapidity, no complicated sample pretreatment, etc. In this study, a new DBDI ion source, based on single electrode technique, was used to detect four weak polar synthetic drugs. The results showed that the protonated molecular ions [ M+H]+of four weak polar synthetic drugs were observed obviously. What′s more, the DBDI ion source was also used for the rapid analysis of Radix Aconiti and Radix Aconiti Preparata pieces without any sample pretreatment. The result showed that the protonated molecular ions [M+H]+ and fragment ions [M+H-60]+of aconitine, hypaconitine, and mesaconitine were detected in Radix Aconiti. And only the fragment ions [M+H-60]+were detected in Radix Aconiti Preparat. The researches indicated that diester aconitine and monoester aconitine were the main effective components of Radix Aconiti Radix and Aconiti Preparat, respectively. The new DBDI ion source provided a fast and reliable method to identify effective components of medicine, showing a broad application prospects in synthetic drugs and herbal medicine research.

ABSTRACT

Objective To study the influence of the combined administration of Bletilla striata and Radix aconiti preparata with different doses or ratios on anhydrous ethanol-induced gastric mucosal injury in mice with gastric ulcer. Methods The uniform design method was adopted on the basis of two factors and seven levels, which aims to investigate the influence of combined oral administration of Bletilla striata and Radix aconiti preparata with different doses or ratios on gastric mucosal injury in the mice with gastric ulcer. The indices like gastric ulcer index and stomach mucosa damage index were used to help selecting the right dose or ratio of decoctions for further study. Results The combined administration of Bletilla striata and Radix aconiti preparata at a certain proportion could reduce gastric ulcer index and stomach mucosa damage index. According to the regression analysis, the gastric ulcer index and stomach mucosa damage index were obviously inhibited by Bletilla striata, while this function decreased with the combined administration of Bletilla striata and Radix aconiti preparata. However, an interactive effect between each other has not been found. The influence of decoctions on the gastric ulcer index and stomach mucosa damage index became less when the compatibility ratio of Radix aconiti preparata increased, which referred to the total dose ranged between 3.64 g/kg and 29.32 g/kg. Conclusions The effect of the combined administration of Bletilla striata and Radix aconiti preparata showed relationship among the doses and ratios on anhydrous ethanol-induced gastric mucosal injury in mice with gastric ulcer.

ABSTRACT

This article was aimed to study the acute toxicities on intragastric administration of differentRadix Aconiti Lateralis Praeparataprocessed products to Beagle dogs. A total of 16 healthy and qualified Beagle dogs were randomly divided into the blank group,Pao-Fu-Pian(PFP) group,Pao-Tian-Xiong(PTX) group andHei-Shun-Pian(HSP) group according to the body weight. The intragastric administration of 4 g crude herb per kg was given. Before medication, 1 h, 24 h, and 3, 7, 14 days after medication, the body weight, food consumption, rectal temperature, electrocardiogram, blood routine and blood biochemistry were measured. The results showed that after medication, all dogs in three experimental groups were depressed. And there were significant differences in the electrolytes of blood. Among them, the HSP group was the most obvious one. The red blood cells, blood sugar and triglycerides of dogs in the PFP group had significant difference. The lymphocytes and blood sugar had significant difference of dogs in the PTX group. However, after the medication of HSP, the lymphocytes of the dogs were decreased significantly. It was concluded that the toxicity of three processed products followed the order of HSP > PFP > PTX.

ABSTRACT

OBJECTIVE To study the mitochondrial toxicity effect of Radix Aconiti Lateralis Praepa?rata(Fuzi)on H9c2 cardiomyocytes. METHODS H9c2 cells were exposed to Fuzi decoction 6.25, 12.5,25,50 and 100 g·L-1 for 24 h. Fluorescence staining and CCK-8 assay were used to detect cell viability. H9c2 cells were exposed to Fuzi decoction 6.25,12.5 and 25 g · L-1 for 24 h,while the effect on mitochondrial membrane potential and reactive oxygen species(ROS)was detected by flow cytometry. The fluorescence molecular probe and laser scanning confocal microscope were used to observe the effect on Ca2+ in cells,Ca2+ and superoxide in mitochondria. The effect on ATP concentration in cells was detected via firefly luciferin and the expression of Pgc-1α,Bcl-2 and Bax mRNA evaluated by real-time PCR,while the expression of Pgc-1α protein was measured by Western blotting. RESULTS H9c2 cell viability was significantly inhibited by Fuzi decoction 12.5-100 g · L-1(P<0.05,P<0.01). The IC50 value was 47.4669 g · L-1,while the 95%confidence limit was 32.5997-69.1145 g · L-1. After treatment with Fuzi decoction 25 g · L-1 ,the fluorescence intensity of ROS in the normal control group increased from 204±67 to 454±78(P<0.05),that of mitochondrial superoxide increased from 5.4±1.8 to 26.8±8.5 (P<0.01),mitochondrial membrane potential decreased from 1.7±0.5 to 0.8±0.4(P<0.05),the fluores?cence intensity of intracellular Ca2+increased from 7.8±0.8 to 22.1±0.5(P<0.05)while that of mitochon?drial Ca2+decreased from 38.0±4.3 to 9.2±1.6(P<0.01),and intracellular ATP concentration decreased from (10.6 ± 0.4)μmol · g-1 to (5.3 ± 1.1)μmol · g-1 protein (P<0.05). qPCR and Western blotting test results showed that compared with the normal control group ,Pgc-1αand Bcl-2 mRNA relative expression level in Fuzi decoction 25 g·L-1 group was decreased from 1.00±0.10 and 1.00±0.10 to 0.09±0.06(P<0.01)and 0.43±0.06(P<0.01),respectively, while the relative expression of Bax mRNA was increased from 1.00 ± 0.03 to 1.17 ± 0.06 (P<0.05),and the expression of Pgc-1α protein was decreased from 0.906±0.034 to 0.541±0.003(P<0.01). CONCLUSION Fuzi has some mitochondrial toxicity to cardiomy?opathy. This effect arises from the combined action of different mechanisms. Mitochondrial toxicity of myocytes may account for the cardiac toxicity of Fuzi.

ABSTRACT

Theultraperformanceliquidchromatographycoupledwithmassspectrometry(UPLC-MS)was used to investigate the metabolic difference of the decoction of Radix Aconiti Preparata ( RAP ) and its co-decoctions with Radix Paeoniae Alba ( RAP-RPA ) or Radix Stephaniae Tetrandrae ( RAP-RST ) in rat intestinal bacteria. The principal component analysis ( PCA) of the relative contents of Aconitum alkaloids after metabolism was performed by SIMCA-P software. The score plots of PCA could successfully distinguish the three groups of RAP, RAP-RPA and RAP-RST. The result indicated that the differences of biotransformation among the groups of PAP, RAP-RPA and RAP-RST were significant. With the loading plot and independent-samples T test, seven relevant markers with the significant differences were found in the group of RAP-RPA, six relevant markers were obtained in the group of RAP-RST. The relative content of four markers in RAP-RPA was higher than that in RAP, and one marker in RAP-RST was higher than that in RAP. The relative contents of other markers were all lower than that in RAP. These markers may be the effective substance for explaining the different effects of Radix Aconiti Preparata before and after combination with Radix Paeoniae Alba and Radix Stephaniae Tetrandrae.

ABSTRACT

OBJECTIVE: To explore the mechanism of compatibility of Radix Aconiti Lateralis Praeparate and Zingiber offiicinale Bosc. by determining the contents of four aconitine alkaloids (hyacinthin, benzoylaconine, benzoylhypaconine and benzoylmesaconine) in decoction made from Radix Aconiti Lateralis Praeparate alone and the co-decoctions made from the two medicinal herbs by HPLC. METHODS: The separation was performed on a Kromasil C18 column (4.6 mm × 250 mm, 5 μm) with a gradient elution of acetonitrile (A) and 0.2% aqueous acetic acid at pH of 6.20 adjusted with triethylamine (B) (0-15 min: 25%-31% A; 15-36 min: 31%-35% A; 36-40 min; 35%-25% A). The column temperature was set at 30°C. The flow rate was 1.0 mL · min-1 and the injection volume was 20 μL. The detection wavelength was set at 240 nm. RESULTS: The four components showed baseline separation. The linearity of this method was good (r≤0.999), and the average recoveries were 96.9%-101.9% with RSD<3%. Compared with the decoction made from Radix Aconiti Lateralis Praeparate alone, the contents of hyacinthin and benzoylmesaconine in the co-decoctions made from Radix Aconiti Lateralis Praeparate and Zingiber offiicinale Bosc. were increased, the contents of benzoylaconine and benzoylhypaconine were decreased, but the sum of the four aconitine alkaloids was increased. CONCLUSION: The compatibility of Radix Aconiti Lateralis Praeparate and Zingiber offiicinale Bosc. can increase the release of aconitine alkaloids. The feasibility of the compatibility of Radix Aconiti Lateralis Praeparate and Zingiber offiicinale Bosc. is elaborated from chemical point of view.

ABSTRACT

Objective To investigate the content change of aconitine in Radix Aconiti Kusnezoffii in different boiling time. Methods The chromatographic procedure was carried out with methanol-water-chloroform-diethylamide (70∶30∶3∶0.2) as mobile phase at a flow rate of 1.0 mL/min, the column temperature at 30 ℃, and UV detector at 240 nm. Results There was a good linear relationship of aconitine within 0.08-0.8 μg, r=0.999 8 (n=6), and the average recovery rate was 97.16%. The content of aconitine increased gradually in 0-2 h, then gradually decreased. And the reduction was evident particularly during 2-4 hours. Conclusion HPLC method is rapid, simple, accurate, reproducible and reliable for determining aconitine. The content of aconitine can be decreased effectively by boiling 4 hours.

Objective Infrared spectroscopy was used to study on both raw material and different degree water processing varieties extract of Radix Aconiti kusnezoffii,to observe the changes of main toxic components in processing,and thus to improve the quality of Radix Aconiti kusnezoffii processed products.Methods Fu Liye transform infrared spectroscopy was adopted to study the infrared spectrum characteristic of raw materials and different degree water processing varieties of.Radix Aconiti kusnezoffii.Results ①Aconitine and hypaconitine infrared spectrum showed that 1717 cm-1,1727 cm-1 and 1711 cm-1 is the C=O stretching vibration peak.It is a diester alkaloids characteristic peaks; ② although absorption peak of all vesicular samples had a certain change,it still existed diester alkaloids absorption peak,indicating the incomplete hydrolysis; ③ boiled aconite processing methods demonstrated diester alkaloids absorption peak shift in the water sample.Conclusion Diester alkaloids in Radix Aconiti kusnezoffii shows a positive relation with its time soaked in water.

ABSTRACT

Objective To explore effects of hcmoperfusion on toxic ingredients in plasma of rabbiis with a-cute intoxication of Radix Aconiti Kusmezoffii Monkshood.Method Sixteen male Japanese Giant Ear Rabbits were randomly divided into acute poisoning(AP)group and acute poisoning + hemoperfusion(AH)group(8 an-hnals in each group).Acute poisoning models were established in rabbits of both groups with intragastric adminis-tration of Radix Aconiti Kusmezoffii Monkshood liquid in dose of 1 mL/kg in order to produce arrhythm which oc-curred within ode hour after intragastric administration was regarded as the criteria of successful animal model.and then hemoperfusion with active carbon was performed for 2 hours in AH group.The pathological chanses of brain,myocardium and hepatic tissues were observed.The plasma concentrations of toxicants including mesaconitine,a-conitine and hypaconitine were measured by using HPLC-MS at 1 h,2 h,3 h,and 6 h after poisoning.Student's T test was used to identify the significance.Results The brain.myocardium and hepatic tissues of the rabbits in AP group showed hyperemia and edema which were attenuated after hemoperfusion.The plasma concentrations of mesaconitine,aconitine and hypaconitine revealed no significant differences between AP group and AH group with-in one hour after poisoning(P>0.05),while at 2 h and 3 h after poisoning,the plasma concentrations of mesaconitine were(2.11±1.08)ng/mL,(2.02±1.46)ng/mL,respectively,aconitine(39.70±9.31)ng/mL,(19.71±16.06)ng/mL,respectively,and hypaconitine(1.70±0.71)ng/mL,(2.12±1.33)ng/mL,respec-fively in AH group,and they were significantly lower than those in AP group(P<0.05).Conclusions The the plasma concentrations of mesaconitine,aconitine and hypaconitine were lower and the histopathological changes were attenuated after hemoperfusion.Hemoperfusion is a good intervention for acute intoxication of Radix Aconiti Kusmezoffii Monkshood.

Chemical compositions were determined by qualitative reactions, the results showed that Radix Aconiti carmichaelii lateralis in Sa Pa containing total alkaloids, diesteralkaloid, lipid, carotenoid, sterol, cumarine, organic acids, free sugars and amino acids. Alkaloid in Radix Aconiti carmichaelii lateralis in Sa Pa was maximum absorbed at wavelength of 231nm. The content of total alkaloid didn’t fluctuate much: from 1.15% (in July) to 1.21% (in August, October), similar to Radix Aconiti carmichaelii lateralis in China. The content of diesteralkaloid of Radix Aconiti carmichaelii lateralis in Sa Pa fluctuated much: from 0.21% (in July) to 0.34% (in October), higher than Radix Aconiti carmichaelii lateralis in China. It is necessary to identify gathering time with high productivity, to establish standards for Radix Aconiti carmichaelii lateralis, processed Radix Aconiti carmichaelii lateralis and to define the content of total alkaloid, diesteralkaloid

ABSTRACT

Study on processing methods of Radix Aconiti carmichaelii lateralis collected at Sa Pa from August 2003 to September 2003. Results: processing method of Radix Aconiti carmichaelii lateralis included these steps: Radix Aconiti were soaked in the MgCl2 and NaCl solutions, boiled, sliced, and dried. Studied samples all contained alcaloid, alcaloid of processed Radix Aconiti had maximum absorbing of ultraviolet spectrum in wavelength of 231nm and 274nm. This analysis could be used for quantitative study. The contents of total alcaloids of processed Radix Aconiti carmichaelii lateralis and diester alcaloids were 0.14-0.17% and 0.017-0.025%, respectively. The product increased amplitude of frog heart beat clearly at concentrations of 20 and 40g/l, but didn’t cause arrhythmia

ABSTRACT

Objective:To optimize the extraction procedure of total alkaloid in Radix Aconiti.Methods:The orthogonal design(L9(34) ) was carried out for the optimum extraction conditions guided by yield of the extract and the content of aconitine,hypaconitine and mesaconitine determined with RP-HPLC method,the mobile phase was MeOH-H2O2-CHCl3-DEA(70∶30∶ 2∶0.1).Results:The best extraction process was as follows:moisten with ammonia test solution,then leach for 24 hours with 15 times of ether.Conclusion:The optimum extraction methods are rational.

ABSTRACT

Aim To develop an HPLC method for the determination of Aconitum alkaloids extracted from Radix aconiti preparata in rats. Methods Waters 2690@996 PAD system was used. The analytical column was a Halsil 100 C18 column (250 mm×4.6 mm ID, 5 μm). The mobile phase was water, methanol and diethyl amine at the ratio of 75∶ 25∶ 0.1. The flow rate was 0.9 mL·min -1. The wavelength of the detector was 240 nm. Results The linear ranges of aconitine in the heart, spleen, lung and kidney were 0.4-100 μg·mL -1, the correlation coefficients were 0.997 2, 0.998 6, 0.999 3 and 0.999 4, respectively. The linear range of aconitine in liver was 2-200 μg·mL -1 and the correlation coefficient was 0.999 0. The linear ranges of hypaconitine in heart, liver, spleen, lung, kidney, brain and spinal cord were 5-100 μg·mL -1, the correlation coefficients were 0.999 4, 0.999 7, 0.999 8, 0.998 4, 0.999 8, 0.999 8 and 0.999 7, respectively. Detection limits (S/N=3) of aconitine and hypaconitine were 0.4 μg·mL -1. The recoveries of aconitine and hypaconitine ranged from 88.7% to 102.2% and 86.5% to 101.3%, respectively, and the RSD of precision of aconitine and hypaconitine was 10%. Conclusion It appears to be an accurate and effective method that can offer reference basis for in toxication of Radix aconiti preparata clinically.
