ABSTRACT

The similarity between plant and microbial cells encourage the use of microbial metabolites of halophilic bacteria for the alleviation of salt stress in plants. In the current research work, a compatible solute ectoine extracted from a moderately halophilic bacteria Chromohalobacter salexigens KT989776 was used to enhance flax germination and primary seedling under different levels of salinity. Two successive experiments including germination in Petri plates under six levels of salinity (0, 3, 5, 7, 9 and 11 dS.m-1) and a pot experiment under three irrigating water salinity levels (2, 3 and 4) with two types of ectoine application (spray and soil addition) were conducted. Germination parameters were recorded for the first experiment while a fresh and dry weight of plants and peroxidase activity in addition to sodium-potassium ratio were estimated in the pot experiment. Also, ectoine accumulation in plants was detected using HPLC. Results of LC-MS proved the production of ectoine by C. salexigens KT989776 and ectoine enhanced significantly all germination parameters of flax seeds, decreased sodium accumulation in the plant, increased potassium content, and lowered peroxidase and phenoloxidase activity. Also, HPLC analysis proved that ectoine was detected in all treated samples while not detected in non-treated control.

ABSTRACT

Objective To compare the rates of clearance of different solutes during continuous veno-venous haemodiafiltration (CVVHDF) between pre-dilution and post-dilution.Methods A study in vitro was carried out using model CRRT system with AN69 filter used,which was applied to perform CVVHDF for solutes clearance.The removed amounts of different solutes including potassium ion (K +),creatinine (Cr),vancomycin,insulin,and interleukin-6 (IL-6) were determined in the groups of control (without dilution),pre-dilution and post-dilution during CVVHDF at the same substitution fluid amount.Each group was repeated 4 times (n =4).Results Post-dilution mode increased K +,Cr,vancomycin and insulin clearances significantly.There was no difference in clearance of IL-6 between the pre-and post-dilution groups.In the control group,insulin and IL-6 levels were decreased extremely.Conclusions In general,the rate of clearance using post-dilution of CVVHDF is higher than that using pre-dilution.Among high molecular weight solutes,the difference in clearance is not significant.The control group demonstrates insulin and IL-6 adsorbed by the filter.

ABSTRACT

Objective To study the effect of continuous venovenous haemodiafiltration (CVVHDF) on the clearances of various solutes in multiple organ dysfunction syndrome (MODS)dogs.Methods Dogs were subjected to hemorrhagic shock plus resuscitation and endotoxemia to set up MODS model,then they were randomly divided into 2 groups: MODS group (M group)and MODS+CVVHDF group (M+C group).A PRISMA pre-dilution system with AN-69 filter was applied.Solute clearance of nitric oxide(NO),urea nitrogen(UN),creatinine (Cr),interleukin-6 (IL-6),interleukin-10 (IL-10),tumor necrosis factor-α (TNF-α) and endotoxin (LP) was determined with same dialysate and ultrafiltrate volume during CVVHDF.The clearance of various solutes (Kd) was calculated by the concentration of the cleared solutes in effluents (E) based on the formula Kd=(ExQE)/P.Results Clearances of NO,UN,Cr were greater than those of IL-6,IL-10,TNF-α,LP during the treatment of CVVHDF.Conclusion CVVHDF could effectively reduce the levels of various solutes in the development of MODS by convection and absorption.
