ABSTRACT

Objective To investigate the genetic data of the 19 X-STR loci in three ethnicities of China ( Han, Gelao,Miao) and to evaluate the application in forensic science. Methods The DNA samples of unrelated individ-ual in Han (n=308), Gelao (n=398), Miao (n=323) ethnicities were amplified using MicroreaderTM19X ID System kit, and the PCR products were analyzed by electrophoresis through 3500XL genetic analyzer. The fragment sizes of alleles were taken subsequently by GeneMapper? ID-X. Allele frequencies and national genetics parameters of the 19 X-STR were analyzed by statistics. The allele frequencies were compared among the three nationalities and were compared with available data of other Han ethnicities from different regions. Results After the Bonferroni correction at a 95% significance level, no significant departures from the Hardy-Weinberg equilibrium was observed. Linkage disequilibrium test showed no significant allelic association between all 19 X-STR loci after Bonferroni’s correction. The cumulative discrimination power in females and in males were greater than 0.999 999 999 99 and 0.999 999 999 94,respectively. The combined power of exclusion in trios and in duos were greater than 0.999 999 999 36 and 0.999 999 52,respectively. The p values, calculated throuth Arle-quin v3.5 software, there were significantly different as detected at loci of X-STR among the different nationalities. Conclusions This panel of X-STR is highly polymorphic in China’s three ethnicities and can be served as a supple-mentary to the current STR system for individual identification.

ABSTRACT

Abstract X chromosome STR typing has emerged recently as a powerful tool, complementary to autosomal STR typing, in solving complex forensic and missing person cases. Investigator® Argus X-12 is a commercial product that allows co-amplification of 12 X chromosomal markers belonging to four linkage groups (LGs). In this study, we analyzed by capillary electrophoresis blood samples from 100 females and 102 males from a population of northern Croatia. Statistical analysis included calculation of allele and haplotype frequencies, as well as forensic parameters. The most informative marker for the northern Croatia population was DXS10135 with PIC=0.9211 and a total of 27 alleles. The least polymorphic marker was DXS8378 with 6 alleles. The proportion of observed haplotypes from the number of possible haplotypes varied from 2.74-8.57% across all LGs, with LG1 being the most informative. Of the 11 tested world populations compared to the population of northern Croatia, significant differences in genetic distance (FST) were found for Greenlandic and all non-European populations. We found that all tested markers are in HWE and can thus be used for match probability calculation. Because of high combined power of discrimination in both men and women, Investigator® Argus X-12 is applicable for the northern Croatia population in routine forensic casework.

ABSTRACT

OBJECTIVES@#To investigate the genetic data of 16 X-STR loci in Henan Han population and to assess the application value in forensic science.@*METHODS@#The DNA of 326 unrelated individuals in Henan Han population were amplified using Goldeneye™ DNA identification system 17X kit, and the PCR products were analyzed by electrophoresis through 3130xl genetic analyzer. The fragment sizes of alleles were analyzed subsequently by GeneMapper® ID-X. Allele frequencies and population genetics parameters of 16 X-STR loci were analyzed statistically and compared with the available data of other Han populations from different regions.@*RESULTS@#Among the 16 X-STR loci, DXS6800 were found to be moderately polymorphic and the other 15 X-STR loci were highly polymorphic. The cumulative discrimination power in females and males were 0.999 999 999 999 992 and 0.999 999 996 577 712, respectively. The combined power of exclusion in trios and in duos were 0.999 999 971 and 0.999 992 574, respectively.@*CONCLUSIONS@#The 16 X-STR loci meet the application requires of forensic genetics, especially for testing the special paternity cases.

ABSTRACT

[Objective] To learn about the genetic diversity,we studied the five X-chromosomal STR (X-STR) loci in Guangdong Han Nationality Groups.[Methods] The five Loci (DXS6803,DXS981,DXS6809,DXS6789,and DXS7132) were amplified in a pentaplex PCR reaction.PCR products were analyzed using capillary electrophoresis and ABI prism 3100 Genetic Analyzer, with GeneMapper ID 3.1 Analysis Software.[Results] A total of 363 individuals (181 unrelated male and 182 unrelated female) from Guangdong Han population were tested,54 alleles were observed for these loci.Polymorphism information content is 0.6935 ~ 0.8177.Power of discrimination in females was 0.8976 ~ 0.9562.Mean exclusion chance for X-STR in standard trios with daughters was 0.7805 ~ 0.8467.[Conclusion] The five loci in the multiplex system provide high polymorphism information for forensic identification and paternity testing, particularly for difficult paternity deficiency cases.

ABSTRACT

This study intends to examine the polymorphism of 5 STR loci inX-chromosome (GATA172D05, HPRTB, DXS8377, DXS101, HumARA) and to evaluate usefulness of them in forensic identification. 100 unrelated Korean men and women were selected. DNA was extracted from these sample and PCR was performed to amplify it. And using automated DNA sequencer and computer program, the genotype and allele frequency of them were investigated and analyzed. The following results were obtained: 1. The genetic analysis of 5 STR loci inX-chromosome was performed with quadruplex PCR for GATA172D05, HPRTB, DXS8377, HumARA and monoplex PCR for DXS101. 2. Polymorphism information content of 5 loci is higher than 0.5, the high information content is observed. The heterozygosity is higher in DXS8377, DXS101, HumARA than others. 3. The power of discrimination is revealed high in all 5 loci in women, but in men DXS8377 and HumARA is higher than others. 4. The mean exclusion chance is revealed high in DXS8377 and HumARA which have more alleles than others in trio case and motherless case. 5. The difference of allele frequency is observed with other population group in DXS8377, DXS101, HumARA of Korean population group. Based on the results of this study, the allele frequency and population data of 5 STR loci inX-chromosome may be useful in forensic investigation.

ABSTRACT

Objective:To investigate genetic polymorphism of four X chromosome short tandem repeats(X-STR)loci(DXS7132、 DXS6789,HPRTB,DXS8378)in Chongqing Han population.Methods:EDTA-blood samples were collected from the unrelated individuals in Chongqing. After being extracted with Chelex-100 method, the DNA samples were amplified by PCR technique. The PCR products were analyzed by PAGE,genotyped with silver stain method.Hardy-Weinberg equilibrium was tested, and then used direct counting method to calculate allele and genotype frequency of these four loci. Results:The polymorphisms of all four STR loci were obtained from 120 unrelated females and 91 unrelated males from Chinese Han group.?2 tests on the genotype frequencies in female reveal that genotype frequencies in females did not depart from Hardy—Weinberg equilibrium. Conclusion:It was suggested that DXS7132,DXS6789,HPRTB,DXS8378 locus are suitable for individual identification and paternity testing, and we provide new selectable STR markers for chromosome X haplotype in forensic application.
