ABSTRACT

OBJECTIVE: To optimize the water extraction technology of classic formula Taohe chengqi decoction. METHODS: Based on single factor test, combined with response surface methodology and information entropy theory, the soaking time, solid-liquid ratio and extraction time were investigated. Using the contents of rhein, amygdalin, cinnamaldehyde and glycyrrhizic acid in Taohe chengqi decoction as indexes, information entropy theory was used to assign weight coefficients to each evaluation index and calculate the comprehensive score. Through Design-Expert 10 software, the interactions of each factor were analyzed. Water extraction technology was optimized, and validation test was also performed. RESULTS: According to information entropy theory, the weight coefficients of rhein, amygdalin, glycyrrhizic acid and cinnamaldehyde were located at 0.097 6, 0.363 2, 0.173 5 and 0.365 7. The results of interaction analysis showed that the material-liquid ratio had a greater impact on the comprehensive score. The optimal water extraction technology of Taohe chengqi decoction were determined as that soaking time was 60 min; the ratio of material to liquid was 1 ∶ 10 (g/mL); total extraction time was 130 min (extracting for 3 times, lasting for 65, 33, 32 min each time). The results of verification test showed that RSD of content of each index component and the comprehensive score was less than 3%. CONCLUSIONS: The optimal water extraction technology is proved to be stable and feasible, which can provide the basis for the further development and utilization of Taohe chengqi decoction.

ABSTRACT

Objective To investigate the effect and mechanism of Taohe Chengqi Decoction(TCD)on intestinal function of severe acute pancreatitis (SAP) rats. Methods Fifty SD rats were randomly divided into blank control group, SAP model group, TCD group (in the dosage of 23.5 mL·kg -1·d -1 by gastric gavage), Octreotide group(in the dosage of 10 μg·kg-1·d-1 by intraperitoneal injection),combined treatment group(TCD in the dosage of 23.5 mL·kg -1·d -1 by gastric gavage, Octreotide in the dosage of 10 μg·kg -1·d -1 by intraperitoneal injection). Except for the blank control group,the rats in the other groups were injected with L-arginine intraperitoneally to establish SAP model. The rats were sacrificed 24 h after the model was successfully established. The serum level of amylase(AMY) in rats was detected by biochemical method. The serum levels of interleukin (IL)-6, IL-10 and tumor necrosis factor (TNF)-α in the fecal flora of rats were measured by enzyme-linked immunosorbent assay (ELISA),and the pathological changes of colorectal tissues and pancreas tissues were observed by hematoxylin-eosin (HE)staining. Results Compared with the blank control group,the levels of serum AMY,TNF-α,IL-6 and IL-10 in SAP model group were significantly increased (P < 0.05), enterococcus and enterobacteria count were increased, while lactobacillus and bifidobacteria count were decreased;the histological structure of pancreas tissues and large intestinal tissues was destroyed, and the proliferation of large amount of inflammatory cells was present. Compared with the SAP model group, serum AMY,TNF-α,and IL-6 levels were decreased and IL-10 level was increased in the three medication groups (P<0.05);enterococcus and enterobacteria count were decreased,while lactobacillus and bifidobacteria count were increased;the pathological features of pancreas tissues and large intestinal tissues were improved;the effect of the combined treatment group was the strongest. Conclusion TCD can improve the intestinal function and intestinal flora of SAP rats,reduce the inflammatory reaction,and prevent SAP from deterioration.

ABSTRACT

Objective]To investigate the thought of manipulating The Three Blood Statsis Removal Decoctions(Taohe Chengqi Decoction, Di Dang Decoction, Di Dang Pills) of ZHANG Zhongjing.[Method]Collect and analyse the explanations of the scriptures involving the Three Blood Statsis Removal Decoctions of ShangHanLun interpreters from past to nowadays, combine with the theory said in Huang Di Nei Jing, knowledge of exegetics and personal view, in order to summarise the original intention of manipulating the Three Blood Statsis Removal Decoctions by ZHANG Zhongjing. [Result] The main purpose of Taohe Chengqi Decoction is to purge heat, since heat is the main reason of stagnation of blood. It applies to the preliminary stage of combination of heat and blood clot. The main purpose of Di Dang Decoction is to attack the rigid blood-heat stagnation in lower-energizer, it is applied to the late stage of combination of heat and blood clot. Di Dang Pill is the milder prescription of Di Dang Decoction, which is applied to the milder situation of blood-heat combination. [Conclusion]Comparing the Three Blood Statsis Removal Decoctions,“ Superficial”and“ deep”words should be used to differentiate the application of Taohe Chengqi Decoction and Di Dang Decoction, while“ light”and“ severe”words should be used to differentiate that of Di Dang Decotion and Di Dang Pills.

ABSTRACT

Objective To analyze the preventive and therapeutic effects of a traditional Chinese medicine Taohe Chengqi decoction on a rat models of hepatic encephalopathy ( HE) induced by thioacetamide ( TAA) .Methods The rat model of HE was established by intraperitoneal injection of TAA,and the effects of Taohe Chengqi decoction were observed by neurobehavioral changes,neurological test,blood ammonia,serum biochemical indexes,and liver and brain pathological examination.Results The rat model of hepatic encephalopathy was successfully established.Low, medium and high dose Taohe Chengqi decoction markedly improved neuro-reflexes and liver and brain pathological damages, and significantly decreased the HE staging and serum biochemical indexes, and the results of high dose group was the best, similar to that of positive drug-treatment group.Conclusions Taohe Chengqi decoction exhibits promising preventive and therapeutic effects on the rat model of TAA-induced hepatic encephalopathy, and these results provide substantial experimetal evidence for its clinical application.

ABSTRACT

Diamicron group. Conclusion Modified Taohe Chengqi Decoction and its diferent extractions maybe effective on raising or stabilizing the activity of Na+-K+-ATPase and Ca2+-ATPase in cadiocyte of diabetic rats, so it could prevent myocardium hypertrophy and patho-change by heart function failure in variably extent. Ethyl-acetate group and n-butyl alcohol group showed rather good effect among these extractions.

ABSTRACT

Objective To investigate the effect of modified Taohe Chengqi Decoction(TCD)and its separated prescriptions on tissue plasminogen activator(tPA)and plasminogen activator inhibitor-1(PAI-1)content in human umbilical vein endothelial cells(HUVEC)with diabetic damage.Methods Purified HUVEC were divided into 10 groups: normal group,model group,rosiglitazone group,high-,middle-and low-dose modified TCD groups,blood-activating prescription group,heat-clearing and purgation prescription group,Qi-strengthening and yin-nourishing group,and TCD group.The serum containing herbal medicine was prepared through seropharmacological method,and was used for co-culture of HUVEC with high glucose and high insulin in vitro.The tPA and PAI contents in the HUVEC culture of different groups were detected.Results The content of tPA was increased in blood-activating prescription group and rosiglitazone group(P0.05).The PAI-1 content in the model group was higher than that in the normal group(P0.05).Conclusion The effect of modified TCD on improving the function of endothelial cells is probably related to the regulatory effect of blood-activating,heat-clearing and purgation herbs on PAI-1.

ABSTRACT

Objective To investigate the anatomical features of different types of unstable fractures of pelvic ring,and to observe the therapeutic effect of open reduction and internal fixation combined with three-stage syndrome differentiation and treatment.Methods Nineteen patients with pelvic fracture were classified according to Tile's classification for pelvic fracture.Of 19 patients,12 were classified into type B(6 being type B1,3 type B2,and 3 type B3),and 7 into type C(3 being type C1,2 type C2,and 2 type C3).All of the patients received pelvic reconstruction through open reduction and internal fixation with titanium plate.After treatment,three-stage syndrome differentiation and treatment were carried out: Taohe Chengqi Decoction was used in the early stage of fractures,Xugu Huoxue Decoction in the middle stage and Yangjin Jiangu Decoction in the late stage.Meanwhile,traumatic ointment was applied on the affected region after the union of operation wound.Results After operation,the follow-up was carried out in all of the patients for 12 ~ 36 months,averaged 18 months.The results of follow-up showed that the fractures and dislocation of all the patients were healed within 3 ~ 6 months,and the patients obtained the normal life and walking function.The results of Majeed's classification showed that 17 were effective,2 ineffective and the effective rate was 89.47%.Conclusion According to the classification of pelvic fractures,it is a better method for pelvic fractures by pelvic reconstruction through different methods of open reduction and internal fixation with titanium plate,and then by three-stage syndrome differentiation and treatment.
