ABSTRACT

Aim To screen and study the expression of long non-coding RNA (IncRNA) in rats with middle cerebral artery occlusion (MCAO) with MCAO treated with Tao Hong Si Wu decoction (THSWD) and determine the possible molecular mechanism of THSWD in treating MCAO rats. Methods Three cerebral hemisphere tissue were obtained from the control group, MCAO group and MCAO + THSWD group. RNA sequencing technology was used to identify IncRNA gene expression in the three groups. THSWD-regulated IncRNA genes were identified, and then a THSWD-regu-lated IncRNA-mRNA network was constructed. MCODE plug-in units were used to identify the modules of IncRNA-mRNA networks. Gene ontology (GO) and kyoto encyclopedia of genes and genomes (KEGG) were used to analyze the enriched biological functions and signaling pathways. Cis- and trans-regulatory genes for THSWD-regulated IncRNAs were identified. Reverse transcription real-time quantitative pol-ymerase chain reaction (RT-qPCR) was used to verify IncRNAs. Molecular docking was used to identify IncRNA-mRNA network targets and pathway-associated proteins. Results In MCAO rats, THSWD regulated a total of 302 IncRNAs. Bioinformatics analysis suggested that some core IncRNAs might play an important role in the treatment of MCAO rats with THSWD, and we further found that THSWD might also treat MCAO rats through multiple pathways such as IncRNA-mRNA network and network-enriched complement and coagulation cascades. The results of molecular docking showed that the active compounds gallic acid and a-mygdalin of THSWD had a certain binding ability to protein targets. Conclusions THSWD can protect the brain injury of MCAO rats through IncRNA, which may provide new insights for the treatment of ischemic stroke with THSWD.

ABSTRACT

Platelet microparticles (PMPs) are membrane particles derived from the platelet membrane that enter into the blood circulation. We sought to explore the therapeutic effects of Tao-Hong-Si-Wu Decoction (THSWD) on angiogenesis in a rat model of cerebral ischaemia-reperfusion (I/R). The protective effect of THSWD on I/R rats was observed morphologically by immunohistochemical expression of VEGF and CD34, along with immunofluorescence results of co-expression of BrdU and vWF. Then, PMPs from different groups of rats were extracted, and cytokine array analysis was used to screen for angiogenesis associated proteins. The results showed that THSWD can promote the expression of VEGF, CD34, BrdU and vWF. Cytokine array analysis revealed the changes in the expression of 29 related angiogenic proteins in the total protein of PMPs, which involved the Notch signalling pathway. Compared with model group, the expression levels of NICD and Hes-1 in the THSWD group were significantly increased. In the context of I/R, the angiogenesis-related proteins of PMPs are different. THSWD may involve the promotion of activation of the Notch signalling pathway to achieve therapeutic effects on cerebral ischaemia.

ABSTRACT

Objective To evaluate the effectiveness and safety of Tao Hong Si Wu decoction in treating Diabetic nephropathy (DN).Methods Randomized controlled trials (RCTs) on Tao Hong Si Wu decoction in treatment of DN were searched in PubMed,EMbase,Cochrane Library,CMCC,VIP,CNKI,CBM.Data were strictly extracted using the fundamental evaluation method recommended in Cochrane Handbook and Meta-analysis was performed using RevMan 5.0.24 software. Results Eleven RCTs of 956 patients of DN were analyzed. The content and methodological quality of these 11 test samples were low, which could not be undertaken high quality research of sensitivity analysis.Meta-analysis results showed that:Tao Hong Si Wu decoction could be reduced to some extent, the early and clinical stage of DN patients with 24 h UPE reduce the Ⅲ,Ⅳ of Scr,and improve renal function to a certain extent. The results also showed that Tao Hong Si Wu decoction could lower TG and CHO,better control FBG and HbAlc of DN patients, besides, serious adverse reactions have not been found during the treatment. Conclusion Tao Hong Si Wu Decoction could be a relatively safe and effective treatment of DN drugs. However,a high probability of selection bias and measurement bias in the included studies was bound to affect the strength of the results.
