ABSTRACT

Objective@#To evaluate the clinical efficacy of invisible orthodontic appliances without brackets for the distal movement of maxillary molars to improve the ability of orthodontists to predict treatment outcomes.@*Methods@#Web of Science, Cochrane Library, Embase, PubMed, Wanfang Database, CNKI Database, and VIP Database were searched for studies investigating the efficacy of invisible orthodontic appliances for distal movement of maxillary molars in adult patients and published from database inception to August 1, 2023. A total of three researchers screened the studies and evaluated their quality and conducted a meta-analysis of those that met quality standards.@*Results@#This study included 13 pre- and postcontrol trials with a total sample size of 281 patients. The meta-analysis revealed no significant differences in the sagittal or vertical parameters of the jawbone after treatment when compared with those before treatment (P>0.05). The displacement of the first molar was MD=-2.34, 95% CI (-2.83, -1.85); the displacement was MD=-0.95, 95% CI (-1.34, -0.56); and the inclination was MD=-2.51, 95% CI (-3.56, -1.46). There was a statistically significant difference in the change in sagittal, vertical, and axial tilt of the first molar before and after treatment. After treatment, the average adduction distance of the incisors was MD=-0.82, 95% CI (-1.54, -0.09), and the decrease in lip inclination was MD=-1.61, 95% CI (-2.86, -0.36); these values were significantly different from those before treatment (P<0.05).@*Conclusion@#Invisible orthodontic appliances can effectively move the upper molars in a distal direction and control the vertical position of the molars. When the molars move further away, there is some degree of compression and distal tilt movement, which is beneficial for patients with high angles. The sagittal movement of incisors is beneficial for improving the patient's profile.

ABSTRACT

SUMMARY: Respiration and water-liquid transportation are controlled by many factors in the lung. The aim of this study was to explore the structure and proteins expression in lungs of Phrynocephalus vlangalii by means of gross anatomy, light microscope observation, scanning electron microscope and immunohistochemistry. Results show that there were many alveoli in the lung and the walls of alveoli and capillaries were very thin. The inner surface of the lung was divided into many cystic chambers by reticular diaphragm, and the network of pulmonary capillaries was dense. Immunohistochemistry showed that AQP1 was mainly expressed in the epithelium of interstitial bronchi, parabronchiole endothelium, capillary endothelium and alveolar epithelial cells. VIP positive nerve fibers are mainly distributed in trachea, bronchial smooth muscle layer, the walls of pulmonary vessels and bronchial vessels and around submucosal glands. CECR2 is distributed in peripheral capillaries and small. Investigations of structure and proteins biology could be relevant with the adaptive strategy to drought and hypoxia environment in Phrynocephalus vlangalii.


La respiración y el transporte de agua y líquido están controlados en el pulmón por muchos factores. El objetivo de este estudio fue explorar la estructura y la expresión de proteínas en los pulmones de Phrynocephalus vlangalii por medio de la anatomía macroscópica, observación con microscopio óptico, microscopio electrónico de barrido e inmunohistoquímica. Los resultados muestran que había muchos alvéolos en el pulmón y que las paredes de los alvéolos y de los capilares eran muy delgadas. La superficie interna del pulmón estaba dividida en cámaras quísticas por el diafragma reticular y se observó una densa red de capilares pulmonares. La inmunohistoquímica mostró que AQP1 se expresaba principalmente en el epitelio de los bronquios intersticiales, el endotelio parabronquial, el endotelio capilar y las células epiteliales alveolares. Las fibras nerviosas VIP positivas se distribuyen principalmente en la tráquea, la capa de músculo liso bronquial, las paredes de los vasos pulmonares y los vasos bronquiales y alrededor de las glándulas submucosas. CECR2 se distribuye en pequeño capilares periféricos. Las investigaciones de la biología de la estructura y las proteínas podrían ser relevantes con la estrategia de adaptación al entorno de sequía e hipoxia en Phrynocephalus vlangalii.

ABSTRACT

ObjectiveTo investigate the protective effect of Guiqi Baizhu prescription combined with oxaliplatin on the intestinal barrier of tumor-bearing mice with gastric cancer by regulating downstream aquaporin 3 (AQP3) and aquaporin 4 (AQP4) through the vasoactive intestinal peptide (VIP)/cyclic adenosine monophosphate (cAMP)/protein kinase A (PKA) signaling pathway. MethodThe gastric cancer cell lines MFC with a density of 1×107/mL were prepared into cell suspension. The tumor-bearing mouse model of gastric cancer was established by inoculating 0.2 mL cell suspension under the right axilla of mice. After successful modeling, mice were randomly divided into 5 groups, namely, model group, oxaliplatin group (10 mg·kg-1), and high, medium, and low-dose oxaliplatin + Guiqi Baizhu prescription groups (17.68, 8.84, 4.42 g·kg-1), with 10 mice in each group, and the remaining 10 mice were set as a blank group. Mice in each group were treated with Chinese medicine, oxaliplatin, or normal saline by gavage or intraperitoneal injection for 14 d. The next day after the last dose, blood was taken from the eyeball to separate serum and take colonic samples. Hematoxylin-eosin (HE) staining was used to observe the changes in tissue morphology. The content of D-lactate acid (D-LA) and diamine oxidase (DAO) in the serum was determined by enzyme-linked immunosorbent assay (ELISA). The mRNA and protein expressions of VIP, cAMP, PKA, AQP3, and AQP4 were detected by Real-time quantitative polymerase chain reaction (Real-time PCR) and Western blot, respectively. ResultCompared with the blank group, the model group showed edema in the colonic submucosa, disordered arrangement of intestinal glands in the mucosal layer, loss of goblet cells, infiltration of inflammatory cells, and villus shedding. However, there were different degrees of improvement in each administration group. As compared with the blank group, the serum levels of DAO and D-LA in the model group were significantly increased (P<0.01). As compared with the model group, the levels of DAO and D-LA in the high-dose oxaliplatin + Guiqi Baizhu prescription group and the level of D-LA in the medium-dose oxaliplatin + Guiqi Baizhu prescription group were decreased (P<0.05, P<0.01). As compared with the oxaliplatin group, the levels of D-LA in the high and medium-dose oxaliplatin + Guiqi Baizhu prescription groups were decreased (P<0.05), and the levels of DAO and D-LA in other administration groups were decreased as well, but the difference had no statistical significance. As compared with the blank group, the mRNA and protein expression levels of VIP, cAMP, PKA, AQP3, and AQP4 in the model group were significantly decreased (P<0.05, P<0.01). As compared with the model group, the mRNA and protein expression levels of VIP, cAMP, PKA, AQP3, and AQP4 in each administration group were increased, and those in the high-dose oxaliplatin + Guiqi Baizhu prescription group were significantly increased (P<0.05, P<0.01), while the protein expression level of cAMP in the medium-dose oxaliplatin + Guiqi Baizhu prescription group were increased (P<0.05). As compared with the oxaliplatin group, the protein expression levels of cAMP in the high-dose oxaliplatin + Guiqi Baizhu prescription group were increased (P<0.05), and the mRNA and protein expressions of these indexes in the other groups were also increased but the differences were not statistically significant. ConclusionGuiqi Baizhu prescription combined with oxaliplatin can regulate AQP3 and AQP4 through the VIP/cAMP/PKA signaling pathway to protect the intestinal barrier of tumor-bearing mice with gastric cancer.

ABSTRACT

ObjectiveTo investigate the effects of spleen-Yin deficiency on gastrointestinal absorption, water metabolism and intestinal flora in rats with spleen-Yin deficiency syndrome. MethodA rat model of spleen-Yin deficiency syndrome was established by using the composite factors, including irregular meat and vegetable diet, weight-bearing fatigue swimming and gavage with warm-heat injury-Yin drugs. The changes of body weight, food intake, water intake and duration of swimming in the blank and model groups were observed. Hematoxylin-eosin(HE) staining was used to observe the histopathological damage of the stomach and colon. Urinary excretion rate of D-xylose was determined by phloroglucinol method. The content of gastrin(GAS) in serum was determined by enzyme-linked immunosorbent assay(ELISA). The relative expression levels of vasoactive intestinal peptide(VIP), aquaporin 3(AQP3) and AQP4 in gastric tissues were detected by Western blot. The relative mRNA expression levels of VIP, AQP3 and AQP4 in gastric tissues were detected by Real-time fluorescence quantitative polymerase chain reaction(Real-time PCR), and the changes of intestinal flora were analyzed by 16S rDNA sequencing. ResultCompared with the blank group, the results of general physical signs showed that the body weight and food intake of rats in the model group were significantly decreased, the water intake was significantly increased(P<0.05, P<0.01), and the duration of swimming was significantly decreased(P<0.01). Pathological examination results showed that in the mucosa of gastric tissues of rats in the model group appeared to be misaligned, the mucosa of colonic tissues could be seen to be obviously thinned or mutilated, and the epithelial cells appeared to be necrotic or even exfoliated. Compared with the blank group, the urinary D-xylose excretion rate of rats in the model group was significantly decreased(P<0.01), and the serum GAS content was significantly decreased(P<0.05). Compared with the blank group, Western blot results showed that the relative expression level of VIP protein in gastric tissues of rats in the model group was significantly decreased, while the relative expression levels of AQP4 and AQP3 proteins were significantly increased(P<0.01). Compared with the blank group, Real-time PCR results showed that the relative expression level of VIP mRNA in gastric tissues of rats in the model group was significantly decreased(P<0.01), and the relative mRNA expression levels of AQP3 and AQP4 were significantly increased(P<0.05, P<0.01). Compared with the blank group, the results of intestinal flora analysis showed that the number of operational taxonomic units(OTUs) and α-diversity increased and β-diversity decreased significantly in the model group, the abundance of Porphyromonadaceae was increased significantly, and the abundance of Oscillibacter_ruminantium was decreased significantly(P<0.05). Spearman correlation analysis showed that Porphyromonadaceae was significantly positively correlated with AQP4 protein level, while Oscillibacter_ruminantium was significantly positively correlated with VIP protein level, and negatively correlated with AQP3 and AQP4 protein levels(P<0.05). Linear discriminant analysis effect size(LEfSe) analysis results showed that there were significant differences in a variety of intestinal bacteria between groups, and the intestinal bacteria of the model group were significantly enriched in the phylum/order/family/genus of Elusimicrobia, Betaproteobacteria, Burkholderiales, Sutterellaceae and Parasutterella(P<0.05). ConclusionSpleen-Yin deficiency syndrome can weaken the digestion and absorption capacity of gastrointestinal tract, and cause the disturbance of water metabolism and intestinal flora. AQP4, AQP3 and VIP protein levels of gastric mucosa are closely related to Porphyromonadaceae and Oscillibacter_ruminantium. And AQP4, AQP3 and VIP may be involved in the regulation of intestinal flora in order to affect the physiological function of spleen governing transportation and transformation.

ABSTRACT

ObjectiveTo investigate the effect and mechanism of Aconiti Lateralis Radix Praeparata-Cinnamomi Cortex in regulating the intestinal function in the rat model of slow transit constipation (STC) due to yang deficiency via the vasoactive intestinal peptide (VIP)/cathelicidin antimicrobial peptide (cAMP)/protein kinase A (PKA)/aquaporin (AQP) pathway. MethodSD rats were randomized into 6 groups (n=6), including a control group, a model group, high-, medium-, and low-dose Aconiti Lateralis Radix Praeparata-Cinnamomi Cortex groups, and a prucalopride group. Other groups except the control group were treated with loperamide hydrochloride combined with ice water by gavage for the modeling of STC due to yang deficiency. The number of fecal pellets, time to the first black stool defecation, fecal water content, intestinal propulsion rate, and score of fecal properties were recorded in each group. At the end of the treatment, the colon was stained with hematoxylin-eosin (HE) to reveal the histopathological changes and Alcian blue/periodic acid-Schiff (AB-PAS) to reveal the secretion of colonic mucus. The enzyme-linked immunosorbent assay (ELISA) was employed to measure the level of VIP in the serum. The mRNA level of AQP in the colon was measured by polymerase chain reaction (Real-time PCR). Immunohistochemical staining was performed to observe the expression of AQPs in the colon and kidney tissues. Western blot was performed to determine the protein levels of cAMP, PKA, and VIP in the colon tissue. ResultCompared with the control group, the model group had longer time to the first black stool defecation, reduced fecal pellets and water content, reduced Bristol Stool Form Scale score and intestinal propulsion rate, and constipation aggravated(P<0.01). Moreover, increased the intestinal lesions, reduced the mucus secretion, reduce the serum VIP level, up-regulated the expression levels of AQP1 in the colon and kidney tissues, inhibited the expression of AQP3 and AQP9(P<0.01)., and down-regulated the protein levels of cAMP, PKA, and VIP in the colon tissue. Compared with the model group, the high-dose Aconiti Lateralis Radix Praeparata-Cinnamomi Cortex group had shortened time to the first black stool defecation, increased fecal pellets and water content, increased Bristol Stool Form Scale score and intestinal propulsion rate, and alleviated constipation symptoms. Moreover, high-dose Aconiti Lateralis Radix Praeparata-Cinnamomi Cortex reduced the intestinal lesions, increased the mucus secretion, elevated the serum VIP level(P<0.01)., down-regulated the expression levels of AQP1 in the colon and kidney tissues, promoted the expression of AQP3 and AQP9(P<0.05,P<0.01), and up-regulated the protein levels of cAMP, PKA, and VIP in the colon tissue. The medium- and low-dose groups had weaker effect than the high-dose group(P<0.01). ConclusionHigh-dose Aconiti Lateralis Radix Praeparata-Cinnamomi Cortex can improve the intestinal motility and balance the intestinal water and fluid metabolism by up-regulating the VIP/cAMP/PKA/AQP pathway, thereby mitigating the constipation symptoms in the rat model of slow transit constipation due to yang deficiency.

ABSTRACT

ObjectiveTo explore the mechanism of the combined therapy of lung and intestine (Mahuangtang + Da Chengqitang) in alleviating pulmonary edema in rats with acute lung injury (ALI) induced by lipopolysaccharide (LPS). MethodWistar rats were randomly divided into blank group, model group, low-, medium-, and high-dose groups with combined therapy of lung and intestine, and positive control group. LPS (10 mg·kg-1) was given (ip) to induce ALI in rats. After modeling, the blank group was given normal saline (25 mL·kg-1), the combined therapy of lung and intestine treatment groups were given (ig) low- (5 g·kg-1), medium- (7.5 g·kg-1), and high-dose (10 g·kg-1) Mahuangtang and Da Chengqitang, and the positive control group was given dexamethasone (5 mg·kg-1). Medications were administered 0, 8, and 16 h after LPS injection for 3 times. Then lung tissue and serum were collected after administration. The lung tissues were stained with haematoxylin-eosin (HE), and the pulmonary edema score was evaluated. The dry/wet (D/W) weight ratio of lung tissues in each group was measured, and the content of serum vasoactive intestinal peptide (VIP) in rats was detected by enzyme-linked immunosorbent assay (ELISA). Western blot was used to detect the protein levels of aquaporin-1 (AQP1), AQP5, VIP, cyclic adenosine monophosphate (cAMP), phosphorylated protein kinase A (p-PKA), and PKA in lung tissues of rats in each group. The level of VIP mRNA in lung tissues of rats was detected by real-time quantitative polymerase chain reaction (Real-time PCR). ResultCompared with the blank group, the model group exhibited obvious lung injury, increased edema score, decreased D/W ratio (P<0.01), declined AQP1, AQP5, cAMP, and p-PKA/PKA in lung tissues (P<0.05, P<0.01), elevated VIP content (P<0.01), and up-regulated levels of VIP protein and mRNA in lung tissues (P<0.05, P<0.01). Compared with the model group, combined therapy of lung and intestine treatment groups showed alleviated lung injury, increased D/W ratio (P<0.01), elevated AQP1, AQP5, VIP, cAMP, and p-PKA/PKA in lung tissues (P<0.05, P<0.01), and up-regulated VIP levels in lung tissues (P<0.05, P<0.01). ConclusionThe combined therapy of lung and intestine can alleviate ALI-induced lung tissue edema, and the mechanism may be related to the activation of the VIP/cAMP/PKA signaling pathway, which further promotes the expression of AQP1 and AQP5 and enhances the water metabolism of lung tissue.

ABSTRACT

Objective:To investigate the curative efficacy of modified Qilang prescription on drug-dependent constipation with Qi and Yin deficiency and the effects on serum vasoactive intestinal peptide (VIP), motilin (MTL), 5-hydroxytryptamine (5-HT), and 5-hydroxytryptamine 4 receptor (5-HT4R). Method:A total of 160 patients diagnosed with drug-dependent constipation were randomly divided into a treatment group (<italic>n</italic>=80, Qilang prescription) and a control group (<italic>n</italic>=80, lactulose oral solution). The treatment lasted for eight weeks. Changes in clinical symptoms, traditional Chinese medicine (TCM) syndrome, and serum VIP, MTL, 5-HT, and 5-HT4R before and after treatment were observed. The clinical efficacies of the two groups were compared. An eight-week follow-up was carried out for the observation of recurrent rate and TCM syndrome. Result:The overall response rate of the treatment group (90.91%) was higher than that (75.00%) of the control group<italic> </italic>(<italic>Z</italic>=-6.514,<italic>P</italic><0.05). There was no significant difference in serum VIP, MTL, 5-HT, and 5-HT4R between the two groups before treatment. After treatment for eight weeks, both groups showed reduced serum VIP level as compared with those before treatment, and the treatment group was inferior to the control group (<italic>P</italic><0.05). The serum MTL levels of the two groups were both higher than those before treatment (<italic>P</italic><0.05), and the treatment group was superior to the control group (<italic>P</italic><0.05). After treatment, the level of 5-HT in the treatment group was higher than that in the control group (<italic>P</italic><0.05). The post-treatment 5-HT4R level in the treatment group slightly increased (<italic>P</italic><0.05), but no significant difference in 5-HT4R levels between the two groups after treatment was observed. During the eight-week follow-up, the recurrence rate in the treatment group was significantly lower than that in the control group at the 2nd and 4th weeks (<italic>P</italic><0.05). There was no significant difference in the recurrence rate between the treatment group [57.14% (40/70)] and the control group [64.81% (35/54)] after eight weeks. Conclusion:Modified Qilang prescription was superior to lactulose in the short- and mid-term efficacy on drug-dependent constipation with Qi and Yin deficiency. No significant difference in the long-term efficacy was observed. The underlying therapeutic mechanism might be related to the regulation of serum VIP, MTL, 5-HT, and 5-HT4R levels.

Phlebitis is the inflammation of vein which is a common complication associated with the use of peripheralintravenous (IV) catheters affecting between 27% and 70% of all patients receiving IV therapy and associationwith up to 10% of S.Aureus bacteremia. Importance factors found to determine phlebitis incidence include thekind of infusion and dwell time of intravenous cannula. The use of the visual infusion phlebitis (VIP) score toolfor the assessment of the early signs of phlebitis along with prompt removal of peripheral intravenous cannulahas been very successful in reducing below the acceptable rate of 5%. However, achieving this goal depends onstrict compliance with guidelines for cannula insertion, documentation and assessment using the VIP tool.This study aimed to increase the use of VIP scoring tool in the intensive care units and wards during a 6 monthsperiod in order to maximize its utility in phlebitis prevention. Nursing audit was carried out, during whichassessment and plan of actions were introduced. The first step aimed to make the nurses aware of VIP scoringtool by the use of posters and training sessions. Second the assessment of nurses’ knowledge and skill relatedto the intravenous cannula insertion and use of VIP score tool for the early detection and prompt action relatedto the care and control of infusion related phlebitis and its documentation.

ABSTRACT

Objective@#To investigate the operating key steps and the initial experience of IUPU (Institute of Urology, Peking University) modified extraperitoneal laparoscopic VIP (Vattikuti institute prostatectomy) technique and the initial follow-up outcomes.@*Methods@#46 patients of prostate cancer underwent the modified extraperitoneal laparoscopic VIP and were reviewed from November 2017 to September 2018. Patients aged 54 to 77 years, with mean age of 65.5 years old. Their preoperative PSA was 0.15 to 69.76 ng/ml with an average of 13.4 ng/ml. All patients were diagnosed by prostate biopsy and Gleason score was 6 to 10 with an average of 7.5. MRI was used to assess their clinical stage before operation. All the modified laparoscopic VIP were done through the extraperitoneal approach by a single surgeon and the follow-up by telephone for the PSA level and continence recovery (urine pad test). The major differences between modified laparoscopic VIP and the routine steps of radical prostatectomy are as follows: use of extraperitoneal approach and traditional laparoscopic operation; after the setup of the extraperitoneal cavity, the bladder neck is separated, isolated and cut down first; the management of DVC: Cut down the super-facial and deep layers of DVC just along the capsule of the prostate apex and it’s unnecessary to suture it. Perioperative data of these patients were collected retrospectively, including operation time, intraoperative blood loss, length of hospital stay, postoperative pathology, and continence outcome after surgery.@*Result@#All 46 patients underwent the IUPU modified extraperitoneal laparoscopic VIP successfully without converting to open surgery. The operation time was 69-143 min with an average of 95.7 min and the estimated blood loss was 20-200 ml with an average of 81.5 ml. It was 3-17 days for the average time to remove the retropubic drainage tube postoperatively, with an average of 4.3 days. The postoperative hospital stay was 3-17 days with an average of 5.3 days. The Foley catheter was removed 14 days postoperatively. Five patients suffered from urine leakage and were managed successfully with conservative treatment. For the postoperative pathological stage, 1 patient was pT2a, 12 patients were pT2c, 12 patients were pT3a and 21 patients were pT3b. The postoperative Gleason score was 6-9 with an average of 7.7. The postoperative follow-up time was 1-11 months with an average of 6.3 months. The PSA was 0.001-0.966 ng/ml with an average of 0.181 ng/ml 1 month postoperatively and the continence rate was 54%(25/46) and 89%(34/38) 1 month and 3 months after operation respectively.@*Conclusions@#The IUPU modified extraperitoneal laparoscopic VIP might be a safe and feasible for the radical prostatectomy. It’s simplified for the operative steps, unnecessary to isolate and suture DVC and doesn’t increase the blood loss. It’s almost left intact for the surrounding tissues of the prostate apex and satisfactory for the early urinary continence. The oncological outcomes are needed to followup further.

Objective To investigate the operating key steps and the initial experience of IUPU (Institute of Urology,Peking University) modified extraperitoneal laparoscopic VIP (Vattikuti institute prostatectomy) technique and the initial follow-up outcomes.Methods 46 patients of prostate cancer underwent the modified extraperitoneal laparoscopic VIP and were reviewed from November 2017 to September 2018.Patients aged 54 to 77 years,with mean age of 65.5 years old.Their preoperative PSA was 0.15 to 69.76 ng/ml with an average of 13.4 ng/ml.All patients were diagnosed by prostate biopsy and Gleason score was 6 to 10 with an average of 7.5.MRI was used to assess their clinical stage before operation.All the modified laparoscopic VIP were done through the extraperitoneal approach by a single surgeon and the follow-up by telephone for the PSA level and continence recovery (urine pad test).The major differences between modified laparoscopic VIP and the routine steps of radical prostatectomy are as follows:use of extraperitoneal approach and traditional laparoscopic operation;after the setup of the extraperitoneal cavity,the bladder neck is separated,isolated and cut down first;the management of DVC:Cut down the super-facial and deep layers of DVC just along the capsule of the prostate apex and it's unnecessary to suture it.Perioperative data of these patients were collected retrospectively,including operation time,intraoperative blood loss,length of hospital stay,postoperative pathology,and continence outcome after surgery.Result All 46 patients underwent the IUPU modified extraperitoneal laparoscopic VIP successfully without converting to open surgery.The operation time was 69-143 min with an average of 95.7 min and the estimated blood loss was 20-200 ml with an average of 81.5 ml.It was 3-17 days for the average time to remove the retropubic drainage tube postoperatively,with an average of 4.3 days.The postoperative hospital stay was 3-17 days with an average of 5.3 days.The Foley catheter was removed 14 days postoperatively.Five patients suffered from urine leakage and were managed successfully with conservative treatment.For the postoperative pathological stage,1 patient was pT2a,12 patients were pT2c,12 patients were pT3a and 21 patients were pT3b.The postoperative Gleason score was 6-9 with an average of 7.7.The postoperative follow-up time was 1-11 months with an average of 6.3 months.The PSA was 0.001-0.966 ng/ml with an average of 0.181 ng/ml 1 month postoperatively and the continence rate was 54% (25/46) and 89% (34/38) 1 month and 3 months after operation respectively.Conclusions The IUPU modified extraperitoneal laparoscopic VIP might be a safe and feasible for the radical prostatectomy.It's simplified for the operative steps,unnecessary to isolate and suture DVC and doesn't increase the blood loss.It's almost left intact for the surrounding tissues of the prostate apex and satisfactory for the early urinary continence.The oncological outcomes are needed to followup further.

ABSTRACT

A total of 268 Bacillus thuringiensis strains obtained from different sources of Argentina were analyzed to determine the diversity and distribution of the cryl, cry2, cry8, cry9 and vip3A genes encoding for lepidopteran-specific insecticidal proteins. Twin strains were excluded. Ten different profiles were detected among the 80 selected B. thuringiensis strains. Two of these profiles (cry1Aa, cry1Ac, cry1Ia, cry2Aa, cry2Ab and vip3Aa (35/80), and cry1Aa, cry1Ab, cry1Ac, cry1Ia, cry2Aa, cry2Ab and vip3Aa (25/80)) pooled 75% of the strains. The existence of this low diversity is rare, since in most of the studied collections a great diversity of insecticidal toxin gene profiles has been described. In addition, the most frequently detected profile was also most frequently derived from soil (70%), stored product dust (59%) and spider webs (50%). In contrast, the cry1Aa, cry1Ab, cry1Ac, cry1Ia, cry2Aa, cry2Ab and vip3Aa profiles were mainly detected in strains isolated from leaves (40%) and dead insect larvae (50%). Six of the identified insecticidal toxin gene profiles were discovered in strains isolated from stored product dust and leaves indicating higher diversity of profiles in these kinds of sources than in others. Some strains with high insecticidal activity against Epinotia aporema (Lepidoptera) larvae were identified, which is important to explore future microbial strategies for the control of this crop pest in the region.


Se analizaron 268 cepas de Bacillus thuringiensis obtenidas de diferentes fuentes de Argentina con el objeto de determinar la diversidad y distribución de genes cryl, cry2, cry8, cry9 y vip3A, que codifican proteínas insecticidas lepidóptero-específicas. Se excluyeron las cepas gemelas. Se detectaron solo diez perfiles diferentes entre los 80 B. thuringiensis seleccionados. Dos de estos perfiles, el cry1Aa, cry1Ac, crylIa, cry2Aa, cry2Ab y vip3Aa (35/80) y el cry1Aa, cry1Ab, cry1Ac, crylIa, cry2Aa, cry2Ab y vip3Aa (25/80), comprendieron el 75% de las cepas seleccionadas. La existencia de esta baja diversidad es una rareza, ya que en la mayor parte de las colecciones estudiadas se ha descrito una gran diversidad de perfiles de genes de toxinas insecticidas. El perfil detectado con mayor frecuencia se obtuvo principalmente de cepas procedentes de suelo (el 70% de los de esa fuente lo tenían), también fue mayoritario entre los procedentes de polvo de producto almacenado (59%) y en los que procedían de telas de arana (50%). En cambio, el perfil cry1Aa, cry1Ab, cry1Ac, crylIa, cry2Aa, cry2Ab y vip3Aa se detectó principalmente en las cepas aisladas de hojas (40%) y de larvas de insectos muertos (50%). Seis de los perfiles identificados fueron encontrados en cepas aisladas de polvo de producto almacenado y de hojas, lo que indica una mayor diversidad de perfiles en estas fuentes que en otras. Se identificaron algunas cepas con alta actividad insecticida contra larvas de Epinotia aporema (Lepidoptera), hallazgo importante para explorar en el futuro estrategias microbianas para el control de esta plaga en la región.

ABSTRACT

Objective To investigate the effect of compound Lactobacillus acidophilus on the levels of serum SP , VIP, 5-HT, IL-6 and TNF-αin patients with diarrhea-predominant irritable bowel syndrome ( IBS) .Methods 82 cases of patients with diarrhea -predominant irritable bowel syndrome ( IBS) from November 2015 to November 2016 in our hospital were selected,and randomly divided into two groups.30 patients in observation group were treated with compound Lactobacillus acidophilus tablets and 52 patients in control group were treated with trimebutine maleate capsule.The levels of serum SP, VIP, 5-HT, IL-6 and TNF-α,quality of life score,clinical curative effect and adverse reaction were observed and compared before and after treatment.Results After treatment,the diarrhea, abdominal distention, defecation frequency and abnormal rate of stool were significantly lower than those before treatment(P <0.05), the levels of SP,VIP,5-HT,IL-6 and TNF-αin serum were significantly lower than those before treatment(P <0.05),and the treatment group were lower than those of the control group.After treatment, the mental status, emotional control and work status scores of the observation group were higher than those of the control group,and the effective rate of the observation group 86.7%was significantly higher than the control group 61.5%,the differences between the two groups were statistically significant (P <0.05).There was no significant adverse events were observed during the treatment.Conclusion Compound Lactobacillus acidophilus is effective in the treatment of diarrhea-predominant irritable bowel syndrome.It can restore the levels of serum SP, VIP, 5-HT, IL-6 and TNF-αin patients with diarrhea-predominant irritable bowel syndrome and improve the quality of life .

ABSTRACT

Objective To explore the effects of vasoactive intestinal peptide (VIP)on the ratio of CD4+CD25+Treg/CD4+T cell and the expression of TGF-β1 in experimental autoimmune encephalomyelitis (EAE)rats. Methods We randomly divided 60 healthy female Wistar rats into normal control group,EAE control group,VIP low-dose group and VIP high-dose group.We used myelin basic protein (MBP)+ complete adjuvant (CFA)to establish EAE model. Since the day of model construction, the low- and high-dose VIP groups received intraperitoneal injection of 4 nmol/kg (0.2 mL)and 16 nmol/kg (0.8 mL)of VIP every other day,respectively;normal control group and EAE group received injection of saline of 0.8 mL for 10 days in a row.We recorded the peak of neurological dysfunction score (NDS)changes in the rats,observed the pathological changes and GFAP+astrocyte activation in the brain at the morbidity peak of rats with HE staining,and detected the ratio of CD4+CD25+Treg/CD4+T in the spleen with FACS and TGF-β1 cytokine level in brain tissue with ELISA.Results The peak nerve dysfunction score was decreased in each VIP dose group.In normal control group,there were decreased inflammatory cell infiltration and decreased number of active astrocytes in the brain tissue.The degree of infiltration of inflammatory cells and astrocyte activation in VIP control groups were significantly lower than those in EAE group.The CD4+CD25+Treg/CD4+T cell ratio of the spleen tissue in each dose VIP treated group rats was higher than that in EAE control group.The cytokine level of TGF-β1 in the brain tissue increased in each VIP dose group in the dose-dependent manner.Conclusion Through up-regulating the ratio of CD4+CD25+Treg/CD4+T cell in the spleen tissue,increasing TGF-β1 content in brain tissue,and inhibiting the infiltration of inflammatory cells and the astrocyte activation,VIP plays an important role in prevention and control of EAE.

Objective:To analyze status quo and trends of VIP services in the tertiary public hospitals of Shang-hai and provide references for health administrative departments. Methods:Health policies of VIP services in tertiary public hospitals were searched and analyzed, and the number of medical institutions, services, prices and service fees were analyzed from 2011 to 2013 . Results:There is a clear demand for VIP services in the tertiary public hospi-tals of Shanghai, and fees for rooms, nursing, and examinations for outpatient and inpatient care are decided by the hospitals. 89. 7% of the tertiary public hospitals in Shanghai offered VIP services, and the trend was steadily grow-ing. The four services that could be decided by hospitals varied greatly, and the service fees for inpatient care in-creased significantly. The total cost of VIP services in the tertiary public hospitals of Shanghai accounted 6. 2% of all costs, and the percentage of income from drugs was lower. Conclusions:VIP services in public hospitals have a his-torical necessity;management should be strengthened in the short term;public hospitals should strengthen their own management and provide VIP services regularly, and health administration departments should strengthen regulation. In the long run, it is suggested that public hospitals should draw lessons from international experiences to form a pat-tern of multi-level medical services and actively carry out cooperation with private medical institutions.

ABSTRACT

Objective To observe the clinical efficacy of electroacupuncture plus umbilical moxibustion in treating gastrointestinal dysfunction after gynecological abdominal operation.Method Totally 112 eligible patients undergone gynecological operation were randomized into group A of 37 cases, group B of 38 cases, and group C of 37 cases. Group A was intervened by electroacupuncture, group B was by umbilical moxibustion, while group C was by electroacupuncture plus umbilical moxibustion. After a treatment course, the symptom scores, restored time of bowel sounds, anal exhaust time, motilin (MTL), gastrin (GAS), and vasoactive intestinal peptide (VIP) contents were observed and compared, and the clinical efficacies were compared between the two groups.Result The symptom scores of the three groups were significantly changed after the intervention (P<0.05). After the treatment, the symptom score of group C was significantly different from that of group A and B (P<0.05). The restored time of bowel sounds and anal exhaust time in group C were significantly different from that of group A and B (P<0.05). The MTL, GAS, and VIP contents of the three groups were significantly changed after the intervention (P<0.05). The MTL, GAS, and VIP contents of group C were markedly different from that of group A and B (P<0.05). The total effective rate was 86.5% in group A, versus 81.6% in group B and 97.3% in group C. The total effective rate of group C was significantly different from that of group A and B (P<0.05).Conclusion Electroacupuncture plus umbilical moxibustion is effective in treating gastrointestinal dysfunction after gynecological abdominal operation.

This article presented a series of cases using vascularized interpositional periosteal-connective tissue ( VIP-CT) flap with guide bone regeneration ( GBR) in peri-implant soft and hard tissue reconstuction at the esthet-ic zone of maxillary. Fifteen cases with bone and soft tissue defects underwent VIP-CT flap with GBR in the implant treatment. And the attached gingiva width was evaluated before treatment and six months and eighteen months after the operation. The width of attached gingival of six months and eighteen months after surgery was significantly dif-ferent from the preoperative value (P<0. 05). However, no statistically significant difference could be found at six months and eighteen months postoperative. The application of VIP-CT flap could increase the width of attached gin-giva around implants and the short-term effects were stable and favorable.

ABSTRACT

Objective To explore the clinical efficacy of acupuncture point embedding therapy in the treatment of slow transit constipation (STC)and the influence on the colon transmission function,plasma substance P(SP), vasoactive intestinal peptide(VIP). Methods A total of 128 slow transit constipation patients were involved in the study and divided randomly into two groups by using stochastic indicator: a treatment group (treated with acupuncture point embedding therapy) and a control group (treated with lactulose oral liquid). The clinical effects, colon transmission function, the influence of SP, and VIP were observed for STC patients. Results In the treatment group, CCS score was (13.52±3.93) before treatment and(8.15±3.77) after the treatment. In the control group CCS score was (14.11±4.24) and (9.01±2.93) respectively before and after the treatment, CCS score in both groups showed significantly improvement after the treatment (P0.05). The plasma SP and VIP level of the two groups after the treatment [treatment group (46.73±1.50) ng/L,(206.10±12.14) ng/L, the control group(40.92±6.25)ng/L,(214.73± 7.62)ng/L] were also improved than before [treatment group was(26.66±0.88)ng/L, (332.68±17.84)ng/L, the control group was (23.32±1.75)ng/L,(306.22±20.01)ng/L], with statistically significant(P<0.05), and the difference between the two groups were also significant(P<0.05).Conclusion The acupuncture point embedding therapy is effective for STC, and it can regulate the level of SP and VIP.

This study was aimed to observe the effect of Bai-Zhu Fu-Ling (BZFL) Decoction in different proportion-ing on VIP and VIPR1 in Crohn's disease (CD) rats with spleen deficiency syndrome, in order to further explore the immunologic mechanism of BZFL Decoction on CD. The CD rat model with spleen deficiency syndrome was estab-lished using exhaustion and hunger. The model rats were treated by BZFL Decoction with different proportioning, and immunohistochemistry (IHC) was used to detect the expression of VIP and its receptor in colon tissues. The results showed that comparing to the blank control group, the level of VIP and its receptor of the model group significantly increased (P< 0.05). Comparing to the model group, the level of VIP and its receptor in BZFL Decoction B5 group (Rhizoma A tractylodis Macrocephalae:Poria = 12:15), B6 group (Rhizoma A tractylodis Macrocephalae:Poria = 15:12) and B7 group (Rhizoma A tractylodis Macrocephalae:Poria = 18:9) was significantly decreased (P< 0.05). It was con-cluded that the effect of BZFL Decoction of B5 group, B6 group and B7 group was better than other groups in VIP and its receptor which can regulate the VIP and its receptor, inhibit the releasing of inflammatory factors and reduce intestinal inflammation injury.

ABSTRACT

Objective To study the changes of plasma gastrin (Gas), substance P (SP) and vascoactive intestinal peptide (VIP) among children with recurrent abdominal pain and their family members,and to explore if there is the rule of the changes.Methods The fasting plasma Gas, SP and VIP were determined by radioimmunoassay method among 30 children with recurrent abdominal pain, and 45 family members including first-degree relatives and second-degree relatives as well as 35 normal healthy children and 20 normal healthy adults. Individuals were divided into five groups:children with recurrent abdominal pain (study group 1), family members with recurrent abdominal pain (study group 2), normal healthy children (control group1), family adults without recurrent abdominal pain (control group2) and normal healthy adults (control group3) . The whole family members of 7 children with recurrent abdominal pain were focused on and analysed.Results 55 of 130 volunteers were with recurrent abdominal pain including 30 children and 25 adults. There were 22 adults with and 20 adults without recurrent abdominal pain in the whole family members of 7 children with recurrent abdominal pain. The fasting plasma Gas content had no difference between study group 1 and control group1 ( 0.05) .The levels of fasting plasma Gas content had no difference between study group 2 and control group 3 (>0.05),but the levels of fasting plasma SP and VIP were both in increase and had difference between study group 2 and control group3. The fasting plasma Gas content had no difference between country group 1 and control group3 ( 0.05) .Conclusions There are the same plasma Gas contents in normal children and adults.However, the plasma SP and VIP contents are higher in normal children than ones in normal adults. The plasma SP and VIP contents are lower in children with recurrent abdominal pain than ones in normal children. The plasma SP and VIP contents are higher in adults with recurrent abdominal pain than ones in normal adults. These suggested that the increases of plasma SP and VIP may have a close relationship with the pathogenesis of children and adults with recurrent abdominal pain. The contents of plasma Gas, SP and VIP were consistent in the whole family members of 7 children with recurrent abdominal pain. It suggested that the secretion of gastrointestinal hormone is disorder in the families with recurrent abdominal pain.
