ABSTRACT

Objective:To optimize the formula of film-forming materials for compound rhizoma corydalis plastics. Methods:Poly-vinyl alcohol 124 (PVA124) and bletilla striata gum were selected as the film-forming materials. The amounts of PVA 124, bletilla striata gum, glycerol and absolute ethanol were used as the influencing factors in an orthogonal test with the composite score of film-forming time and appearance as the indices to screen out the best composition of film-forming materials. Results:The best composition of film-forming materials was as follows:8 g PVA 124, 1 g bletilla striata gum, 2 ml glycerol and 15 ml ethanol. Conclusion:The op-timized film-forming materials can be used for the preparation of compound rhizoma corydalis plastics.

ABSTRACT

OBJECTIVE To investigate the protective effect of total alkaloids of Rhizoma Corydalis (TARC) on experimental gastric mucosal lesions in rats. METHODS Gastric mucosal lesions were induced in rats by injecting acetic acid under gastric mucosal. From the 2nd day post the preparation of the rat model, cimetidine 400 mg · kg-1 or TARC 20, 40 and 80 mg · kg-1 was ig delivered for 15 d in different groups. Two days after the last delivery, gastric juice volume and total acidity were measured. Histopathology of stomach tissues was observed by HE staining. The area of gastric ulcer area was measured and the ulcer index and ulcer inhibitory rate were calculated. The expression of epidermal growth factor (EGF) and EGF receptor (EGFR) was detected by immunohistochemical staining. RESULTS Comparing with shame group, the eare of gastric ulcer and ulcer index were increased signifi?cantly in the model group(P<0.01), suggesting that the model rats were prepared properly. Compared with the model group, the ulcer area in rats of cimetidine and TARC 80 mg·kg-1 groups was decreased by 39.9%and 23.7%,respectively. The ulcer index was decreased by 52.3%and 30.5%,respectively. There was no significant difference between the cimetidine group and TARC 80 mg · kg-1 group, in the ulcer area or index. Compared with model group, EGF protein expression of cimetidine and TARC 40 and 80 mg · kg-1 groups was increased by 81.8%,24.2%and 57.6%,respectively while EGFR protein expression was increased by 45.9%,16.2%and 29.7%,respectively(P<0.05,P<0.01). Deciduous and necrotic gastric mucosal and a large amount of inflmmatory cells infiltration were observed in model group, and gastric mucosal lesions were improved in cimetidine and TARC 40 and 80 mg · kg-1 groups. CONCLUSION TARC has protective effect on gastric mucosal lesions in rats. The mechanism may be related to the increase of EGF and EGFR protein expression.

ABSTRACT

Objective:To optimize the ethanol extraction process of Rhizoma Corydalis and Salvia miltiorrhiza, and study the effect of comminution degree of the medicinal materials on the effective constituent. Methods:Using the alcohol concentration, alcohol vol-ume, extraction time and extraction times as the impacting factors, the content of tetrahydropalmatine and salvianolic acid as the indi-ces, the extraction process was optimized by orthogonal test of L9(34). Results:The best extraction conditions were as follows:Rhizo-ma Corydalis was crushed into coarse powder, the material / liquid ratio was 1∶6 with 75% ethanol as the extraction solvent, extracting twice with 1. 5h each. Conclusion:The extraction method is appropriate and simple, and can be used in the industrial production.

ABSTRACT

Since alkaloids are the major active constituents of Rhizoma corydalis (RC), a convenient and accurate analytical method is needed for their identification and characterization. Here we report a method to profile the alkaloids in RC based on liquid chromatography-tandem quadrupole time-of-flight mass spectrometry (LC-Q-TOF-MS/MS). A total of 16 alkaloids belonging to four different classes were identified by comparison with authentic standards. The fragmentation pathway of each class of alkaloid was clarified and their differences were elucidated. Furthermore, based on an analysis of fragmentation pathways and alkaloid profiling, a rapid and accurate method for the identification of unknown alkaloids in RC is proposed. The method could also be useful for the quality control of RC.

ABSTRACT

Objective To optimize the alcohol extraction technique of the medical herbs such as Rhizoma Corydalis and Radix Angelicae Dahuricae. Methods To optimize the alcohol extraction technique with L9(34) orthogonal test. The content of the tetrahydropalmatine in Rhizoma Corydalis, which was used as evaluating criteria, was determined by HPLC. Results The optimized extraction technique was as follows: the medicinal material was extracted for 3 times with 3 times amount of 75% alcohol,1.5 hours per time. Conclusions The optimized technique is reasonable and the extraction rate was high.

ABSTRACT

Objective To choose the bestway of extracting procedure of Rhizoma corydalis. Methods The optimal extraction process is selected with the orthogonal design. The content of corydalis B and the yield of extracts were used to evaluate the factor levels. Results The ideal extraction process is:extracted with 75% alcohol for 2 times, first 6 times amount, soaked 0.5 h, refluxed 1.5 h, then 4 times amount, refluxed 1.5 h. Conclusions This extractive condition is the best way.

ABSTRACT

Objective To develop a HPLC method for the determination of protopine in Rhizoma Corydalis Decumbentis Capsules. Methods A HPLC was performed on a Hypersil ODS column( 4.6? 250 mm, 5 ? m) . The mobile phase was methanol - 0.1 % water solution of triethylamine (65∶ 35). Determination wavelength was 290 nm and flow rate was 1.0 mL/min. The theoretical plates should over 4000 according to the content of protopine. Results A good linearity was shown in the range of 0.3168~ 1.5840 ? g, r=0.9996. The average recovery was 100.6 % , RSD=2.68 % . Conclusion The method is simple, effective and can be used for the quality control of Rhizoma Corydalis Decumbentis Capsules.

ABSTRACT

Objective: To explore the effect of processing on the content of tetrahydropalmatine in Rhizoma Corydalis. Methods: HPLC was used with chromatographic condition included: C 18 column, mobile phase of methanol water phosphate buffer (70∶30∶1.8) and detection wavelength at UV 280nm. Results: The content of tetrahydropalmatine in the Rhizoma Corydalis processed with vinegar is the highest among all processed products. The recovery is 99.07% and RSD =1.81%, respectively. Conclusion: The processing with vinegar can increase tetrahydropalmatine content of Rhizoma Corydalis.

ABSTRACT

In this paper, the effects of rhizoma corydalis turtschaninovii f.yanhusuo (large leaf type5 LLYH) on experimental myocardial ischemia and coronary blood flow are reported.LLYH was showed to be active in increasing the tolerance of mice to monobaric hypoxia and prolonging survival time of the animal under the high oxygen consumption condition induced by large dose of isoproterenol. It decreased the severity of myocardial ischemic necrosis induced by large dose of isoproterenol in mice and reduced the extent of myocardial infarction in Wistar rats 12 h after ligation of the root of th,e left coronary artery. In anesthetized open chest cats, coronary blood flow was increased by LLYH infusion with slight decrease in heart rate and blood pressure.
