ABSTRACT

Objective: Network pharmacology and molecular docking technology were used to study the material basis and possible mechanism of Shaoyao Decoction in treatment of ulcerative colitis (UC). Methods: TCMSP and TCMID were used to obtain the potential active components and drug targets of Shaoyao Decoction. GeneCard and OMIM were used to search disease targets. The common targets obtained by matching drug targets and disease targets were imported into String to construct a PPI network, and Cyto NCA plug-in was used to screen key targets. The network diagram was drawn by connecting the key targets with the corresponding components, so as to screen the key components. GO and KEGG enrichment analyses were performed on key targets. SYBYL-X2.0 software was used to dock the molecules of the key components with the key targets. The rat UC model was replicated in vivo. After the intervention of Shaoyao Decoction, the disease activity index was observed, the colonic pathological damage was evaluated, and the levels of TNF-α, IL-4 and CXCR4 were detected by ELISA. Results: A total of 424 potential active components were found in Shaoyao Decoction. The key components included quercetin, palmitic acid, catechin, and procyanidins, etc. Its 41 key targets for UC were mainly related to the positive regulation of transcription, the negative regulation of apoptosis process, the signal transduction, and other biological processes. The key targets played a role in treating UC through signaling pathways such as TNF, HIF-1, cancer pathway, TLR, PI3K-Akt, et al. Molecular docking results showed that key components had good binding activity with corresponding targets. Shaoyao Decoction improved colon pathological damage, down-regulated the level of TNF-α and CXCR4, and up-regulated the level of IL-4 in vivo. Conclusion: Shaoyao Decoction has the characteristics of "multi-components, multi-targets, multi-pathways" in treatment of UC, which lays a foundation for further study of its mechanism.

ABSTRACT

Objective To investigate the clinical effect of traditional Chinese medicine rhubarb peony decoction combined with laparoscopic surgery in the treatment of elderly patients with acute suppurative cholecystitis. Methods From March 2016 to April 2017,52 patients with acute suppurative cholecystitis who were surgically treated in Anji Branch of Zhejiang Province Traditional Chinese Medical Hospital were selected in this study.According to different treatment methods, the patients were divided into control group and observation group, with 26 cases in each group. The control group was given laparoscopic surgery, the observation group was given traditional Chinese medicine rhubarb peony decoction based on laparoscopic surgery. The clinical treatment effect, hospitalization time, leukocyte recovery time, antipyretic time, antibiotic use time, occurrence of complications, and patients' satisfaction with treatment were observed. Results The patients' satisfaction rate with the treatment of the observation group was 96.15%,which was significantly higher than 73.08% of the control group (x2 =18.093,P =0.001).The hospitalization time, antibiotic use time, defervescence time and leukocyte recovery time of the observation group were (6.2 ± 0.9) d,(4.4 ± 0.6) d,(3.4 ± 0.7) d,(4.1 ± 1.1) d,respectively, which were significantly shorter than those in the control group (t =14.706,13.425,12.604,13.102;P =0.003,0.001,0.002,0.001).The treatment effective rate, incidence rate of complication in the observation group were 96.15%,7.69%,respectively,which were significantly better than those in the control group (x2 =96.15,9.082;P =0.001,0.001).Conclusion Laparoscopic surgery combined with traditional Chinese medicine rhubarb peony decoction for elderly patients with acute suppurative cholecystitis can significantly shorten the length of hospital stay, defervescence time, leukocyte recovery time and antibiotic use,and reduce the incidence of postoperative complications, improve the patients' clinical effect of surgery. In addition, this method can also improve the patients' satisfaction with the treatment, which can be used in clinical practice.

ABSTRACT

Objective: To observe the efficacy of combined Chinese and Western medicines in the treatment of senile functional constipation. Methods: One hundred patients with senile functional constipation were divided into treatment group and control group. There were 50 in each group. The patients in the treatment group were treated with Danggui Shaoyao Decoction and Cisapride Tablets, while the patients in the control group were treated with Cisapride Tablets only. The efficacy in the two groups were compared after three courses. Results: The total effective rate of the treatment group was 92% and the control group's was 78%, the former was higher than latter with the comparatively significant difference (P < 0.05). Conclusion: The results indicate that the combination of Chinese and Western medicines is highly effective in the treatment of senile functional constipation.

ABSTRACT

Objective To determine the content of berberine hydrochloride in Shaoyao decoction. Methods HPLC was performed on an Agilent XDB C18 column by using acetonitrile-0.05 mol/L potassium dihydrogen phosphate-triethylamine (30∶70∶0.14) as mobile phase at a flow rate of 1.0 mL/min, the column temperature was 25 ℃ and the UV detection wavelength was 345 nm. Results Berberine hydrochloride showed a good linearity redationship in the range of 0.07~1.40 ?g. The average recovery was 98.63% (n=6, RSD=1.21%). The average content was 1.55%. Conclusion The method is simple and accurate, with good repeatability, and can be used for the quality control of Shaoyao decoction.
