ABSTRACT

Objective To study the relationship of different histological types and degrees of infiltration of gastric adenocarcinoma on lymph node metastasis.Methods The clinical data of 1 615 cases of gastric adenocarcinoma were analyzed retrospectively,and the rate and number of lymph node metastasis in different histological types and with different depths of invasion were statistically analyzed.Results There were 46 227 lymph nodes collected from 1615 cases,with an average of 28.62 per case. Lymph node metastasis was found in 1046(64.77%) cases of gastric cancer and 10 173(22.01%) lymph nodes. Different TNM stages showed different rates of metastasis: T1(M)2.78%,T1(SM)13.61%,T2 55.59%,and T3 84.84% respectively,while the metastatic rate for histolgically well-differentiated carcinoma was 50.25%,and for low-differentiated earcinoma was 78.95%,and both had statistically significant difference(P

ABSTRACT

Objective To observe the effects of VEGF antisense oligonucleotide transfected to SGC-7091 cell line on expression of VEGF and survivin of the cell.Methods The VEGF-ASODN was synthesised artificially.There were five groups in this experiment: control group,missense group and three various concentration groups.After transfection,RNA copies were detected by real-time PT-PCR,VEGF protein in cell and culture supernate was detected by ELESA,survivin protein was measured by Western-blot,and apoptosis was detected by FCM.Growth of cells was evaluated by growth curve.ResultsVEGF-ASODN tranfection reduced the VEGFmRNA and VEGF protein expression in gastric cancer cells and supernate remarkably.It also reduced survivin protein expression and increased apoptosis in gastric cancer cells.Conclusions Transfection with VEGF-ASODN to gastric cancer cells SGC-7901 can reduce the expression of VEGF,and can increase apoptosis and suppress the proliferation of gastric cancer cells.

ABSTRACT

Objective To evaluate the clinical significance of the intraoperative detection of peritoneal micrometastases of gastric cancer.Methods In selected 50 cases of gastric cancer in which no obvious peritoneal metastasis was found preoperatively or during laparotomy,Douglas′s pouch peritoneal biopsy was undertaken intraoperatively,then HE and CK-20 immunohistochemistry staining of the specimens was performed.The expression of CK-20 mRNA in peritoneal irrigation fluid was also determined by RT-PCR.Results HE staining of all cases was negative.The positive rate of CK-20 immunohistochemistry staining was 24.0 %(12/50),and 36.0 %(18/50) with RT-PCR method.The positive rate of CK-20 mRNA was significantly related with the histological type,the depth of invasion and the number of lymph node metastasis(P

ABSTRACT

Objective To investigate the correlated factors of peritoneal dissemination and lympah node metastasis from gastric cancer, the prognosis of patients with peritoneal dissemination and N_2 lymph node metastasis, and the impact of palliative surgery on the prognosis. Methods Based on our database built from 1994, the clinicopathologic data and the outcomes of the follow-up were analyzed respectively. ABSTRACT

Objective To investigate the clinical and pathological characteristics, diagnosis and treatment of gastric cancer with ovarian metastasis. Methods The clinical data of 17 cases of gastric cancer with ovarian metastasis, confirmed by surgery and pathology, were analyzed retrospectively. Results The average age of the patients was 48.41 years, and the first appearance of symptoms and signs often were of metastatic ovarian cancer. The main ultrasonographic findings were either a complex type of mass with both solid and cystic characteristics or only solid, and most of them were accompanied by intraperitoneal fluid accumulation. Bilateral metastatic ovarian cancer was more common(13 cases). The preoperative accurate diagnosis of this disease was difficult, so that the misdiagnostic rate was 64.7% in this series. Operation was done in all the patients, but prognosis was poor. The median survival time was only 11.6 months. Conclusions The prognosis of gastric cancer with ovarian metastasis is poor. It is of importance to inspect the stomach in cases of bilateral ovarian cancer. Radical resection of the primary disease focus together with hysterectomy and bilateral adnexectomy should be performed. Postoperative comprehensive therapy is conducive to improve the prognosis of gastric cancer with ovarian metastasis.

ABSTRACT

Objective To explore the significance of using cytologic and RT-PCR methods to examine(peritoneal) washings and peritoneal tissues of gastric cancer patients in prediction of peritoneal micrometastasis.Methods The peritoneal washings of 38 patients with gastric cancer and 5 patients with benign gastric(lesions) were collected and,at the same time,a small amount of omentum and peritoneum were removed for control.CEAmRNA expression of free cells in peritoneal washings were detected by RT-PCR method and(also) cytology of the washings were performed.Results The CEAmRNA expression rate of peritoneal washings and peritoneal tissues were 36.8%(14/38) and 39.5%(15/38)respectively.Both were more(sensitive) than that of cytologic examination 26.3%(10/38).TNM staging,depth of invasion,lymph node metastasis,and serosal involvement were related to the expression rate of CEAmRNA.Conclusions mRNA of CEA is more sensitive and specific than cytologic examination for detecting free cancer cells in peritoneal cavity.It is an effective method for detecting peritoneal micrometastases in gastric cancer patient.

ABSTRACT

Objective To investigate the expression and clinical significance of tumor infiltrating dendritic cells(TIDCs) within gastric tumor tissues.Methods Immunohistochemistry(IHC),in-situ hybridization(ISH) and flow cytometry were applied to detect the expression of S100,CD83 mRNA and CD83 on DCs in 45 gastric adenocarcinoma tissues.The co-relationship of the S100 and CD83 expression with clinical(pathological) features was analyzed.Results IHC showed that S100 expression was unevenly distributed(within) 42 cancer tissue and CD83 was only expressed in tumor-adjacent tissue and normal tissue.ISH showed that CD83 mRNA was limitedly expressed within 7 samples.S100 expression had no significant(correlation) with clinical pathological features,while CD83 reversely correlated with TNM stages.Detected by flow cytometry,CD83 was expressed in low level in all 45 gastric cancer tissues and negative correlations were found with lymph node metastasis and TNM stages of gastric cancer(r=-0.879,P

ABSTRACT

Objective To investigate the role of loss of heterozygosity (LOH) of SMAD2 and its relationship with clinicopathological parameters of primary gastric carcinoma. Methods Fifty cases of primary gastric carcinoma were monitored by polymerase chain reaction -single strand conformation polymorphism (PCR- SSCP) and silver staining to detect SMAD2 LOH. Results The incidence of LOH was 40.0%(22/45) at D18S450 and 33.3%(16/48)at D18S460 . ABSTRACT

Objective To investigate an effective method for detecting lymphatic micrometastasis in gastric carcinoma.Methods RT-PCR technique was applied to examine MMP-7 mRNA in 94 lymph nodes in 24 cases of gastric carcinoma.Results Routine pathological method detected lymphatic metastasis in 54 lymph nodes,while MMP-7 mRNA RT-PCR showed positive in 78 lymph nodes. When re-examination for the 28 negative lymph nodes in the initial pathological examination and positive in RT-PCR, 8 lymph nodes with metastasis were found by routine pathological method.2 of 5 patients whose lymph nodes were negitive in pathological examination,but positive in MMP-7mRNA were found liver metastasis 16 and 22 months after radical gastrectomy. Conclusions MMP-7 mRNA RT-PCR is a sensitive method for detecting lymphatic micrometastasis for patients with gastric carcinoma. It is a great help for eluvating the postoperative prognosis and supplemental treatmemt.

ABSTRACT

Objective To investigate the effect of TGIF on apoptosis of TGF-?-induced gastric canceer cell.Methods After TGIF was stably transfected into gastric cancer cell line BGC823,apoptosis was examined with flow cytometry,and the expressions of TGIF,caspase8 and caspase9 were analyzed with Western blot.Results The apoptosis rate of BGC823 cells with the treatment of TGF-? obviously increased,and was accompanied by activation of caspase9 but not caspase8.Moreover,compared with controls,pcDNA3.1-TGIF-transfected BGC823 cells significantly decreased the sensitivity of TGF-?-induced apoptosis.Conclusions TGF-? induced the apoptosis of gastric cancer cell BGC823 via caspase9 pathway;furthermore,TGIF could inhibit TGF-?-induced apoptosis.

Objective To study the relationship between lymph node micrometastasis and clinicopathological parameters. Methods Immunohistochemical method was adopted to detect CK19,CK20 and CD44v6 in 509 lymph nodes from 53 cases of gastric cancer, in which, conventional pathological diagnosis showed no lymph node metastasis. ABSTRACT

Objective To study the significance and expression of gelatinase A in human gastric cancers (GC). Method Gelatinase A expression in 36 cases of GC was examined by immunohistochemistry method. Results Expression of gelatinase A in GC (31/36) was much higher than that in paratumor tissues (7/18) or normal gastric tissues (2/14). There was no obvious correlation between the total gelatinase A staining and tumor size, Lauren typing . Intense expression of gelatinase A was freguently found in the advanced cancer, especially in GC with serosal invasion and lymph node metastasis, while expression of gelatinase A was not found in the early stage GC. Gelatinase A was shown to be expressed in the extracellur matrix of tumor tissues. The staining positive cells were mainly macrophages. Conclusions Gelatinase A expression could be used as a marker for evaluating the invasion and metastasis of GC. Macrophages may play an important role in this process.

Objective To study the significance of expression of hypoxia-inducible factor 1 alpha(HIF-1?) in gastric mucosal diseases. Methods Using tissue chip technique to creat tissue chip in 120 cases of vary gastric mucosal disease,and using S-P immunohistochemical methods to detect the expressions of HIF-1? in these tissue chips. Results The positive rates of HIF-1? was 28.3% in 120 tissue chips of gastric mucosal diseases. ABSTRACT

Objective To study the relationship between the type of histopathology and prognosis of contained mucous adenocarcinoma(CMAC) of stomach and its clinical significance. Methods Eighty-seven cases of CMAC of stomach were observed by means of pathomorphology, histochemistry, immunohistochemistry and follow up etc. Results The malignant behavior of the cancer was significant difference according to the nature and quantity of the CAMC and the primary pathological type of the cancer.The clinical type in simple mucous carcinoma was mainly Borrmann type I;in tubular papilla mucous adenocarcinoma was mainly Borrmman type II; in signet-ring cell mucous carcinoma was mainly Borrmman type IV,and in mixed cell mucous carcinoma was mainly Borrmman type III. There was significant difference in the lymphatic metastasis and survival rate between the 4 groups. Conclusions Correct differentiation of the pathological type of contained mucous adenocarcinoma of stomach is important for guading the treatment and predicting the prognosis.Cathepsin D expression can help for understanding the biological behavior of CMAC of the stomach.

ABSTRACT

Objective To study the role of mutation of p300/CBP gene in the development of human gastric carcinoma (GC) . Methods Mutations analysis of the p300/CBP gene were carried out in 30 fresh GC samples using a combination of polymerase chain reaction single-strand conformation polymorphism (PCR-SSCP) and DNA sequencing,and compared with gastric mucosa distance from the GC. Results p300/CBP genes mutations were detected in 4 of 30 fresh GCs. All the mutation were point mutations, including T to G in nucleotide 4741, 4773,4847 and 4881 in genomic DNA of p300. The mutation site was in the non-coding area in nucleotid 4741;whereas in the others were in the coding area, leading to the change of the amino acids, induding Ser to Arg in nucleotide 4773, Val to Ala in nucleotide 4847 and 4881, respectively. Of the 30 patients, 4 cases with the single site mutation were discovered, 2 cases had the first two sites mutation, while the others had all the four sites mutation. Conclusions Two to four poiuts mutation of p300/CBP gene are presented in GC.

ABSTRACT

Objective To determine the correlation between the expression of interleukin 8 (IL 8) and vascularity in human gastric carcinoma speciemens,and to provide a possible new clinical way for anti angiogenesis of tumor . Methods IL 8 mRNA expression was examined with reverse transcription polymerase chain reaction (RT PCR).IL 8 protein secretion level and its specific expression were determined by enzyme linked immunosorbent assay (ELISA).The number of blood vessels in the gastric carcinoma was determined by antibody against factor Ⅷ associated antigen and immunohistochemical analysis. Results Most of the tumors (26/29,89.7%) and metastatic lymph nodes (17/20,85%) expressed IL 8 at higher levels than that in corresponding normal mucosa of resection end of stomach and negative metastatic lymph node.The expression of IL 8 protein and IL 8 mRNA were at the same level.The level of IL 8 mRNA in the gastric carcinoma tissue correlated strongly and positively with vascularization (P= 0.001).Conclusions There is a positive correlation between IL 8 mRNA expression and the vascularity of gastric carcinoma.IL 8 produced by tumor cells may up regulate the microvascularization in the gastric cancer, and enhance the growth,spreading and lymph node metastasis in human gastric carcinoma.So decreasing the IL 8 expression might inhibit the microvascularization,growth and metastasis of gastric cancer.
