ABSTRACT

OBJECTIVE@#To investigate iron accumulation level over the whole brain and explore the possible neuromechanism of medication-overuse headache (MOH) using quantitative susceptibility mapping (QSM).@*METHODS@#Thirty-seven MOH patients and 27 normal control subjects were enrolled in the study for examinations with both a multiecho gradient echo magnetic resonance (MR) sequence and brain high resolution structural imaging. A voxel-based analysis was performed to detect the brain regions with altered iron deposition, and the quantitative susceptibility mapping values of the positive brain regions were extracted. Correlation analysis was performed between the susceptibility values and the clinical variables of the patients.@*RESULTS@#In patients with MOH, increased susceptibility values were found mainly in the bilateral substantia nigra (SN) (MNI coordinate: 8, -18, -14; -6, -16, -14) as compared with the normal control subjects (P < 0.001), but these alterations in iron deposition were not significantly correlated with the clinical variables of the patients (P > 0.05). The susceptibility value in the left SN had an area under curve (AUC) of 0.734, and at the cut-off value of 0.077, its diagnostic sensitivity was 72.97% and its specificity was 70.37% for distinguishing MOH from normal controls; The susceptibility value in the right SN had an AUC of 0.699 with a diagnostic sensitivity of 72.97% and a specificity of 62.96% at the cut-off value of 0.084.@*CONCLUSION@#Increased iron deposition occurs in the bilateral SN of MOH patients, which provides a new insight into the mechanism of mesocorticolimbic dopamine system dysfunction in MOH. QSM technique can be used as a non-invasive means for quantitative analysis of brain iron deposition in migraine neuroimaging.

ABSTRACT

Objective To study the correlation between novelty seeking(NS) personality of healthy young individuals and fractional anisotropy (FA) with voxel-based analysis (VBA) of diffusion tensor imaging(DTI),and explore the microstructure of white matter of personality. Methods Total 333 healthy right handed volunteers completed the Chinese version of the TPQ and the Beck Depression Inventory(BDI),and then finished 3D T1 structure scan,DTI scan. All preprocessed DTI datum was analyzed with SPM8 soft ware in partial correlation analysis, using age and depression scores as controlled variable. The correlation studied between NS and FA (a voxel significance threshold P < 0.001 and a cluster extent threshold of 30 voxels). Results NS score of men and women were 14.55 ± 4.66 and 14.08 ± 5.42 points, and the difference between the two groups was not statistically significant (t =0.81,P=0.46).BDI score of men were higher than that of women (t =3.10,P=0.002),which were 9.23 ± 7.90 and 6.76 ± 5.79 points respectively.NS correlated negatively with FA of the right inferior frontal gyrus, and the right supramarg(i)nal gyrus(P < 0.001).Conclusion NS personality has specific microstructure background of white matter.

ABSTRACT

PURPOSE: To generate a Korean specific brain template, especially in patients with Alzheimer's disease (AD) by optimizing the voxel-based analysis. MATERIALS AND METHODS: Three-dimensional T1-weighted images were obtained from 123 subjects who were 43 cognitively normal subjects and patients with 44 mild cognitive impairment (MCI) and 36 AD. The template and the corresponding aprior maps were created by using the matched pairs approach with considering differences of age, gender and differential diagnosis (DDX). We measured several characteristics in both our and the MNI templates, including in the ventricle size. Also, the fractions of gray matter and white matter voxels normalized by the total intracranial were evaluated. RESULTS: The high resolution template and the corresponding aprior maps of gray matter, white matter (WM) and CSF were created with the voxel-size of 1 x 1 x 1 mm. Mean distance measures and the ventricle sizes differed between two templates. Our brain template had less gray matter and white matter areas than the MNI template. There were volume differences more in gray matter than in white matter. CONCLUSION: Gray matter and/or white matter integrity studies in populations of Korean elderly and patients with AD are needed to investigate with this template.
