ABSTRACT

To explore the main target and signal pathway of Simiao Yongan Decoction in the treatment of psoriatic arthritis(PsA) by network pharmacology, so as to reveal the intervention mechanism of Simiao Yongan Decoction in the treatment of psoriatic arthritis. The platform of pharmacology technology of traditional Chinese medicine system(TCMSP) was used to predict and screen the active ingredients of Simiao Yongan Decoction, and GeneCards database was searched to obtain the disease target related to the psoriatic arthritis. Protein interaction network model was constructed with STRING platform; drug-component-target-disease network map was constructed with Cytoscape Software; Wayne Diagram of common target of Simiao Yongan Decoction and psoriasis arthritis was drawn with the help of ClusterProfiler R Software. At the same time, the genetic ontology(GO) enrichment analysis and the Kyoto encyclopedia of genes and genomes(KEGG) pathway analysis were conducted. Through database analysis, 1 128 targets related to 70 main active components of Simiao Yongan Decoction and psoriatic arthritis were selected. On this basis, the interaction network between Simiao Yongan Decoction and psoriatic arthritis was constructed, and 38 common targets were screened out. By GO and KEGG enrichment analysis, 135 signal pathways related to the main components of Simiao Yongan Decoction were selected. It was found that Simiao Yong-an Decoction may play a role in the treatment of psoriatic arthritis through antiviral effect, anti-inflammatory repair, protection of vascular endothelial cells, regulation of immunity and other multiple targets. The mechanism of Simiao Yongan Decoction in the treatment of psoriatic arthritis from multi-component, multi-target and multi-pathway was revealed, which provided a research direction for screening its subsequent clinical effect evaluation indexes.

ABSTRACT

Objective: To study the chemical constituents of Simiao Yongan Decoction. Methods: The silica gel, ODS, Sephadex LH-20, and AB-8 macroporous adsorption resin column chromatography were used to isolate and purify the chemical constituents from Simiao Yongan Decoction. The structures of the constituents were identified on the basis of physiochemical properties, NMR, MS, and other data. Results: Twenty-two compounds were isolated and identified as 5(S)-5-carboxystrictosidine (1), harpagoside (2), geniposide (3), glycyrrhetinic acid (4), glycyrrhizic acid (5), hyperoside (6), liquiritin (7), isoliquiritoside (8), liquiritigenin (9), isoliquiritigenin (10), luteolin (11), quercetin (12), 2-(3-hydroxy-4-methoxyphenyl)ethyl O-α-arabinopyranosyl-(1→6)-O-α- rhamnopyranosyl-(1→3)-O-β-glucopyranoside (13), angroside C (14), acteoside (15), cinnamic acid (16), ferulic acid (17), (E)-aldosecologanin (18), protocatechuic acid (19), stigmasterol (20), hentriacontanol (21), and daucosterol (22). Conclusion: Compounds 1-3, 5, 6, 8, 11, 13-15, 18-21 are isolated from Simiao Yongan Decoction for the first time.

ABSTRACT

Objective Network pharmacology method was adopted in this study to explore the active compounds and mechanism of Simiao Yongan Decoction for angiogenesis. Methods Chemical components and targets related to the four traditional Chinese medicine (TCM) herbs were searched through the Traditional Chinese Medicine Systems Pharmacology Database and Analysis Platform (TCMSP). Compound-target network were constructed to explore the mechanism of Simiao Yongan Decoction. Through OMIM database, TTD database, and PharmGkb database, the targets of angiogenesis were selected, and then the network diagram of disease-target interaction was constructed; The nuclear target of drug target and disease target interaction diagram was screened, ClueGO was used to carry out GO analysis of the nuclear target, and KEGG database was used to carry on the related pathway enrichment of the nuclear target. Results Selecting the oral bioavailability (OB) ≥ 30% and drug likeness (DL) ≥ 0.18 as filter condition, 120 active ingredients and 155 corresponding protein targets were screened from Simiao Yongan Decoction. Through the evaluation of network topological characteristics, such as degree, medium degree, and proximity to the center, 197 targets were selected to serve as the nuclear target in atherosclerosis. A total of 942 gene ontology (GO) entries were screened out, including 685 biological process entries, 164 molecular function entries, and 93 cellar component entries. The KEGG database was used for the enrichment of the selected targets, and 20 pathways were screened which had effect on angiogenesis. Conclusion This study revealed the main active compounds and possible mechanism of Simiao Yongan Decoction for treatment of angiogenesis and verified the characteristics of multi-components, multi-targets, and integral regulation for Simiao Yongan Decoction, predicting the main possible mechanism for the treatment of angiogenesis, which provided theoretical basis for the study of active ingredients and experimental research.

ABSTRACT

Objective To observe the effects of serum containing Simiao Yongan decoction (SYD) on secretion of extracellular matrix (ECM) of rabbit vascular smooth muscle cells (VSMC) induced by high insulin/high glucose and its mechanism. Methods Serum containing SYD was prepared by serum pharmacological method, and the proliferation of rabbit VSMC was induced by high concentrations of glucose and insulin. The experiments were divided into following groups: blank group [normal glucose DMEM containing 10% fetal bovine serum (FBS)], normal group (normal glucose DMEM containing 10%FBS+normal serum), model group (high glucose DMEM containing 10%FBS+100 U/L insulin+normal serum), western medicine group (high glucose DMEM containing 10%FBS+100 U/L insulin+serum containing simvastatin), traditional Chinese medicine group (high glucose DMEM containing 10% FBS +100 U/L insulin+serum containing SYD). The content of hydroxy proline (Hyp) in cell culture supernatant was detected by digestion method. The reverse transcription-polymerase chain reaction (RT-PCR) was used to assay the effects of serum containing drug on the mRNA expressions of collagen-Ⅳ(COL-Ⅳ), matrix metalloproteinase-2 (MMP-2), tissue inhibitors metalloproteinase-1 (TIMP-1) and the MMP-2/TIMP-1 ratio of rabbit VSMC. Results Compared with the model group, the contents of Hyp in cell culture supernatant in traditional Chinese medicine group and western medicine group were significantly reduced (both P 0.05). Conclusion Serum containing SYD can reduce the secretion of ECM of the rabbit VSMC during the high state of glucose and insulin, and the mechanisms are possibly related to reducing the synthesis of Hyp, influencing the mRNA expression levels of COL-Ⅳ, MMP-2, TIMP-1, and adjusting the balance of MMP-2/TIMP-1.

ABSTRACT

Objective:To observe whether the Simiao Yongan Decoction regulate the nuclear factor-?B(NF-?B) expression and related factors in atherosclerosis(AS)rabbit model,thus restrain the occurrence of atherosclerosis and plaque formation.Methods:56 male Japanese rabbits were randomly divided into normal group,model group,Simvastatin group and Simiao Yongan Decoction group,in addition to the normal group,other groups were established aortic atherosclerotic plaque model.From the1st day of experiment,the corresponding drug intervention began,10 weeks later,the rabbits were killed over the weekend after the anesthesia line access aortic NF-?B immunohistochemical staining,and the experimental dynamic 0,3,6,10 weeks in serum TNF-?,IL-1 and MCP-1 content.Results:The expression of NF-?B p65 subunit in model group was rich,and compared with it,the expression of NF-?B p65 subunit in treatment groups decreased,and the Simiao Yongan Decoction can obviously inhibit the expression of NF-?B p65 subunit.In model group,the expressions of IL-1,TNF-?and MCP-1 in serum increased significantly at different time points(P

ABSTRACT

AIM: To investigate the method for using semi-bionic extraction (SBE) on Simiao Yongan Decoction as an example. METHODS: Orthogonal design was used to optimize the extraction conditions of SBE with the evaluation markers such as chlorogenic acid, ferulic acid, glycyrrhizic acid, total polysaccharide, ethanol soluble extractives and dried extract. RESULTS: The optimized SBE conditions were as follows: The medical material was powdered through a sieve with 4 meshes) and extracted with 20% ethanol for three times, the pH of the three extractions were in proper order 3.50, 7.50, 8.50 ; the solvent volume was 10,8,8 times, respectively, and the extraction time was 1.5,1.0,1.0 h, respectively. CONCLUSION: The optimum extraction conditions are stable and practicable, and surpass the original SBE extraction conditions.
