ABSTRACT

Objective To observe the clinical efficacy and safety of sodium glycididazole injection combined with concurrent chemoradiotherapy in treatment of primary advanced cervical cancer.Methods A total of 84 patients with primary advanced cervical cancer were selected from November 2010 to November 2016 in Qinghai Provincial People's Hospital.All the patients were divided into the control group (n =42) and the experimental group (n =42) according to random number table method.The control group was treated with routine chemoradiotherapy,and the experimental group was treated with sodium glycididazole injection based on the control group.The curative effects,levels of serum tumor markers such as squamous cell carcinoma associated antigen (SCC),tissue polypeptide specific antigen (TPS),soluble human major histocompatibility complex-Ⅰ molecular chain related gene A protein (sMICA),hypoxia inducible factor-1α (HIF-1α),human epidermal growth factor receptor 2 (Her-2) and adverse reactions of the two groups were compared.Results Four months after treatment,the total effective rates of the experimental group and the control group were 73.81% (31/42) and 50.00% (21/42).The total effective rate in the experimental group was higher than that in the control group,and the difference was statistically significant (x2 =5.048,P =0.025).Four months after treatment,the levels of SCC [(2.67±0.76) ng/ml vs.(4.10±0.94) ng/ml],TPS [(56.85±6.39) U/L vs.(86.69±12.62) U/L],sMICA [(90.31 ±11.25) ng/L vs.(129.46±16.60)ng/L],HIF-1α [(55.47±7.04) ng/Lvs.(68.07±7.97) ng/L] and Her-2 [(44.64±4.96) U/L vs.(53.07±6.56) U/L] in the experimental group were lower than those in the control group,with statistically significant differences (t =7.667,P < 0.001;t =13.671,P <0.001;t =12.653,P<0.001;t =7.679,P<0.001;t =6.643,P<0.001).The occurrence rates of bone marrow suppression of the experimental group and the control group were 23.81% (10/42) and 21.43% (9/42),the occurrence rates of gastrointestinal reaction were 19.05% (8/42) and 14.29% (6/42),the occurrence rates of abnormal electrocardiogram were 4.76% (2/42) and 2.38% (1/42),and the tatal adverse reaction rates were 47.62% (20/42) and 38.10% (16/42).The difference of the total adverse reaction rates between the experimental group and the control group was not statistically significant (x2 =0.778,P =0.378).Conclusion The clinical efficacy of sodium glycididazole injection combined with concurrent chemoradiotherapy is definite for primary advanced cervical cancer.This treatment regimen can reduce serum levels of SCC,TPS,sMICA,HIF-1α and Her-2,and not increase the incidence of adverse reactions.

ABSTRACT

Objective To assess the efficacy and safety of radiotherapy combined with sodium glyci-didazole (CMNa)in treating nasopharyngeal carcinoma by conducting a Meta-analysis.Methods Randomized clinical controlled studies (RCTs)of radiotherapy plus CMNa versus radiotherapy alone in the treatment of nasopharyngeal carcinoma were retrieved from China Biology Medicine disc,China National Knowledge Infrastruc-ture,digital journal of Wanfang Data,China Science and Technology Journal Database,PubMed and Cochrane Library.According to the inclusion and exclusion criteria,data extraction and quality assessment were done by two researchers independently,and clinically important measures were selected.Outcomes were pooled with Rev-Man 5.2.Results The rate of complete remission (CR)in the radiotherapy combined CMNa group was better than that in the radiotherapy alone group in 1 .41 multiple to primary lesion,RR =1 .41 ,95%CI:1 .30-1 .52, Z =8.20,P =0.00.To lymphonodi cervicales metastasis,the rate of CR in the radiotherapy combined CMNa group was better than that in the radiotherapy alone group in 1 .45 multiple,RR =1 .45,95%CI:1 .30-1 .62, Z =6.50,P =0.00.There were no significant differences in the incidence rates of adverse reactions of skin (RR =0.97,95%CI:0.87-1 .08,Z =0.57,P =0.57 ),mucosa (RR =1 .03,95%CI:0.93-1 .15,Z =0.65, P =0.52 )and decrease in white blood cells (RR =0.84,95% CI:0.56-1 .25,Z =0.87,P =0.38). Conclusion In the treatments of nasopharyngeal carcinoma,the rates of CR of primary lesion and lymphonodi cervicales metastasis in radiotherapy combined with CMNa group are superior to the radiotherapy alone group. However,the occurrence rates of adverse reactions in the two groups have no obvious differences.

ABSTRACT

Objective To study the radioseusitization effect of gold nanoparticles modified by sodium glycididazole.Methods The sodium glycididazole was connected to gold nanoparticle,in dimension of about 18 nm,that had been modified with polyethylene glycol.The nanoparticle-swallowing efficiency of lung adenocarcinoma A549 cells was observed by a scanning electron microscope.Cells were divided into four groups:sodium glycididazole group,gold nanoparticles group,sodium glycididazole-gold nanoparticles group,and no drug control group.The radiosensitivity was detected by MTT and colony formation assays.Results Sodium glycididazole-gold nanoparticles could enter the cell cytoplasm and nucleus.The concentration of 0.003 mg/ml gold nanoparticles and sodium glycididazole-gold nanoparticles had no obvious cytotoxic effect.After irradiation of 2,4,6,8 Gy,the cell survival of the sodium glycididazole-gold nanoparticle group was significantly lower than that of the other three groups (F =4.8,14.5,5.7,7.6,P <0.05) and the D0 and Dq values of the sodium glycididazole-gold nanoparticle group were significantly lower than those of other three groups.Conclusion Sodium glycididazole-gold nanoparticles can enhance the radiosensitivity of lung adenocarcinoma cells.

ABSTRACT

ObjectiveTo evaluate the efficacy and clinical safety of sodium glycididazole (CMNa)in thoracic esophageal squamous carcinoma.Methods From June 1,2008 to October 13,2009,66pathologically proved thoracic esophageal squamous carcinoma (stage Ⅱa-Ⅲ,stage Ⅳ with metastases only in supraclavicular lymph nodes,by AJCC 6th ed) were randomized into radiotherapy plus CMNa (A) or radiotherapy plus placebo (B) group.Radiotherapy was given by conventional schedule:1.8-2.0 Gy per fraction,5 times per week to a total dose of 66 Gy/6.6-7.2w.CMNa was given intravenously 800 mg/m2 3 times a week in solution of 100 ml saline within 30 minutes.Radiotherapy was started 30-60 minutes after completion of infusion.Patients of Group B received placebo in saline solution.A total of 66 patients were enrolled ( Group A:32 ; Group B:34 ),and four patients were unanalyzable,remaining 31 patients in each Group.Baseline factors were balanced.ResultsFollow-up rate was 97%.Group A vs.Group B:the overall response rate was 93.5% vs.67.7% ( x2 =6.61,P =0.01 ),2-year overall survival was 39.9% vs.29.9% ( x2 =0.62,P =0.433 ),2-year cancer specific survival was 43.1% vs.26.8% ( x2 =0.30,P =0.878),and 2-year progression-free survival was 30.1% vs.27.9% ( x2 =0.02,P =0.586).No severe side effects observed.All patients tolerated CMNa infusion well.Conclusions CMNa is tolerable and effective as a hypoxic radiosensitizer,and its combination with radiotherapy can improve short term effect.However,survival is not improved within our follow-up period.

ABSTRACT

OBJECTIVE:To evaluate the radiosensitization effect of docetaxel versus sodium glyci-didazole.METHODS:A total of 105 patients with intermediate or advanced stage nasopharyngeal carcinoma were randomly assigned to receive docetaxel 40 mg once a week plus radiotherapy or sodium glycididazole 1 000 mg three times a week(Monday,Wednesday and Friday) plus radiotherapy for a total of 7 weeks,or radiotherapy only(control group).RESULTS:There were significant differences between docetaxel group or sodium glycididazole group and control group in sensitization efficiency and radiation dose on tumor regression(P0.05).CON-CLUSION:Lower dose docetaxel is better than sodium glycididazole in radiosensitization efficiency for advanced nasopharyngeal carcinoma,especially in controlling the metastasis,yet it has minimal side effect.
