ABSTRACT

Resveratrol (RES), a natural polyphenolic phytochemical, has been suggested as a putative anti-aging molecule for the prevention and treatment of Alzheimer's disease (AD) by the activation of sirtuin 1 (Sirt1/Sir2). In this study, we tested the effects of RES and Sirt1/Sir2 on sleep and courtship memory in a Drosophila model by overexpression of amyloid precursor protein (APP), whose duplications and mutations cause familial AD. We found a mild but significant transcriptional increase of Drosophila Sir2 (dSir2) by RES supplementation for up to 17 days in APP flies, but not for 7 days. RES and dSir2 almost completely reversed the sleep and memory deficits in APP flies. We further demonstrated that dSir2 acts as a sleep promotor in Drosophila neurons. Interestingly, RES increased sleep in the absence of dSir2 in dSir2-null mutants, and RES further enhanced sleep when dSir2 was either overexpressed or knocked down in APP flies. Finally, we showed that Aβ aggregates in APP flies were reduced by RES and dSir2, probably via inhibiting Drosophila β-secretase (dBACE). Our data suggest that RES rescues the APP-induced behavioral deficits and Aβ burden largely, but not exclusively, via dSir2.

RESUMEN La enfermedad por coronavirus del 2019es una preocupación a nivel mundial, su propagación está relacionada a características propias del virus y a la toma de acciones gubernamentales, de salud pública y hábitos sociales. Utilizar herramientas matemáticas y computacionales ayuda a tener un panorama de la situación actual y a futuro de esta problemática. Con el objetivo de reflejar el comportamiento de esta enfermedad mediante simulación con dinámica de sistemas y evaluar la cantidad de contagios activos, muertes y recuperados en Paraguay en el mes de febrero del 2021 se utilizó el software Vensim aplicando una modificación del modelo básico de epidemiología de Kermack y McKendrick que considera a la población de susceptibles, infectados y recuperados. La simulación de esta dinámica ha presentado diferencias de 758 casos activos, 216 muertes y 37009 recuperados respecto a lo reportado, siendo los casos activos la aproximación más importante del estudio.


ABSTRACT The 2019 coronavirus disease is a worldwide concern, its spread is related to the characteristics of the virus and the taking of government, public health and social habits. Using mathematical and computational tools help to have an overview of the current situation and a future of this problem. In order to reflect behavior of this disease through simulation with system dynamics and evaluating the number of active infections, deaths and recoveries in Paraguay in the month of February 2021, the Vensim software was released by applying a modification of the basic model of epidemiology of Kermack and McKendrick who consider the population of susceptible, processed and recovered. The simulation of this dynamic has presented differences of 758 active cases, 216 deaths and 37009 recovered with respect to what was reported, with active cases being the most important approximation of the study.


RESUMO A doença do coronavírus 2019 é uma preocupação mundial, sua disseminação está relacionada às características do vírus e à toma de ações governamentais, de saúde pública e hábitos sociais. O uso de ferramentas matemáticas e computacionais ajudam a ter uma visão geral da situação atual e futura deste problema. Para refletir o comportamento desta doença através de simulação com dinâmica de sistemas e avaliar o número de infecções ativas, óbitos e recuperações no Paraguai no mês de fevereiro de 2021, foi utilizado o software Vensim, aplicando uma modificação do modelo básico de epidemiologia de Kermack e McKendrick que considera a população de suscetíveis, infectados e recuperados. A simulação dessa dinâmica apresentou diferenças de 758 casos ativos, 216 óbitos e 37.009 recuperados em relação ao informado, sendo os casos ativos a aproximação mais importante do estudo.

@#The management of emerging infectious diseases has always been given a high priority in public health. Identification of the epidemiological characteristics and transmission patterns of emerging infectious diseases is of great significance to contain the disease transmission and reduce the damages to public health and socioeconomic developments. Currently, infectious disease dynamics models are mainly established based on infectious disease surveillance data to predict the epidemiological patterns and trends of emerging infectious diseases; however, many model-based predictions fail to achieve the expected results due to the presence of multiple uncertain factors during the integrated management of infectious diseases. This review describes the basic principles and variables of common infectious disease dynamics models, including the susceptible-infected-recovered ( SIR ) model, susceptible-infected-removed-susceptible ( SIRS ) model, susceptible-exposed-infected-removed ( SEIR ) model and improved SEIR model, compares the advantages and disadvantages of these models, and summarizes the advances of the infectious disease dynamics models in the prediction of trends in incidence of emerging infectious diseases, so as to provide insights into the effective application of infectious disease dynamics models in the management of infectious diseases.

ABSTRACT A new coronavirus denominated first 2019-nCoV and later SARS-CoV-2 was found in Wuhan, China in December of 2019. This paper compares three mathematical methods: nonlinear regression, SIR, and SEIR epidemic models, to track the covid-19 disease in nine countries affected by the SARS-CoV-2 virus, to help epidemiologists to know the disease trajectory, considering initial data in the pandemic, mainly 100 days from the beginning. To evaluate the results obtained with the three methods one-way ANOVA is applied. The average of predicted infected cases with SARS-CoV-2, obtained with the mentioned methods was: for United States of America 1,098,508, followed by Spain with 226,721, Italy with 202,953, France with 183,897 United Kingdom with 182,190, Germany with 159,407, Canada with 58,696, Mexico with 50,366 and Argentina with 4,860 in average. The one-way ANOVA does not show a significant difference among the results of the projected infected cases by SARS-CoV-2, using nonlinear regression, SIR, and SEIR epidemic methods. The above could mean that initially any method can be used to model the pandemic course.


RESUMEN Un nuevo coronavirus denominado primero 2019-nCoV y más tarde SARS-CoV-2 fue encontrado en Wuhan, China en diciembre de 2019. El objetivo de este trabajo es comparar tres métodos matemáticos: regresión no lineal, modelos epidemiológicos SIR y SEIR, para rastrear la enfermedad del COVID-19 en nueve países infectados por el virus SARS-CoV-2, con el propósito de ayudar al epidemiólogo a conocer el curso de la pandemia, considerando principalmente sus primeros 100 días. Para evaluar los resultados obtenidos de la aplicación de los tres métodos, se aplicó ANOVA de una vía. El número promedio de casos infectados con SARS-CoV-2, obtenidos con los tres métodos descritos son: para Estados Unidos 1,098,508, seguido de España con 226,721, Italia con 202,953, Francia con 183,897 Reino Unido con 182,190, Alemania con 159,407, Canadá con 58,696, México con 50,366 y Argentina con 4,860 en promedio. El ANOVA de una vía no muestra diferencias significativas entre los resultados de los casos infectados proyectados por SARS-CoV-2, utilizando la regresión no lineal y los métodos SIR and SEIR. Lo anterior podría señalar que cualquiera de los tres métodos estudiados puede modelar el curso de la pandemia en las condiciones descritas para cada uno.

ABSTRACT

Las matemáticas en epidemiología y en general en las ciencias biológicas constituyen, además de una herramienta, una forma de pensar y estructurar descripciones, explicaciones y predicciones de procesos. Por ello, tanto en epidemiología como en otras áreas del conocimiento biológico, las matemáticas son utilizadas para modelar. El objetivo de este artículo es presentar como los modelos matemáticos se utilizan en la teoría epidemiológica. En este artículo nos centraremos en un modelo en particular, el modelo SIR, utilizado para describir la evolución de epidemias. Se presenta sus características fundamentales desde el punto de vista matemático y se discute el papel de los diferentes parámetros. Además, este modelo se aplica, a modo de ejemplo, a la evolución del Covid-19 en Chile.


Mathematics in epidemiology and in general in biological sciences constitute, in addition to a tool, a way of thinking and structuring descriptions, explanations and predictions. Thus, both in epidemiology and in other areas of biological knowledge, the mathematics is used for modeling. The objective of this article is to present how mathematical models are used in epidemiological theory. In this article will focus on a particular model, the SIR model, used to describe the evolution of epidemics. Its fundamental characteristics from the mathematical point of view are presented and the role of different parameters. In addition, this model is applied, by way of example, to the evolution of Covid-19 in Chile.

ABSTRACT

Background: Cochlear Implantation (CI) has become an important modality of treatment for children with severe to profoundpre-lingual sensorineural hearing loss who do not benefit from hearing aids (HAs). The final outcome is not totally predictable,as there are a large number of factors which either alone or in combination will play their roles in the final outcome of CI.Aim of the Study: This study aims to evaluate prospectively the relative impact of multiple pre-, peri-, and post-operative factorson the final outcome of the CI in pre-lingual hearing impaired children aged 5 years under “Sruthitharangam” free cochlearimplant program of Government of Kerala.Materials and Methods: This study was conducted at Government Medical College, Kozhikode (GMC-KKD), Kerala, fromJanuary 2014 to January 2015. The study group consisted of 60 patients screened from the patients who have attendedAuditory verbal habilitation (AVH) categories of Auditory Performance (CAP) test, Meaningful auditory integration scale (MAIS)and Speech intelligibility rating test (SIR) at GMC-KKD, Kerala. Counseling of parents was done regarding regular follow-upsand therapy/support to the child at home.Observations and Results: Pearson correlation test and Spearman correlation test were done to check the correlation betweenage at which HA was first fitted and MAIS scores. Correlation between the age at which HA first fitted and MAIS was negative.As the age at which HAs were fitted increases, the MAIS score decreases. This indicates the significance of using the residualhearing and stimulation of auditory nerve as early as possible. Pearson correlation and Spearman correlation tests were appliedto check the correlation between age of surgery and MAIS score and found that there was negative correlation existing betweenage of surgery and MAIS scores. This meant, as the age at which surgery was done increases, the MAIS score decreases.Pearson correlation test and Spearman correlation tests were applied to check the MAIS scores and duration of AVH with HAs.Conclusions: A Cochlear implant was not a passive sensory aid or sensory substitution device that simply replaces a damaged ordefective cochlea to restore normal hearing but requires prolonged period of aural rehabilitation that involves perceptual learning,adaptation, and readjustment of their attention. The various risk factors that affect the auditory gain and speech perception eitheracting singly or in combination and the statistical analysis of the present study showed are the age at implantation, duration ofauditory deprivation, and the residual hearing which have a direct impact on the outcome over a period of 1 year.

ABSTRACT

Yttrium-90 (90Y) radioembolization therapy can directly deliver a high dose of radiation to the tumor being targeted,which has been demonstrated to be effective in treating patients with hepatic malignancy.Compared with transarterial chemoembolization,90Yradioembolization is less frequently associated with toxicities such as abdominal pain,fever,nausea,and vomiting.However,the efficacy and safety of 90Y radioembolization therapy are largely reliant on careful preprocedural evaluation and diligent follow-up.This review discusses the details of essential preprocedural work-up and postprocedural follow-up for patients treated with 90Y radioembolization.

ABSTRACT

Objective To investigate the expression and clinical significance of Sir2?related enzymes?1(SIRT1)in colorectal cancer. Methods The expression of SIRT1 was evaluated by immunohistochemistry in 86 selected cases of primary Colorectal cancer and 30 samples of normal rectum tissues besided carcinoma. The relationship of the expression and clinical pathological features were analyzed. Results The positive expression rate of SIRT1 in Colorectal cancer tissue was 88.4%,and significantly higher than in normal rectum tissue besided carcinoma(P<0.001). There were no correlation between the expression of SIRT1 and sex,age and tumor diameter,and significant positive correlation between the expression of SIRT1 and TNM stage (P<0.05),lymph node metastasis(P<0.05),infiltrating depth(P<0.05),and negagtive correlation between the expression of SIRT1 and tumor differentiation(P<0.05). Conclusion The positive expression rate of SIRT1 in colorectal cancer tissue was significantly higher than in normal rectum tissue besided carcinoma and intimate correlation with tumor differentiation,TNM stage,lymph node metastasis and Infiltrat?ing depth. Conclusion SIRT1 may be an important assistance gene to diagnose and judge prognosis,and may improve diagnostic rate and auxiliari?ly judge prognosis as a important Colorectal cancer marker.

ABSTRACT

Several proteins associated with apoptosis were analyzed to investigate the progress of presbycusis based on cell apoptosis. Four proteins of caspase-3, XIAP, Sir2 T1 and cytochrome C which were associated with apoptosis were selected. Influence and progress of four proteins on presbycusis were analyzed. Apoptosis is the key factor which causes presbycusis, including apoptosis of cochlear hair cells, cochlear spiral ganglion cells and primary auditory cortical neuron. There is a close relationship between the four proteins and apoptosis of hearing related cells. These four kinds of protein factors played a role in the progress of presbycusis and are expected to serve as a target for the treatment of presbycusis.

El método holmesiano o sherlockiano según se le quiera designar, se advierte en la obra de Pifano y se sostiene en los siguientes enunciados: 1 Ciudadosa observación mediante el empleo combinado de los sentidos, asociada a un conocimiento intiutivo y razonamiento deductivo. 2 Prestar atención a los detalles y mantener alerta a las discrepancias. 3 Considerar las diversas opciones presentes en un caso. 4 Aprender a diferenciar y conocer lo que es importante de lo que no lo es. El Maestro Pifano dio a conocer a sus alumnos lo que consideraba importante al fusionar el método clínico condicente a formular un diagnóstico (historia clínica y anamnesis cuidadosa, examen físico integral e integrado, e investigaciones complementarias) con el método epidemiológico que situaba al paciente en un espacio geográfico, en un tiempo determinado e insertado en una comunidad donde concurrían los diferentes elementos que determinaban su condición de enfermo. En suma, en su concepción, el estudio integral del enfermo era indisoluble de su ambiente, del biotopo donde se desarrollaban los fenómenos de la ecología asociada al grupo a la cual pertenecía-soma, psiquis y mundo externo

Through the discussion we will show how these elements were present in the work of Félix Pifano, M.D. Master Pifano unveiled their students what was considered important to merge the leading clinical method to make a diagnosis (medical history and careful anamnesis, physical examination, integral and integrated and complementary procedures) with the epidemiological method that placed the patient in a geographic area, at a time determined and inserted in a community where the various elements that determined their condition of ill person. In short, in its conception, the comprehensive study of the patient was indissoluble of its environment, the biotope where unfolded the phenomena of ecology associated to the group to which he belonged-soma, psyche and world external

ABSTRACT

OBJECTIVES: In 1995, an outbreak survey in Gozan-dong concluded that an association between fiberglass exposure in drinking water and cancer outbreak cannot be established. This study follows the subjects from a study in 1995 using a data linkage method to examine whether an association existed. The authors will address the potential benefits and methodological issues following outbreak surveys using data linkage, particularly when informed consent is absent. METHODS: This is a follow-up study of 697 (30 exposed) individuals out of the original 888 (31 exposed) participants (78.5%) from 1995 to 2007 assessing the cancer outcomes and deaths of these individuals. The National Cancer Registry (KNCR) and death certificate data were linked using the ID numbers of the participants. The standardized incidence ratio (SIR) and standardized mortality ratio (SMR) from cancers were calculated by the KNCR. RESULTS: The SIR values for all cancer or gastrointestinal cancer (GI) occurrences were the lowest in the exposed group (SIR, 0.73; 95% CI, 0.10 to 5.21; 0.00 for GI), while the two control groups (control 1: external, control 2: internal) showed slight increases in their SIR values (SIR, 1.18 and 1.27 for all cancers; 1.62 and 1.46 for GI). All lacked statistical significance. All-cause mortality levels for the three groups showed the same pattern (SMR 0.37, 1.29, and 1.11). CONCLUSIONS: This study did not refute a finding of non-association with a 13-year follow-up. Considering that many outbreak surveys are associated with a small sample size and a cross-sectional design, follow-up studies that utilize data linkage should become standard procedure.

ABSTRACT

Cancer microsphere technology is the latest trend in cancer therapy. It helps the pharmacist to formulate the product with maximum therapeutic value and minimum or negligible range side effects. Cancer is a disease in which the abnormal cells are quite similar to the normal cells, with just minute genetic or functional change. A major disadvantage of anticancer drugs is their lack of selectivity for tumor tissue alone, which causes severe side effects and results in low cure rates. Thus, it is very difficult to target abnormal cells by the conventional method of the drug delivery system. Microsphere technology is probably the only method that can be used for site-specific action, without causing significant side effects on normal cells. This review article describes various microspheres that have been prepared or formulated to exploit microsphere technology for targeted drug therapy in various cancers. We looked at the usefulness of microspheres as a tool for cancer therapy. The current review has been done using PubMed and Medline search with keywords.

ABSTRACT

La insulinorresistencia y el síndrome de insulinorresistencia constituyen la causa primaria mas importante dentro de la etiopatogenia del hígado graso no alcohólico, conjuntamente con la obesidad y la diabetes mellitus tipo 2. La insulinorresistencia puede ser causa y a la vez consecuencia de la enfermedad hepática. La prevalencia del hígado graso no alcohólico documentada en trabajos realizados en países desarrollados es la siguiente: el hígado graso no alcohólico simple tiene una prevalencia de un 20 a 23 por ciento, aumentando en los pacientes con hipertransaminasemia persitente, en los que se puede observar una prevalencia entre 21 y 63 por ciento, y la prevalencia de esteatohepatitis oscila entre un 2 y un 3 por ciento de la población. Es bueno señalar que esta prevalencia va en aumento conjuntamente con el aumento de la prevalencia del síndrome de insulinorresistencia, la obesidad y la diabetes mellitus tipo 2. El hígado graso no alcohólico constituye la causa mas frecuente de hipertransaminasemia persistente, de hepatopatía crónica y de cirrosis hepßtica idiopática. La edad igual o superior a 40 años, la presencia de insulinorresistencia, de síndrome de insulinorresistencia, de obesidad y de diabetes mellitus tipo 2, ensombrecen el pronóstico de los pacientes con hígado graso no alcohólico, por lo que la presencia de estos factores antes mencionados son considerados factores de mal pronóstico en el paciente con hígado graso no alcohólico(AU)


Insulin resistance and the insulin resistance syndrome are the more significant main cause in pathogeny of non-alcoholic fatty liver, together with obesity and type 2 diabetes mellitus. Insulin resistance may be the cause and the consequence of hepatic disease. Prevalence of non-alcoholic fatty liver documented in papers performed in developing countries is at follow: from a 20 percent to 23 percent, increase in patients presenting with persistent high level of transaminase, in which it is possible to observe a prevalence between 21 percent and 63 percent, and prevalence of steatohepatitis fluctuates between 2 percent and 3 percent of population. We must to mark that this prevalence is increasing together with the prevalence increase of insulin resistance syndrome, obesity, and type 2 diabetes mellitus. The non-alcohol fatty liver is the more frequent cause of a persistent high level of transaminase, of chronic liver disease, and of idiopathic liver cirrhosis. An age similar or higher than 40 years, presence of insulin resistance, insulin resistance syndrome, obesity, and type 2 diabetes mellitus darken the prognosis of patients presenting with non-alcoholic fatty liver, so the presence of above mentioned factors is considered as a poor prognosis factors in patient presenting non-alcoholic fatty liver(AU)

@#Objective To investigate the related factors of speech intelligibility in the prelingually deafened children after cochlear implantation. Methods 47 prelingually deafened children who had received cochlear implantation and their families were investigated with questionnaires and analyzed with Fisher test. Results The age when cochlea implanted, the age when hearing aided, the time after the cochlear implantation and the time the cochlea opened were related with their speech intelligibility rating. Conclusion Suitable time for implantation, the hearing aid and making reasonable rehabilitation programme play a crucial role in the hearing rehabilitation of prelingually deafened children after cochlear implantation.

Nicotinamide at millimolar concentrations affects cell survival in various conditions, and is being utilized therapeutically in many human diseases. However, the effect of an acute treatment of nicotinamide at such high dose on gene expression and cellular metabolism has rarely been determined previously. In this study, we found that levels of O-N-acetylglucosamin(O- GlcNAc)ylated proteins including Sp1 acutely decreased upon treatment of 10 mM nicotinamide. Concomitantly, Sp1 protein level decreased rapidly through accelerated proteasome-mediated proteolysis. Cotreatment of glucosamine or 2-deoxyglucose, which inhibits protein deGlcNAcylation, effectively blocked the decrease induced by nicotinamide. Interestingly, the decline in the levels of Sp1 and protein O- GlcNAcylation was only transient lasting for two days post treatment, and this pattern matched closely the rapid fluctuation of the cellular [NAD(+)]. Our results suggest a possible link between cellular nicotinamide metabolism and protein O-GlcNAcylation, and an existence of cellular [NAD(+)] homeostasis.

ABSTRACT

Objective To screen cell growth and senescence-related genes of the parasitic pmtist Trichomonas vaginalis,we launched an EST program and isolated two cDNA clones from a T.vaginalis cDNA library,which showed high homology in deduced amino acid sequences to yeast Sir2 and designated as TvSir2 and TvSir2-like.Method The cDNA sequence of TvSIR2 had a length of 1034 base pairs (bp) with an open reading frame of 915 bp,and TvSIR2-like,1214 bp with an open reading frame of 1116 bp.Result The two deduced amino acid sequences shared all the three conserved cole domains with yeast Sir2 and its homologues,suggesting that the two clones were Sir2 homologues. A cDNA fragment from each cDNA clone was subvloned into the expression vector pET-41a.The expression of the fusion proteins in E.coli BL21 stains was induced by isopropylthio-β-D-galactoside (IPTG).Two anti-sera were prepared by immunizing two guinea pigs with the purified fusion proteins, Western-blot analysis demonstrated that each anti-serum reacted with the corresponding recombinant protein and detected a clear band (TvSir2,34 000 Mr;TvSir2-like,42 000 Mr)in protein extracts of the protist.Immunofluolescence techniques showed that TvSir2 and TvSir2-like proteins were both localized in the legions of perinuelear (ER) and Golgi complex.Conclusion Our data suggest that TvSir2 and TvSir2-like were two members of Sir2 family.Their biological functions in the protist would be further studied.

ABSTRACT

This study was carried out to evaluate the relationship between the biological lead exposure indices and air lead concentrations measured by personal air samplers. The 72 occupationally lead exposed workers were observed and the biological lead exposure indices chosen for this study were blood lead(PuB), urine lead(PbU), zinc protoporphyrin in whole blood(ZPP), gamma-aminolevulinic acid in urine(ALAU), gamma-aminolevulinic acid dehydratase activity(ALAD), coproporphyrin in urine(CPU) and hemoglobin(Hb). The workers were divided into four groups by air lead concentrations: Group I; under 0.05 mg/m3, Group II; 0.05-0.10 mg/m3, Group III; 0.10-0.15 mg/m3 and Group IV; and over 0.15 mg/m3. For evaluation the relationship between the biological lead exposure indices and air lead concentrations was used as correlation coefficients. The results obtained were as follows: 1. In Group I, II, III and IV, the mean value of PbB were 25.45+/-1.84 microgram/dl, 27.87+/-3.53 microgram/dl, 31.21+/-1.76 microgram/dl and 47.02+/-13.96 microgram/dl. Between Group IV and other groups showed statistically significant difference(p<0.05). 2. There was an increasing tendency of PbB, PbU, ALAU and ZPP according to the increase the mean air lead concentration, while ALAD has decreasing tendency. CPU and Hb did not show any constant tendency. 3. Correlation coefficients between PbB, PbU, ZPP, ALAU, ALAD, CPU, Hb and air lead concentration were 0.95, 0.83, 0.89, 0.72, -0.83, 0.51 and -0.45 respectively, and regression coefficient between PbB(Y) and PbA(X) was Y=126.8746X+16.9996(P<0.01).

ABSTRACT

Total RNA was isolated from Trichomonas vaginalis and Tv-Sir2-like cDNA was amplified by RT-PCR and cloned into pGEM-T Easy plasmid. A fragment of Tv-Sir2-like cDNA was subcloned into the expression vector pET-41b and expressed in E.coli BL21 with induction of IPTG. The full-length of Tv-Sir2-like cDNA was cloned and sequenced. The prokaryotic expression system of pET-41b/Tv-Sir2-like was constructed. The fusion protein of Tv-Sir2-like was expressed in E.coli BL21, occupying 30% of the total bacterial protein after being induced by IPTG for 5 h. SDS-PAGE analysis showed that the fusion protein was about Mr 59 000. The recombinant protein of Tv-Sir2-like is efficiently expressed in E.coli BL21.
