ABSTRACT

@#Objective To develop and verify a sulfosalicylic acid spectrophotometric method for the determination of trace iron ions in diphtheria toxin medium,and apply it preliminarily.Methods The maximum absorbance of the complex of iron and sulfosalicylic acid was scanned by full spectrum;A method for the determination of iron ion in culture medium was developed by linear regression between the absorbance of the complex and the content of iron ion,and the stability,accuracy and precision of the method were verified.The effects of Ca~(2+),Mg~(2+),K~+,Na~+ and reactants on the method were investigated.Spectrophotometric method with sulfosalicylic acid was used to determine trace iron in the self-made and commercial medium for diphtheria toxin production.Sulfosalicylic acid spectrophotometry,ferrizine colorimetry and o-phenanthroline spectrophotometry were used to detect iron content in two kinds of culture media(beef trypsin digestion liquid and 5% polypeptone),and the detection results of the three methods were compared.Results The complex of iron and sulfosalicylic acid showed the maximum absorbance at the wavelength of 425 nm;There was a good linear relationship between the absorbance and concentration of iron ion in the range of 1~0.05 μg/mL,the detection limit was 0.05 μg/mL,and the standard equation was:Y=0.027 9 X+0.046 1,R~2> 0.99;The coefficients of variation(CVs) of A_(425) value of each concentration of standards measured every 5 min were less than 5%;Low(0.05 μg/mL),medium(0.5 μg/mL)and high concentration(1 μg/mL) of Fe~(3+) standard solutions were continuously determined for 3 times.The CVs of 9groups of each concentration measured in parallel were all less than 5% and the recovery rates were higher than 95%;Ca~(2+),Mg~(2+),K~+,Na~+,15 μL of sulfo salicylic acid(20%) and 50 μL of ammonia hydroxide(1:1) showed no interference in the method;The results of toxin-producing medium were consistent with those of diphtheria bacteria;The results of the three detection methods were consistent.Conclusion The developed spectrophotometric method with sulfo salicylic acid can determine the content of trace iron ions in diphtheria toxin medium accurately and effectively.

ABSTRACT

Objective: To investigate the feasibility of four methods for macromolecule protein determination in Chinese materia medica (CMM) injection. Methods: The sensitivity, specificity, and protein test results of sulfosalicylic acid, Coomassie brilliant blue, Lowry, and size exclusion chromatograph (SEC) methods on five kinds of CMM injections were compared. Results: Sulfosalicylic acid method had low sensitivity, Coomassie brilliant blue and Lowry methods had poor specificities, and SEC method had high sensitivity and good specificity. No protein was detected in five kinds of CMM injection by SEC. Conclusion: SEC is suitable for the determinatoin of macromolecule protein in CMM injection.

ABSTRACT

A study was carried out on growth, photosynthetic pigments, and yield attributing and seed characteristics of Arachis hypogaea L. (Cv. SB-11). The treatments comprised of different concentrations (5, 50, 100 and 200 ppm) of Sulfosalicylic acid (SSA). The results showed that 50 ppm concentration of foliar applied SSA effectively promoted the growth parameters, photosynthetic pigments (chl a, chl b, total chl, carotenoids and chlorophyll stability index), yield parameters and seed protein and seed oil contents of groundnut. Similarly significant decrease in severity of tikka disease was reported with 50 ppm SSA. SSA 100 ppm profound better influence on the number of branches, height of plant and the shelling percentage. It could be concluded that SSA proved to be beneficial in promoting plant growth and yield and in inducing systemic resistance against tikka disease in groundnut.

ABSTRACT

Objective: To determine the reliability and accuracy of the sulfosalicylic acid test in the fast detection and semi-quantitative analysis of proteinuria. Methods: Prospective, descriptive study at Hospital Universitario San Vicente de Paúl in Medellín, Colombia. Ninety eight women with proteinuria and 129 without it were included. Turbidity test with sulfosalicylic acid was compared to 24-hour-proteinuria. Results: The likelihood ratios (LR) for turbidity test results of 1+, 2+, 3+, 4+, and 5+ were 0.4, 0.7, 3.4, 6.7, and 39.1, respectively. Interclass correlation between the three evaluators of turbidity was 0.966. With the cut point set at 4+, sensitivity was 41.1% (CI 95%: 30.6-51.5), and specificity was 97.7% (CI 95%: 94.7-100). In hypertensive patients, positive predictive value was 95% (CI 95%: 87-100) and negative predictive value was 53.3% (CI 95%: 42.5-64). Conclusion: Turbidity test is an easy, fast, cheap, and reproducible analysis. Because of its high specificity it is ideal to study pregnant women, particularly those with hypertension, for proteinuria.


Objetivo: determinar la confiabilidad de la prueba del ácido sulfosalicílico en la detección rápida y semicuantificación de la proteinuria. Métodos: estudio descriptivo, prospectivo llevado a cabo en el Hospital Universitario San Vicente de Paúl, Medellín, Colombia. Se incluyeron 98 mujeres gestantes con proteinuria y 129 sin proteinuria. La prueba de turbidez con ácido sulfosalicílico se comparó con la medición de proteinuria en 24 horas. Resultados: los coeficientes de probabilidad (likelihood ratio) para los resultados de la prueba fueron los siguientes: 1+ = 0,4; 2+: = 0,7; 3+ = 3,4; 4+ = 6,7 y 5+ = 39,1. La correlación entre los tres evaluadores de la turbidez fue 0,966. Con un punto de corte de 4+ la sensibilidad fue 41,1% (IC 95%: 30,6- 51,5), la especificidad fue 97,7% (IC 95%: 94,7-100). Cuando la prueba se aplicó solamente a las pacientes hipertensas el valor predictivo positivo fue 95% (IC 95%: 87-100) y el valor predictivo negativo fue 53,3% (IC 95%: 42,5-64). Conclusiones: la prueba de turbidez con ácido sulfosalicílico es fácil, rápida, económica y reproducible. Por su baja sensibilidad no es útil como método de tamización pero por su alta especificidad es ideal para la confirmación rápida de proteinuria en las gestantes hipertensas.
