ABSTRACT

Photoperiod plays an important role in transformation from vegetative growth to reproductive growth in plants. CONSTANS (CO), as a unique gene in the photoperiod pathway, responds to changes of day length to initiate flowering in the plant. In this study, the expression level of FaCONSTANS (FaCO) gene under long-day, short-day, continuous light and continuous darkness conditions was analyzed by real-time quantitative PCR. We constructed the over-expression vector p1300-FaCO and infected into Arabidopsis thaliana by Agrobacterium-mediated method. We constructed the silencing vector p1300-FaCO-RNAi and infected into Festuca arundinacea by Agrobacterium-mediated method. The expression of FaCO gene was regulated by photoperiod. The over-expression of FaCO promoted flowering in wild type of Arabidopsis thaliana under long day condition and rescued the late flowering phenotype in co-2 mutant of Arabidopsis thaliana. Silencing FaCO gene in Festuca arundinacea by RNAi showed late-flowering phenotype or always kept in the vegetative growth stage. Our understanding the function of FaCO in flowering regulation will help further understand biological function of this gene in Festuca arundinacea.

ABSTRACT

MicroRNAs(miRNAs,miR) is a class of 21~22 nt small non-protein-coding single strand RNA molecules. In the liver, the phenotypic alternation or expressed abnormality of many miRNAs can influence the hepatitis virus replication at the gene level, which would provocate the development from hepatitis to hepatic fibrosis, cirrhosis, and even primary hepatic carcinoma on target. And so,several miRNAs are confirmed to be a new class of related gene to carcinoma.Furthermore, the dysfunction or abnormal quantity of miRNAs will promote the formation of fatty liver disease by interfering with signal transduction of insulin, inducing insulin resistance and then influencing normal metabolism of endosomatic lipid,Therefore, the specific medicine or methods, which might retrieve the expression or function of miRNAs in the hepatocytes, will provide a new valuable method to manage human liver disease.

ABSTRACT

Aquaporins (AQP), abundant in diversity, are the member of the major intrinsic proteins (MIPs) family with extraordinary ability to transport water, and thus, play important roles in modulation of water relations in plants. The recent advances in classification, structural characteristics, and physiological functions during plant growth and development as well as the various regulation modes of the activity and the effects of several abiotic stresses (water stress and salt stress) and phytohormones (ABA, GA and ethylene) on the gene expression of AQP were reviewed.

ABSTRACT

Objective:To screen the high performance, specific and nontoxic anti HBV antisense oligonucleotide fragment. Methods:Taking 2.2.15 cells as cell model, the antisense oligodeoxynucleotides(ASON)toward the HBV regulation genesenhancerⅡ(HBV EFⅡ)were designed and synthesized. The role played by ASON in inhibiting the secretion and expession of HBsAg and HBeAg by the host cells was detected by ELISA method. The effect of ASON on prolifezation and metabolism of the cells was detected by MTT method. Results:The inhibitory rate of HBsAg by ASON was 92% and 75%,respectively,indicating that itsdifference from that of noncomplementary sequence control group(inhibitory rate was 11%)had considerable statistical significance( P 0.05).Conclusion:ENⅡ is one of the most important target oriented sequencial selective regions of research on anti HBV nature of antisense oligonucleotide.
