ABSTRACT

Objective: To establish an HPLC method for the simultaneous content determination of seven constituents in Compound Yi’anning Pills (CYP). Methods: The determination was performed on Shim-pack GIST C18 column with mobile phase consisted of acetonitrile-0.1% phosphoric acid (gradient elution: 0-5 min, 5% acetonitrile; 5-12 min, 5%-12% acetonitrile; 12-17 min, 12%-20% acetonitrile; 17-30 min, 20% acetonitrile; 30-36 min, 20%-50% acetonitrile; 36-45 min, 50%-80% acetonitrile; 45-70 min, 80% acetonitrile) at the flow rate of 1.0 mL/min. The detection wavelength was set at 203 nm for notoginsenoside R1, ginsenoside Rg1, and ginsenoside Rb1, 220 nm for schisantherrin A, 250 nm for schisandrin, 268 nm for salvianolic acid B, and 270 nm for tanshinone IIA. The column temperature was set at 35 ℃ and the injection volume was 10 μL. Results: The linear range of detection concentration was 0.015-0.150 mg/mL for notoginsenoside R1, 0.077-0.766 mg/mL for schisandrin, 0.006-0.062 mg/mL for salvianolic acid B, 0.009-0.092 mg/mL for buddleodide, 0.050-0.503 mg/mL for ginsenoside Rg1, 0.067-0.670 mg/mL for ginsenoside Rb1, and 0.011-0.108 mg/mL for tanshinone IIA. Average recoveries respectively were 99.5%, 99.8%, 99.2%, 98.9%, 96.9%, 98.8%, and 97.1%. Conclusion: The method is simple, effective, and accurate, which can be used for simultaneous determination of seven active constituents in CYP.

ABSTRACT

Objective: To optimaize the ethanol extraction technique for Schisandrae Fructus and Salviae Miltiorrhizae Radix and confirm the optimum extraction process parameters by multi index comprehensive weighted score evaluation. Methods: Alone and mixed extraction was carried out by using 90%, 70%, and 50% ethanol relux extraction. Orthogonal test method was uesd to optimize the process with ethanol concentration, solvent ratio, extraction time, and extraction times as detecting factors and schisandrin, schisantherrin A, deoxyschisandrin, schisandrin B, Tanshinone IIA, and salvianolic acid B as examining indexes, the detection was carried out by HPLC and DAD detector. The data were analyzed by multi index comprehensive weighted score evaluation. Results: The effective composition contents of mixed extract from Schisandrae Fructus and Salviae Miltiorrhizae Radix were more than alone extracting. The best extraction conditions were solvent ratio of 1:8 with 90% ethanol reflux extraction for three times, and 1.5 h each time. Conclusion: The mixed extracting is simple and appropriate, the extraction yield of active ingredients by mixed extracting is more than alone extracting. Different materials with different wavelength can effectively eliminate the interference at the same time, the accuracy is higher.

ABSTRACT

Objective To establish a HPLC method for the determination of deoxys chizandrin and schisantherrin A in Fructus Schisandrae Chinensis. Methods The as say was conducted on a YWG-C18 column with methonal-water (77 ∶23) as mobile phase. The flow rate was 1.0 mL/min and detection wavelength was 230 nm. Results Satisfactory linearities of deoxyschizandrin and schisantherrin A were shown in the range of 0.125~1.25 ?g and 0.08~0.8 ?g,and the recovery was 100.1 %an d 100.0 %(RSD being 0.15 %and 0.28 %。Schisandra sphenanthcra),and 99.7 %an d 101.2 %(RSD being 0.32 %and 0.67 %。Schisandra chinensis) (n=5). Conclusion The HPLC method was simple,curate,highly sensitive and reproducible. It may b e used for the quantitative determination of deoxyschizandrin and schisantherrin A in Fructus Schisandrae Chinensis.
