ABSTRACT

Objective To investigate the effects of local application of NGF combined with Insulin on wound healing with Ⅱ degree deep scald and the expression of HIF-1α and VEGF in diabetic rats. Methods 60 male Wistar rats were used in the study by intraperitoneal injection of streptozotocin (STZ). Deep partial thickness scalding was engendered on the back of the rats after one month. Then these rats were random divided into group B (scalding of the rats with diabetes, n = 15), C (insulin control, n = 15), D ( NGF control, n = 15) and E ( NGF and insulin supplementation, n = 15). And group A ( nomal control,n = 15) was created by normal rats of partial thickness scalding. The wound area, wound healing rate, and grey of HIF-1α and VEGF immunohistochemistry staining were determined on the 3rd, 7th, 15th, and 21st day post scalding days (PSDs). Results Compared to those in groups A, B, C and D, the wound healing rate in group E increased significantly since the 7th PSD [(25.33 ± 2. 32 ) %, P < 0.05]. The wound healing rate in group C and D increased significantly compared with group B [( 16. 68 ± 1.95 ) %, ( 18.29±1. 70)%, P <0.05]. Compared to group A, the group D increased significantly since the 15th PSD [(54. 84 ±3.60)%, P <0. 05]. The expression of HIF-1α and VEGF in group E began to increase slower than other groups after the 7th day ( P <0. 05 ). Conclusion Local application of NGF combined with Insulin could be beneficial to the regeneration of capillary vessel in the burn wounds of the rats with diabetes,as well as to accelerate wound healing by redress anoxia and decrease the expression of HIF-1α and VEGF.

ABSTRACT

ObjectiveTo explore the effect of celecoxib on the tendon adhesion and healing after anastomosis.Methods54 New Zealand white rabbits were randomly assigned to one of 3 ( celecoxib, ibuprofen, and saline) groups. The deep flexor tendon was transected, followed by a primary repair. The care was begun the day after surgery and was continually provided for 14 days.Celecoxib was given[20 mg/( kg · d) ],ibuprofen was given [75 mg/( kg · d) ],or the same volume of saline solution was given respectively. At the 4th and 8th week the animals were killed and assessed by general observation, histologically observation, and biomechanical testing.ResultsGeneral observation, celecoxib group and ibuprofen group showed good shape of tendons, smooth surface, and covered with a film - like tissue, and it was easy to be separated.Saline group showed that tendon and surrounding tissue medium had dense adhesions, and it was difficult to be separated.Histologically observation showed between tendon and paratenon had clear gap in celecoxib and ibuprofen group. The mature fiber cells gradually arranged rules and in the same direction.Saline between the tendon and paratenon gradually emerged a gap,but more fibroblasts and arranged in irregular.Biomechanical testing showed that tendon sliding resistance appeared in celecoxib and ibuprofen group.Compared with saline group, the differences were statistically significant (0.354 ± 0.078/0.382 ±0.121 vs 0.521 ±0.126,P <0.05;0.075 ±0.035/0.097 ±0.043 vs 0.414 ±0.110,P <0.01).UTS (ultimate tensile strength had statistical significance among celecoxib, saline groups and ibuprofen group.(36.812 ±6.388 vs 24.899 ±4.667,P <0.05;34.297 ±8.132 vs 24.899 ±4.667,P <0.01 ;54.515 ±4.688/59.037 ± 6.606 vs 42.418 ± 5.594,P < 0.01 ).ConclusionsCelecoxib can effectively prevent tendon adhesion, and does not affect the tendon healing.

ABSTRACT

Objective To observe the effects of aloe juice on skin wound healing in scalded rats and scavenging the oxygen free radical in their bodies. Methods 24 rats were divided into 3 groups, namely, aloe group, burn cream group and blank control group. And burn wounds ( Ⅱ degree deep) were created with a diameter of 2. 6cm in their backs, respectively. Smear Burn Ointment and Aloe Vera juice, were used to observe the time of the appearance of their epithelial tissues and the time of the wound healing, and the content of SOD, GSH-PX and MDA in healed skin surfaces were detected. Results The time of the appearance of epithelial tissues[(4.6 ±0. 56)d ,(16.2±2.6),(5.4±0.6)d,(18. 1 ±3.4)d,(6.8± 0. 3) d , (22. 3 ± 3.2 ) d], the wound healing and the content of SOD [( (98.07 ± 6. 22) vs(83.97 ± 6. 34), (57. 50 ± 9. 43 )], GSH-PX [(243.21 ± 20. 18)vs (208.25 ± 24. 52), ( 139. 88 ± 22. 70)] and MDA [(4. 89 ±2. 12) vs (6. 93 ± 3.05 ), (8.98 ± 2. 14)] in aloe group have significant difference compared with the blank group( P <0.01, 0.001, 0.005), but no significant difference compared with the burn cream group. Conclusion Aloe juice has the effects of promoting the skin wound healing of scalded rats ,scavenging the oxygen free radical in their bodies and protecting cells.
