ABSTRACT

AIM To establish a GC-MS method for the simultaneous content determination of sixteen pesticide residues in Lycii Fructus and perform safety assessment.METHODS The analysis was performed on DB-5MS chromatographic column(30 m×0.25 mm,0.25 μm)subjected to the programmed heating,with splitless injection of 1.0 μL dissolved sample at a flowing rate of 1.0 mL/min.Other parameters were as follows:injection port temperature of 250℃,electron impact ionization(EI),electron energy of 70 eV;ion source temperature of 230℃,multi-reaction monitoring mode,and collision gas.of high-purity N2.Pesticide residues with relatively high dietary risk were analyzed and discussed with regard to residue levels,dietary intake risk,risk ranking and cumulative exposure assessment.RESULTS Sixteen pesticides showed good linear relationships within their own ranges(r≥0.994 4),whose average recoveries were 70%-114%,with the RSDs of less than 2%.The highest average cyfluthrin residue of 0.999 2 mg/kg in Lycii Fructus of production regions and the highest average cypermethrin residue of 0.088 4 mg/kg in Lycii Fructus commodities were both detected.In Lycii Fructus of production regions with chronic hazard index(HI)value of 0.012 9 and acute HI value of 0.065 5 and their commodities with chronic HI of 0.001 2 and acute HI of 0.005 4,the pesticide residue of cypermethrin was the leading cause of chronic and acute dietary risk,and additionally,pyridaben within maximum residue limit(MRL)was the only detectectable highly toxic pesticide among the other most concerning pestcides of deltamethrin,pyridaben,chlorpyrifos,dichlorvos and methidathion.CONCLUSION There exist pesticide residues within MRL values in some samples of Lycii Fructus and the use of cypermethrin should be well-controlled.

ABSTRACT

Objective To establish the HPLC fingerprint and pesticide residue detection methods for different kinds of rhubarb, and evaluate the quality of rhubarb comprehensively. Methods 20 batches of three types of rhubarb were collected and analyzed by high-performance liquid chromatography. The mobile phase was methanol-0.1 % phosphoric acid solution; gradient elution; column temperature of 35 ℃; detection wavelength of 254 nm; flow rate 1.0 ml/min. And cluster analysis was performed on the results. Direct extraction method was used and high-performance liquid chromatography-tandem mass spectrometry and gas chromatography-tandem mass spectrometry were established, 33 prohibited pesticides from different sources and origins of rhubarb were detected. Results The similarity among the fingerprint spectra of three sources of rhubarb, namely Rheum palmatum L., Rheum tanguticum Maxim.ex Balf., and Rheum officinale Bail1., and their control fingerprint spectra was>0.95. 20 batches of rhubarb samples were divided into 3 categories by cluster analysis. 33 prohibited pesticides were detected in rhubarb samples from different regions. Conclusion The quality of three kinds of rhubarb was significantly different. The established HPLC fingerprint and the method of banning agricultural residues were stable, reliable, simple and accurate, which could provide a basis for quality control evaluation of rhubarb.

ABSTRACT

Biofuel production from agricultural residues presents an innovative solution to the global energy challenge. This study delves into the potential of using such residues as a renewable feedstock, addressing the pressing need to transition from conventional fossil fuels. By evaluating various agricultural residues' types and characteristics, a comprehensive assessment of their worldwide availability and potential yield was undertaken. Emphasizing sustainable and eco-friendly approaches, the research underscores closed-loop systems, efficient utilization of co-products, and the imperative of a holistic life cycle assessment (LCA) for biofuel production. The LCA revealed a significant reduction in greenhouse gas emissions, emphasizing water conservation and waste reduction during the process. Despite the evident potential, there are identifiable challenges, primarily technological research gaps, economic constraints, infrastructural limitations, and regulatory hurdles. Yet, the undeniable benefits include a notable reduction in carbon footprint, effective resource management, and a bolstered economy, especially for agrarian communities. Policies promoting sustainable farming practices, incentivizing research and development, and fostering collaborations are recommended. Such a framework can enhance biofuel infrastructure, necessitate regular monitoring, and optimize the biofuel production process. Conclusively, while challenges persist, with cohesive policy recommendations and technological innovations, agricultural residues can pivot as the linchpin in a sustainable energy future.

ABSTRACT

Pests and pathogens significantly reduce crop yields and cost the global economy USD 540 billion annually. The phytomicrobiome is becoming a cornerstone of a comprehensive rethink of agricultural management practices, with microbiome-assisted plant breeding focusing on three principles: minimal soil disturbance, continuous soil cover with crops, cover crops, or a mulch of crop residues, and crop rotation. Plant diseases, phytomicrobiomes, and agricultural practices can all affect plant health. Conservational agricultural practices like minimum tillage and no-tillage have been implemented to reduce anthropogenic activity and preserve microbial diversity. Mulching is a common practice in agriculture to stop moisture loss, maintain soil temperature, control weed growth, and stop soil erosion. Monoculture farming is the practice of cultivating a single crop continuously over several growing seasons on the same field. Intercropping systems encourage the growth of beneficial fungi, such as mycorrhiza, endophytes, saprophytes, decomposers, and bioprotective fungi, and can benefit forest ecosystems by creating disease-suppressive soils. By combining conservation tillage and crop rotation, farmers can reduce disease pressure by disrupting the life cycles of soil-borne pathogens linked to particular crops or genotypes. Composed manure and plant residues can control plant pathogens like Pythium, but the effect is thought to be due to microbial competition or plant host resistance. Green manure can control plant diseases brought on by pathogens in Rhizoctonia, Verticillium, Sclerotinia, Phythophthora, Pythium, Aphanomyces, and Macrophomina, but manure-derived fertilizers may contain antibiotic resistance genes and mobilomes, which could pose risks to both human and animal health. Climate change impacts crop yields by reducing crop physiology and productivity, increasing pathogen diversity, and affecting pathogen genetic traits and speciation. The state of the environment and the availability of suitable hosts significantly impact the ability of these pathogens to survive and spread. However, PGPRs are unpredictable and soil warming can interfere with their effectiveness. Interdisciplinary collaborations between plant biologists, microbiologists, climatologists, and agronomists are required to create effective strategies to reduce the effects of climate change on plant health, crop production, and ecosystem stability.

La proteína proteasa 3CLpro del SARS-CoV-2 es una enzima crucial para la replicación viral, razón por la cual se convierte en un blanco terapéutico de gran importancia. El timol (2-isopropil-5-me-tilfenol), un compuesto natural que se encuentra en el tomillo (Thymus vulgaris), exhibe potencial actividad antiviral contra la proteasa 3CLpro. En este estudio, usando acoplamiento molecular con AutoDockTools-1.5.6, se evaluaron las energías de interacción molecular entre el timol y los residuos de aminoácidos en el sitio activo de la proteína proteasa 3CLpro. Luego, con la teoría cuántica de Átomos en Moléculas (QTAIM) y la de Interacciones no covalentes (NCI) se analizaron los tipos de interacciones moleculares entre los residuos de aminoácidos identificados y el timol. Los cálculos cuánticos se llevaron con el software Orca-5.0.3, utilizando el método DFT con el funcional M06-2X y el conjunto base aug-cc-pVDZ en fase gaseosa. Los resultados de acoplamiento molecular indican que el timol se une a la proteína 3CL con una energía de interacción igual a -3,784 kcal/mol. El análisis QTAIM indica la presencia de puntos críticos de enlace entre el timol y los residuos HIS41 y CYS145. Además, se observa la formación de un enlace de hidrógeno entre el grupo OH del timol y el residuo CYS145, lo cual es corroborado por los análisis ELF (Electron Localization Function) y NCI (Non Covalent Interactions). Finalmente, el método NCI confirma la presencia de interacciones de Van der Waals con el residuo HIS41. Los resultados sugieren que el mecanismo de inhibición de la actividad de la proteína 3CLpro es controlado por interacciones moleculares tipo puente de hidrógeno e interacciones débiles.


The protease 3CLpro of the SARS-CoV-2 is a crucial enzyme for viral replication, becoming a highly important therapeutic target. Thymol (2-isopropyl-5-methyl-phenol), a naturally occurring compound found in thyme, exhibits potential antiviral activity against the 3CLpro protease. In this study, using molecular docking with AutoDockTools-1.5.6, the molecular interaction energies between thymol and amino acid residues in the active site of the protein protease 3CLpro were evaluated. Then, with the Atoms in Molecules (QTAIM) and Non-covalent Interactions (NCI) theories, the types of molecular interactions between identified amino acid residues and thymol were analyzed. Quantum calculations were carried out with the Orca-5.0.3 software using the DFT method with the M06-2X functional and the aug-cc-pVDZ basis set in the gas phase. The molecular docking results indicate that thymol is linked to the 3CL protein with an interaction energy equal to -3.784 kcal/mol. QTAIM analysis indicates the presence of critical binding sites between thymol and residues HIS41 and CYS145. In addition, the formation of a hydrogen bond between the OH group of thymol and the CYS145 residue is observed, which is corroborated by the ELF and NCI analyses. Finally, the NCI method confirms the presence of Van der Waals interactions with the HIS41 residue. The results suggest that the mechanism of inhibition of the activity of the 3CLpro protein is controlled by molecular interactions such as hydrogen bonding and weak interactions.

Em seguida, com a teoria quantica de atomos em moleculas (QTAIM) e da interacões no-covalentes (NCI), foram analisados os tipos de interações moleculares entre os resíduos de aminoácidos identificados e o timol. ABSTRACT

Chicken meat is highly preferred protein food worldwide. To meet the demand, huge poultry farms are established and using antibiotics as prophylaxis and treatment against the bacterial diseases. Uncontrolled usage of antibiotics has led to development of antibiotic resistance in poultry and antibiotic residues in poultry chicken. Fifty one chicken meat samples were collected from various retail outlets. Antibiotic residues were quantified by HPLC, total microbial load was measured by growth of bacteria on growth medium and antibiotic resistant profile of Escherichia coli, Salmonella spp, Staphylococcus aureus and Campylobacter spp was determined by well diffusion method. Except neomycin, all tested antibiotics were present in the range of 10-978 ppm, the average microbial load was in the range log 10 of 7.32 per gram of chicken sample, E. coli, Salmonella spp, Staphylococcus aureus and Campylobacter spp were resistant to several antibiotics studied. Hence there is a need of appropriate usage of antibiotics in poultry and proper handling of chicken during farming and slaughtering.

ABSTRACT

A hyperuricemic rat model induced by adenine and ethambutol was established to investigate the anti-hyperuricemia activity and its mechanism of the flavonoid extract from saffron floral bio-residues. Sixty-seven SD rats were randomly divided into control group, model group, positive control group, and flavonoid extract groups(with 3 doses), respectively, and each group contained 11 or 12 rats. The hyperuricemic model was established by continuous oral administration of adenine(100 mg·kg~(-1)) and ethambutol(250 mg·kg~(-1)) for 7 days. At the same time, the positive control group was given allopurinol(20 mg·kg~(-1) per day) and the flavonoid extract groups were given the flavonoid extract at doses of 340, 170 and 85 mg·kg~(-1) per day, respectively. On day 8, rat serum, liver, kidney, and intestinal tissues were collected, and the levels of uric acid in serum and tissue, the xanthine oxidase activities and antioxi-dant activities in serum and liver were evaluated, and the kidney histopathology was explored. In addition, an untargeted serum metabolomics study was performed. According to the results, the flavonoid extract effectively reduced the uric acid levels in serum, kidney and ileum and inhibited the xanthine oxidase activities and elevated the antioxidant activities of serum and liver in hyperuricemic rat. At the same time, it reduced the levels of inflammation factors in kidney and protected renal function. Moreover, 68 differential metabolites of hyperuricemic rats were screened and most of which were lipids and amino acids. The flavonoid extract significantly retrieved the levels of differential metabolites in hyperuricemic rats, such as SM(d18:1/20:0), PC[18:0/18:2(92,12Z)], palmitic acid and citrulline, possibly through the following three pathways, i.e., arginine biosynthesis, glycine, serine and threonine metabolism, and histidine metabolism. To sum up, the flavonoid extract of saffron floral bio-residues lowered the uric acid level, increased the antioxidant activity, and alleviated inflammatory symptoms of hyperuricemic rats, which may be related to its inhibition of xanthine oxidase activity and regulation of serum lipids and amino acids metabolism.

ABSTRACT

To study the residue and dietary risk of propiconazole in Panax notoginseng and the effects on physiological and bioche-mical properties of P. notoginseng, we conducted foliar spraying of propiconazole on P. notoginseng in pot experiments. The physiolo-gical and biochemical properties studied included leaf damage, osmoregulatory substance content, antioxidant enzyme system, non-enzymatic system, and saponin content in the main root. The results showed that at the same application concentration, the residual amount of propiconazole in each part of P. notoginseng increased with the increase in the times of application and decreased with the extension of harvest interval. After one-time application of propiconazole according to the recommended dose(132 g·hm~(-2)) for P. ginseng, the half-life was 11.37-13.67 days. After 1-2 times of application in P. notoginseng, propiconazole had a low risk of dietary intake and safety threat to the population. The propiconazole treatment at the recommended concentration and above significantly increased the malondialdehyde(MDA) content, relative conductivity, and osmoregulatory substances and caused the accumulation of reactive oxygen species in P. notoginseng leaves. The propiconazole treatment at half(66 g·hm~(-2)) of the recommended dose for P. ginseng significantly increased the activities of superoxide dismutase(SOD), peroxidase(POD), and catalase(CAT) in P. notoginseng leaves. The propiconazole treatment at 132 g·hm~(-2) above inhibited the activities of glutathione reductase(GR) and glutathione S-transferase(GST), thereby reducing glutathione(GSH) content. Proconazole treatment changed the proportion of 5 main saponins in the main root of P. notoginseng. The treatment with 66 g·hm~(-2) propiconazole promoted the accumulation of saponins, while that with 132 g·hm~(-2) and above propiconazole significantly inhibited the accumulation of saponins. In summary, using propiconazole at 132 g·hm~(-2) to prevent and treat P. notoginseng diseases will cause stress on P. notoginseng, while propiconazole treatment at 66 g·hm~(-2) will not cause stress on P. notoginseng but promote the accumulation of saponins. The effect of propiconazole on P. notoginseng diseases remains to be studied.

ABSTRACT

Based on the data of 56 kinds of diseases and drug use in 100 kinds of cultivated Chinese herbal medicines, this paper used frequency analysis method to count the types of diseases and their drug use characteristics, and systematically analyzed the status of drug registration and monitoring standards for disease prevention and control of Chinese herbal medicines. The results showed that 14 diseases such as root rot, powdery mildew, and drooping disease were common in the production of Chinese herbal medicines. Among the 99 pesticides reported, 67.68% were chemically synthesized, 23.23% were biological pesticides, and 9.09% were mineral pesticides. Among the reported pesticides, 92.93% of them were low toxic, with relative safety. However, 70% of the production drugs were not registered in Chinese herbal medicines, and the phenomenon of overdose was serious. The current pesticide residue monitoring standards does not match well with production drugs in China. Although the matching degree between Maximum Residue Limit of Pesticide in Food Safety National Standard(GB 2763-2021) and production drugs is more than 50%, there are few varieties of Chinese herbal medicines covered. The matching degree between Chinese Pharmacopoeia(2020 edition), Green Industry Standard of Medicinal Plants and Preparations(WM/T2-2004), and production drugs is only 1.28%. It is suggested to speed up the research and registration of Chinese herbal medicine production and further improve the pesticide residue limit standard combined with the actual production, so as to promote the high-quality development of Chinese herbal medicine industry.

ABSTRACT

Objective To explore the residual level of FPMs in indoor dust samples in Shenzhen from 2020 and 2021, and to analyze its temporal distribution characteristics. Methods In the present study, indoor dust samples (n=193) from residential buildings in Shenzhen. were collected to analyze the temporal variation characteristics of FPMs. Ultra-performance liquid chromatography-tandem mass spectrometry (UPLC-MS/MS) was applied to determine the concentrations of FP and its four major metabolites (fipronil-sulfone, fipronil-sulfide, fipronil-desulfinyl, and fipronil-amide; abbreviated as FP-SFO, FP-SFI, FP-DES, and FP-AM) in the samples. The sum of the concentrations of FP and its four metabolites was represented as additive mass concentration (ΣFPMs). Additionaly, Wilcoxon test was performed to determine the temporal distribution differences of FPMs’ concentrations. Results From 2020 to 2021, the concentration of ΣFPMs for the in door dust samples in Shenzhen ranged from 0.51 to 4 415 ng/g (median: 18.8ng/g). FP, FP-SFO AND FP-SFI were the major target analytes in the sample with detection rates of 90.60%,86.20% and 75.40%, respectively. The detection rates of other metabolites were low (≤ 44.3%). Analysis of the temporal variation trend of FPMs’ concentrations showed that there was no significant difference in the levels of ΣFPMs between warm season(spring and summer)and cold season(autumn and winter)in the indoor dust samples from 2020 to 2021(2.38 vs 2.84ng/g , P > 0.05). However , the concentrations of FP-SFI and ΣFPMs in the indoor dust samples collected from 2021 showed an significantly increasing trend compared with 2020(1.02 vs 1.89 , 17.80vs. 20.10 ng/g , P < 0.05). Conclusion From 2020 to 2021 , the detection level of FPMs in indoor dust in Shenzhen is relatively high and shows an upward trend , with no obvious seasonal difference. However, whether the residual level of FPMs in indoor dust poses a risk to human health needs further study.

@#Objective To develop and verify a quantitative real-time PCR method for determination of the content of host cell DNA residues in severe acute respiratory symptom coronavirus 2(SARS-CoV-2) inactivated vaccine(Vero cells),in order to better control the safety of products.Methods DNA was extracted from inactivated SARS-CoV-2 vaccine(Vero cells) bulk by magnetic bead separation method,and the DNA residues of host cells were quantitatively analyzed by probetype PCR.The linear range,repeatability,intermediate precision,quantitative limit,specificity,durability and accuracy of the developed method were verified,and the host cell DNA re sidues of 5 batches of inactivated SARS-CoV-2 vaccine(Vero cells)were determined by this method.Results DNA standard curve showed good linearity in the range of 300~0.003 pg/μL(each R~2> 0.99);Relative standard deviations(RSD) of repeatability and intermediate precision verification were less than 20%;The quantitative limit was 0.001 pg/μL;Sample dilution and purified liquid dilution had no interference to detection;The results of 60 min incubation at 53,55,57 ℃ and 56,60,64 min incubation at 55 ℃ showed no significant difference;The recoveries of accuracy verification were 79%~83%,RSD <5%.This method had good adaptability in detecting DNA residues in the bulk of inactivated SARS-CoV-2 vaccine(Vero cells).Conclusion The quantitative realtime PCR method for determination of host cell DNA residues in inactivated SARS-CoV-2 vaccine(Vero cells) has been successfully developed,of which the linearity and range,repeatability,intermediate precision,quantitative limit,specificity,durability and accuracy meet the acceptable standards,and are suitable for the detection and quality control of host cell DNA residues in inactivated SARS-CoV-2 vaccine(Vero cells).

ABSTRACT

In recent years, the traditional Chinese medicine(TCM)industry has experienced rapid development, resulting in a significant amount of Chinese medicinal residues generated during the industrial manufacturing process. Currently, the main methods of handling Chinese medicinal residues include stacking, landfilling, and incineration, which lead to substantial resource waste and potential environmental pollution. With "carbon peak" and "carbon neutrality"( "Dual Carbon")becoming national strategic goals, the TCM industry is ushering in a new wave of "low-carbon" trends, and the high-value utilization of Chinese medicinal residues has become a breakthrough for implementing a low-carbon economy in the TCM sector. From the perspective of a low-carbon economy, this article reviewed literature in China and abroad to summarize the microbial transformation technology, enzymatic conversion technology, biomass pyrolysis, gasification, hydrothermal liquefaction, and other high-value utilization technologies for Chinese medicinal residues. It also overviewed the applications of Chinese medicinal residue in feed additives, organic fertilizers, edible mushroom cultivation substrates, preparation of activated carbon for wastewater treatment, and new energy batteries. Considering the current status of resource utilization of Chinese medicinal residues, feasible strategies and suggestions for resource development and utilization were proposed to improve the quality and efficiency of the Chinese medicinal resource industry chain and promote green development, thereby providing research ideas and theoretical basis for achieving carbon peak and carbon neutrality goals.

ABSTRACT

Moutan Cortex(MC) residues produced after the extraction of MC can be re-extracted for active components and used to produce organic fertilizer and animal feed. However, they are currently disposed as domestic waste, which pollutes the environment. This study analyzed the chemical composition of the medicinal material, residues, and residue compost of MC by UPLC-UV-Q-TOF-MS. Furthermore, the nutrient composition of MC residues and the residue compost was analyzed. The results showed that:(1)MC residues had lower content of chemicals than the medicinal material, and content of paeonol, gallic acid, and galloylglucose in MC residues were about 1/3 of that in the medicinal material. The content of chemicals were further reduced after residue composting, and the quantitative compounds were all below the limits of detection.(2)Compared with MC residues, the residue compost showed the total nitrogen, total phosphorus, total potassium, and organic matter content increasing by 122.67%, 31.32%, 120.39%, and 32.06%, respectively. Therefore, we concluded that the MC residues can be used to re-extract active compounds such as paeonol, gallic acid, and galloylglucose. ABSTRACT

Objetivo: objetivo identificar na literatura científica as boas práticas utilizadas em ambientes hospitalares sobre o processo de gerenciamento de resíduos em saúde. Método: trata-se de uma revisão integrativa, realizada nas bases de dados LILACS, MEDLINE/PubMed, Scopus e Web of Science, em abril de 2022. Utilizaram-se os descritores Waste Management, Medical Waste, Solid Waste Processing, Hospitals, combinados com o operador booleano AND. Foram encontrados 95 artigos e selecionados 12. Resultados: as boas práticas utilizadas pelos profissionais em ambientes hospitalares que auxiliam no processo de gerenciamento de resíduos em saúde foram: implementação de política de gestão sobre resíduos em saúde e presença de orçamento especial para a gestão de resíduos, estabelecimento de subcomitês de descarte em diferentes departamentos para monitorar o descarte de resíduos médicos, presença de comitê de gerenciamento de resíduos de saúde, comitê de proteção contra radiações e de comitê de prevenção e controle de infecção, disponibilização de equipamentos de proteção individual, fichas de informação de gerenciamento de resíduo químico perigoso hospitalar, diretrizes de prevenção e controle de infecção e de instrumentos educativos, além de treinamentos específicos sobre a temática e disponibilização de locais de armazenamento de resíduos separados de resíduos perigosos. Conclusão: a gestão segura dos RSS se mostra cada dia mais muito importante no que diz respeito ao meio ambiente e à saúde pública. Porém, entre os artigos citados percebe-se a implementação de boas práticas mas, de forma incipiente.


Objective: objective: to identify in the scientific literature the good practices used in hospital environments on the process of waste management in health. Method: This is an integrative review, carried out in the LILACS, MEDLINE/PubMed, Scopus and Web of Science databases in April 2022. The descriptors Waste Management, Medical Waste, Solid Waste Processing, Hospitals, combined with the Boolean operator AND were used. 95 articles were found and 12 were selected. Results: the good practices used by professionals in hospital environments that assist in the process of waste management in health were: implementation of management policy on waste in health and presence of special budget for waste management, establishment of disposal subcommittees in different departments to monitor the disposal of medical waste, presence of health waste management committee, radiation protection committee and infection prevention and control committee, provision of personal protection equipment, information sheets for management of hazardous chemical waste in hospital, guidelines for infection prevention and control and educational instruments, besides specific training on the subject and provision of separate waste storage sites for waste from hospital waste dangerous. Conclusion: the safe management of SSR is becoming increasingly important in terms of the environment and public health. However, the articles mentioned include the implementation of good practices, but in a nascent way.


Propósito: identificar en la literatura científica las mejores prácticas utilizadas en los ambientes hospitalarios en el proceso de manejo de residuos en salud. Método: Se trata de una revisión integrada realizada en las bases de datos LILACS, MEDLINE/PubMed, Scopus y Web of Science en abril de 2022. Se utilizaron los descriptores Gestión de Residuos, Residuos Médicos, Procesamiento de Residuos Sólidos, Hospitales, combinados con el operador Y booleano. Se encontraron 95 artículos y se seleccionaron 12. Resultados: las buenas prácticas utilizadas por los profesionales en los entornos hospitalarios que ayudan en el proceso de gestión de los desechos en la salud fueron: aplicación de la política de gestión de los desechos en la salud y presencia de un presupuesto especial para la gestión de los desechos, establecimiento de subcomités de eliminación en diferentes departamentos para supervisar la eliminación de los desechos médicos, presencia de comités de gestión de los desechos sanitarios, comité de protección radiológica y comité de prevención y control de las infecciones, suministro de equipo de protección personal, información sobre la gestión de los desechos químicos peligrosos hospitalarios herramientas educativas, así como capacitación específica sobre el tema y suministro de sitios de almacenamiento de desechos separados para desechos peligrosos. En conclusión, la gestión segura de las denuncias de infecciones de transmisión sexual es cada vez más importante en lo que respecta al medio ambiente y la salud pública. Sin embargo, entre los artículos citados figura la aplicación de buenas prácticas, pero de manera incipiente.

Abstract Fipronil is an insecticide and acaricide widely used in agriculture and domestic animals worldwide. Ecotoxicology studies have shown that, even at the low concentrations used on target species, fipronil and its degradation products have a significant impact on non-target species, either by direct toxicity or indirect effects affecting the food chain. The negative effects of fipronil on non-target species of terrestrial and aquatic invertebrates and indirect effects on food chains have led to its use being banned or severely restricted in numerous countries, including all of the European Union, China, and the United States. Some of the species highly susceptible to fipronil are of great economic and ecological importance, including crayfish, brown shrimp, and bees. In particular, the impact on decimating bee hives worldwide is an example of fipronil´s undesirable effects on agriculture. Other species affected by fipronil -for which there are few studies- are biological predators of the same pests controlled by fipronil. Considering all the impacts on non-target species, the commercialization and indiscriminate use of fipronil in agriculture seem irresponsible. In Colombia, as of September 2021 and pressed by local beekeepers, the Instituto Colombiano Agropecuario (ICA) prohibited its use on avocado, coffee, citrus, and passiflower plantations. However, as long as its use is not prohibited in all agricultural applications, farmers could divert its use and continue using it in any other type of plantation. This paper describes the impact of fipronil on some of the beneficial invertebrate species of outmost economic and ecological importance.


Resumen El fipronil es un insecticida y acaricida ampliamente utilizado en la agricultura y los animales domésticos en todo el mundo. Los estudios ecotoxicológicos han demostrado que, aún a las bajas concentraciones utilizadas en las especies destino, el fipronil y sus productos de degradación tienen un impacto significativo en las especies no-blanco, ya sea por efectos tóxicos directos o indirectos que afectan la cadena alimentaria. Los peligros para las especies no-blanco de invertebrados terrestres y acuáticos y los efectos indirectos sobre las cadenas alimentarias han llevado a prohibir o severamente restringir su uso en numerosos países, incluyendo toda la Unión Europea, China y los Estados Unidos. Algunas de las especies altamente susceptibles al fipronil son de gran importancia económica y ecológica, incluyendo cangrejos de río, camarones y abejas. En particular, el impacto en la diezma de las colmenas de abejas en todo el mundo es un ejemplo de los efectos indeseables que tiene el fipronil en la agricultura. Otras especies afectadas por el fipronil -para las que existen pocos estudios- son los depredadores biológicos de las mismas plagas controladas por el fipronil. Teniendo en cuenta su impacto sobre las especies no-blanco, la comercialización y el uso indiscriminado de fipronil en la agricultura parecen irresponsables. En Colombia, a partir de septiembre de 2021 y presionado por los apicultores locales, el Instituto Colombiano Agropecuario (ICA) prohibió su uso en plantaciones de aguacate, café, cítricos y pasifloras. Sin embargo, mientras no esté prohibido su uso en todas sus aplicaciones agrícolas, los agricultores pueden desviar su uso y seguir utilizándolo en cualquier tipo de plantación. Este artículo describe el impacto del fipronil en sólo unas pocas especies de invertebrados beneficiosos de gran importancia económica y ecológica.


Resumo Fipronil é um inseticida e acaricídio amplamente utilizado na agricultura e animais domésticos em todo o mundo. Estudos de ecotoxicologia têm demonstrado que em baixas concentrações utilizadas em espécies-alvo, o fipronil e seus produtos de degradação têm um impacto significativo em espécies não-alvo, seja por efeitos diretos de toxicidade ou efeitos indiretos que afetam a cadeia alimentar. Os perigos para espécies não-alvo de invertebrados (terrestres e aquáticos) e efeitos indiretos sobre as cadeias alimentares levaram ao seu uso ser banido ou severamente restrito em vários países, incluindo toda a União Europeia, China e Estados Unidos. Algumas das espécies altamente suscetíveis ao fipronil são de grande importância econômica e ecológica, incluindo lagostim, camarão marrom e abelhas. Em particular, o impacto na dizimação das colmeias de abelhas em todo o mundo é um exemplo dos efeitos indesejáveis que o fipronil tem sobre a agricultura. Outras espécies afetadas pelo fipronil, para as quais há poucos estudos, são predadores biológicos das mesmas pragas controladas pelo fipronil. Considerando todos os impactos sobre espécies não-alvo, a comercialização e o uso indiscriminado do fipronil na agricultura parecem irresponsáveis. Na Colômbia, a partir de setembro de 2021 e pressionada por apicultores, a ICA proibiu seu uso em plantações de abacate, café, cítricos e maracujá. No entanto, desde que seu uso não seja proibido em todas as suas aplicações agrícolas, os agricultores podem desviar seu uso e continuar a usá-lo em qualquer tipo de plantação. Este artigo descreve o impacto do fipronil em apenas algumas espécies com grande importância econômica e ecológica.

ABSTRACT

Aims: The objective of this study is to monitor the evolution of organochlorine residues levels in kola nuts collected from various actors in the eastern producing region of Côte d'Ivoire. Study Design: Kola nuts samples were collected from farmers, collectors and urban stores in the Eastern of Côte d’Ivoire. Place and Duration of Study: Health Department of Hydrology Health and Toxicology, Training and Research Unit of Pharmaceutical and Biological Sciences, Abidjan, Côte d’Ivoire, running 2016-2019. Methodology: A total of 225 samples were analyzed using Gas chromatography–tandem mass spectrometry (Agilent 7010B Triple Quadrupole GC/MS System). Results: Data showed that all 21 pesticides analyzed were detected in kola nuts samples. Statistical analysis indicated no significant difference in OCPs sub-group used by actors during the three-crop studied. The mean levels of HCHs, DDTs and cyclodienes were ranged from 5±1– 136.67±77.3 µg/kg FW, 5±1 – 116.67±63.2 µg/kg FW and 5±1 - 145±63.2 µg/kg FW, respectively. Otherwise, farmer’s levels of OCPs were lower than those detected in kola nuts from collectors and urban stores samples. Thus, compared to the MRL set by the World Health Organization/Food and Agricultural Organization, the farmer’s samples are lower than limits fixed, unlike the contents registered with collectors and urban stores. In the latter, nearly 80% to 100% of kola nuts collected are contaminated with hexachlorinated residues (HCHs) and heptachlor. Conclusion: There is the need to keep monitoring ecotoxicological chemical substances in kola nuts produced in Côte d’Ivoire and take steps that ensure health safety of end users. Care should be taken since residues could pose chronic health risk for adults and children.

Este estudio se realizó con el objetivo de desarrollar y validar un método para la determinación de 30 medicamentos veterinarios en muestras de trucha y langostino. El método utiliza extracción en fase sólida dispersiva (dSPE) con C18 y detección por cromatografía líquida acoplada a espectrometría de masas. Se determinó linealidad, veracidad (porcentaje de recuperación), repetitividad y reproducibilidad intralaboratorio (porcentaje de desviación estándar relativa (% RSD)), límites de detección (LoD), límites de cuantificación (LoQ), selectividad e incertidumbre. La recuperación varió de 70 a 120% y la repetibilidad y la reproducibilidad fueron menores de 20% de la desviación estándar relativa. La selectividad fue adecuada, sin picos interferentes. Las relaciones iónicas cumplieron con los criterios de confirmación. Los coeficientes de determinación (R2) fueron mayores de 0,99, con excepción de la sulfaquinoxalina en langostino (R2 = 0,97). Los LoD y los LoQ variaron entre 0,6 µg/kg y 12,8 µg /kg y los valores de incertidumbre entre 6 µg/kg y 49 µg/ kg. Se analizaron adicionalmente 6 muestras de diferentes mercados de Lima y se detectaron trazas de algunos medicamentos incluidos en el ensayo. El método es adecuado para el análisis de residuos de medicamentos veterinarios y se recomienda su aplicación en los programas nacionales de monitoreo de la inocuidad de truchas y langostinos provenientes de acuicultura.


The study was aimed at developing and validate an analysis method to determine residues of 30 veterinary drugs in rainbow trout and shrimp specimens. The method involves extraction in dispersive solid phase with C18 and the subsequent detection through liquid chromatography coupled to mass spectrometry. Validation was done through determination of linearity, trueness (% of recovery), repeatability and intralaboratory reproducibility, limits of detection (LoD), limits of quantification (LoQ) selectivity and uncertainty. Recovery ranged from 70 to 120% and repeatability and intralaboratory reproducibility were lower than 20%. Selectivity was adequate, without interference peaks. Likewise, the ionic relationships met the confirmation criteria. The linearity was adequate, with determination coefficients (R2) above 0.99, except for sulfaquinolaxin in shrimp specimens (R2 = 0,97). LoD and LoQ varied from 0,6 µg /kg to 12,8 µg / kg. Limits of uncertainty ranged from 6 µg /kg to 49 µg /kg. The method was used to analyze 6 samples from different markets in Lima (Peru), identifying traces of some drugs included in the study. Our results show that the method is adequate for the analysis of veterinary drug residues and allow us to recommend its application in national monitoring programs, to assess the safety of rainbow trout and shrimp specimens from aquaculture.


O estudo foi realizado com o objetivo de desenvolver e validar um método para a determinação de 30 medicamentos veterinários, em amostras de truta e camarão. O método utiliza extração dispersiva em fase sólida com C18 e detecção por cromatografia líquida acoplada à espectrometria de massas. Foram determinados a linearidade, a veracidade (recuperação percentual), a repetibilidade, a reprodutibilidade intra-laboratorial, os limites de detecção (LoD) e de quantificação (LoQ), a linearidade, a selectividade e a incerteza. A recuperação variou de 70 a 120%, a repetibilidade e reprodutibilidade estiveram abaixo do 20% do desvio padrão relativo. A selectividade fio adequada, sem picos de interferentes. As proporções de íons atenderam aos critérios de confirmação. Os coeficientes de determinação (R2) foram superiores a 0,99, com excepção da sulfanoxalina em camarão (R2 = 0,97). LoD e LoQ variavam entre 0,6 µg /kg e 12,8 µg /kg e valores de incerteza entre 6 µg /kg e 49 µg / kg. Seis amostras de mercados do Lima foram adicionalmente analisadas e foram detectados vestígios de alguns medicamentos incluídos no estudo. O método é adequado para o análise de resíduos de medicamentos veterinários e sua aplicação é recomendada em programas nacionais de controlo da segurança da truta e do camarão provenientes da aquicultura.

ABSTRACT

Chlorpyrifos (CPF) is a pesticide widely used in Colombia´s agriculture, including crops, farm animals and pets, despite it has been banned for use in the European Union and the United States. Studies demonstrate that even low blood levels of CPF -which do not inhibit blood acetylcholinesterase- can lead to child developmental and neurological disorders such as smaller head circumference and brain alterations, and psychomotor and cognitive deficits related to learning ability, attention and memory. In adults, CPF is an endocrine disruptor and breast carcinogen. High direct and indirect economic costs have been associated with CPF exposure. Not only farmers and their families -who have the highest exposures- but the general population consuming crops sprayed with CPF are also at risk. For these reasons CPF was recently banned by the European Union (2020) and the USA (2021). Pesticide regulation policies vary greatly depending on which and how scientific studies are used to assess health risks. Pesticide evaluations funded by the chemical industry should be rectified to avoid conflicts of interest. Furthermore, political alignment with the interests of the industry should not take precedence over independent scientific evidence. It is discouraging, to say the least, that until stricter health laws are passed in Colombia, CPFs and related pesticides will continue to be imported from those countries that have already banned them. Colombian scientists should raise their voice to challenge blind acceptance of profits over unintended consequences, and efforts to prevent pesticide´s abuse should be encouraged.


El clorpirifos (CPF) es un pesticida ampliamente utilizado en la agricultura de Colombia, incluidos cultivos, animales de granja y mascotas, a pesar de haber sido prohibido en la Unión Europea y Estados Unidos. Los estudios han demostrado que incluso niveles bajos de CPF en sangre -que no inhiben la acetilcolinesterasa sanguínea- pueden provocar trastornos neurológicos y del desarrollo infantil, como menor circunferencia de la cabeza y alteraciones cerebrales, y déficits psicomotores y cognitivos relacionados con la capacidad de aprendizaje, la atención y la memoria. En adultos, el CPF es un disruptor endocrino y causante de cáncer de mama. Altos costos económicos directos e indirectos se han asociado con la exposición al CPF. No solo los trabajadores agrícolas y sus familias, que están más expuestos, sino también la población en general que consume cultivos rociados con CPF también están en riesgo. Por estas razones el CPF fue prohibido recientemente por la Unión Europea (2020) y los EE. UU. (2021). Las políticas de regulación de plaguicidas varían mucho según los estudios científicos escogidos para evaluar los riesgos para la salud. Las evaluaciones de plaguicidas financiadas por la industria química deben rectificarse para evitar conflictos de interés. Además, ante la evidencia científica independiente no debería prevalecer la alineación política con los intereses de dicha industria. Es desalentador, por decir lo menos, que hasta que se aprueben leyes de salud más estrictas en Colombia se seguirán importando CPF y pesticidas relacionados desde aquellos países que ya los han prohibido. Los científicos colombianos deben alzar la voz para desafiar la aceptación ciega de ganancias por encima de las consecuencias no deseadas en salud pública, y se deben alentar los esfuerzos para prevenir el abuso de pesticidas.


Clorpirifós (CPF) é um pesticida registrado amplamente utilizado na agricultura colombiana, incluindo lavouras, animais de fazenda e animais de estimação, apesar de ter sido proibido na União Europeia e nos Estados Unidos. Estudos têm demonstrado que mesmo níveis baixos de CPF no sangue -que não inibem a acetilcolinesterase sanguínea-podem levar a distúrbios neurológicos e de desenvolvimento em crianças, como menor perímetro cefálico e alterações cerebrais, além de déficits psicomotores e cognitivos relacionados à capacidade de aprendizagem, atenção e memoria. Em adultos, o CPF é um desregulador endócrino e cancerígeno da mama. Altos custos econômicos diretos (devido ao tratamento) e indiretos (devido à perda de produtividade) têm sido associados à exposição ao CPF. Não apenas os trabalhadores agrícolas e suas famílias, que têm as maiores exposições, mas a população em geral que consome culturas pulverizadas com CPF também estão em risco. Por essas razões, o CPF foi recentemente proibido pela União Europeia (2020) e pelos EUA (2021). As políticas de regulamentação de pesticidas variam muito, dependendo de quais (e como) os estudos científicos são usados para avaliar os riscos à saúde. As avaliações de pesticidas financiadas pela indústria química devem ser retificadas para evitar conflitos de interesse. Além disso, o alinhamento político com os interesses da indústria não deve ter precedência sobre as evidências científicas independentes. É desanimador - para dizer o mínimo - que até que leis de saúde mais rígidas sejam aprovadas na Colômbia, o CPF e tóxicos relacionados continuarão a ser importados dos países que já os proibiram.

Identifying the key proteins among different types of human disease-causing coronaviruses is essential for the molecular mechanism and thereby designing potential drug molecules. Eight selected proteins of seven types of disease-causing coronaviruses, viz.SARS-CoV-2 (severe acute respiratory syndrome coronavirus2), SARS-CoV (severe acute respiratory syndrome coronavirus), MERS-CoV (middle east respiratory syndrome coronavirus), Human coronavirus OC43, Human coronavirus HKU1, Human coronavirus 229E and Human coronavirus NL63, were chosen for the comparison. Further, an attempt has been made to explore the most important host-pathogen interactions with a special focus on spike (RBD) protein region as this region deemed to be functionally most important. Epitope region was also identified which helps in the design of epitope-based vaccines. The structural comparison carried out among the seven types of human coronaviruses has revealed the molecular level details on the similarity among this series. This study has facilitated the identification of the important residues in the studied proteins which control the key functions such as viral replication and transmission. Thus, exploring the protein space in the family of coronaviruses, provide valuable insights into the molecular basis associated with the role of proteins and viral infections, which is expected to trigger the identification of the drug targets for coronaviruses infections, in a rational way.
