ABSTRACT

Objective@#To investigate the level and timeliness of CD4+T lymphocyte cell (CD4 cell) counts at first measurement among newly reported HIV/AIDS cases in Taizhou City, Zhejiang Province from 2012 to 2021, so as to provide insights into improving the management of HIV/AIDS cases. @*Methods@#Demographic data and first measurement of CD4 cell counts of newly reported HIV/AIDS cases in Taizhou City from 2012 to 2021 were collected from the HIV/AIDS Comprehensive Control System of Chinese Disease Prevention and Control Information System. The level and time of CD4 cell counts at first measurement were descriptively analyzed, and factors affecting CD4 cell counts at first measurement were identified using a multivariable logistic regression model. @*Results@#A total of 4 834 newly reported HIV/AIDS cases were recorded in Taizhou City from 2012 to 2021, including 3 889 men (80.45%), 2 005 cases at ages of 20 to 39 years (41.48%), and 2 130 farmers (44.06%). There were 1 664 cases (34.42%) with CD4 cell counts of <200/mm3 at first detection, 2 576 (53.29%) with CD4 cell counts of 200/mm3 to 499/mm3, and 594 (12.29%) with CD4 cell counts of ≥500/mm3. The proportion of CD4 cell counts of <200/mm3 showed a tendency towards a rise from 2012 to 2021 (χ2trend =4.250, P<0.001). There were 3 465 cases (71.68%) that had an interval of ≤14 days between the first detection of CD4 cell counts and confirmatory HIV test, 740 (15.31%) that had an interval of >14 to 30 days, 315 (6.52%) that had an interval of >30 to 90 days, 135 (2.79%) that had an interval of >90 to 180 days, and 179 (3.70%) that had an interval of >180 days. The proportion of an interval of ≤14 days appeared a tendency towards a rise from 2012 to 2021 (χ2trend=6.874, P<0.001). Multivariable logistic regression analysis identified women (OR=0.630, 95%CI: 0.529-0.751), age of ≥20 years (OR: 1.958 to 3.218, 95%CI: 1.216-5.412), other or unknown routes of HIV infection (OR=1.785, 95%CI: 1.100-2.896), and identification by medical institutions (OR=1.779, 95%CI: 1.497-2.114) as factors affecting CD4 cell counts of <200/mm3 at first measurement. @*Conclusions@#The timely detection of CD4 cell counts at first measurement gradually increased with year among newly reported HIV/AIDS cases in Taizhou City from 2012 to 2021; however, there were still 34.42% of cases with CD4 cell counts of <200/mm3. Gender, age, route of HIV infection, and sample source were factors affecting CD4 cell counts of <200/mm3 at first measurement.

ABSTRACT

@#[Abstract] Autophagy, as an important intracellular metabolic pathway, has been proved to be ubiquitous in many kinds of cells. Its functional impairment can easily cause lots of diseases, such as cancer, leukemia, liver disease, diabetes and heart disease. In particular, autophagy is important for the development, differentiation and regulation of immune function of T lymphocytes. Abnormal autophagy of T lymphocytes can cause immune dysfunction and lead to diseases, such as inflammation, infection and autoimmunity. In view of the important role of autophagy in regulating T lymphocyte function and disease, this article illustrates the research progress on autophagy regulating the homeostasis, survival, proliferation, senescence, metabolism, immune function of T lymphocytes and many diseases, including tumors, in recent years.

ABSTRACT

Autoimmune thyroid disease (AITD) is a complex organ-specific autoimmune disease, associated with many factors such as genetic susceptibility, environmental factors, immune system disorders, and intestinal microbiota imbalance, which seriously affect the quality of life. At present, oral medicine, iodine 131 treatment and surgical treatment are mainly adopted in clinical parctice of western medicine. Although they can exert a certain curative effect, they still have surgical risks and side effect. Modern research shows that, traditional Chinese medicine(TCM) has the characteristics of stable curative effect, multi-targeted regulation and less side effect. It has definite efficacy and unique advantages in the prevention and treatment of AITD. Helper T lymphocyte cell 17 (Th17) mediate inflammation to induce immune promotion, while regulatory T lymphocyte cell (Treg) mediate immunosuppression, Th17 and Treg cooperate to maintain the balance of the immune microenvironment. During AITD progress, Inflammatory Th17 can be high, and the levels of mediated immunosuppressive Treg are relatively decreased. The restoration of balance between the two plays a key role in the inflammatory and immune processes of AITD. In recent years, based on Th17/Treg cell axis, a large number of clinical and experimental studies on the intervention of TCM on Th17/Treg balance in AITD have been carried out in the field of TCM, and some results have been achieved. Studies have shown that intervention in the Th17/Treg signaling axis is an important mechanism for the treatment of autoimmune thyroid diseases. This paper summarizes and analyzes the previous studies on the intervention effect of Chinese medicine monomer, Chinese medicine composition and Chinese medicine compound on Th17/Treg cell axis in AITD, mainly from the aspects of intervention related inflammatory factor secretion, regulation of antibody titer and the expression of related genes of related genes. These studies will help people to understand the mechanism of TCM in interfering with the Th17/Treg balance in AITD more accurately and comprehensively, and provide references for the rational application of TCM in the prevention and treatment of autoimmune thyroid diseases in clinical practice.

ABSTRACT

Objective:To investigate the effects of TLR4 knockout on immune cells and adipokines in mouse visceral adipose tissue.Methods: Cells were isolated from the spleen and epididymal adipose tissue of 20-week-old male wild type mice C57BL/6 and TLR4-/-.The expression of F4/80,CD11b,CD11c,CD3,CD4 and CD8 in these cells were analyzed by flow cytometry.The expression of IL-6,HMGB1,TNF-α,adiponectin and resistin in epididymal adipose tissue were detected by qPCR.Results: Compared with wild type C57BL/6 mice,the percentage of M1 macrophage which marked F4/80+CD11b+CD11c+ in spleen and epididymal adipose tissue of TLR4-/-mice increased(P<0.05) greatly,while that of M2 macrophage which marked F4/80+CD11b+CD11c-was decreased(P<0.05)significantly.This trend was more remarkable in epididymal adipose tissue than in spleen(P<0.05).In epididymal adipose tissue,the percentage of CD4+T cells decreased but that of CD8+T cells increased in TLR4-/-mice.Moreover,the high-level expressions of IL-6,HMGB1,resistin were found in epididymal adipose tissue of TLR4-/-mice.However,the expressions of TNF-α and adiponectin decreased obviously.Conclusion: TLR4 knock-out could lead to a disorder in adipokine and immune cells in visceral adipose tissue.

ABSTRACT

Objective To study the effects of B cells from lupus prone Triple congenic (TC) mouse model on the differentiation and development of T cell subsets. Methods The spleen size and B cell numbers were measured, and surface CD40, CD86 and Ⅰ-Ab molecules on B cells as well as CD4+T cell subsets were detected using flow cytometry when the spontaneous systemic lupus erythematosus (SLE) model TC mice and control B6 mice were 6 months old. In addition, the chimera of TC B cells and B6 CD4+T cells or chimera of B6 B cells and B6 CD4+ T cells were transferred into B6.Rag-/- mice via intravenous injection. Then, T cell subsets in the spleen of recipient B6.Rag-/-mice were observed 7 days after cell transplantation. Results TC mice had significantly bigger spleen [(5337±934) mg] and more CD19+B cell number [(276.0±48.7)×107] than control B6 mice [spleen weight: (91±4) mg; B cell number: (6.4±0.3)×107](P0.05). The recipient B6.Rag-/-mice transplanted with TC B cells had significantly more Th1 subset [(54.1±2.8)%] and IL-21+CD4+T cell population [(14.3±1.0)%], but less Th17 subset [(2.05±0.09)%] in spleen than the recipient B6.Rag-/-mice administered by B6 B cells [Th1 subset: (39.5±1.1)%; IL-21+CD4+T cell population:(7.5±1.2)%;Th17 subset:(6.45±1.10)%](P<0.01). Conclusion The B cells of lupus-prone TC mice exhibit a markedly hyper-activation in spleen, and promote CD4+T cells differentiation preferentially into Th1 subset and IL-21+CD4+T cell population, which may further contribute to SLE pathogenesis.

ABSTRACT

Objective To explore the proportional changes of regulatory B lymphocyte cells (Bregs) and suppressor T lymphocyte cell (Ts) in the peripheral blood of patients with Hashimotos' thyroiditis (HT).Method Sixty-one cases of HT and 38 healthy subjects with matched age and gender were recruited.Flow cytometry technology was used to detect the proportional changes of CD19+ CD24+ CD27 + B cells,CD19+ CD24hi CD38hi B cells,CD19+ IL-10+B cells,and CD8+CD28-T cells.Result Compared with control group,CD19+CD24+CD27+B cells in HT group increased significantly(P<0.05).With the increase of thyroid peroxidase antibody,CD19+ CD24hi CD38hi B cells were decreased significantly.With the increase of antithyrogloblin antibody (TGAb),CD8 + CD28-T cells were decreased markedly (P<0.05).TT3 was positively related with CD19 + CD24hi CD38 hiB cells while TT4 had a negative correlation with CD8+ CD28-T cells and positive correlation with CD19+ CD24+ CD27+ B cells.Logistic regression analysis showed that TGAb and FT4 were independent risk factors for decreased CD8+ CD28-T cells in lymphocyte percentage(P<0.05).TSH was an independent risk factor for the decreased proportion of CD19+ CD24hi CD38hiB cell.FT4 was the independent risk factors for increased CD19+CD24+CD27+B cells(P<0.05).Conclusion The percentage of Ts cells in patients with HT was lowered,while the different phenotypic Bregs cells showed various changes.Thyroid autoantibodies and thyroid function were closely associated with the immunological changes in HT patients.
