ABSTRACT

B chromosomes are extra chromosomes from the normal chromosomal set, found in different organisms, highlighting their presence on the group of fishes. Callichthys callichthys from the upper Paraná River has a diploid number of 56 chromosomes (26 m-sm + 30 st-a) for both sexes, with the presence of a sporadically acrocentric B chromosome. Moreover, one individual presented a diploid number of 57 chromosomes, with the presence of a morphologically ill-defined acrocentric B chromosome in all analyzed cells. The physical mapping of 5S and 18S rDNA shows multiple 5S rDNA sites and only one pair of chromosomes with 18S sites in C. callichthys, except for two individuals. These two individuals presented a third chromosome bearing NORs (Ag-staining and 18S rDNA) where 5S and 18S rDNA genes are syntenic, differing only in position. The dispersion of the 18S rDNA genes from the main st-a chromosome pair 25 to one of the chromosomes from the m-sm pair 4 would have originated two variant individuals, one of which with the ill-defined acrocentric B chromosome. Cromossomos B são cromossomos extras ao conjunto cromossômico normal, encontrado em diferentes organismos, com destaque para sua presença no grupo de peixes. Callichthys callichthys do alto rio Paraná tem um número diploide de 56 cromossomos (26 m-sm + 30 st-a) para ambos os sexos, com a presença esporádica de um cromossomo B acrocêntrico. Além do mais, um indivíduo apresentou número diploide de 57 cromossomos, com a presença de um cromossomo B acrocêntrico morfologicamente mal definido em todas as células analisadas. O mapeamento físico do DNAr 5S e 18S mostrou múltiplos sítios de DNAr 5S e apenas um par de cromossomos com sítio para o DNAr 18S em C. callichthys, com exceção para dois indivíduos. Estes dois indivíduos apresentaram um terceiro cromossomo portador das RONs (Ag-RONs e 18S rDNA), onde os genes DNAr 5S e 18S são sintênicos, diferindo apenas na posição. A dispersão dos genes DNAr 18S do par de cromossomos principal st-a 25 para um dos cromossomos do par m-sm 4 teria originado dois indivíduos variantes, um dos quais com cromossomo B acrocêntrico mal definido. ABSTRACT

B chromosomes are supernumerary elements present in about 15 percent of eukaryotic species and are most frequently heterochromatic, behave parasitically, show a transmission rate higher than standard (A) chromosomes, and can provoke harmful effects on carriers. In the current work, Prochilodus lineatus individuals carrying eight and nine B chromosomes were obtained by induced crossing performed involving breeders with different B chromosome numbers in their cells. The high B chromosome numbers found in the offspring were recorded for the first time in this species. The use of cytogenetic techniques applied in the present study revealed that regardless of the increase in number of B chromosomes in the genome of these individuals, those elements did not presented active genes, and showed their normal heterochromatic characteristic.

ABSTRACT

Cytogenetic analyses of the location of 18S and 5S ribosomal DNAs, and the base composition of B chromosomes of Iheringichthys labrosus from Tibagi River, Paraná, Brazil, are provided. AgNORs were observed in the terminal position on the long arm of a subtelocentric chromosome pair. CMA3-positive staining was observed in some chromosomes, which besides being associated with NORs, were all DAPI-negative. Chromosome B showed a strong fluorescence with CMA3. The concomitant use of 18S and 5S rDNA probes using the FISH technique revealed 18S ribosomal cistrons in a pair of subtelocentric chromosomes, on the long arm in the terminal position, coinciding with the AgNOR. The 5S sites were found in another subtelocentric pair, on the long arm in the interstitial region, near the centromere. The findings of the present study suggest that, although there are some more conserved cytogenetic characteristics, populations of I. labrosus may show their own characteristics.


Foram realizadas análises citogenéticas em Iheringichthys labrosus do Rio Tibagi, Paraná, Brasil com a localização cromossômica dos DNAs ribossômicos 18S e 5S e a composição de bases de seus cromossomos B. As AgNORs foram observadas em posição terminal, no braço longo de um par de cromossomos subtelocêntricos. Marcações CMA3 positivas foram observadas em alguns cromossomos e associadas com as RONs. Porém, todas estas marcações apresentaram-se DAPI negativas. O cromossomo B mostrou-se fortemente fluorescente com CMA3. O uso concomitante das sondas de DNAr 18S e 5S, através da técnica de FISH, revelou os cístrons ribossômicos em um par de cromossomos subtelocêntricos, em posição terminal do braço longo, coincidindo com a AgNOR. Os sítios 5S foram observados em outro par subtelocêntrico, em posição intersticial do braço longo, próximo ao centrômero. Os resultados observados no presente estudo sugerem que, embora existam algumas características citogenéticas mais conservadas, as populações de I. labrosus podem mostrar suas próprias características.

ABSTRACT

A cytogenetic study was conducted on an Astyanax eigenmanniorum population from the Caetano Stream (18º 44' 56" S/ 048º 18' 39" W) - in Uberlândia, MG, Brazil - showing a modal diploid number of 48 chromosomes in the standard male and female karyotypes. However, in several specimens it was also possible to observe metaphases with one or two B chromosomes, increasing the diploid number to 49 or 50 chromosomes, respectively. The supernumerary chromosomes were totally heterochromatic and highlighted after C-banding. The silver-stained nucleolus organizing regions (Ag-NORs) were located in at least five chromosomes of the standard karyotype, thus characterizing a multiple NOR system in the species. This is the first occurrence of an A. eigenmanniorum population with 2n = 48 chromosomes, bearing supernumerary chromosomes.
