ABSTRACT

Objective To observe the mechanism of action of panax quinquefolium diolsaponins (PQDS) on anti-ventricular remodeling in rats. Methods The model of pressure-loading ventricular remodeling was established through the method of rat’s abdominal aorta deligation. The male Wistar rats were divided into 5 groups,including sham operation group,remodeling model group,Benazepril group,and low and high dose groups of PQDS. The rats were treated with PQDS (with dose of 50 and 100 mg?kg-1 i.g) for 6 weeks. The rats in sham operation group and remodeling model group were treated with normal sodium (with dose of 10 mL?kg-1?d-1 i.g) for 6 weeks. The rats in masculine medicine group were treated by Benazepril (with dose of 10 mg?kg-1 i.g) for 6 weeks. After 6 weeks of treatment,myocardial morphological parameters,the content of malondialdehyde (MDA) and activity of superoxide dismutase (SOD) in serum,the contents of prostacycline (PGI2),thromboxane A2 (TXA2),nitric oxide (NO) and angiotensinⅡ (AngⅡ) in plasma were determined. At the same time,the contents of endothelium (ET) in plasma and myocardium were also determined. Results Compared with remodeling model group,the ventricular weight and cardiac coefficient were decreased significantly in the rats treated with PQDS (P0.05). Conclusion PQDS has protective effects on ventricular remodeling through increasing anti-oxidase activity,reducing the damage of free radicals and vasoactive substance on myocardium and correcting disequilibrium of PGI2/TXA2 in ventricular remodeling etc.

ABSTRACT

Objective To observe the relationship between vasoactive substances and blood pressure in old patients with hypertension.Method Venous blood was drawn from 35 old hypertension patients when blood pressure was normal and abnormal,respectively.Epinephrine(E),norepinephine(NE),endothelins(ET),angiotensin ll (Angll) and ealcitonin-gene-retated peptide were determined by radio-immunity.The above five cytokines were also determined in 33 old healthy people.The cytokines levels were compared between the normal and abnormal blood pressure in hypertension patients and also between the old hypertension patients and healthy persons.The correlation between blood pressure and cytokines were analyzed.Results The level of ET,Angll,E and NE were obviously increased in old hypertension patients when their blood pressure was abnormal,while CGRP obviously decreased.No difference was found in the five cytokines between two groups when the blood pressure were normal.The correlation analysis showed that systolic pressure had positive correlation with ET,AnglI and E(r=0.304,0.337,0.587,P=0.031,0.019,0.036)and negative correlation with CGRP(r=-0.416,P=0.028)in old hypertension patients when the blood pressure was abnormal.And the diastolic pressure had positive correlation with AnglI and E(r=0.357,0.333;P=0.039,0.028).ET had positive correlation with Angll and E(r=0.361,0.315;P=0.041,0.029).Conclusion The levels of vasoaetive substances significantly changed in old hypertension patients with the blood pressure change.Among those cytokines,the substances that contract blood vessel increased most obviously.The vasoactive substances played an important role in the progress of blood pressure rising.

ABSTRACT

OBJECTIVE: To observation the effects of Glycosaminoglycan from Scallop Skirt (SS-GAG) on the production of vasoactive substance in the endothelial cells injured by hydrogen peroxide(H2O2). METHODS: In H2O2-induced en-dothelial cell oxidative damage model group which was established with human umbilical vein endothelial cell (HUVEC, ECV304) cultured in vitro, SS-GAG groups (at different concentrations) and the normal group, levels of 6-keto-PGF1a, Thromboxane-B2 and Endothelin (ET) in the culture fluid were detected by radioimmunity assay and the content of nitric oxide was determined by nitrate reductase method. ABSTRACT

OBJECTIVES AND METHODS: To directly examine the function of the endothelial cell(EC) and smooth muscle cell in umbilical arteries acquired from preeclampsia patients between June 1998 to November 1999, using a conventional tension measurement and bioassay experiment. RESULTS: Relaxation responses to EC-dependent relaxing agents including bradykinin and A23187 in human umbilical artery rings were significantly decreased in preeclampsia(p<0.01). Relaxation responses to EC-independent agents(SNP and SNAP) were also inhibited in umbilical artery rings acquired from preeclampsia patients(p<0.01). To test the change of endothelial cell function in preeclampsia without involvement of smooth muscle dysfunction, we used human umbilical artery and rabbit femoral artery as a donor and detector, respectively, in bioassay experiment. Relaxation responses to EC-dependent agents(A23187 and bradykinin) showed similar results to conventional tension measurement (p<0.01). Relaxation responses to 8-bromo-cGMP in human umbilical artery rings were also significantly decreased in preeclampsia(p<0.01). CONCLUSIONS: It can be concluded that increased vascular resistance in preeclampsia is not only due to the disturbance of endothelial function, but also to smooth muscle dysfunction.

ABSTRACT

BACKGROUND: The gastroepiploic artery is not only an alternative graft but also may be considered an important primary graft for coronary revascularization. However, the long-term patency of the gastroepiploic arterial graft is yet to be determined and the incidence of perioperative spasm and long-term patency of a coronary graft may be affected by the properties of the graft response to certain vasoactive substances. The reactivity of the gastroepiploic artery to vasoactive substances has not been studied extensively and the results of the studies are contradictory. MATERIAL AND METHOD: This study was designed to test the reactivity of human gastroepiploic artery to four constrictors and four relaxants. The middle sections of the human gastroepiploic arteries were collected from the patients undergoing gastrectomy and the arterial rings with intact endothelium were suspended in organ baths for isometric tension recording. RESULT: Epinephrine, norepinephrine, and potassium chloride induced the maximum constriction to higher forces (7.0+/-1.1g, 6.6+/-0.9g, and 6.5+/-1.1g) than 5-hydroxytryptamine did (3.8+/-1.7g, p<0.05). Nitroprusside and histamine induced almost full relaxation in the gastroepiploic arteries preconstricted with norepinephrine. There was no significant difference between two relaxants regarding maximum relaxation force. Acetylcholine induced the maximum relaxation to weaker force when compared with nitroprusside and histamine (p<0.05), and isoproterenol was the weakest of the relaxants (p<0.05 compared with acetylcholine). CONCLUSION: The gastroepiploic artery has a strong capacity of endothelium-dependent relaxation which could have an important influence on long-term patency. The gastroepiploic artery exhibits a potent contractility to catecholamines and the enhanced contractility may facilitate vasospasm in the presence of high circulating levels of catecholamines. Nitroprusside, a potent relaxant in gastroepiploic artery, might be beneficial for the treatment of gastroepiploic arterial graft spasm. The gastroepiploic arterial graft with intact endothelium may respond weakly to beta-adrenoceptor agonist and 5-hydroxytryptamine.

ABSTRACT

AIM: To observe the influence of Toufengyu Dropping Pill(Rhizome chuanxiong,Radix angelicae dahuricae,etc) on neuropeptide and vasoactive substances in rat's migraine model. METHODS: Rat's migraine model caused by nitroglycerin were given Toufengyu Dropping Pill and rat's behavior and blood levels of NO,CGRP,ET,TXB_2 and 6-keto-PGF_(1a) were measured. RESULTS: The experimental results showed that Toufengyou Dropping Dill could obviously decrease rat's positive reaction number,the blood levels of NO,CGRP,TXB_2 and 6-keto-PGF_(1a),and increase the blood plasma level of ET,and rectify the imbalance of ET/NO ratio. CONCLUSION: Toufengyou Dropping Pill can affect the contents of neurotransmitter,neuropeptide in rat's migraine,then decrease the algoenic and inflammatory substances,relieve the neurogenic inflammation,and increase the pain threshold value,finally be quite effective for migraine.

ABSTRACT

Objective To investigate the difference of the morbidity rate of hypertension among patients with sleep apnea syndrome (SAS) in different stages, and the change in the blood levels of some vasoactive substances and their correlations with severity of SAS, in order to further explore the mechanism of hypertension caused by SAS. Methods 63 patients suspected to suffer from SAS were enrolled, and they were divided into normal, mild, moderate, and severe SAS groups according to respiratory disorder index (RDI) obtained by polysomnography (PSG) monitoring. The diagnosis of hypertension was made by the measurement of blood pressure and 24-hour dynamic measurement of blood pressure. Blood levels of angiotesnin Ⅱ (Ang Ⅱ), endothelin-1 (ET-1), throboxane B2 (TXB2), calcium gene related protein (CGRP), 6-keto-protagladin factor1?(6-keto-PGF1?) were determined with radioimmunoassay. Results The mobidity rate of hypertension was significantly higher in moderate and severe SAS group than that in the normal group (P0.05). Correlative analysis showed that the blood levels of Ang Ⅱ, ET-1, TXB2 were positively correlative to RDI (r=0.594, 0.523, 0.574, respectively, P0.05). Conclusion SAS is significantly related to the incidence of hypertension. Vasoactive substances with vasoconstrictive effect such as Ang Ⅱ, ET-1, TXB2 play an important role in the pathogenesis and development of hypertension in SAS patients than those with vasodilative effect such as CGRP and 6-keto-PGF1?.
