ABSTRACT

Objective To address the absence of matrix specified for the determination of intrinsic uniformity in the current standard, and to investigate the effect of source distances on intrinsic spatial linearity, the intrinsic uniformity and intrinsic spatial linearity of 16 probes in eight SPECT devices were measured and analyzed with different matrices and source distances, in order to determine the optimal measurement conditions. Methods According to the standard Specification for Testing of Quality Control in Gamma Cameras and Single Photon Emission Computed Tomograph (SPECT) (WS 523—2019), the intrinsic uniformity was measured using 64 × 64 and 256 × 256 matrices and the intrinsic spatial linearity was measured using of 1.7 and 3 m source distances. Results When intrinsic uniformity was measured with the 64 × 64 matrix, more than 50% of the probes showed lower values. When intrinsic spatial linearity was measured with the 3 m source distance, more probes showed lower values. Conclusion The 64 × 64 matrix is recommended for the determination of intrinsic uniformity and a source distance of >5 FOV is recommended for the measurement of intrinsic spatial linearity.

ABSTRACT

A rich diversity of wild medicinal plant resources is distributed in China, but the breeding of new plant varieties of Chinese medicinal plants started late and the breeding level is relatively weak. Chinese medicinal plant resources are the foundation for new varieties breeding, and the plant variety rights(PVP) are of great significance for the protection and development of germplasm resources. However, most Chinese medicinal plants do not have a distinctness, uniformity, and stability(DUS) testing guideline. The Ministry of Agriculture and Rural Affairs has put 191 plant species(genera) on protection lists, of which only 30 are medicinal species(genera). At the same time, only 29 of 293 species(genera) plants in the Protection List of New Plant Varieties of the People's Republic of China(Forest and Grass) belong to Chinese medicinal plants. The number of PVP applications and authorization of Chinese medicinal plants is rare, and the composition of variety is unreasonable. Up to now, 29 species(genera) of DUS test guidelines for Chinese medicinal plants have been developed. Some basic problems in the breeding of new varieties of Chinese medicinal plants have appeared, such as the small number of new varieties and insufficient utilization of Chinese medicinal plant resources. This paper reviewed the current situation of breeding of new varieties of Chinese medicinal plants and the research progress of DUS test guidelines in China and discussed the application of biotechnology in the field of Chinese medicinal plant breeding and the existing problems in DUS testing. This paper guides the further application of DUS to protect and utilize the germplasm resources of Chinese medicinal plants.

ABSTRACT

Targeting the poor powder characteristics of the contents in Hewei Jiangni Capsules, this study characterized the powder properties of the contents and employed particle design technique for improving the content quality. The content composite particles of Hewei Jiangni Capsules prepared by the particle design technique were evaluated by scanning electron microscopy(SEM), followed by infrared ray(IR), content uniformity, and in vitro dissolution detection. It was found that there was a good correlation between the crushed particle size of slices and the crushing time, and the calcined Haematitum was responsible for the poor content uniformity. After the fine powder of calcined Haematitum was super-finely ground for 8.5 min and those of the other contents in the capsule for 1 min, they were prepared into the composite particles, whose property characterizations were compared with those of the physical mixtures. The content uniformity of the prepared composite particles was significantly improved, and the preparation process was stable and reliable. The adoption of particle design technology to correct the poor uniformity of the physical mixture, solve the pharmaceutical defects of Hewei Jiangni Capsules, and improve the quality of prescriptions has provided important reference for the clinical application and development of Chinese medicinal preparations.

ABSTRACT

Objective:To discuss the problems existing in the implementation of the Chinese health industry standard WS 523-2019 by testing SPECT device. Methods:Under the WS 523-2019 standards, a total of 10 SPECT devices were tested with regard to their SPECT reconstructed spatial resolution (SRSR), system planar sensitivity (SPS), system spatial resolution (SSR), whole body system spatial resolution (WSSR), intrinsic uniformity (IU), intrinsic count rate performance (ICR), intrinsic spatial resolution (ISR) and intrinsic spatial linearity(ISL).Result:Under the requirements of WS 523-2019 standards for qualified limits, there are 3 devices with ISL unqualified and the rest of the performances qualified. The new standards basically can meet the clinical requirements and reflected the overall performance of SPECT.Conclusions:The distance between the radiation source and the surface of the detector has great influence on the spatial resolution. In the measurement of ISL, there must be a lead grid separately in the x and y directions. The lead grids with the parallel slits shall be positioned on the detector with the center slit centered on the detector. It is suggested to add rotation center in the new standards.

ABSTRACT

Spatial distribution uniformity is the critical quality attribute(CQA) of Ginkgo Leaves Tablets, a variety of big brand traditional Chinese medicine. The evaluation of the spatial distribution uniformity of active pharmaceutical ingredients(APIs) in Ginkgo Leaves Tablets is important in ensuring their stable and controllable quality. In this study, hyperspectral imaging technology was used to construct the spatial distribution map of API concentration based on three prediction models, further to realize the visualization research on the spatial distribution uniformity of Ginkgo Leaves Tablets. The region of interest(ROI) was selected from each Ginkgo Leaves Tablet, with length and width of 50 pixels, and a total of 2 500 pixels. Each pixel had 288 spectral channels, and the number of content prediction data could reach 1×10~5 for a single sample. The results of the three models showed that the Partial Least Squares(PLS) model had the highest prediction accuracy, with calibration set determination coefficient R_(pre)~2 of 0.987, prediction set determination coefficient R_(pre)~2 of 0.942, root mean square error of calibration(RMSEC) of 0.160%, and root mean square error of prediction(RMSEP) of 0.588%. The classical least-squares(CLS) model had a greater prediction error, with the RMSEP of 0.867%. Multivariate Curve Resolution-Alternating Least Square(MCR-ALS) model showed the worst predictive ability among the three models, and it couldn't realize content prediction. Based on the prediction results of PLS and CLS models, the spatial distribution map of APIs concentration was obtained through three-dimensional data reconstruction. Furthermore, histogram method was used to evaluate the spatial distribution uniformity of API. The data showed that the spatial distribution of APIs in Ginkgo Leaves Tablets was relatively uniform. The study explored the feasibility of visualization of spatial distribution of Ginkgo Leaves Tablets based on three models. The results showed that PLS model had the highest prediction accuracy, and MCR-ALS model had the lowest prediction accuracy. The research results could provide a new strategy for the visualization method of quality control of Ginkgo Leaves Tablets.

ABSTRACT

The spatial distribution uniformity of valuable medicines is the critical quality attribute in the process control of Tongren Niuhuang Qingxin Pills. With the real world sample of the mixed end-point powder of Tongren Niuhuang Qingxin Pills as the research object, hyperspectral imaging technology was used to collect a total of 32 400 data points with a size of 180 pix×180 pix. Spectral angle matching(SAM), classical least squares and mixed tuned matched filtering(MTMF) were used to identify the spatial distribution of rare medicines. MTMF model showed higher identification accuracy, therefore the spatial distribution of the blended intermediates was identified based on the MTMF model. The histogram method was also used to evaluate the spatial distribution uniformity of rare medicines. The results showed that the standard deviation was 4.78, 6.5, 3.48, 1.96, and 3.00 respectively for artificial bezoar, artificial musk, Borneol, Antelope horn and Buffalo horn; the variance was 22.8, 42.3, 12.1, 3.82, and 9.00, and the skewness was 1.26, 1.71, 0.06,-0.86, and 1.04, respectively. The final results showed that the most even blending was achieved in concentrated powder of Borneol, Antelope horn and Buffalo horn, followed by artificial bezoar, and last artificial musk. A visualization method was established for quality attributes of distribution uniformity in blending process of Tongren Niuhuang Qingxin Pills. It could provide evidences of quality control methods in the mixing process of big brand traditional Chinese medicine.

ABSTRACT

Cotton has received attention of geneticists since more than a century. Gossypium hirsutum, the predominantly cultivated cotton species worldwide, has a narrow genetic base. It is important to broaden its genetic base through introgression of novel alleles from related species. Here, we report the development and characterization of a backcross population derived from the hybridization of a ‘synthetic’ (derived by crossing and chromosomal doubling of nonprogenitor Gossypium species) and natural tetraploid upland cotton. ‘Synthetic’ was observed to be male-sterile and thus, was used as the female parent. A total of 7434 flowers were pollinated to obtain 1868 BC1F1 seeds by direct and reciprocal crosses. Characterization of the experimental plant material was conducted in the field for several morphological traits such as pubescence on the stem, leaf, petiole and bract, presence/absence of petal spot, petal margin colouration and stamen filament colouration. Genetic analysis revealed that petal margin colouration phenotype was governed by a single dominant gene, whereas the petal spot and filament colouration phenotypes manifested segregation distortion. None of the BC1F1 plants was devoid of trichomes thus demonstrating that presence of trichomes is dominant over their absence. Modern upland cotton cultivars are usually devoid of petal spot, petal margin colouration and stamen filament colouration. These floral anthocyanin pigmentation characteristics, if fixed in the cotton germplasm, may serve as diagnostic features for the identification of cultivars during DUS testing as well as in the maintenance breeding programmes

ABSTRACT

ABSTRACT: The objective of this research was to determine the optimal plot size and the number of replications to evaluate the fresh matter of ryegrass sown to haul. Twenty uniformity trials were conducted, each trial with 16 basic experimental units (BEU) of 0.5 m2. At 117, 118 and 119 days after sowing, the fresh matter of ryegrass in the BEUs of 5, 10 and 5 uniformity trials, respectively, were determined. The optimal plot size was determined by the maximum curvature method of the variation coefficient model. Next, the replications number was determined in scenarios formed by combinations of i treatments (i = 3, 4, ... 50) and d minimum differences between means of treatments to be detected as significant at 5% of probability by the Tukey test, expressed in experimental mean percentage (d = 10, 11, ... 20%). The optimal plot size to determine the fresh matter of ryegrass seeded at the haul is 2.19 m2, with a variation coefficient of 9.79%. To identify as significant at 5% probability, by the Tukey test, differences between treatment means of 20%, are required five, six, seven and eight replications, respectively, in ryegrass experiments with up to 5, 10, 20 and 50 treatments.

ABSTRACT

ABSTRACT: The construction of the hydroelectric power plant of Ilha Solteira, in state of São Paulo, was initiated in the 1960s, when an average, 8.60 m of soil depth was removed, resulting in a degraded area. A plan for the recovery of the area started in 2005 in Selvíria /MS with the use of plant species adapted to the Cerrado biome. This study aimed to evaluate the soil macrofauna of an area under recovery by using different types of soil cover (1- bare soil (control); 2- native Cerrado vegetation; 3- specie Astronium fraxinifolium; 4- Astronium fraxinifolium + Canavalia ensiformis; 5- Astronium fraxinifolium + Raphanus sativus; 6- Astronium fraxinifolium + Brachiaria decumbens + sewage sludge). Soil macrofauna was evaluated in 2005, 2006 and 2007 using the direct collection method and manual counting. Number of species, diversity and uniformity index were determined. Principal component analysis (PCA) and cluster analysis were used for data interpretation. Results showed that treatment 6 (Astronium fraxinifolium+ Brachiaria decumbens+ sewage sludge) increased the soil macrofauna population by approximately 4 to 6 times more than the other types of cover after three years of evaluation. And the PCA and cluster analysis showed the approximation of the data between treatment 6 and Cerrado, which represents the most appropriate treatment for the recovery of the degraded soil.

ABSTRACT

ABSTRACT: The aim of this study was to determine the optimal plot size for evaluating the fresh weight of black oats (Avena strigosa Schreb) and the common vetch (Vicia sativa L.) in scenarios comprising combinations of the number of treatments, number of replications and levels of precision. Fifteen uniformity trials were conducted with single-crop and intercropped black oats and vetch. Fresh weight was evaluated in 540 basic experimental units (BEU), each of 1 m × 1 m (36 BEU per trial). The Smith index of soil heterogeneity (1938) was estimated. Plot size was determined using the HATHEWAY method (1961), in scenarios comprising combinations of i treatments (i = 5, 10, 15 and 20), r replications (r = 3, 4, 5, 6, 7 and 8) and d levels of precision (d = 2%, 4%, 6%, 8%, 10%, 12%, 14%, 16%, 18% and 20%). To evaluate the fresh weight of monocropped or intercropped black oats and vetch in a completely randomized or randomized complete block design, with from 5 to 20 treatments and five replications, plots of 10 m2 are sufficient to identify, at a probability of 0.05, significant differences between treatments of 10% of the overall mean value of the experiment.

ABSTRACT

Objective: To explore the law of temperature change in hot air drying process, and provide a theoretical basis for improving the drying efficiency and uniformity of Erzhi Pills (EP). Methods: Heat and mass transfer models were established to simulate the temperature distribution of the drying process of EP with no hole bottom and screen hole bottom based on COMSOL Multiphysics. The drying process of EP layer at 60, 80 and 100 ℃ was studied. The effective water diffusion coefficient (Deff) of EP layer was calculated by Fick’s second law plate model. A probe temperature sensor was used to monitor the temperature changes of the intermediate surface and interior of EP layer during the drying process to verify the drying model. Results: The surface and internal temperature of EP layer showed a trend of first sharp rise and then slow rise in the drying process of different hot air temperature. The inner temperature range of the drying model with no hole bottom and screen hole bottom was 55.3-56.0 ℃ and 57.5-57.7 ℃ respectively. The uniformity of drying temperature of EP can be improved by using drying plate with screen hole. The Deff range of the two model experiments were 0.76 × 10-7-2.82 × 10-7 m2/s and 1.15 × 10-7-3.94 × 10-7 m2/s, respectively. The experimental results showed that the model could well reflect the temperature variation of EP layer drying process within a certain drying temperature range. Conclusion: The change of drying temperature was directly reflected through the reliable hot air drying model of EP layer. The study provides reference for the optimization of EP drying process.

ABSTRACT

Objective: To prepare the new diclofenac potassium cataplasms, establish a quality research method and evaluate the preparation quality by the established method. Methods: A high performance liquid chromatography(HPLC)was performed on an ODS column(the column temperature 35℃)using the methanol-4% glacial acetic acid solution(80: 20, V/V)as mobile phase, and the detection wavelength was set at 276 nm. Diclofenac potassium was extracted with methanol, and the adhesion, content and uniformity of potassium diclofenac were measured. The release of diclofenac potassium from the cataplasms was determined in accordance with the fourth method of the XD release methods in the Appendix of the Chinese Pharmacopoeia. Results: The average maximum number of steel ball stuck in the cataplasms in the initial adhesion test was No. 6. Under the HPLC conditions, potassium diclofenac showed good linearity within the concentration range of 400-800 μg/ml, with the average sample recovery rate 1.33 % and RSD< 1.93%(n=6). The methodological studies for the drug release test for the diclofenac potassium cataplasms showed that the diclofenac potassium showed a good linearity within the range of 1-50 μg/ml in the drug release test, and the precision and recovery well satisfied the requirements of Pharmacopoeia. The content uniformity of the cataplasms was in accordance with the Pharmacopoeia. The release amounts of the cataplasms in 2, 5 and 8 hours were 20%-45%, 40%-80% and more than 70% of the labeled amount, respectively, and the release curve followed the first-order release equation. Conclusion: The established HPLC method is sensitive, accurate, easily operable and reproducible, which could be used for the quality control of diclofenac potassium cataplasms. The prepared diclofenac potassium cataplasms were of uniform content and showed characteristics of the sustained release, which is expected to be developed to a new preparation of diclofenac potassium.

ABSTRACT

Principal components is a multivariate statistical technique of synthesis of information or reduction of the number of variables, with the potential to expose non-obvious relationships between characters, thus contributing to a more efficient interpretation of the information contained in a set of data. Its application to six traits associated to sexual maturity (age and body weight at first egg, weight of the first and the first ten eggs, number of days required to lay the first ten eggs and coefficient of variation of their weights) allowed to characterize the structure of intra-population variability in hens of the experimental three-way cross Campero Casilda. The first two components -PC1 and PC2- explained 56% of the total phenotypic variance. The joint consideration of both components allowed identifying a group of birds characterized for being older and heavier at sexual maturity, with a more regular onset of the productive phase, with uniform and heavier eggs, a conjunction of desirable characters for the beginning of the cycle. The importance of this identification lies in the recognized usefulness of principal components analysis as a strategy to generate biological selective indexes.

ABSTRACT

ABSTRACT: This study aimed to evaluate the optimum plot size for the papaya crop by using the modified maximum curvature method under soil and climatic conditions of the Recôncavo Baiano. The experiment comprised a uniformity test using the CNPMF-L78 strain developed by Embrapa Mandioca and Fruticultura, planted at a spacing of 3 m x 2 m, with 16 central rows and 22 plants per row, totaling 352 plants and an area of 2,112 m2. The following parameters were evaluated: plant height and diameter; height of insertion of the first fruits; precocity; number of commercial fruits per plant; productivity; length, diameter, weight, firmness, internal cavity diameter, pulp thickness, and soluble fruit solids. Each plant was considered as a basic unit, with an area of 6 m2, thus making up 352 basic units whose adjacent units were combined to form 11 pre-established parcel arrangements with rectangular and row formats. The optimal plot size is seven plants perpendicular to the crop rows, that is, seven rows with one plant in each row, corresponding to the area of 42 m2, considering spacing of 3 m between rows and 2 m between papaya plants in the soil and climatic conditions of the Recôncavo Baiano.

ABSTRACT

Objective: To establish a quantitative analysis model for online monitoring of the blending uniformity of hydroxychloroquine sulfate raw and auxiliary materials, so as to accurately and quickly determine the blending endpoint. Methods: A mixture of excipients and hydroxychloroquine sulfate raw material was prepared with a labeling percentage of 70%-130%. The near-infrared spectrum was collected; and the standard normal variate transformation and frst derivative by Norris smoothing were used for spectra pretreating, with 8 372-9 045 cm-1, 5 616-6 058 cm 1 used as the spectral bands. A quantitative analysis model was developed using partial least squares regression. The quantitative analysis model was used to predict the labeling percentage of hydroxychloroquine sulfate in the blending process of raw and auxiliary materials, and the blending endpoint was verified by high-performance liquid chromatography (HPLC) analysis. Results: Five primary factors were used to establish the model. The root mean square error of calibration was 0.96 and the correlation coefficient of calibration set (Rc) was 0.998. The root mean square error of prediction was 0.97 and the correlation coefficient of validation set (Rp) was 0.998. The root mean square error of cross-validation was 1.56 and the correlation coefficient of cross-validation (Rcv) was 0.995. The prediction results of the near-infrared model was consistent with the results of HPLC verification. Conclusion: The near-infrared model in this study can be used for online quantitative analysis of the blending uniformity of hydroxychloroquine sulfate, and it can accurately and quickly determine the blending endpoint.

ABSTRACT

Objective:To establish a mathematical model of total statistical moment, information amount and one-time steady-state feeding quantity of HPLC fingerprint of traditional Chinese medicines (TCMs) and their classical prescriptions, and to carry out experimental verification. Method:The total statistical moment principle, Hardy-Weinberg equilibrium principle and one-time steady-state feeding quantity calculation method were used to study the total statistical moment parameters and one-time steady-state feeding quantity of 15 batches of original medicinal materials, decoction pieces and compound preparations of Taohong Siwutang, these original medicinal materials were from three producing areas. Result:In addition to the total first-order moment, RSD of total zero-order moment, total second-order moment, information entropy and information amount of each medicinal material, decoction piece and compound were all greater than 10%. In the case of single original medicinal material, except for Angelicae Sinensis Radix (142.34 kg), the one-time steady-state feeding quantity calculated according to the RSD change of the accumulated information amount of the whole prescription was the largest (59.10 kg), and according to ratio of the whole prescription, one-time steady-state feeding quantity was the largest (958.57 kg). In the case of processed products, the one-time steady-state feeding quantity of the whole prescription was the largest (73.18 kg) except for Carthami Flos processed with wine (83.28 kg), while according to the whole prescription, one-time steady-state feeding quantity of each processed products was the same as that of the original medicinal material. The one-time steady-state feeding quantity of the compound consisted of the original medicinal materials or processed products was often higher than that of the single medicinal material. Conclusion:The components of TCM are controlled by genetic polymorphism, the chromatographic characteristics (types of components) of Chinese medicine is similar and the intensity of action (content of components) is different. The stability of total first-order moment and information entropy indicates that the overall chemical structure of TCM multi-component system has a stable imprinting trend. The one-time steady-state feeding quantities of each medicinal material in Taohong Siwutang and this compound are far greater than the prescription amount, which can provide reference for the industrial production.

ABSTRACT

Objective To explore the feasibility of application of the Monte Carlo method to simulate the whole body dose distribution in patients with total body X (γ) ray irradiation by comparing the actual measurement results. Methods A Monte Carlo model of a 6 MV Elekta Synergy Clinical linear accelerator was established by MCNPX.According to the relationship between the CT value and the density of the material, the CT of the ATOM physical phantom was converted into a voxel phantom for MCNPX calculation. The dose distribution of the whole body was simulated in the total body X (γ) ray irradiation. The simulated results were compared with the measurement values of the thermoluminescence dosimetry at different positions in the ATOM physical phantom to analyze the differences. Results The difference between the depth dose curve and the off-axis dose curve and the actual measurement values calculated by the 6 MV accelerator treatment head model in the water tank was less than 2%,with the maximum dose depth of approximately 1.5 cm and field size of 10 cm× 10 cm,which were consistent with the actual measurement values. The maximum difference between the simulated results at different locations in the body and the thermoluminescence dosimeter was approximately 4%,and the simulated results of MCNPX were almost in good agreement with the results of thermoluminescence. Conclusions The whole body dose distribution in patient with total body X (γ) ray irradiation can be accurately simulated by MCNPX.Monte Carlo simulation makes it possible to optimize the uniformity of the total body dose during the total body irradiation process.
