ABSTRACT

Objective To investigate the expression and significance of Ezrin and ICAM-1 in gastric carci- noma. Methods The expression of Ezrin and ICAM-1 were examined by S-P immunohistochemistry assays in 78 ca- ses of gastric carcinoma.30 cases of nonmalignant tissue and 30 cases of normal stomach. The relationship between them and their pathological characteristics were analyzed. Results ①The expression rates of Ezrin and ICAM-1 were obviously higher in carcinoma group than that of nonmalignant group and normal group(P<0.01),in the lym- phatic metastasis group than that in non lymphatic metastasis group(P<0.05),and in T3.T4 group and than that in T1,T2 group(P<0.05).②The expression level of ICAM-1 was positively related with that of Ezrin(r=0.46. P=0.00).③The test of the expression of Ezrin and ICAM-1 simutaniously could obviously increase the sensitivity and specificy for detecting the metastasis of stomach malignant tumor(P<0.05).Condusion The over expression of Ezrin and ICAM-1 is related to the pathogenesis and invasion as well as metastasis of stomach carcinoma. The combined detection of two markers can obviously increase the sensitivity and specificy for predicting metastasis of stomach carcinoma.

ABSTRACT

Objective To report the surgical treatment results of cardiac and esophageal cancer of remnant stomach. Methods From 1980 to 2002, 30 patients with cardiac and esophageal cancer of remnant stomach were treated surgically. Results Primary cardiac and esophageal carcinoma of remnant stomach developed in male patients more than females. Of 30 patients, 21(70%) underwent subtotal gastrectomy with Billroth II reconstruction at first time. The mean interval between two operations was 13.5 years. Clinical symptoms frequently included abdominal distension, pain and hemorrhage. Thirty patients received radical resection. Conclusion Cancer in remnant stomach should be suspected in patients undergoing subtotal gastrectomy 5 years and over have clinical symptoms of upper gastrointestinal tract. Early diagnose and early curative resection should be done. For cardiac cancer of remnant stomach, total gastrectomy with “P” loops and Roux-en-Y esophagojejunostomy reconstruction were recommended. This procedure have advantage in retaining enough food storage preventing reflux esophagitis and allowing resection of esophagus long enough to avoid cancer remnant. For esophageal cancer of remnant stomach, colon reconstruction after the resection of esophageal cancer is recommended.

ABSTRACT

PURPOSE: Recent studies have shown that the expression of mucin is organ- and cell-type specific and it is increasingly possible that its change could result from oncogene activation. To evaluate histogenesis and prognostic factors for gastric carcinoma, we studied the oncoprotein expression in gastric cancer cells classified by mucin phenotye. MATERIALS AND METHODS: Mucin histochemistry and immunohistochemistry for ras, c-erbB2, and p53 oncoprotein expression were performed in 101 surgically resected gastric carcinoma specimens. PAS-Con A, GOS, and HID-AB staining techniques were employed in identifying mucosubstances, RESULTS: Of the 101 specimens studied, 73(72.3%) revealed as having mixture of various mucin-secreting cancer cells. Overall, ras immunoreactivity was observed in 72(71.3%), c-erbB2 in 7(6.9%), and p53 in 47(46.5%). Of the 73 mucus-secreting carcinomas, the surface mucous cell type were shown in 65 (89.0%), the pyloric gland cell type in 48(65,8%), the sialomucin type in 47(64.4%), and the sulfomucin type in 54(74.0%). There was significant association between mucin secretion and ras expression, but not c-erbB2 and p53 expression. There was no significant association between mucin secreting cell types and Lauren classification. Ras expression was correlated with serosal invasion, lymph node metastasis and poor prognosis. CONCLUSION: The phenotypic expression by mucin histochemistry may be not more important for studying of histogenesis in gastric carcinoma than Lauren classification. Ras expression is a poor prognostic indicator and may be correlated with phenotypic expression of surface mucous cell and intestinal cell type in gastric carcinoma.

ABSTRACT

We report a case of disseminated cutaneous metastatic carcinoma from stomach carcinoma in a 66-year-old male who complained chiefly of widespread cutaneous nodules. He had been admitted to the Department of Internal Medicine for stomach carcinoma. A radiological examination revealed a diffuse stomach wall thickening from the fundus to the antrum, and enlargement of paraaortic and pericaval lymph nodes during an abodominal CT scan. The clinical findings manifested above may suggest cutaneous metastasis from stomach carcinoma. A histological examination of the skin and stomach biopsies showed atypical cells with pleomorphic, hyperchromatic nuclei and bubble-like cytoplasm, which are consistent with poorly differentiated adenocarcinoma. Immunohistochemical stainings showed that these cells were positive for epithelial membrane antigen (EMA), carcinoembryonic antigen (CEA) and low molecular weight cytokeratin.

ABSTRACT

This study was carried out to investigate the effects of ATP on cultured human stomach carcinoma MGC-803 cells. After ATP(0.16—0.23 mg/ml, 24—120 hrs)treatment, inhibition of cell growth rate was observed. The inhibition rate reached 80% after 96 hr exposure to ATP. At the same time, anti-tubulin immunofluorescent staining showed increase of organized microtubules (MT) in ATP-treated MGC-803 cells, in contrast with the poor MT immunofluorescence in control MGC-803 cells. By using rhodamie phalloidin combined staining, increased number of actin bundles, took the place of actin dots and patches which were seen in control MGC-803 cells. Immunofluorescent staining of fibronectin in ATP-treated MGC-803 cells was also increased evidently as compared with the control MGC-803 cells. Concomitant with the improvement of MT and stress fiber organization, the cell Concohas become more flattened. It appeared that the increase of MT and stress fibers was closely related to the recovery of cell morphology in ATP treated tumor cells. It is therefore concluded that ATP not only may inhibit tumor cell proliferation, but also may induce differentiation of cell morphology of tumor cells through its action on cytoskeletal structures.
