ABSTRACT

Background: Cathartic colon belongs to the category of 'constipation' in Traditional Chinese Medicine, and its pathogenesis is related to deficiency of kidney temperament and weak promotion ability, which has become a hot spot of global medical attention. Aims: To investigate the effect and possible mechanism of Jiawei Shenqi-wan (JSQW) on intestinal transmission function and pathological changes of interstitial cells of Cajal (ICCs) in rats with cathartic colon. Methods: Forty rats were randomly divided into blank group, model group, prucalopride group and JSQW group. Fecal moisture content, fecal particle number and intestinal transit rate were detected. The pathological changes of ICCs were observed under transmission electron microscope. RT-qPCR was used to detect the mRNA expressions of water channel proteins (AQP3 and AQP9) and SCF/c-kit pathway. Immunohistochemistry was used to measure the expressions of α-smooth muscle actin (α-SMA), inducible nitric oxide synthase (iNOS) and 5-hydroxytryptamine (5-HT)

ABSTRACT

Objective To observe the effect of Jingui Shenqi Wan (JSW) on the content of calcium channel protein CaV1.3 of guinea pigs with ototoxic deafness induced by gentamicin (GM). Methods Forty healthy guinea pigs were randomly divided into normal group, model group and high-, middle-, and low-dose JSW treatment groups. The model group was given intramuscular injection of GM ( 120 mg/kg) per day for 10 continuous days. The high-, middle-, and low-dose JSW treatment groups were given intramuscular injection of GM (120 mg/kg) and intragastric administration of JSW in the dosage of 20.3, 13.5, 6.08 g·kg-1·d-1 respectively per day for 10 days. Immuno histochemistry was used to detect the mean optical density value of CaV1.3 expression in cochlear hair cells. Results There were significant differences of the optical density value of CaV1.3 in cochlear hair cells between the model group and normal group (P<0.01), and between the model group and high-dose JSW treatment group (P<0.05). Conclusion JSW has certain effect on preventing and treating guinea pig with ototoxic deafness induced by GM, thus to protect the hearing function.

ABSTRACT

Shenqi-wan, Oriental herbal medicine formulation, has been traditionally used for delayed mental and physical development in children, complications of diabetes, and glomerulonephritis. In the present study, the protective effect of the aqueous extract of Shenqi-wan against traumatic brain injury (TBI) in the rat hippocampal dentate gyrus was investigated. For this study, step-down avoidance task, terminal deoxynuclotidyl transferase (TdT)-mediated dUTP nick end labeling (TUNEL) assay, Bax immunohistochemistry, and 5-bromo-2'-deoxyuridine (BrdU) immunohistochemistry were conducted. In the present results, it was shown that apoptotic cell death and cell proliferation in the dentate gyrus were significantly increased following TBI in rats and that the aqueous extract of Shenqi-wan suppressed the TBI-induced increase in apoptosis and cell proliferation in the dentate gyrus. Based on the present results, it is possible that the aqueous extract of Shenqi-wan has a neuroprotective effect on TBI-induced neuronal cell death.
