ABSTRACT

Objective: To detect the expressions of periostin, matrix metalloproteinase-8 (MMP-8) and tissue inhibitor of matrix metalloproteinase-2 (TIMP-2) in gingival tissue of the patients with chronic periodontitis, and to explore the possible role of periostin in the occurrence and development of chronic periodontitis and its related biological mechanism. Methods: A total of 23 normal gingival tissues (control group) and 19 inflammatory gingival tissues (case group) were selected. The expressions of periostin, MMP-8 and TIMP-2 in gingival tissue of the patients in two groups were detected by immunohistochemical SP method. The correlation between the expressions of periositn and MMP-8 and TIMP-2 in gingival tissue of the patients in two groups were detected by Spearman analysis. Results: The positive expression rate of periostin in gingival tissue of the patients in case group was lower than that in control group (P<0. 05). The correlative analysis results demonstrated that the expression of periostin had negative relationship with the expression of MMP-8 (r=-0.51, P<0. 05) in control group, and positive relationship with the expression of TIMP-2 (r=0. 50, P<0. 05); in case group, the expression of periostin had negative relationship with the expression of MMP-8 (r=-0.482, P<0. 05), and positive relationship with the expression of TIMP-2 (r= 0.655, P<0. 05). Conclusion: Periostin can be used as a preliminary indicator for maintaining the structural stability of gingival tissue, and become a research target for periodontal tissue regeneration.

ABSTRACT

Background The scarring of conjunctival filtering blebs after glaucomatous surgery is a leading cause of operation failure.Exploring the balance between matrix metalloproteinase-2 (MMP-2) and tissue inhibitor of matrix metalloproteinase-2 (TIMP-2) in conjunctival filtering bleb is very important for the study on pathogenesis of postoperative scarring. Objective This study was to evaluate the role of MMP-2 and TIMP-2 in wounding healing of subconjunctival tissue after filtering surgery in rats. Methods Sixty-three clean male SD rats were divided into normal control group and postoperative 1-day,3-day,5-day,7-day,14-day and 28-day group. The drainage tube was monocularly implanted into the anterior chamber of the rats to establish the filtering surgery models, and the operative response of the eyes was examined under the slit lamp microscope. The animals were sacrificed in corresponding time points, and the frozen sections of eyeballs were prepared, and the expressions of MMP-2 and TIMP-2 in conjunctival and subconjunctival tissues were detected using immunofluorescence technique.Western blot was employed to assay the dynamic expressions of MMP-2 and TIMP-2 proteins in conjunctival and subconjunctival tissues in different groups. The use and care of the rats complied with the Instruction Notions with Respect to Care for Laboratory by State Ministry of Science and Technology. Results Filtering blebs were formed in all the operated eyes 1 day after surgery and remained for 7-17 days, with the average survival time of 12 days.Western blot assay revealed that the expression levels of MMP-2 protein in the filtering blebs of the normal control group were 121.67 ±4.37,and the expressions were gradually elevated from the postoperative 3-day group (183.67±5.61) until the postoperative 7-day group (230.50±8.48) and then gradually declined till the postoperative 28-day group (172.33 ± 8.43),showing a significant difference among the groups (F=280.18,P<0.05).In addition, the expression levels of TIMP-2 protein in filtering blebs of the normal control group were 102.50 ±6.25.The expression levels were gradually raised in postoperative 3-day group (162.67±7.00) and peaked in the postoperative 5-day group (232.00± 11.03),and then the levels gradually reduced till the postoperative 28-day group (150.50±6.41),with a significant difference among the groups (F =145.34,P < 0.05).Immunofluorescent staining showed that MMP-2 and TIMP-2 were weakly expressed in the conjunctival epithelium of the normal rats, while in the operated rats, strong fluorescence for MMP-2 and TIMP-2 was seen in both conjunctival epithelium and subconjunctival tissues of filtering blebs. Conclusions The expression trends of MMP-2 and TIMP-2 in the filtering blebs are consistent to the fibrosis process of conjunctival tissue, indicating that MMP-2 and TIMP-2 participate in the scar formation of conjunctival filtering bleb after glaucoma filtering surgery.

ABSTRACT

The study of glomerular mesangial cells of normal rats showed that angiotensin Ⅱ (ATⅡ) down-regulated the expression of MMP-2 mRNA, did not have significant effect on TIMP-2 mRNA. And in consequence, ATⅡ down-regulated the ratio of MMP-2 mRNA to TIMP-2 mRNA.

ABSTRACT

BACKGROUND: The pathogenesis of transition from viral myocarditis to dilated cardiomyopathy is elusive, although the incidence of dilated cardiomyopathy in human is increasing. METHODS: To clarify the role of the tissue inhibitor of matrix metaloproteinase-2 (TIMP-2) in this event, we performed immunohistochemistry, immunoblotting and immunoassay of matrix metalloproteinase-9 (MMP-9) and TIMP-2 in the serum and heart tissue of mice, which were inoculated with 4000 plaque-forming units of coxsackie B virus. RESULTS: The MMP-9 was expressed in damaged cardiomyocytes, and the TIMP-2 was expressed in mainly interstitial connective tissue between cardiac muscle bundles by immunohistochemistry. The level of serum MMP-9 was higher in the complicated than non-complicated group (p<0.001), but the level of TIMP-2 was much lower in complicated than non-complicated group (p<0.05). These findings were similar to the results of immunohistochemistry and immunoblotting in tissues. CONCLUSIONS: These results suggest that an imbalance in the level of MMP-9 and its inhibitor might activate cardiac complication in viral myocarditis.

ABSTRACT

Objective To study the relationships between serum matrix metalloproteinase-2 (MMP-2) and tissue inhibitor of matrix metal]oproteinase-2 (TIMP-2) with bone metabolic markers and bone mineral density (BMD) in postmenopausal Chinese women. Methods In this study,192 postmenopausal Chinese females aged 48-65 were selected.Serum MMP-2,TIMP-2,bone alkaline phosphatase (BAP),osteocalcin, bone cross-linked C-telopeptides of typeⅠcollagen (CTx) and urine bone cross-linked N-telopeptides of typeⅠcollagen (NTx) were measured by ELISA.And the ratio of MMP-2/TIMP-2 was calculated.BMD was measured by dual energy X- ray abserptiometry.According to the criteria of WHO,these women were assigned to 3 groups : the normal, the low BMD and the osteoporosis groups. Results (1) Serum MMP-2 level was significantly higher in postmenopausal women with osteoporosis (1388?121)?g/L than that in age-matched normal controls (1126?141)?g/L (P0.05).(2) Notable negative relations were found between MMP-2,ratio of MMP-2/TIMP-2 with BMD of lumbar spine and Ward's triangle, serum BAP and osteocalcin (all P0.05).(3) In osteoporosis women, notable negative correlations between MMP-2,ratio of MMP-2/TIMP-2 with BMD of lumbar spine, femoral neck and Ward's triangle, serum BAP and osteocalcin were found (all P
