ABSTRACT

OBJECTIVE To study the improvement effect of total flavonoids from Rosa multiflora root on vascular injury in rheumatoid arthritis (RA) model rats and its potential mechanism. METHODS Female Wistar rats were randomly divided into normal control group, model group, aspirin group (positive control, 30 mg/kg), low-dose and high-dose groups of total flavonoids from R. multiflora root (4.15, 8.30 g/kg, by crude drug), with 10 rats in each group. Except for the normal control group, the RA model was induced in other groups by collagen induction and high-fat diet. After 14 days of modeling, they were given corresponding drug solution/0.5% sodium carboxymethyl cellulose solution intragastrically, once a day, for 36 consecutive days. The total body score, arthritis index (AI) and swollen joint count (SJC) of the rats were evaluated regularly. After the last medication, serum levels of interleukin-6 (IL-6), intercellular adhesion molecule-1 (ICAM-1) and vascular cell adhesion molecule- 1 (VCAM-1) were determined. The pathological morphological changes in the vascular tissue of thoracic aorta were observed; the protein expression of Toll-like receptor 4 (TLR4) and the protein phosphorylation levels of Janus kinase 2 (JAK2) and signal transduction and activator of transcription 3 (STAT3) in vascular tissue of thoracic aorta were measured. RESULTS Compared with the normal control group, serum levels of IL-6, ICAM-1 and VCAM-1, protein expression of TLR4, and the protein phosphorylation levels of JAK2 and STAT3 in vascular tissue of thoracic aorta were increased significantly in model group (P< 0.01). The atherosclerotic plaque (atheroma), cholesterol crystal, lymphocyte infiltration and a small number of unbroken foam cell aggregation could be seen in the vascular tissue of thoracic aorta. Compared with the model group, total body score (except for the low-dose group), AI and SJC were decreased significantly in groups of total flavonoids from R. multiflora root on the 28th day (P<0.05 or P<0.01); total body score,AI and SJC were decreased significantly in low-dose group of total flavonoids from R. multiflora root on the 49th day (P<0.05 or P<0.01); the other quantitative indicators in serum and vascular tissue were significantly reversed in groups of total flavonoids from R. multiflora root (P<0.05 or P<0.01), and pathological damage of vascular tissue was significantly relieved. CONCLUSIONS Total flavonoids from R. multiflora root can significantly improve vascular injury in RA model rats, and its mechanism may be related to reducing the protein expression of TLR4 in vascular tissue and inhibiting the activation of IL-6/JAK2/ STAT3 signaling pathway.

ABSTRACT

The pink coloration of the teeth can be caused by dif-ferent factors. In the forensic field, the phenomenon called post mortem pink teeth has been described as a sign associated with violent deaths of various etiology. In clinical practice, it is also possible to ob-serve patients with pink teeth, frequently caused by trauma or iatrogenesis from orthodontics, whose production mechanism is due to different etiopatho-genesis, highlighting internal dentin resorption, ex-ternal cemento-dentinal resorption and dentin calci-fications. This article presents the case of an adult individual with a history of bone-dental trauma due to a road accident who, after a long time, attends the Dental Emergency and Patient Guidance Service of the Faculty of Dentistry of the University of Bue-nos Aires, where an ostensible pink coloration was detected in the lower right canine as an exploratory finding. The situation motivated a detailed clinical and radiographic approach, inquiring about the probable factors that intervened in its generation and development (AU)

ABSTRACT

Written by João Guimarães Rosa and published in 1961, the short story "My uncle the Jaguar" remains a unique fictional text in the Brazilian Literature. By means of a monologue in a dialogical situation and written in an oral Portuguese entangled with expressions and words in Tupi, a becoming-jaguar of a previous jaguar-killer is progressively enacted. This jaguar killer finally recognizes the jaguars as his real folks, as his kinfolk. The article emphasizes the untamable character of Rosa's short story and develops its antimetaphysical and political implications.

ABSTRACT

ABSTRACT

Abstract: Focusing on the novel "Grande Sertão: Veredas", the article articulates the trauma and desire notions to the figure of Diadorim, thinking about fiction as a response to the emergence of desire in its relationship with a traumatic core and refractory to the symbolic. Trauma is also addressed in its relationship with analytical technique, its particular mode of operation with time (emphasizing the Freudian use of the expression Nachtraglichkeit) and in its relation to the subversion of the linguistic code taken to the extreme by Rosa, who signs a work recognized throughout the world for the physiognomy of a unique language.

ABSTRACT

OBJECTIV E To optimize the s alt-processing technology of Rosa laevigata ,and to study high performance liquid chromatography(HPLC)fingerprints and chromaticity values of R. laevigata before and after salt-processing. METHODS The comprehensive scoring method was adopted to optimize the salt-processing technology of R. laevigata using appearance character , moisture and polysaccharide content as index. Fingerprints were established by HPLC method before and after salt-processing ,and chromaticity values (L*,a*,b*)of the powder before and after salt-processing were determined. The multivariate statistical analysis was carried out for raw product and salt-processing product of R. laevigata by using common peak areas and chromaticity values as index. RESULTS The optimal salt-processing technology of R. laevigata was to mix it with appropriate amount of salt water ,place them in the preheated frying wok at 140 ℃,fry them for 25 min,and rotate frying wok 20 times/min. Ten common peaks were calibrated by HPLC fingerprints before and after salt-processing ,and 3 components were identified ,such as gallic acid ,catechin and ellagic acid. The chromaticity values L*,b* and E* changed significantly after salt-processing. The multivariate statistical analysis method could distinguish raw products and salt-processing products into two categories ,in which peaks 1,5,6 and 10 and chromaticity values b* and E* were important characteristic factors. CONCLUSIONS The optimized salt-processing technology is stable and reliable ,and the established fingerprint has good repeatability and stability. Fingerprint and chromaticity values combined with multivariate statistical analysis can provide reference for the identification and quality analysis of R. laevigata before and after salt-processing.

ABSTRACT

Abstract: Climatic variation throughout the day influences the ecophysiological performance of plants at different growth stages and phases. Therefore, this work aimed to evaluate the effect of climatic variation on ecophysiological aspects of T. roseoalba and H. heptaphyllus at different hours of the day and indicate the ideal time for measuring ecophysiological variables in these species. The research was carried out in a greenhouse at the forest nursery of the Federal University of Paraíba, Campus II, in the municipality of Areia, Paraíba state, Northeastern Brazil. The experimental design was completely randomized, consisting of 10 evaluation times throughout the day (from 8 am to 5 pm), with 1 h hour interval between each evaluation. Temperature and air relative humidity inside and outside the greenhouse were evaluated to understand the effect on gas exchange (net assimilation rate of CO2, stomatal conductance, transpiration rate, internal concentration of CO2, and vapor-pressure deficit) and chlorophyll a fluorescence (initial, maximum, and variable fluorescence, photochemical quenching, and electron transport rate). Data were submitted to canonical correlation analysis and principal component analysis to verify the relationship between climatic and ecophysiological variables. For both species, higher correlation was found between internal and external relative humidity with all the ecophysiological variables analyzed, except for initial fluorescence. Thus, climatic factors influenced the photosynthetic performance of T. roseoalba and H. heptaphyllus plants, and 8 am to 9 am is indicated for carrying out ecophysiological evaluations in both species.

ABSTRACT

Objective:To investigate the antioxidant activity and chemical composition of 75% ethanol extract of Rosa cymosa roots and its different polar parts. Method:The 75% ethanol extract of R. cymosa roots was divided into dichloromethane, ethyl acetate, n-butanol and water parts by organic solvent extraction. In vitro antioxidant activity of each fraction was determined by 1,1-diphenyl-2-picrylhydrazyl (DPPH) and 2,2'-azino-bis(3-ethylbenzothiazoline-6-sulfonic acid) (ABTS) free radical scavenging assays, as well as ferric reducing antioxidant power (FRAP) test. The contents of total triterpenes, total phenols, total tannins and condensed tannins in each fraction were determined by spectrophotometry. SPSS 24.0 software was used to conduct Pearson correlation analysis between the antioxidant activity of each fraction and the content of the main components, and then the main active fraction and the main active components were determined. The chemical constituents of the active fraction was analyzed by ultra performance liquid chromatography-quadrupole-time-of-flight mass spectrometry (UPLC-Q-TOF-MS/MS), and the structures of the main chromatographic peaks were predicted. Result:Each fraction of R. cymosa roots had certain antioxidant activity, and there was a significant dose-effect relationship within a certain concentration range, but the antioxidant activity of different polar parts was different. In DPPH and ABTS free radical scavenging tests, the antioxidant activity of each fraction and vitamin C (VC, the positive drug) was ranked as ethyl acetate fraction>VC>n-butanol fraction>ethanol extract>water fraction>dichloromethane fraction. In FRAP test, the activity of ethyl acetate fraction was weaker than that of VC, and the other order was unchanged. The contents of total triterpenes, total phenols, total tannins and condensed tannins in ethyl acetate fraction were 3.81%, 50.33%, 3.32%, and 39.79%, in n-butanol fraction were 0.88%, 41.42%, 2.25% and 23.55%, in ethanol extract were 2.90%, 41.95%, 3.43% and 20.14%, in water fraction were 0, 26.80%, 16.90% and 7.57%, and in dichloromethane fraction were 21.23%, 12.90%, 1.59%, and 6.17%, respectively. Correlation analysis results showed that the contents of total phenols and condensed tannins were positively correlated with the antioxidant activity, the contents of total triterpenes were negatively correlated with the antioxidant activity, and the correlation between total tannins and antioxidant activity was not obvious. A total of 26 compounds were identified from the ethyl acetate fraction by UPLC-Q-TOF-MS/MS, including 11 condensed tannins, 4 hydrolysable tannins, 6 triterpenes, 3 flavonoids, 1 benzoic acid derivative and 1 chlorogenic acid analogue. Conclusion:Ethyl acetate fraction is the main antioxidant active site of R. cymosa roots, and phenols mainly composed of condensed tannins are the main active components. The results can provide experimental basis for the development of natural antioxidants.

ABSTRACT

The aim of this paper was to study the protective effect of total flavonoids from Rosa multiflora(TF-RM) on the injury of HUVEC induced by oxidized low density lipoprotein(ox-LDL). SPF male SD rats were randomly divided into blank group, simvastatin group(1.8 mg·kg~(-1)·d~(-1)) and TF-RM group(2.5 g·kg~(-1)·d~(-1)), with 10 rats in each group. They were intragastrically administered with drugs for 7 days, and then blood was collected from the abdominal aorta to prepare drug-containing serum. The HUVEC injury model was established through ox-LDL induction, and added with 15% simvastatin, 5% TF-RM, 10% TF-RM, 15% TF-RM drug-containing serum and blank serum, respectively. Reactive oxygen species(ROS) was determined by flow cytometry. Nitric oxide(NO) content was determined by nitrate reductase method. The contents of ET-1, P-selectin, E-selectin, ICAM-1, VCAM-1, IL-1β, IL-6 and TNF-α were determined by ELISA. The expression of Lox-1 protein was determined by Western blot. Compared with the blank group, ROS level in HUVEC and the contents of ET-1, P-selectin, E-selectin, ICAM-1, VCAM-1 and IL-1β in HUVEC were significantly increased(P<0.05), NO decreased significantly(P<0.01),Lox-1 protein expression increased significantly(P<0.05), and TNF-α and IL-6 showed an increasing trend. Compared with the model group, TF-RM significantly reduced ROS level in HUVEC and ET-1, P-selectin, E-selectin, ICAM-1, TNF-α, IL-1β content in supernatant(P<0.05), significantly increased NO content(P<0.01), and inhibited Lox-1 protein expression(P<0.05). VCAM-1, IL-6 contents showed a decreasing trend. Serum containing TF-RM acts on lectin-like oxidized low-density lipoprotein receptors, and exerts a protective effect on vascular endothelial cells by reducing cell oxidative damage, regulating vasoactive substances, and reducing adhesion molecules and inflammatory cascades.

ABSTRACT

Utilization of plant resources for treatment of Helicobacter pylori infections is one of the appealing approaches as rapid emergence of antibiotic-resistant strains is occurring throughout the world. Ethanol extract and its fractions from Hibiscus rosa-sinensis red flower were assessed for antibacterial and urease inhibitory activities towards forty-three clinical strains and two reference strains of H. pylori. The ethyl acetate fraction exhibited the most potent bacteriostatic activity with minimum inhibitory concentrations (MICs) of 0.2-0.25 mg/mL and minimum bactericidal concentrations (MBCs) of 1.25-1.5 mg/mL against all test strains, including forty-three strains resistant to one to four antibiotics, azithromycin (MICs, 8-256 µg/mL), erythromycin (MICs, 8-128 µg/mL), levofloxacin (MICs, 8-256 µg/mL), and/or metronidazole (MICs, 8-256 µg/mL). The fraction had similar antibacterial activities toward these test strains suggesting the preparation and the antibiotics do not have a common mechanism of anti-H. pylori activity. The fraction also had stronger effects on biofilm formation, morphological conversion, and urease activity of H. pylori than the other fractions and the ethanol extract. These flower preparations were non-toxic to three human cell lines, and nine compounds were also isolated and identified from the ethyl acetate fraction. In vivo research needs to be conducted to confirm the potential usefulness of H. rosa-sinensis flower and its constituents for effective prevention and treatment of H. pylori disease.

ABSTRACT

ABSTRACT: Cedrela fissilis is a species of great genetic diversity, with low population density and seminal propagation, which causes difficulties in the vegetative propagation process. This research evaluated the vegetative rescue and propagation of stem cutting rooting originated from epicormic and canopy sprouts of C. fissilis. For this, the induction of epicormic sprouts was evaluated 52 days after the complete girdling and semi-girdling 20 and 40 cm from the ground, and no girdling treatment, during spring (2018), summer (2018) and autumn (2019). The variables evaluated were, survival (%), sprouting (%), number, length (cm) and diameter (mm) of sprouts. The cuttings were made from spring epicormic sprouts, divided in two categories: 10 cm cuttings placed vertically in pits and 5 cm cuttings placed horizontally in furrows. The canopy sprouts were collected in the summer, then cut in apical and intermediate cuttings (15 cm). After 60 days, the cuttings were evaluated in survival (%), rooting (%), callus (%), average number and length of roots (cm). Results showed that only the complete girdling produced sprouts (average >67%) with no difference between 20 and 40 cm heights, with a greater number of sprouts during spring. The cuttings from epicormic sprouts, planted vertically in pits presented higher percentage of rooting (44%) than cuttings planted horizontally in furrows (17%). Cuttings from the canopy had inconsiderable rooting (apical - 2%; intermediate - 0%). The girdling periods influences the number of epicormic sprouts and its use for cutting was more efficient in rooting.

ABSTRACT

Objective: This study was performed to evaluate the use of PEEK abutments versus zirconium abutments with lithium disilicate superstructure on the esthetic acceptance and peri-implant clinical parameters. Zirconium abutments were found to have high surface roughness even after polishing. This causes a remarkable collapse of the soft tissue papilla that is a primary factor in the Pink Esthetic Score(PES), resulting in failed esthetic restoration in the anterior esthetic zone. Material and Methods:Twenty patients who needed a single implant restoration in the esthetic zone were included in this study. Eighteen patients completed screening, baseline, three-month, six-month, and twelve-month follow-up visits. At the screening visit, the patients were randomly allocated into two groups: Zirconia abutments (A) Group, and PEEK abutments (B) Group, both of which were restored with IPS e.max CAD superstructure. Patients Satisfaction was assessed by visual analogue scale (VAS). Pink esthetics score (PES), modified plaque index (mPI) and modified gingval index (mGI) were also assessed. Results: All implants were successfully osseo-integrated with a 100% survival rate over one year. Patient satisfaction was significantly higher for the PEEK group than the zirconium group. The pink esthetic score showed no statistical significance between both groups. The modified plaque index was significantly lower for the PEEK group than the zirconium group. The modified gingival index showed no statistical difference between both groups. Conclusion: PEEK revealed to be a versatile material to replace zirconium for implant abutments, due to its lower plaque affinity and higher patient satisfaction (AU)

(AU)

ABSTRACT

H. impetiginosus belongs to the Bignoniaceae family; it has a great potential for economic exploitation and can be used in landscaping of urban areas, reforestation, recovery of degraded areas, and folk medicine. The experiment was carried out to evaluate the effect of light and temperature regimes on the germination and vigor of Handroanthus impetiginosus seeds at the Seed Analysis Laboratory of UFRPE/UAG. The seeds were subjected to light regimes: white, far red, red, and no light at 15°C, 20°C, 25°C, 30°C, 35°C, and 40°C, using a completely randomized experimental design in a factorial scheme (4 × 6), with four repetitions of 25 seeds. The different light regimes did not influence the seed germination of H. impetiginosus. The highest germination percentage (92%) and germination speed index (7.94) were obtained at temperatures 28.2°C and 29.2°C, respectively, both under red light. The longest seedling length was also obtained from the seeds subjected to red light regime at 25°C. The temperatures of 15°C and 40°C inhibited the germination of H. impetiginosus seeds. H. impetiginosus seeds are classified as neutral photoblastics, and constant temperatures of 28.2°C and 29.2°C provided maximum germination.

ABSTRACT

Objective: The use of medicinal plants may be an alternative method for the control of Candida spp. Responsible for human infections. This study evaluated the antifungal effect of Schinus terebinthifolius extract (Brazilian Peppertree) on C. albicans, C. dubliniensis, C. glabrata, and C. krusei planktonic cultures and bio films. Material and Methods: Minimum inhibitory concentration (MIC) and minimum fungal concentration (MFC) of the plant extract were determined by the broth microdilution method. Biofilms formed in microplate wells were exposed to the extract for 5 min (50, 100 and 200 mg/mL) or 24 h (25, 50 and 100 mg/mL). After determination of colony-forming units per milliliter (CFU/mL), the data were analyzed by one-way ANOVA and Tukey's Test (P ≤ 0.05). Results: Different MIC (mg/mL) were found, such as 0.39 (C. dubliniensis), 1.56 (C. albicans), and 3.13 (C. glabrata and C. krusei). Besides, MFC (mg/mL) of 0.78 (C. dubliniensis) and 3.13 (C. albicans, C. glabrata and C. krusei) were also observed. Regarding the biofilms, significant reductions (log10) were found after 5 min and 24 h exposure to the plant extract, compared to the control group. However, C. dubliniensis was significantly affected only in 24 h treatment. Conclusion: S. terebinthifolius extract presented a significant antifungal effect on C. albicans, C. dubliniensis, C. glabrata, and C. Krusei both in planktonic cultures and biofilms (AU)
