ABSTRACT

Whiteflies are a severe threat to soybean production in the tropics. This study aimed to evaluate the soybean resistance level of the whitefly Bemisia tabaci in controlled and uncontrolled environments that is associated with plant age, damage intensity, and trichome density. The research was conducted under two conditions: non-sprayed (NS) and sprayed (SP). This study used 50 soybean genotypes arranged in a randomized block design with three replicates. The whitefly population was derived from natural infestations. The results showed that the highest wild population of B. tabaci occurred at 40 days after planting (DAP), i.e., 126.08 adults/plant in the NS environment and 22.57 adults/plant in the SP environment. The peak damage intensity occurred at 50 DAP, 20.71% in the NS environment, and 17.15% in the SP environment. In the NS environment, there were six resistant genotypes (including the resistant control G100H), 25 moderate, and 19 susceptible genotypes. In the SP environment, 19 genotypes were resistant, 22 genotypes were moderate, and nine genotypes were susceptible, respectively. Six soybean genotypes showed consistent resistance to B. tabaci in NS and SP environments. The low density of leaf trichomes in soybean may influence the high resistance to B. tabaci. The resistant genotypes identified in this study could be utilized in breeding programs for B. tabaci resistance.

ABSTRACT

Artemisia stolonifera is a relative of A. argyi. The two species are difficult to be distinguished due to the similarity in leaf shape and have even less distinctive features after processing. This study aims to establish a method to quickly distinguish between them. At the same time, we examined the reasonability and applicability of the specific polymerase chain reaction(PCR) method. The C/T single nucleotide polymorphism was detected at the position 202 of the sequence, based on which specific primers were designed to identify these two species. The PCR with the specific primer JNC-F and the universal primer ITS3R produced a specific band at 218 bp for A. argyi and no band for A. stolonifera, which can be used to detect at least 3% of A. argyi samples mixed in A. stolonifera samples. The PCR with the specific primer KY-F and the universal primer ITS3R produced a specific band at 218 bp for A. stolonifera and no band for A. argyi, which can be used to detect at least 5% of A. stolonifera samples mixed with A. argyi. The limit of detection of the established method was 5 ng DNA. The established PCR method can accurately distinguish between A. stolonifera and A. argyi, which provides an experimental basis for the quality control of A. stolonifera and determines whether the herbs are adulterated.

ABSTRACT

Aim: The study was aimed at evaluating some pharmacognostic parameters and investigates the anti-diabetic activity of ethanol extract of Triumfetta cordifolia leaf Methods: The pharmacognostic profiling of Triumfetta cordifolia leaves was carried out using some standard pharmacognostic tools for crude drug standardization such as qualitative and quantitative microscopy, analytical evaluation and phytochemical screening. The plant material was extracted using cold maceration method in ethanol and fractionation was carried out using n-hexane, ethylacetate and butanol. The acute toxicity study was done following standard method. Diabetes in albino wistar rats was intraperitoneally induced using 120 mg/kg body weight of alloxan monohydrate. The diabetic rats where treated with 200 and 400 mg/kg body weight of the crude extract and 400mg/kg of each of the fractions. Glibenclamide was used as the standard drug (5 mg/kg) and diabetic rats without treatment as negative control. The procedure was also similarly performed using the non-diabetic rats. The administration of all treatments was done orally, once daily for 21 days and blood sera of the blood samples from rats across the groups were collected at the end of the treatment period and the concentrations of Aspartate aminotransferase (AST), Alkaline phosphatase (ALP) and Alanine aminotransferase (ALT) were evaluated. Results: The results of the qualitative microscopic evaluation of Triumfetta cordifolia leaf revealed paracytic stomata, unicellular trichomes, wavy wall epidermal cells and prismatic calcium oxalate. Quantitative microscopic study gave 23.67 ± 0.58 stomata number, 0.023 ± 0.00058 stomata index, 18.33 ± 1.53 Palisade ratio and 10.67 ± 0.58 vein-islet number while the analytical standard revealed 9.3 total ash, 3.5 water soluble ash and 1.34 acid insoluble ash. The phytochemical analysis revealed the presence of alkaloids, glycosides, Tannins, flavonoids steroids and terpenoids in the Triumfetta cordifolia leaves ethanol extract (TCEE). A significant reduction (P?0.05) in fasted blood sugar level of diabetic rats was observed during treatment with Triumfetta cordifolia leaves extract and the blood sugar level lowering potential was comparable to the glibenclamide’s group. There was improvement of body weight in TCEE treated groups and ethylacetate fraction group. The Leaf extract of Triumfetta cordifolia showed a high significant (P?0.05) ameliorating potential on liver’s degenerating hepatocytes evidenced by the comparable reduction in AST, ALT and ALP levels with the glibenclamide’s and diabetic’s groups. Conclusion: The overall results showed that Triumfetta cordifolia leaf possesses blood sugar lowering and liver hepatocytes regenerating potentials while the pharmacognostic profiling of the plant can serve as a reference and guide for future researchers.

ABSTRACT

Plant trichomes are special structures that originate from epidermal outgrowths. Trichomes play an important role in plant defense against pests and diseases, and possess economic and medicinal values. Study on molecular mechanism of plant trichomes will contribute to the molecular design breeding and genetic improvement of crops. In recent years, the regulation mechanism of trichome development has been basically clarified in the model plant Arabidopsis thaliana, while great progresses are also found in other plant species. In this review, we focus on the developmental regulation of trichome formation from gene and phytohormones levels in Arabidopsis and cotton (with unicellular trichomes), as well as in tomato and Artemisia annua (with multicellular trichomes). The research progress associated with trichomes is also introduced in other typical monocotyledons and dicotyledons. Finally, the research and application of plant trichomes are prospected.

ABSTRACT

On the surfaces of plants, there are many small protrusions of epidermal origin-trichomes. Trichomes are normally divided into two general categories: non-glandular and glandular. Glandular trichomes, an important secretory tissue in plant, can synthesize a variety of secondary metabolites specially. In recent years, with the deepening of research on medicinal plants, the importance of glandular trichomes research has been increasing prominent significance. A variety of medicinal ingredients were synthesized, modificated, and storaged in glandular trichomes. The growth situation of glandular trichomes can directly affect the quality of medicinal materials. Here we provide an update on the methods and technologies which have been used to investigate glandular trichomes. In additon, we also review the literature on the structure, biochemistry, and gene regulatory of glandular trichomes.

ABSTRACT

Glandular trichomes play a major role in the morphological characterization of the Bignoniaceae. Due to their great diversity of forms and functions, this study aimed to inventory the glandular trichomes present in the aerial vegetative axis of Amphilophium magnoliifolium, Martinella obovata and Stizophyllum riparium, analyze their structure and register the participation of ants in these plants. Fresh samples from the nodal region, petiole and from medium to apical regions of the leaflet blade were fixed and processed according to usual methods in light and scanning electron microscopies. The glandular trichomes found were: peltate, capitate, stipitate, and patelliform/cupular. Peltate trichomes are the most abundant ones and present the most uniform distribution. Patelliform/cupular trichomes occur at specific regions, such as prophylls, leaflet blade and nodal regions. Martinella obovata is the only species that presents capitate and stipitate trichomes, which are widely distributed along the entire aerial vegetative axis. Ants were found in all species, mainly at nodal regions. The occurrence of the capitate-type trichome is reported for the first time to the genus.


Tricomas glandulares desempenham papel importante na caracterização morfológica de Bignoniaceae e, devido à grande diversidade de formas e funções que esses tricomas apresentam, o objetivo desse estudo foi inventariar os tricomas glandulares presentes no eixo vegetativo aéreo de Amphilophium magnoliifolium, Martinella obovata. e Stizophyllum riparium, bem como analisar sua estrutura e registrar a participação de formigas nessas espécies. Amostras frescas da região mediana à apical das lâmina foliolar, pecíolo e nó foram fixadas e processadas de acordo com os métodos usuais para microscopia de luz e eletrônica de varredura. Os tricomas glandulares encontrados nas espécies foram: peltados, capitados, estipitados e pateliformes/cupuliformes. Dentre esses tipos o mais abundante foi o tricoma peltado, apresentando também uma distribuição mais uniforme que os demais. Os tricomas pateliformes/cupuliformes estavam em regiões mais específicas, como profilos, lâmina foliolar e na região nodal. Martinella obovata foi a única espécie que apresentou os tricomas capitados e estipitados, amplamente distribuídos ao longo de todo eixo vegetativo aéreo. Em todas as espécies foram encontradas formigas, principalmente nas regiões nodais. A ocorrência do tricoma capitado é relatada pela primeira vez para o gênero.

In the present study, the effects of two CaCl2.2H2O levels (440 and 1320 mg L-1) and two subcultures were evaluated on in vitro shoots of Lavandula angustifolia cv. Provence Blue. Ca2+ content of the apical, middle and basal portion of shoots was determined. Increasing CaCl2.2H2O level in the culture medium increased tissue Ca2+ content and decreased hyperhydricity. Shoot-tip necrosis also decreased with 1320 mg L-1 CaCl2.2H2O, but it did not occur in the second subculture. The middle and basal portion had higher Ca2+ content than apical portion. In non-hyperhydric tissues, there were smaller and more juxtaposed cells. Scanning electron microscopy of the leaves demonstrated that trichomes from in vitro leaf surface occurred in smaller quantities.

We present the leaf anatomy of seven amphibious species of Aeschynomene L. (Papilionoideae, Leguminosae), interpreting their structures and ecological functions, and also, providing information on which their taxonomy can be based, especially of morphologically similar species. We evaluated Aeschynomene americana, A. ciliata, A. evenia, A. denticulata, A. fluminensis, A. rudis and A. sensitiva. The anatomy corroborates the separation of the series Americanae, Fluminenses, Indicae and Sensitivae, with the shape of the petiole, types of trichomes and quantity of vascular units in the petiole as main characteristics to delimit the species. The petiole shape varies from cylindric in A. americana, A. sensitiva and A. fluminensis, to triangular in A. evenia and quadrangular in A. rudis, A. denticulata and A. ciliata. We observed four types of trichomes: hydathode trichome, long conic trichome, short conic trichome and bulb-based trichome. The hydathode trichome was the most common, except for A. americana and A. fluminensis. Species with higher affinity with water share similar adaptive characteristics, including hydathode trichomes described for the first time for the genus. This article adds unseen descriptions for the genus and on the adaptation factors of the amphibious species.


Neste trabalho é apresentada a anatomia foliar de sete espécies anfíbias de Aeschynomene L., interpretando estruturas e suas funções ecológicas e fornecendo informações que subsidiam sua taxonomia, principalmente de espécies semelhantes morfologicamente. Foram avaliadas Aeschynomene americana, A. ciliata, A. evenia, A. denticulata, A. fluminensis, A. rudis e A. sensitiva, pertencentes ao grupo Papilionoideae - Leguminosae. A anatomia corrobora a separação das séries Americanae, Fluminenses, Indicae e Sensitivae, sendo o formato do pecíolo, tipos dos tricomas e quantidade de unidades vasculares no pecíolo as principais características para delimitação das espécies de Aeschynomene. O formato do pecíolo variou de cilíndrico para A. americana, A. sensitiva e A. fluminensis, triangular para A. evenia e quadrangular para A. rudis, A. denticulata e A. ciliata. Foram observados quatro tipos de tricomas, são eles: tricoma-hidatódio, tricoma cônico longo, tricoma de base bulbosa e cônico curto. O tricoma-hidatódio foi o mais comum, ausente somente em A. americana e A. fluminensis. Espécies com maior afinidade pela água compartilham características adaptativas similares, incluindo tricoma-hidatódio descrito pela primeira vez para o gênero. Este trabalho acrescenta descrições inéditas para o gênero e sobre fatores adaptativos das espécies anfíbias.

Objective:To study the leaf epidermis of wild and micropropagated Dioscorea bulbifera Linn. (D. bulbifera) in order to document useful diagnostic features that may be employed for correct crude drug identification and to clear any taxonomic uncertainties in the micropropagated medicinal plant. Methods:Growth responses of micropropagated D. bulbifera were observed on Murashige Skoog medium supplemented with 6-benzylamino purine (1.0 mg/L)+α-naphthaleneacetic acid (0.2 mg/L)+cysteine (20 mg/L) using nodal segments as explants. Leaves of the wild and micropropagated plants were studied microscopically. Results: More than 80%shoot regeneration and formation of 10%-30%whitish-brown callus were observed within 3 weeks. The highest root proliferation was obtained from Murashige Skoog medium of 6-benzylamino purine (0.05 mg/L) and α-naphthaleneacetic acid (0.01 mg/L) with mean root length of (27.00±1.25) mm and elongated single shoot of mean length (38.00±11.09) mm. Leaf epidermal features that revealed similarities between the wild and micropropagated plants included amphistomatic condition, presence of mucilage, glandular unicellular trichome with multicellular head, polygonal cells with smooth walls, stomata type and shape. Slight variations included thick cuticular wall with closed stomata in wild plant compared to thin walled opened stomata in the in vitro plant. Opening of stomata accounted for larger average stomata sizes of (7.68±0.38) μm and (6.14±0.46) μm on the adaxial and abaxial surfaces, respectively of the micropropagated plant compared to the wild. Conclusions: The diagnostic features obtained in the study could serve as a basis for proper identification for quality control for standardization of the medicinal plant.

Eugenia pyriformis Cambess., known as uvaia, is a species of Myrtaceae native to Brazil. Its leaves are used in folk medicine to treat gout because they possess the property of inhibiting xanthine oxidase, an enzyme involved in the conversion of xanthine into uric acid. The objective of this work was to study the leaf and stem morpho-anatomy of E. pyriformis, in order to contribute to what is known about the Brazilian flora, and this medicinal plant and potential vegetal drug. Samples of mature leaves and young stems were fixed and sectioned by freehand, or embedded in glycol methacrylate and sectioned with a microtome, and then stained. In addition, microchemical tests and scanning electron microscopy were performed. The leaf is simple, symmetric, elliptic-lanceolate, with an acute apex and base, and an entire margin. The epidermis is uniseriate and coated with a moderately thick cuticle. The stomata are anomocytic and inserted at the same level as the adjacent cells. Unicellular non-glandular trichomes are abundant on the abaxial surface. The mesophyll is dorsiventral. In transverse section, the midrib is plano-convex and the petiole is circular, and both of these structures have a single bicollateral vascular bundle. In the stem, the vascular cylinder consists of external phloem, xylem and internal phloem, traversed by narrow rays. Phenolic compounds, druses and prismatic crystals of calcium oxalate are also present in the leaf and stem.

ABSTRACT

Connarus suberosus is a typical species of the Brazilian Cerrado biome, and its inflorescences and young vegetative branches are densely covered by dendritic trichomes. The objective of this study was to report the occurrence of a previously undescribed glandular trichome of this species. The localization, origin and structure of these trichomes were investigated under light, transmission and scanning electron microscopy. Collections were made throughout the year, from five adult specimens of Connarus suberosus near Botucatu, São Paulo, Brazil, including vegetative and reproductive apices, leaves and fruits in different developmental stages, as well as floral buds and flowers at anthesis. Glandular trichomes (GTs) occurred on vegetative and reproductive organs during their juvenile stages. The GTs consisted of a uniseriate, multicellular peduncle, whose cells contain phenolic compounds, as well as a multicellular glandular portion that accumulates lipids. The glandular cell has thin wall, dense cytoplasm (with many mitochondria, plastids and dictyosomes), and a large nucleus with a visible nucleolus. The starch present in the plastids was hydrolyzed during the synthesis phase, reducing the density of the plastid stroma. Some plastids were fused to vacuoles, and some evidence suggested the conversion of plastids into vacuoles. During the final activity stages of the GTs, a darkening of the protoplasm was observed in some of the glandular cells, as a programmed cell death; afterwards, became caducous. The GTs in C. suberosus had a temporal restriction, being limited to the juvenile phase of the organs. Their presence on the exposed surfaces of developing organs and the chemical nature of the reserve products, suggest that these structures are food bodies. Field observations and detailed studies of plant-environment interactions, as well as chemical analysis of the reserve compounds, are still necessary to confirm the role of these GTs as feeding rewards.


Connarus suberosus es una especie típica del bioma Cerrado brasileño, y sus inflorescencias junto con los ramos vegetativos jóvenes están densamente cubiertos por tricomas dendríticas. El objetivo de este estudio es presentar la ocurrencia de un tricoma glandular sin descripción previa para esta especie. La localización, origen y estructura de estos tricomas se observó mediante microscopia óptica, así como electrónica de transmisión y de barrido. Los tricomas glandulares (TGs) se producen en los órganos vegetativos y reproductivos, en sus fases jóvenes. Los TGs consisten en un pedúnculo uniseriado y multicelular, cuyas células contienen compuestos fenólicos, así como un sitio en el cual las células glandulares acumulan compuestos pécticos, almidón y lípidos. La acumulación de aceite en el citosol es impresionante, pues ocupa una gran parte del volumen celular. La porción glandular presenta células con paredes delgadas, un denso citoplasma y un núcleo grande con nucleolo visible. El citoplasma contiene gran cantidad de mitocondrias, plástidos y dictiosomas. El almidón presente en los plástidos se hidroliza durante la fase de síntesis, de tal modo que reduce la densidad del estroma plastidial. Algunos plástidos se fusionan con las vacuolas, además hay pruebas de la conversión de los plástidos en vacuolas. Durante las etapas finales de la actividad de los TGs, se observó un oscurecimiento del protoplasma en algunas de las células glandulares lo que sugiere la existencia de la muerte celular programada; después de lo anterior, los TGs se convierten en caducas. Por lo tanto la aparición de TGs en las superficies expuestas de los órganos en desarrollo en C. suberosus y la naturaleza química de los productos de reserva sugieren que estas estructuras son cuerpos alimenticios. Hay evidencias sobre el papel de estos tricomas en las interacciones planta-hormiga, especialmente en el bioma Cerrado.

ABSTRACT

Variações nas características morfológicas das plantas podem ter efeito imediato sobre a preferência alimentar, escolha de sítios de oviposição e vulnerabilidade de herbívoros a inimigos naturais. A longo prazo, podem ter consequências sobre a dinâmica populacional desses organismos, já que atuam sobre seu desenvolvimento, sobrevivência e reprodução. Neste trabalho, avaliou-se o desenvolvimento de Polyphagotarsonemus latus (Banks) em cinco espécies de Capsicum com diferentes densidades de tricomas. Testou-se a hipótese de que o crescimento populacional de P. latus nessas plantas apresenta relação negativa com o aumento da densidade de tricomas nas folhas. Observou-se um padrão decrescente na taxa intrínseca de crescimento (r m) de P. latus com o aumento da densidade de tricomas nas folhas. O menor r m foi obtido em folhas de C. praetermissum (r m = 0,46), que apresenta a maior densidade de tricomas entre as espécies de Capsicum avaliadas. Os maiores valores de r m foram observados em espécies de Capsicum com densidade intermediária de tricomas nas folhas, Capsicum frutescens (r m = 0,77) e Capsicum chinense (r m = 0,76). Os tricomas de Capsicum spp. atuaram como fator limitante ao desenvolvimento de P. latus à medida que se apresentaram em maiores densidades e mais bem distribuídos na superfície foliar. Foi discutido ainda o papel da defesa morfológica e química de plantas de Capsicum sobre o desenvolvimento de P. latus.


Plant morphology traits can affect the feeding preference, oviposition choice and the vulnerability of herbivores to natural enemies. At long term, these plant variations may influence herbivore population dynamics due to the interference on their development, survival and reproduction. In this paper, the development of the broad mite Polyphagotarsonemus latus (Banks) was evaluated on five species of Capsicum with different density of trichomes. We tested the hypothesis that P. latus population growth has a negative relationship with the increase trichome density on leaves. It was assessed a decrease in the intrinsic growth rate (r m) of P. latus with the increase of trichome density on the leaves tested. The lower r m was measured in leaves of C. praetermissum (r m = 0.46), which has the highest trichome density among the assessed Capsicum species. The highest values of r m were observed on Capsicum species with intermediate densities of trichomes on leaves such as Capsicum frutescens (r m = 0.77) and Capsicum chinense (r m = 0.76). Capsicum spp. trichomes acted as a limiting plant trait for the development of P. latus when they were in higher densities and better distributed on the leaf surface. We also discuss the role of morphologic and chemical plant defenses on P. latus development.

Avaliou-se a influência da face foliar, do comportamento geotrópico do inseto e do tipo de tricoma foliar sobre a oviposição de Bemisia tabaci (Genn.) biótipo B em folhas de tomateiro. Para avaliação desses efeitos foram realizados vários ensaios nos quais as fêmeas foram confinadas em caixas de acrílico. Folíolos de tomateiro foram fixados no topo e na base das caixas, de tal forma a permitir o acesso dos insetos às faces adaxial e abaxial de maneira isolada e concomitante (com e sem chance de escolha). Os insetos sempre depositaram maior número de ovos quando a superfície foliar se encontrava no topo da caixa e, de maneira geral, preferiram depositar seus ovos na face abaxial dos folíolos. Para avaliar o efeito do tipo de tricoma (glandular e não-glandular) sobre a oviposição de B. tabaci, a face abaxial dos folíolos de tomateiro foi oferecida a cerca de 30 fêmeas para oviposição em cada caixa de acrílico. Foram oferecidos folíolos previamente lavados com etanol a 70 por cento (de forma a eliminar os exsudatos glandulares) e sem lavagem prévia. A maior parte da oviposição ocorreu ao lado dos tricomas não-glandulares, demonstrando que o inseto discrimina o tipo de tricoma.


The objective of this work was to evaluate the influence of the leaf surface, the insect geotropic behavior and the type of foliar trichome on Bemisia tabaci (Genn.) biotype B oviposition on tomato leaves. Bemisia tabaci females were confined in acrylic boxes in which tomato leaflets were fixed at the bottom and top part of the boxes to allow insects to access the leaf surface to be tested (adaxial and/or abaxial) in both no-choice and free choice tests. Oviposition was always higher when the leaf was offered at the top of the box and preferably at the abaxial leaf surface. The effects of leaf trichomes (glandular and non glandular) on B. tabaci oviposition was evaluated by offering the abaxial surface of tomato leaflets to females after a 70 percent ethanol wash to remove glandular exsudates against a control treatment (without a ethanol wash). Oviposition was concentrated mostly near to non glandular trichomes, showing whitefly females can discriminate among the trichomes.

Yacón é a denominação popular da erva perene nativa dos Andes, Smallanthus sonchifolius (Poepp.) H. Rob. (Asteraceae), cultivada em diferentes países como recurso alimentar e medicinal. Raízes tuberosas e partes vegetativas aéreas são freqüentemente empregadas como coadjuvante no tratamento do diabetes. Este trabalho objetivou estudar os caracteres anatômicos de folha e caule, a fim de fornecer informações para a identificação microscópica no controle de qualidade farmacognóstico. O material botânico foi fixado e preparado de acordo com técnicas usuais de microscopia de luz e eletrônica de varredura. A folha é anfiestomática, com predominância de estômatos anomocíticos. Tricomas tectores e glandulares estão presentes, sendo os primeiros pluricelulares e unisseriados. Os tricomas glandulares são curtos e pluricelulares, com duas séries de células. O mesofilo é dorsiventral e a nervura central possui vários feixes vasculares colaterais em disposição cêntrica. O caule jovem apresenta epiderme persistente, colênquima angular, parênquima cortical, endoderme e cilindros contínuos de floema e xilema. Dutos secretores encontram-se na folha e no caule. O conjunto dos caracteres observados, como a presença de tricomas tectores e glandulares, dutos secretores e endoderme com estrias de Caspary, é útil para o controle de qualidade dessa espécie.


Yacon is the common name of the perennial herb native to the Andes, Smallanthus sonchifolius (Poepp. ) H. Rob. (Asteraceae), cultivated in different countries as food and medicinal resource. Fleshy roots and aerial vegetative organs are often used in the treatment of diabetes. This work has aimed to study the anatomical characters of the leaf and stem, aiming to supply information for microscopic identification of the pharmacognostic quality control. The botanical material was fixed and prepared according to standard techniques of light and scanning electron microscopy. The leaf is amphistomatic, predominantly with anomocytic stomata. Non-glandular and glandular trichomes are present, being the former multicellular and uniseriate. The glandular ones are short and multicellular, with two series of cells. The mesophyll is dorsiventral and the midrib has various collateral vascular bundles in centric arrangement. The young stem has remaining epidermis, angular collenchyma, cortical parenchyma, endodermis and continuous cylinders of phloem and xylem. Secretory ducts are found in the leaf and stem. The assembled characters, such as non-glandular and glandular trichomes as well as secretory ducts and endodermis with Casparian strips, are useful for the quality control of this species.

Conhecida comumente como erva-de-colégio, erva-grossa e sussuaiá, Elephantopus mollis Kunth (Asteraceae) é uma espécie herbácea perene, de base sublenhosa, ramos curtos e flores arroxeadas. Suas folhas são empregadas como emoliente, resolutivo, sudorífico e no tratamento de bronquite, tosse e gripe na medicina popular. O objetivo deste trabalho foi estudar caracteres anatômicos foliares e caulinares, a fim de fornecer subsídios aplicáveis à identificação da planta medicinal. Folhas adultas e fragmentos de caules foram fixados, seccionados e corados com azul de astra/fucsina básica ou com azul de toluidina. Testes microquímicos e análise ultra-estrutural de superfície foram também executados. A folha possui epiderme uniestratificada, recoberta por cutícula estriada e, em vista frontal, tem células com contorno ondulado. Ocorrem tricomas tectores e glandulares. Os primeiros são eretos, pluricelulares e unisseriados. Os tricomas glandulares são capitados e bisseriados. O mesofilo é dorsiventral e a nervura central é percorrida por feixes vasculares colaterais dispostos em arco aberto. O caule, em estrutura secundária inicial, possui epiderme unisseriada com cutícula estriada e tricomas semelhantes aos da folha. No córtex, observam-se colênquima anelar alternado com clorênquima e endoderme com estrias de Caspary. Calotas esclerenquimáticas apõem-se ao floema. Este é formado pelo câmbio vascular em direção à periferia e o xilema é gerado de modo centrípeto. A medula é parenquimática. Pequenos cristais de oxalato de cálcio estão presentes na folha e no caule.


Commonly known as erva-de-colégio in Portuguese, Elephantopus mollis Kunth (Asteraceae) is a perennial herb, with slightly lignified base, short branches and purple flowers. Its leaves are used as emollient, vulnerary and diaphoretic, and to treat bronchitis, coughs and influenza in folk medicine. The aim of this work was to study leaf and stem anatomical characters, in order to expand the knowledge for this medicinal plant identification. Adult leaves and stem fragments were fixed, sectioned and stained either with astra blue/basic fuchsine or toluidine blue. Microchemical essays and scanning electron analysis were also carried out. The leaf has wavy-contoured cells in surface view, forming a single layer and coated with striate cuticle. Non-glandular and glandular trichomes occur. The former is erect, multicellular and uniseriate. The glandular ones are capitate and biseriate. The mesophyll is dorsiventral and the midrib is traversed by collateral vascular bundles arranged in open arc. The stem, in initial secondary growth, has uniseriate epidermis coated with striate cuticle and trichomes similar to the leaf. In the cortex, annular collenchyma alternates with chlorenchyma and an endodermis with Casparian strips is found. Sclerenchymatic caps adjoin the phloem, which is formed outwards by the vascular cambia, while the xylem is produced inwards. The pith is parenchymatic. Small calcium oxalate crystals are present in the leaf and stem.

Using collection specimens, we measured the density and wing area of trichomes in 37 species of the bromeliad genus Tillandsia, specifically the abaxial proximal, abaxial distal, adaxial proximal and adaxial distal parts of the leaf. The product of the trichome "wing" area by the number of trichomes (means) produced a pure number (T) that was correlated to ecological features. The correlation was positive with respect to arid environments (xeric Tillands) and negative with respect to humid environments (mesic Tillands). Bulbous, and particularly myrmecophytic species and species with tanks, represented particular categories. Other intermediate types were identified based on the T number, totalling five ecological types. In comparison with other systems of ecological typification for Tillands and other Bromeliaceae, the present system offers measurable data whose analysis is reproducible.


Medimos el número por milímetro cuadrado y el área del “ala” (parte móvil) de los tricomas en las partes adaxial próxima y distal, y adaxial próxima y distal, de la hoja de 37 especies de bromelias del género Tillandsia. El producto del área del ala para el número de los tricomas (promedio) produjo un número puro (T). Hallamos que T se correlaciona con las características ecológicas de las tilandsias investigadas. La correlación es positiva con respecto a ambientes áridos (especies xéricas) y negativa con respecto a los ambientes húmedos (especies mésicas). Las especies con bulbo, y particularmente las asociadas con hormigas y especies con de tanque representan categorías particulares. Identificamos otros tipos intermedios, agradando así cinco tipos ecológicos. En comparación con otros sistemas de tipificación ecológica, este sistema ofrece la ventaja de ser reproducible y cuantitativo.

Os objetivos deste trabalho foram avaliar alguns aspectos biológicos das fases de ovo e de ninfa de Bemisia tabaci (Gennadius) biótipo B e avaliar a não-preferência para oviposição em testes com e sem chance de escolha e sua correlação com o número e tipo de tricomas nas cultivares de algodoeiro (BRS Ipê, BRS 186-Precoce 3, BRS Acala, BRS Verde, BRS 200-Marrom, BRS Cedro, BRS Ita 90-2 e BRS Aroeira). Os experimentos foram realizados em câmaras climatizadas (28 ± 2°C, 70 ± 10 por cento UR e fotofase de 14h), e em casa-de-vegetação. A viabilidade dos ovos não foi afetada pelas cultivares de algodoeiro, porém, a sobrevivência no período de ovo a adulto foi influenciada pelo hospedeiro. Não houve efeito das cultivares na duração da fase de ovo, do segundo, terceiro e quarto ínstares nem no período de ovo a adulto. As ninfas que se desenvolveram na cultivar BRS Ipê tiveram o primeiro ínstar alongado, diferentemente daquelas desenvolvidas nas demais cultivares. As cultivares BRS Aroeira, BRS Verde e BRS Ita 90-2 apresentaram baixo número de ovos nos testes com e sem chance de escolha, indicando um possível mecanismo de resistência, mas não foi possível estabelecer uma correlação entre a densidade de tricomas e a não-preferência para oviposição.


The purposes of this work were to evaluate some biological aspects of Bemisia tabaci (Gennadius) biotype B at egg and nymphal stages and to evaluate the non-preference for oviposition and its correlation with the number and type of trichomes on the cotton cultivars BRS Ipê, BRS 186-Precoce 3, BRS Acala, BRS Verde, BRS-200 Marrom, BRS Cedro, BRS Ita 90-2 and BRS Aroeira. The experiments were conducted in climatic chambers at 28 ± 2°C, 70 percent RH and photophase of 14h, and in greenhouse. Egg fertility was not affected by the cotton cultivars but survival in egg-adult period was influenced by the host plant. There was no influence of cultivars neither on the duration of egg stage, nymphs at 2nd, 3rd and 4th instars nor on the duration from egg to adult, but nymphs reared on the cultivar BRS Ipê had their 1st instar extended. Low number of eggs was detected on the cultivars BRS Aroeira, BRS Verde and BRS Ita 90-2 in both experiments with and without oviposition choice, indicating a possible mechanism of resistance, but no correlation could be established between trichome densisty and oviposition non-preference.

Trials were carried out with seven soybean genotypes, to evaluate the resistance to Bemisia tabaci Genn. biotype B, between October 2000 and January 2001, at the Faculdade de Ciências Agrárias e Veterinárias – UNESP Jaboticabal, SP, Brazil. Oviposition non-preference (free choice and nochoice tests) and antibiosis were evaluated as well as the correlation between the density of the trichomespresent in the genotypes’ leaflets and oviposition preference. Among the genotypes tested, BR-82 12547 and PI 229358 showed non-preference type resistance to oviposition of the silverleaf whitefly, and oviposition was positively correlated with the total density of trichomes. Thus, PI 227687 with the biggest quantity of trichomes was the most oviposited genotype, whereas BR-82 12547 and PI 229358 with thesmallest quantities of trichomes were the least oviposited genotypes. The antibiosis test was accomplished with five genotypes: BR-82 12547, IAC 100, IAC 78-2318, PI 229358 and PI 227687, selected from the oviposition non-preference test. IAC 100 affected negatively B. tabaci biotype B, by prolonging the ninfal period (12,8 days) and the reducing adults’ emergence (80,0%).


Este trabalho foi realizado com o objetivo de avaliar em sete genótipos de soja, os tipos de resistência à mosca-branca Bemisia tabaci Genn. biótipo B, bem como a correlação entre a densidade dos tricomas presentes nos folíolos dos genótipos estudados e a preferência de oviposição da espécie. Osensaios foram conduzidos na área experimental e no Laboratório de Resistência de Plantas a Insetos doDepartamento de Fitossanidade, da Faculdade de Ciências Agrárias e Veterinárias – Campus de Jaboticabal- UNESP, Jaboticabal, SP, entre outubro de 2000 e janeiro de 2001. Dentre os genótipos testados, BR-82 12547 e PI 229358 apresentaram resistência do tipo não–preferência para oviposição à mosca branca; a oviposição foi correlacionada positivamente com a densidade total de tricomas. Assim, PI 227687, com a maior quantidade de tricomas, foi o genótipo mais ovipositado, enquanto que BR-82 12547 e PI 229358, com as menores quantidades de tricomas, foram os menos ovipositados. O ensaio de antibiose foi realizado utilizando-se cinco genótipos: BR-82 12547, IAC 100, IAC 78-2318, PI 229358 e PI 227687, selecionados do ensaio de não-preferência para oviposição. IAC 100 destacou-se por afetar negativamente B. tabaci biótipo B, prolongando o período ninfal (12,8 dias) e reduzindo a emergência de adultos (80,0%).

ABSTRACT

The aphid Aphis gossypii Glover is associated with several vegetable and ornamental greenhouse crops, especially chrysanthemum (Dendranthema grandiflora Tzvelev). The effect of different temperatures and chrysanthemum cultivars (Yellow Snowdon, White Reagan and Dark Splendid Reagan) on developmental time and survival of A gossypii were evaluated. The trials were carried out at environmental chambers, at constant temperatures: 15, 20, 25 and 30°C, 70±10 percent RH and photophase 10h. Nymphs of A. gossypii were kept individually on chrysanthemum leaf discs (2 cm diameter) of different cultivars in glass petri dishes, with 10ml of sterile solution of agar/water 1 percent. The increase of temperature from 15°C to 30°C, caused a significant reduction in the developmental time of the instars, and the first instar was the most affected. The survival of the nymphs of 1st and 2nd instars was affected by all chrysanthemum cultivars evaluated. These cultivars presented different densities of trichomes/mm² on the leaf (11.3, 16.6 and 21.6 trichomes, respectively), which provided higher survival to nymphs maintained on Yellow Snowdon cultivars (88.5 and 96.6 percent for the 1st and 2nd instars, respectively). The nymphal period decreased from 13.5 days at 15°C to 5.0 days at 30°C. Temperature and chrysanthemum cultivars affected the developmental time and the survival of the initial instars of A. gossypii, and this information is important for A. gossypii management in greenhouse crops.


O pulgão Aphis gossypii Glover está associado a culturas de grande importância econômica em cultivos protegidos, especialmente o crisântemo. Neste trabalho, avaliou-se o efeito de diferentes temperaturas e cultivares comerciais de crisântemo (Yellow Snowdon, White Reagan e Dark Spendid Reagan) sobre o desenvolvimento e a viabilidade de ninfas de A. gossypii. O experimento foi conduzido sob temperaturas constantes de 15, 20, 25 e 30°C, UR de 70±10 por cento e fotofase de 10h. Ninfas de A. gossypii foram mantidas individualizadas em discos foliares (2 cm de diâmetro) das cultivares de crisântemo, em placas de Petri, contendo 10 ml de solução estéril de agar/água a 1 por cento. O incremento da temperatura de 15°C para 30°C reduziu significativamente a duração dos ínstares, sendo o 1° ínstar o mais afetado. A sobrevivência das ninfas de 1° e 2° ínstar foi afetada pelas cultivares de crisântemo Yellow Snowdon, White Reagan e Dark Spendid Reagan as quais apresentaram diferentes densidades de tricomas/mm² na folha (11,3; 16,6 e 21,6 tricomas, respectivamente). Foi obtida maior sobrevivência para ninfas mantidas na Yellow Snowdon (88,5 e 96,6 por cento para o 1° e o 2° instares, respectivamente). O período ninfal decresceu de 13,5 a 15°C para 5,0 dias a 30°C. A temperatura e as cultivares de crisântemo afetaram o tempo de desenvolvimento e a sobrevivência dos instares iniciais de A. gossypii, sendo isso um importante parâmetro para a adoção de táticas de manejo desse pulgão em condições de ambientes protegidos.
