ABSTRACT

Objective:To explore the prescription and medication law of Traditional Chinese Medicine (TCM) compounds in the treatment of vascular dementia (VD) based on patent database.Methods:TCM compounds with patents about VD were retrieved from Chinese patent announcement website of the State Intellectual Property Office and CNKI. The retrieval time was from the establishment to the databases to 31 st, March 2022. The frequency, clusteringand association analysis were carried out with the help of TCM inheritance auxiliary platform (V2.5). The medication law was analyzed. Results:154 TCM compound patents for the treatment of vascular dementia were screened, involving 227 kinds of Chinese materia medica. Among them, Acori Tatarinowii Rhizoma (44 times, 28.57%) was used more frequently, and the common medicinal pair was Salviea Miltiorrhizae Radix et Rhizoma- Acori Tatarinowii Rhizoma (17 times, 11.03%). The medicinal property was mainly warm, the taste was mainly sweet, and the meridian was mainly liver meridian. Those with high confidence based on association rules were " Corni Fructus -Acori Tatarinowii Rhizoma" (0.90), " Corni Fructus -Rehmannize Radix et Praeparata" (0.90). Based on the complex network, it was concluded that the core drugs were 14 groups such as " Rehmannize Radix et Praeparata- Cistanches Herba- Corni Fructus". The new prescriptions extracted by entropy cluster analysis included 7 groups such as " Rehmannize Radix et Praeparata, Cistanches Herba, Corni Fructus and Asparagi Radix". Conclusion:The treatment of VD by TCM compounds with national patents is mainly based on tonifying deficiency, promoting blood circulation and removing blood stasis, eliminating phlegm and dampness, expelling wind and dredging collaterals, opening orifices and resuscitation, which can provide reference for clinical practice and new drug research and development.

ABSTRACT

OBJECTIVE:To comprehensi vely evaluate the application progress of network pharmacology methods in TCM compound prescription research ,and to provide reference for modernization of TCM compound prescription research. METHODS : Taking“network pharmacology ”and“TCM compound prescription ”as keywords ,the literatures were retrieved from CNKI , Wanfang database and VIP during May 2006 to May 2020. Screening literature ,the databases ,analysis platforms ,and software used of the literature were summarized ;on the basis of quantitative analysis ,the application of network pharmacology in the research of traditional Chinese medicine compound were summarized. RESULTS & CONCLUSIONS :There were a total of 761 valid literatures ,among which the number of literatures that could be retrieved in 2019 reached 313. In the modernization research of TCM compound prescription ,network pharmacology methods were mainly usedmechanism ,material basis of pharmacodynamics , compatibility law ,compound optimization ,and“effect-toxic”network. Commonly used databasesand platforms included traditional chinese medicine information database (TCMSP and TCMID ),therapeutic target database (TTD,OMIM),drug targets and target prediction platform (Drugbank,SwissTargetPrediction,TargetNet,PharmMapper),network pharmacology analysis and prediction software and platform (CytoScape),etc. Network pharmacology method was widely used in the field of TCM compound prescription research ,and provided new ideas and methods for the modernization of TCM compound prescription research. In the future,the related research can be combined with the pharmacokinetic parameters ,the efficacy of active compounds and related basic experiments ,use the weighted method to carry out network pharmacology analysis ,and integrate the information of multiple databases to improve the scientificity of research results.

ABSTRACT

OBJECTIVE: To systematically evaluate the efficacy and safety of TCM compound preparation for tonifying kidney and activating blood circulation, and to provide evidence-based reference for rational drug use in the clinic. METHODS: By retrieving Cochrane library, PubMed, Embase, CBM, CNKI, VIP and Wanfang database, randomized controlled trials (RCTs) about TCM compound preparation for tonifying kidney and activating blood circulation (trial group) versus calcium or non-calcium agents (control group) in the treatment of postmenopausal osteoporosis were included. After literature screening, data extraction and quality evaluation with bias risk evaluation tool and Jadad scale of Cochrane system evaluator manual 5.1.0, Meta-analysis was conducted by using Stata 12.0 software, and trial sequential analysis (TSA) was conducted by using TSA 0.9 software. RESULTS: Totally 18 RCTs were included, involving 1 408 patients. The results of Meta-analysis showed that total response rate [RR=1.35,95%CI(1.17,1.54),P<0.000 1] and bone density[SMD=0.24,95%CI(0.16,0.32),P<0.000 1] of trial group were significantly higher than those of control group; blood calcium [SMD=-0.05,95%CI(-0.09,0.00), P=0.033] of trial group was significantly lower than that of control group. There was no statistical significance in the levels of urine creatinine [SMD=-1.60,95%CI(-5.94,2.74),P=0.470], urinary calcium/urine creatinine ratio [SMD=-0.05,95%CI(-0.14,0.04),P=0.295], urinary hydroxyproline/urine creatinine ratio [SMD=-0.16,95%CI(-1.04,0.72),P=0.726], ALT [SMD=0.51,95%CI(-3.26,4.28),P=0.790], AST [SMD=0.23,95%CI(-5.22,4.77),P=0.929], serum alkaline phosphatase [SMD=-0.22,95%CI(-0.68,0.25),P=0.361], serum phosphate [SMD=-0.02,95%CI(-0.11,0.07),P=0.639], urea nitrogen [SMD=-0.19,95%CI(-0.70,0.31),P=0.453], estradiol [SMD=0.62,95%CI(-0.28,1.52),P=0.177], IL-6 [SMD=-1.78,95%CI(-4.86,1.30),P=0.258] or VAS [SMD=0.55,95%CI(-1.03,2.13),P=0.496] between 2 groups. No server ADR was found in 2 groups. TSA showed that there were extract evidences for total response rate of TCM compound preparation in the treatment postmenopausal osteoporosis. CONCLUSIONS: TCM compound preparation for tonifying kidney and activating blood circulation shows significant therapeutic efficacy for postmenopausal osteoporosis, and can improve serum calcium and bone density with good safety.

ABSTRACT

Ulcerative colitis (UC) is a common chronic non-specific intestinal inflammation, which has a long course and is difficult to cure, and the incidence is increasing year by year. Helper T cells 17 (Th17) are one of immune-promoting cells, while regulatory sputum cells (Treg) are immunosuppressive cells, and Th17 cells and regulatory T cells together maintain the balance of the body's immune microenvironment. During progression of UC, the population of sputum helper cells 17 (Th17 cells) that cause inflammation generally increases, while the number of sputum regulatory T cells that inhibit Th17 cell activity decreases. Among them, Th17 cells mediate immune response, regulatory T cell-mediated immunosuppression, and the balance between the two plays a key role in the inflammation and immune process of ulcerative colitis. Although Western medicine treatment of UC has certain effects, however, The frequency and severity of side effects, inconvenient dose adjustments, and partial price excessions limit their clinical application.As a traditional medicine in China, traditional Chinese medicine(TCM) has multi-target, multi-link and multi-channel treatment characteristics, and has unique advantages and broad prospects in anti-ulcerative colitis. In recent years, the TCM field has taken Th17/Treg balance as the entry point, and carried out a large number of clinical and experimental studies on the intervention of TCM in Th17/Treg balance in UC, and achieved certain results. Clinical and experimental evidence clearly indicates intervention in Th17/Treg. It is an important mechanism of action of TCM in the treatment of ulcerative colitis. This paper mainly summarizes and analyzes the intervention effects of TCM monomer, component or active ingredient and TCM compound on Th17/Treg balance in UC, which is helpful for people to understand Chinese medicine intervention in UC more accurately and comprehensively. The mechanism of action of Th17/Treg balance provides a reference for the clinical design of a treatment plan for anti-ulcerative colitis.

ABSTRACT

The cure of tumors is a difficulty in the world, and both the quality of life and the survival rate of patients remain low. Therefore, it is very meaningful to find a drug target to inhibit the occurrence and development of tumors. In recent years, autophagy or self-phagocytosis has become a hotspot of medical research. It can remove damaged or excess organelles from cells, be survived from external environmental pressures, and affect the survival, metabolism, differentiation, aging and death of tumor cells. The biological behavioral process plays important roles in remodeling and maintaining the dynamic balance of cell survival, especially in close relations to tumor development. Autophagy is also a double-edged sword in effect on a single tumor cell and the entire tumor. When the autophagy of the tumor cells is abnormal, or the cells are unable to remove the damaged substances in time under the conditions of hypoxia and nutrient deficiency, autophagy is beneficial to the proliferation and survival of the tumor cells. Contrarily, moderate autophagy acts as an inhibitor of tumors and has an anti-tumor effect. Traditional Chinese medicine (TCM) has a long history of controlling tumors, with the advantages of low toxicity and multiple targets. Through overall and local therapies, it has a comprehensive therapeutic effect in cancer. With the deepening of tumor autophagy research, in addition to western medicine researches on tumor autophagy, there are also domestic and foreign researches on the autophagy in single herb and TCM compounds. The latest insights into the molecular mechanism of autophagy have led to the discovery of potential drug targets. At the same time, TCM researches have made some progress in tumor autophagy. The authors review the research progress of autophagy in TCM and the research progress of effect of TCM in regulating tumor autophagy, in the hopes to provide useful reference for effect of TCM in the treatment of autophagy.

ABSTRACT

To differentiate traditional Chinese medicines (TCM) derived from congeneric species in TCM compound preparations is usually challenging. The roots of(PG),(PQ) and(PN) are used as popular TCM. They contain similar triterpenoid saponins (ginsenosides) as the major bioactive constituents. Thus far, only a few chemical markers have been discovered to differentiate these three species. Herein we present a multiple marker detection approach to effectively differentiate the threespecies, and to identify them in compound preparations. Firstly, 85 batches of crude drug samples (including 32 PG, 30 PQ, and 23 PN) were analyzed by monitoring 40 major ginsenosides in the extracted ion chromatograms (EICs) using a validated LC-MS fingerprinting method. Secondly, the samples were clustered into different groups by pattern recognition chemometric approaches using PLS-DA and OPLS-DA models, and 17 diagnostic chemical markers were discovered. Aside from the previously known Rf and p-F, ginsenoside Rscould be a new marker to differentiate PG from PQ. Finally, the above multiple chemical markers were used to identify thespecies in 60 batches of TCM compound preparations.

ABSTRACT

[Objective] The better optimization of traditional Chinese medicine for the treatment of respiratory syncytial virus, in the past five years is re-viewed in this paper, also the related domestic experimental research results, and according to the pathogenesis of RSV, it further summarizes the current domestic research for the main direction of the disease. [Methods] The article from the two aspects of compound Chinese medicine and traditional Chinese medicine monomer, according to the pathogenesis of respiratory syncytial virus section, discusses the experiment in vivo and in vitro related rich research results. [Results] It is found by inducing and summarizing, whether individual or compound traditional Chinese medicine, for respiratory syncytial virus re-search has made great achievement, but with experimental study limited to suppress the virus more value-added; replication and inhibiting inflammatory infiltrates the two big aspects, lacking of deeper side of molecular biology research, making the traditional Chinese medicine in the treatment of respiratory syncytial virus research appear too single, not a broader recognition by the industry. [Conclusion] We need to be sure the safety of traditional Chinese medicine for the treatment of respiratory syncytial virus and curative effect, but at the same time must recognize that because of the complexity of its com-position and design of experiment of traditional Chinese medicine, the irrationality of the existence of many factors, it makes progress with many problems, so we must be aware of their own insufficiency, more in-depth explore its mechanism of action, with al kinds of research direction in the future for us.

ABSTRACT

[Objective]To disclose the mechanism of Leweiyin treating functional dyspepsia(FD) through experimental research. [Methods]Wistar rats are randomly divided into 7 groups: normal control, natural recovery, Madinlin, Shenling Baizhu pill, low dosage of Leweiyin, mediate dosage of Leweiyin, high dosage of Leweiyin, 6 in each group. Respectively administer to them with distilled water, Madinlin, Shenling Baizhu pill liquid, low, mediate and high dosage of Leweiyin at 2ml/100g, once every day, successively administer for 14 days one day after self model-making. Observe the gastric mucosa,measure gastro-intestinal hormones SP and VIP. [Results] Leweiyin can effectively increase SP and decrease VIP in experimental rats gastric antrum. [Conclusion] Leweiyin can increase gastro-intestinal tract excitement, regulate the contractility of gastro-intestinal smooth muscle, which may be the main mechanism for Leweiyin to treat FD.

Objective: To observe the effect of polysaccharide ingredients from six TCM complex prescriptions on releasing the cytokine(CK) level of mouse celiac macrophages (M?), to explore the possible mechanism and direct the extractionand separation of active components for above compounds. Methods: The total polysaccharide ingredients of six complex prescriptions were prepared: Siwu Decoction, Sijunzi Decoction, Liuweidihuang Decoction, Guizhi Decoction, Longdanxiegan Decoction and Yupingfeng Pulveres, then the mouse celiac M? were incubated together in 96 holes board. Furthermore, levels for the latter to release CK, including IL-1?, IL-6, IL-8 and TNF-?, were measured.
