ABSTRACT

Objective To explore the mechanism of human umbilical cord mesenchymal stem cell (HUC-MSC) alleviating ischemia-reperfusion injury (IRI) of liver cells through mitochondrial transfer. Methods Normal human liver cell line L02 was divided into the blank control group, oxygen-glucose deprivation (OGD) group, experimental control group, and L02 and HUC-MSC co-culture group (L02+HUC-MSC group). L02+HUC-MSC group was further divided into 10:1 co-culture subgroup (group A), 4:1 co-culture subgroup (group B), 2:1 co-culture subgroup (group C), 1:1co-culture subgroup (group D) and 1:2 co-culture subgroup (group E) according to different co-culture ratio of L02 and HUC-MSC. The apoptosis rate and relative reactive oxygen species (ROS) level of L02 cells were detected by flow cytometry. The MitoTracker positive rate of L02 cells was detected by flow cytometry. The mitochondrial transfer from HUC-MSC to L02 cells was observed by laser confocal microscope. Results The apoptosis rate and relative ROS level of L02 cells in the OGD group were significantly higher than those in the blank control group (both P < 0.05). Compared with the OGD group, the apoptosis rates of L02 cells in group B, C, D and E were significantly decreased (all P < 0.05), and the relative ROS level of L02 cells in group E was significantly declined (P < 0.05). The MitoTracker positive rate of L02 cells did not significantly differ between group A and experimental control group (P>0.05), whereas the MitoTracker positive rates of L02 cells in group B, C, D and E were significantly higher than that in the experimental control group in a concentration-dependent manner (all P < 0.05). Under the laser confocal microscope, mitochondrial transfer fromHUC-MSC to L02 cells could be observed through tunneling nanotube (TNT). Conclusions HUC-MSC may alleviate cell apoptosis and reduce ROS level of liver cells after IRI via direct mitochondrial transfer between cells.

ABSTRACT

Objective To investigate the effect of Beagle's cranium on shockwave blocking after free field explosion, and whether a quantitative relationship exists between intra- and extracranial pressure. Methods Eighteen Beagle dogs were used in the free field explosion experiment. The explosion source was 1 kg and 3.5 kg column-shaped trinitrotoluene (TNT), which was located 1.5 m above the land to rule out the possibility of reflex. Measurment instruments were located 1-6 m from the explosion source. Head of Beagle dog was located at the same location as that of measurment instruments. After Beagle dog was anaesthetised, pressure sensor was planted under parietal bone. One side of the sensor borehole faced to the brain tissue, and the other side of the sensor borehole faced to the cranium to measure the pressure from the skull direction. All Beagle dogs were recumbent fixed, the head was positively faced to the source of explosion. Pressures from the bone and from the brain tissue were measured respectively. Results One dog died after the explosion and the remaining 17 dogs were alive. Twenty-four hours later, Tarlov scores of the alive dogs were graded 4-5, and neurofunctional score in large animals was more than 13. The peak explosive wave pressure was 207(47.6-207) kPa with a mean of 122 kPa. The maximal impact pressure impulse reached to 88 Pa·s. The pressure curve from the cranium direction of Beagle dog was similar to that from the brain tissue, but the pressure curve from the brain tissue direction changed more gently. A logarithmic trend existed of the peak pressure of the shockwave from the cranium direction and the external shockwave at the same position (correlation coefficient = 0.804). Conclusions In free field explosion, shockwave can enter the brain through cranium, chest and abdomen. The cranium has some blocking effect on the shockwave, and partial shockwave pressure can be uploaded to the brain tissue through the chest and abdomen, but because of the blocking of the body tissue, the change curve is relatively smooth.

ABSTRACT

Objective To explore the value of using sPESI score and hs-TnT in the evaluation of short-term prognosis in hemodynamically stable pulmonary embolism. Methods We collected 99 patients with hemodynamically stable PE from our department. According to the sPESI score and hs-TnT, patients were divided into high risk group (sPESI score≥1) and low risk group (sPESI score=0), positive group (hs-TnT≥0.014 ng/mL) and negative group (hs-TnT<0.014 ng/mL) . Then all patients were treated and followed up for 30 days. The sensitivity, specificity, positive predictive value and negative predictive value of adverse events of prognosis were calculated, and ROC curve was drawn to analyze the values in different grouping methods for the prognostic evaluation.Results Thirteen adverse events occurred in all patients. The single test showed that sensitivity, specificity, positive predictive value, and negative predictive value of hs-TnT for predicting adverse events were respectively 84.6%,55.2%,22%,and 96%. When sPESI was used alone, sensitivity, specificity, positive predictive value and negative predictive value were 92.3%, 48.8%, 21.1% , and 97.7% . Results of combined testing showed 100% sensitivity, 29% specificity, 17.6% positive predictive value, and 100% negative predictive value. The ROC curve area of the sPESI, hs-TnT, sPESI and hs-TnT are 0.832 (95% CI, 0.705-0.958), 0.825 (95%CI, 0.694-0.957),0.872 (95%CI, 0.773-0.971) . Conclusions PESI and hs-TnT have clinical value in evaluating the short-term prognosis of hemodynamically stable pulmonary embolism. sPESI combined with hs-TnT has higher significance, especially in patients with low-risk PE.

ABSTRACT

Objective To investigate the effect of brain natriuretic peptide (BNP) ,uric acid (UA) ,lactic dehydrogenase(LDH) and troponin T (TnT)in the diagnosis and prognosis observation of cardiovascular disease .Methods A total of 271 cases of cardio-vascular diseases and 271 cases of non-cardiovascular diseases were selected as the cardiovascular diseases group and control group respectively ,moreover the cardiovascular diseases group was further divided into the Uyghur cardiovascular diseases group and Han cardiovascular diseases group according to the nationality type .The levels of BNP ,LDH ,UA and TnT were detected in all cases . Then the detection results were statistically analyzed .Results The levels of BNP ,UA ,LDH and TnT in the cardiovascular diseases group were significantly higher than those in the control group ,the difference was statistically significant (P<0 .05);the LDH and TnT levels in the Uyghur cardiovascular diseases group were higher than those in the Han cardiovascular diseases group ,while the BNP and UA levels in the Han cardiovascular diseases group were higher than those in the Uyghur cardiovascular diseases group , the difference was not statistically significant (P>0 .05);the decrease after 1 week treatment in the patients with heart failure and coronary heart disease was most significant ,which showed statistical difference compared with before treatment (P<0 .05);the UA level after 1 week treatment in heart failure patients had statistical difference compared with before treatment (P<0 .05) .Conclu-sion The combined detection of BNP ,LDH ,UA and TnT has an important clinical significance in early diagnosis ,treatment and prognosis of cardiovascular diseases .

ABSTRACT

Objective To investigate the diagnostic value of High-Sensitive Troponin T(hs-TnT)in acute myocardial infarction(AMI). Methods One hundred and sixty nine patients with serum hs-TnT concentration≥0.014 μg/L in early hospitalization were enrolled in this study.The ROC curve was used to compare the concentra-tion of hs-TnT with four heart enzyme(CK,CK-MB,LDH,AST)on the diagnostic efficacy to AMI. The differ-ence of hs-TnT in different clinical data groups were investigated using Mann-Whitney U rank test.Then the correla-tion between hs-TnT and Gensini score of Coronary angiography was investigated using the Spearman rank correla-tion test.Results The concentration of hs-TnT in patients with chest pain was significantly higher than that in non-AMI group(P<0.05).The AUC of each ROC curve was hs-TnT(0.806)>CK-MB(0.792)>CK(0.780)>AST (0.704)> LDH(0.684). The optimal diagnostic point of hs-TnT was 0.152 ug/L(sensitivity 0.659,specificity 0.894,Yuden index 0.553).There was a positive correlation between hs-TnT and Gensini scores in men,age>65 years old and the chest tightness group(P < 0.05). Conclusion The hs-TnT is better than four heart enzyme in early diagnosis of AMI and benefit early treatment of AMI.

ABSTRACT

Degradation of 2,4,6-trinitrotoluene (TNT), a nitroaromatic explosive found in the soil and ground water, was investigated using Pseudomonas aeruginosa in in vitro experiments. Biodegradable abilitiy of this bacteria was performed with 50 and 75 mg L−1 TNT concentrations in a defined liquid medium for 96 h time period. Treatment of TNT in supernatant samples taken at 0, 6, 12, 24, 48, 72 and 96 h from agitated vessels was followed by reverse-phase high-performance liquid chromatography (HPLC). In cultures supplemented with 50 and 75 mgL−1 TNT, after 96 h of incubation 46% and 59% reduction were detected respectively. Two metabolites as degradation intermediates with nitrite release into the medium, 2,4-dinitrotoluene (2,4-DNT) and 4-aminodinitrotoluene (4-ADNT), were elucidated by thin layer chromatography (TLC) and gas chromatography-mass spectrometry (GC-MS). These findings clearly indicate that Pseudomonas aeruginosa can be used in bioremediation of TNT contaminated sites.

ABSTRACT

White-rot fungus Irpex lacteus degraded TNT significantly in proportion to the culture time. After 48 h incubation, about 95% of TNT was degraded. Two reduced metabolites were identified as 4-amino-2,6-dinitrotoluene (4-ADNT) and 2-amino-4,6-dinitrotoluene (2-ADNT) which was further degraded.
