ABSTRACT

The expressions of vascular endothelial growth factor(VEGF)and pigment epithelium derived factor(PEDF)in the tubulointerstitium were determined by immunohistochemistry in the streptozotoein-induced diahetic rats.The mRNA expressions of VEGF and PEDF in normal rat kidney (NRK) were measured by using RT-PCR under different high glucose conditions.The results showed that VEGF expression increased and PEDF expression decreased in the kidney tissue of the streptozotocin-induced diabetic rats.Hiish glucose increased VEGF expression and decreased PEDF expression in N RK cells in a time-and dose-dependent manner.The unbalanced expressions of VEGF and PEDF may play an important role in the occurrence and progression of diabetic nephropathy.

ABSTRACT

Objective: To investigate the development of tubulointerstitial fibrosis caused by partial unilateral ureter obstruction(PUUO) and the improvement of tubulointerstitial fibrosis after obstruction was relieved. Methods: PUUO was induced by the method of embedding upper half of the ureter into ipsilateral psoas.Obstruction was relieved by dissecting the ureter from psoas groove and returning its normal anatomical position.Pathology examination were performed,and collagenⅠand Ⅲ were semi-quantified by immunohistochemistry assay. Results: Massive accumulation of TypeⅠand Ⅲ collagen in tubulointerstitium leads to renal fibrosis.Renal fibrosis can recover if obstruction be relieved within 4 weeks. Conclusion: The results indicated tubuloinsterstitisl fibrosis develop progressively after PUUO,and fibrosis reduce by obstruction relief.

ABSTRACT

To investigate the pathogenic mechanism of interleukin 6(IL-6) in the tubulointerstitial lesions of patients with active lupus nephritis(LN). Methods Uninary IL-6 from 42 active LN cases and the renal tissues sections from 15 of them were examined by performing ELISA and in situ hybridization with IL-6 cDNA probe labelled by digoxingenin respectively. Results In 36/42 (85.7%) of active LN patients, uninary IL-6 content was higher than or equal to 5 pg/mg ,cr, while no one case did in normal controls. Uninary IL-6 content was highly correlated with the uninary ft-m and NAG excretion respectively. In normal tubules and interstitium, IL-6 mRNA was undetectable. For all sections from 15 active cases, IL-6 mRNA was found in tubular and interstitial cells, and the distribution correlated with the extent of tubulointerstitial lesions. Conclusion IL-6 is probably involved in tubulointerstitial lesions of patients with active lupus nephritis.

ABSTRACT

Objective To observe the effect of ligustrazine (TMP) on the expression of connective tissue growth factor (CTGF) and osteopontin (OPN) in the renal tubulointerstitium of diabetic nephropathy (DN) rats. Methods Wistar rats were used to establish the models through intraperitoneal injection of onedose streptozotocin (STZ) and nephrectomy. Rats were divided into 5 groups at random: model group, blank control group, Lotensin (1.7 mg? kg- 1? d- 1) group, of high-and lowdosage of TMP groups (150, 50 mg? kg- 1? d- 1). Except the rats in blank group and model group, the rats received corresponding medicines according to the experimental design for 12 weeks.Volume of urine protein within 24 hours was examined in the 4th, the 8th and the 12 th week, OPN mRNA expression in the renal tubulointerstitium was detected by RT- PCR and CTGF was detected by means of immunohistochemistry. Aim To observe the effect of mycophenolate mofetil(MMF) on expressions of MCP-1 and CD68 in renal tubulointerstitial injury of diabetic rats and explore the mechanism of MMF′s protective role.Methods Diabetes was induced in uninephrectomized male Wistar rats by peritoneal injection of STZ (65 mg?kg-1). Rats were randomly divided into three groups:control group (NC),diabetic group (DM) and treated group (DM+MMF) with MMF(15 mg?kg-1?d-1).This study lasted for 8 weeks. 24 h urinary protein,blood glucose and the ratio of left kidney weight/body weight were determined after 8 weeks.The renal tubulointerstitial morphological change was observed,immunohistochemical method was used to analyze expressions of MCP-1 protein and CD68.
