ABSTRACT

Objective To observe the effects of Purpura decoction and its disassembled prescriptions on peripheral blood reticulated platelets (RP) and serum thrombopoietin (TPO) of idiopathic thrombocytopenic purpura (ITP) model mice, to explore the action mechanism and core structure of the prescription. Methods Totally 84 BalB/c mice were randomly divided into normal group, model group, prednisone group, Purpura decoction group, Buyi Qixue group, Buqi Huoxue group and Buxue Huoxue group. ITP mouse model was made by injecting intraperitoneally guinea pig-antimouse platelet serum. From the 5th day of modeling, mice were intragastrically administered. The normal group and model group were fed with normal saline 0.2 mL per mouse. Purpura decoction group was fed with Purpura decoction 0.43 mL per mouse. Buyi Qixue group was fed with Buyi Qixue decoction 0.36 mL per mouse. Buqi Huoxue group was fed with Buqi Huoxue decoction 0.27 mL per mouse. Buxue Huoxue group was fed with Buxue Huoxue decoction 0.3 mL per mouse. Prednisone group was fed with prednisone acetate suspension 0.2 mL per mouse. Ten days later, took the blood from the eyeball and isolated serum, the mouse peripheral platelet, RP and TPO were observed. Results After treatment, the platelet count of treatment groups was improved. Compared with before treatment, Purpura decoction group,Buqi Huoxue group and prednisone group had significant difference in platelet count (P<0.05). The RP of model group was significantly higher than the normal group (P<0.05), but RP absolute value was lower than the normal group (P<0.05). Compared with the model group, RP of treatment groups reduced (P<0.05), and RP was negatively correlated with platelet count (P<0.05). TPO level of the model group was higher than that of the normal group (P<0.01). TPO of treatment groups declined significantly, and had significant differences with that of the model group (P<0.05). Conclusion Core structure of Purpura decoction is qi tonifying and blood activating drug. The mechanism of treating ITP is related with lowering RP and TPO.

ABSTRACT

Objective To study the disease pathogenesis of systemic lupus erythematosus patients with thrombocytopenia ,and discuss reticulated platelets (RP) in SLE merger thrombocytopenia disease of diagnosis and treatment in the clinical application val-ue .Methods 55 patients diagnosed with SLE patients and 50 cases of healthy check-up ,low platelet count patients of SLE patients treated before and after the inspection by Sysmex XE-5000 automatic blood cell analyzer immature platelet fraction (IPF) and other peripheral blood cell parameters .Results Group of patients with SLE and normal controls of IPF test results respectively was (5 .30 ± 3 .75)% ,(2 .74 ± 1 .05)% ,both comparative difference was statistically significant (P<0 .05);SLE associated with throm-bocytopeniagroupofIPF[(10.14±3.66)% ],platelets(PLT)[(67.2±13.5)×109/L],meanplateletvolume(MPV)[(12.64± 0 .92)fL] ,platelet distribution width(PDW)[(18 .24 ± 1 .70)fL] compared with normal control group and SLE did not reduce plate-let group ,differences had statistical significance(P<0 .05);SLE associated with thrombocytopenia in patients with IPF respectively before and after treatment was (9 .76 ± 1 .82)% and (5 .86 ± 0 .96)% ,both comparative difference was statistically significant (P<0 . 05) .Conclusion The main cause of SLE patients with thrombocytopenia may be associated with peripheral blood platelet destruc-tion ,IPF′s result can reflect the bone marrow platelet production ,Help in the auxiliary diagnosis of disease and prognosis judg-ment .

ABSTRACT

OBJECTIVE:To study the association between the reticulated platelets(RP) percentage and aspirin resistance(AR) with the percentage of reticulated platelets in whole blood representing the platelets turnover rate to illuminate the influence of platelets turnover rate on AR.METHODS:The RP percentage and arachidonic acid(AA) and adenosine diphosphate(ADP)-induced platelet aggregation in 80 patients after receiving aspirin(100 mg?day) for no less than 7 days were determined.The patients were divided into AR group,aspirin semi-resistance(ASR) group,and aspirin sensitive(AS) group,and the RP percentage in the three groups were compared.RESULTS:RP percentages in AR,ASR,and AS groups were(4.11?2.62)%,(2.23?1.79)%,and(2.18?1.24)%,respectively,showing signficant differneces between AR group and AS group(P0.05).CONCLUSION:Increased platelets turnover may induce AR,therefore,the patients with coronary artery disease should receive aspirn for multiple daily(antiplatelet therapy) or to take controlled release or sustained release aspirin.The safety and efficacy of these therapies should be confirmed in clinical trials.

ABSTRACT

BACKGROUND: Analysis of reticulated platelets (RPs) is useful for discriminating the causes of thrombocytopenia and monitoring the thrombopoiesis. In the patients with severe thrombocytopenia, we evaluated the thrombopoiesis-discriminating ability of several indices applying forward scatter (FSC) and thiazole orange (TO) fluorescence in addition to the percentage of reticulated platelets (RPs%). METHODS: Forty cases with decreased thrombopoiesis, twenty cases with increased thrombopoiesis and twenty cases with liver cirrhosis were selected. By flow cytometry with two analytic methods, dependent on or independent of the staining of CD41-PE as a platelet marker, the primary parameters including RPs% were measured and the applied parameters were calculated from them. And we compared the diagnostic efficiency of each parameter and analyzed the purity of platelet light scatter gate. RESULTS: The purity of platelet light scatter gate was significantly lower in patients with severe thrombocytopenia than in healthy persons with normal platelet counts (P<10(-6)), so the use of CD41-PE for platelet gating improved the diagnostic efficiency of RPs%. Compared to the primary parameters, the applied parameters originated from RPs%, FSC and TO fluorescence improved diagnostic efficiency significantly (RPs%: 55%, RPs%xs delta MFI: 80%) between decreased and increased thrombopoiesis groups. CONCLUSIONS: In the patients with severe thrombocytopenia, the estimate of the thrombopoiesis by a flow cytometric analysis can be more predictable by using platelet markers and by considering the fluorescence intensity of TO together with the RPs%.
