ABSTRACT

Objective:To investigate the effective substance of the choleretic effect of Gardeniae Fructus, and analyze the relationships between its choleretic effect and the HPLC fingerprint chromatogram. Method:HPLC method was applied to establish the fingerprint chromatography of 8 batches of Gardeniae Fructus at different harvest periods. The flow, the content of bile acid, bilirubin and cholesterol in bile were tested, and then the principal component analysis was used to comprehensively evaluate the total choleretic effects of Gardeniae Fructus. After the relationships between the relative peak area of the common peaks and the choleretic effects were explored using grey relationship analysis method, the spectrum-effect relationship of Gardeniae Fructus was established. Result:The order of the contribution of the chemical components to the choleretic effect at the common peaks was as follow(r>0.8):P9>P14>P26>P4>P30>P6>P1>P10>P5>P24. Among all peaks, the full wavelength scanning results implied that the peaks 9,14 and 4 might be iridoids, and the peaks 26,30 and 24 might be crocins. By comparing with the standard substances, the peak 9 was finally identified as geniposide. Conclusion:The choleretic effect of Gardeniae Fructus may be the results of multiple components and pathways, and the main components in Gardeniae Fructus with the choleretic effect was from geniposide. In conclusion, these results provide a reference for investigating the material basis of choleretic effect of Gardeniae Fructus.

ABSTRACT

Objective To investigate the expression of programmed death receptor-1 (PD-1) in CD8+ T cells and FoxP3+CD4+ cells in patients with primary biliary cholangitis (PBC).Methods The peripheral blood and clinical data of 69 PBC patients in Peking Union Medical College Hospital and 58 health controls (HC) were collected. They were divided into initial treatment group and follow-up group according to whether they were treated or not.Patients in the treatment group were further divided into the refractory group and stable group according to treatment response.Flow cytometry was used to detect the expression of PD-1 in CDS+T cells and FoxP3+CD4+ cells.T-test and Person correlation analysis were used for data analysis. Results The PD-1 expression in peripheral blood mononuclear cells (PBMCs) of 69 PBC patients (12±9)% was lower than that of HC (20±12)% (t=-3.687,P<0.01).The percentage of PD-1+ in FoxP3+ CD4+T cells was significantly increased in PBC (5.6±3.7)% than HC (7.4±2.4)% (t=2.048,P<0.01).The proportion of CD8+T cells,PD-1 expression in CD8+T cells and the proportion of FoxP3+CD4+ cells weren't correlated with clinical parameters (P>0.05).There was a negative correlation between the expression of PD-1 cells in FoxP3+CD4+ cells and GLOBE score (r=-0.307,P<0.05).Conclusion The expression of PD-1 in peripheral CD8+T lymphocytes of PBC patients is lower than that of HC,and decreases more significantly in the refractory group. The expression of PD-1 on FoxP3+CD4+T cells is higher than that in HC,and is negatively correlated with the prognostic GLOBE score. It suggests that PD-1/PD-L1 pathway participates in the immune mechanism of PBC.

ABSTRACT

Biofilm is a bacterial lifestyle ubiquitously in natural environments. Bacterial biofilm leads to drug resistance, a main reason why many infectious diseases are difficult to control. Due to the prominent points of biofilms implicated in infectious disease and the spread of multi-drug resistance, it is urgent to discover new antibacterial agents that can regulate biofilm formation and development. This review introduces chemical agents that could modulate bacterial biofilm formation and development.

ABSTRACT

A comprehensive review was conducted to evaluate characteristics of the innovative research fund in a military hospital during past five years. We created a special index system according to the features of this innovative research fund:short supporting period, less amount of funding, and strict limit to the pre-study with the combination of common Research Performance Indicators. We use this index system to evaluate all finished projects and scored their performance.Other factors, such as research area, age of researchers, and job title, were also combined to analyze the relationship between funded research performance and fund characteristics. We found that all projects with higher performance score in past 5 years were conducted by young researchers, while this meets our original intention that using this fund to support junior scientists. In addition,we also found that the process of project assessment was fire and scientific, and funding for key research disciplines provided high level research outcomes.
