ABSTRACT

Objective To investigate the effects of morroniside extracted from root-bark of Sambucus williamsii on TNF-α-induced MC3T3-E1 cell inflammation, and explore its mechanism of action including Caspase, MAPK, and NF-κB signaling pathway. Methods MC3T3-E1 cells were cultured in medium containing TNF-α (50 ng/mL) and different doses of morroniside. The cell viability was examined by MTT assay, and the levels of IL-1β and IL-6 in the supernatants were identified by ELISA. The expressions of Caspase-3, Caspase-9, p-ERK, p-JNK, p-p38, p-IκBα, IκBα, and NF-κB on the protein level was tested by Western blotting. Results MTT assay results showed that morroniside could promote the proliferation of MC3T3-E1 cells and protect the MC3T3-E1 cells induced by TNF-α. ELISA results showed that the expressions of IL-1β and IL-6 were inhibited. Meanwhile, morroniside could reduce the expression of Caspase3, Caspase9, p-ERK, p-JNK, p-p38, and p-IκBα protein, and increase IκBα protein expression while there was no significant difference in the expression of NF-κB p65. Conclusion Morroniside has protective effect on TNF-α-induced MC3T3-E1 cells inflammation. The possible related mechanism is that morroniside inhibits the release of inflammatory cytokines, the activation of MAPK and NF-κB pathway, and the expression of Caspase, thereby inhibiting the apoptosis of MC3T3-E1 cells.

ABSTRACT

Objective: To study the chemical constituents from the roots of Sambucus williamsii. Methods: Silica gel, ODS, and preparative HPLC were used to isolate the compounds. Their chemical structures were elucidated on the basis of spectral data. Results: Eighteen compounds were isolated, and they were identified as 7α-O-ethylmorroniside (1), 7β-O-ethylmorroniside (2), dehydromorroniside (3), loganin (4), 7-dehydrologanin (5), 7-formyloysecologanin (6), sweroside (7), coniferyl alcohol 9-O-β-D-glucopyranoside (8), 3-methoxy-4-(2-glycerol)-phenylpropanol (9), (7R, 8R)-7, 8-dihydro-9'-hydroxyl-3'-methoxyl-8- hydroxymethyl-7-(4-hydroxyl-3-methoxyphenyl)- 1'-benzofuranpropanol-9'-O-β-D-glucopyranoside (10), (7R, 8R)-4, 7, 9, 9'- tetrahydroxy-3-methoxy-8-O-4'- neoligan-3'-O-β-D-glucopyranoside (threo) (11), (7R, 8R)-3-methoxy-8, 4'-oxyneoligna-3', 4, 7, 9, 9'- pentol (threo) (12), 5-(1'-hydroxyethyl)-methyl nicotinate (13), 3-(hydroxyacetyl) indole (14), 4'-hydroxy-N-(4-hydroxy-3- methoxybenzoyl)-3', 5'-dimethoxybenzamide (15), 3-methoxyl-1H-pyrrole (16), (1S, 3S)-1-methyl-1, 2, 3, 4-tetrahydro-β-carboline-3- carboxylic acid (17), and syringic acid-4-O-α-L-rhamnopyranoside (18). Conclusion: Compounds 8-10 and 13-17 are firstly isolated from the plants in Caprofoliaceae, and furthermore, compounds 1-4, 6, 11, and 18 are isolated from the plants of Sambucus L. for the first time.

ABSTRACT

Objective To determine the content of rutin in fruits,stems,and leaves of Sambucus williamsii Hance,and provide an essential reference for the exploitation and utilization of the resources. Methods The content of rutin was analyzed on an Agilent ZORBAX Eclispse XDB C18 column(150 mmí4. 6 mm,5 μm). The mobile phase was methanol-0. 1%phosphoric acid (4555),the column temperature was set at 25 ℃,the flow rate was 0. 8 mL·min-1,and the detection wavelength was set at 360 nm. Results The linear range of rutin was 0. 105-0. 525 μg (r=0. 999 6),the average recovery was 97. 86%,and RSD was 0. 49%. The content of rutin in stems, leaves, and fruits of Sambucus williamsii Hance was 0. 171, 0. 522, and 0. 165 mg·g-1 ,respectively. Conclusion The stems,leaves and fruits of Sambucus williamsii Hance all contain rutin,but at a significant different content level. The leaves contain the highest level of rutin.

ABSTRACT

This study was aimed to optimize the purification technology for Sambucus williamsii Hance. With the morroniside as a marker, the purification technology for Sambucus williamsii Hance was optimized by different types of macroporous resin. The results showed that the optimum purification technology was that, the extract of less than 1:250 (morroniside:resin) was adsorbed and the AB-8 resin was washed with distilled water, and then the morroni-side was eluted from the macroporous resin with 10% ethanol. And the content of the morroniside was more than 50%. It was concluded that the purification technology was simple, reliable, repeatable and suitable for industrial production.

ABSTRACT

Objective: To observe the promotion of Sambucus williamsii (SW) root barks for fracture healing. Methods: Rabbit models with radius fracture were established. The rabbits were divided into control, model, Dieda Pill negative control, and low-, mid-, and high-dose (0.17, 0.34, and 0.68 g/kg) of SW groups. Drugs were added into forage and were given to rabbits 2 d after modeling for 36 d. The effect of SW root barks on serum biochemical indexes and histomorphology of fracture healing was observed. Results: Compared with the model group, a significant increase of alkaline phosphatase (ALP) and acid phosphatase (ACP) was observed in high-and mid-dose SW groups in the initial stage of fracture healing of 12 d after administration (P < 0.05, 0.01). A significent increase of serum bone gla protein (BGP) was observed in the high-and mid-dose SW groups in the middle stage of fracture healing of 24 d after administration (P < 0.05, 0.01). X-ray revealed that deep periosteal reaction density was found, osteotylus was higher and more in the mid-dose SW group, and the density was similar to bone density; Light microscope revealed that trabecular bone appeared in new born bone tissue, and the substance of bone was encapsulated with I-section new born bone tissue. Conclusion: SW root barks have the active fraction for fracture healing by promoting the synthesis and secretion of ALP and BGP of osteoblast, promoting the synthesis of ACP in osteoclast, increasing the quantity of osteoblasts, promoting the absorption of necrotic bone, and speeding up the maturation calcification of bone matrix.

ABSTRACT

Objective: To study the chemical constituents in 60% ethanol extract from the stems of Sambucus williamsii. Methods: Six compounds were isolated and purified by macroporous resin D-101, silica gel, Sephadex LH-20, Toyopearl HW-40, ODS column chromatographies, and RP-HPLC. Results: Their structures were elucidated by spectroscopic methods and identified as 23-hydroxy-3-oxo-28-nor-12, 16-dien oleanane (1), 3-oxo-oleanolic acid (2), oleanolic acid (3), betulinic acid (4), palmitic acid (5), and 1-octacosanol (6). Conclusion: Compound 1 is a new nortriterpenoid named sambucusan A. Compounds 2 and 6 are isolated from this plant for the first time.
