ABSTRACT

The chemical composition of Mangifera indica L. cv. "Kent" leaves was determined by HPLC-ESI-QTOF-MS/MS. Polyphenolic compounds characterized as benzophenone derivatives were the main components found in extracts (1, maclurin 3-C-(2-O-galloyl)-D- glucoside isomer; 2, maclurin 3-C---D-glucoside; 3, iriflophenone 3-C---D-glucoside; 5, maclurin 3-C-(2,3-di-O-galloyl)---D-glucoside; 6, iriflophenone 3-C-(2-O-galloyl)---D-glucoside; 7, methyl-iriflophenone 3-C-(2,6-di-O-galloyl)---D-glucoside) and xanthones (4, mangiferin and 8, 6-O-galloyl-mangiferin). The estrogenic and antioxidant effects of aqueous extracts from Mangifera indica L. cv. "Kent" leaves on ovariectomized rats were determined by uterotrophic assay and malondialdehyde (MDA) levels in erythrocytes, bone, liver, and stomach. We conclude that the polyphenolic compounds from extracts act as exogenous antioxidant agents against oxidative damage in ovariectomized rats.


La composición química de las hojas de Mangifera indica L. cv. "Kent" se determinó por HPLC-ESI-QTOF-MS/MS. Compuestos polifenólicos caracterizados como derivados de benzofenona fueron los componentes principales encontrados en los extractos (1, isómero de la maclurina 3-C-(2-O-galoyil)-D-glucósido; 2, maclurina 3-C-ß-D-glucósido; 3, iriflofenona 3-C-ß-D-glucósido; 5, maclurina 3-C-(2,3-di-O-galloíl)-ß-D-glucósido; 6, iriflofenona 3-C-(2-O-galloil)-ß-D-glucósido; 7, metil-iriflofenona 3-C-(2,6-di-O- galloyl)-ß-D-glucósido) y xantonas (4, mangiferina y 8, 6-O-galoyil-mangiferina). Los efectos estrogénicos y antioxidantes de los extractos acuosos de hojas de Mangifera indica L. cv. "Kent" en ratas ovariectomizadas se determinaron mediante ensayo uterotrófico y la medición de los niveles de malondialdehído (MDA) en eritrocitos, huesos, hígado y estómago. Concluimos que los compuestos polifenólicos de los extractos actúan como agentes antioxidantes exógenos contra el daño oxidativo en ratas ovariectomizadas.

ABSTRACT

OBJECTIVE: To evaluate the Tabernaemontana catharinensis ethyl acetate fraction hypoglycemic and antioxidant activity through the peripheral glycemic dosage and enzymatic tests. Methods: Male rats were divided into 6 groups: control, diabetic control, control extract 50, diabetic extract 50, control extract 80, diabetic extract 80. In diabetic group animals alloxan (150mg/Kg) was administered to induce Diabetes Mellitus. The animals were beheaded following 15 days of treatment with extract or distilled water and the blood was collected in order to perform oxidative stress tests. Results: The diabetic control group showed high levels of glucose, increased levels of thiobarbituric acid and superoxide dismutase activity, and decreased activity of catalase and glutathione peroxidase enzymes. The diabetic animals that received 50mg/Kg and 80mg/Kg of extract showed a decrease in thiobarbituric acid levels and an increase of glutathione peroxidase activity when compared to the diabetic control group. It was observed that only animals treated with 80mg/Kg of extract had positive results regarding superoxide dismutase. Conclusions: The Tabernaemontana catharinensis ethyl acetate fraction when orally administered for 14 consecutive days at doses of 50mg/Kg and 80mg/Kg reduces the oxidative stress induced by alloxan administration


OBJETIVO: Avaliar a ação hipoglicemiante e antioxidante da fração acetato de etila do extrato de Tabernaemontana catharinensis através da dosagem glicêmica periférica e testes enzimáticos. MÉTODOS: Ratos machos foram divididos em seis grupos: controle, controle diabético, controle extrato 50, diabético extrato 50, controle extrato 80, diabético extrato 80. Nos animais dos grupos diabéticos foi induzida Diabetes Mellitus pela administração de 150mg/Kg de aloxana. Após 15 dias de tratamento com a fração acetato de etila de Tabernaemontana catharinensis ou água destilada, os animais foram decapitados e o sangue foi coletado para realização dos testes de estresse oxidativo. RESULTADOS: O grupo controle diabético apresentou níveis elevados de glicose, aumento dos níveis de ácido tiobarbitúrico e atividade da superóxido dismutase, e diminuição da atividade das enzimas catalase e glutationa peroxidase. Os animais dos grupos diabéticos tratados com 50 e 80mg/Kg do extrato apresentaram redução nos níveis de ácido tiobarbitúrico e aumento da atividade de glutationa peroxidase quando comparado ao grupo controle diabético. Apenas os animais que receberam o extrato na dose de 80mg/Kg obtiveram resultados positivos em relação ao superóxido dismutase. CONCLUSÕES: A fração acetato de etila de Tabernaemontana catharinensis, quando administrada por 14 dias consecutivos, via oral, nas doses de 50 e 80mg/Kg, promove redução nos níveis de estresse oxidativo gerado pela administração de aloxana

ABSTRACT

As folhas de Mangifera indica L são importantes como fonte de compostos fenólicos, especialmente mangiferina, que apresentam propriedades antidiabética, hipolipemiante, antioxidante e anti-inflamatória. O estudo teve como objetivo avaliar os efeitos do extrato etanólico de folhas de M. indica e da mangiferina isolada sobre a lesão aterosclerótica em camundongos ApoE-/-. Métodos: Camundongos ApoE-/- com 15 semanas de idade foram divididos aleatoriamente em 4 grupos de acordo com o tratamento, por gavagem, durante 56 dias: controle (veículo, dimetil sulfóxido); E200 (200 mg/kg/dia de extrato da folha de M. indica), E400 (400 mg/kg/dia de extrato da folha de M. indica); M40 (40 mg/kg/dia de mangiferina). Parâmetros sanguíneos foram dosados utilizando-se kits enzimáticos e as lesões ateroscleróticas foram avaliadas pelo método en face. Resultados: O extrato seco apresentou 17% de mangiferina. Os níveis sanguíneos de colesterol total, frações HDLc e LDLc e triacilgliceróis, bem como o percentual de deposição lipídica no arco aórtico e aorta torácica não diferiram significativamente entre os grupos (p>0,05). Conclusão: A administração do extrato de folhas de M. indica e da mangiferina em camundongos ApoE-/- não afetou a lipidemia e não diminuiu as lesões ateroscleróticas pré-existentes.


Mangifera indica L leaf are an important source of phenolic compounds, especially mangiferin, that exhibits antidiabetic, hypolipidemic, anti-oxidant and anti-inflammatory activities. This study aimed to evaluate the effects of mangiferin and ethanolic extract of M. indica leaf on atherosclerotic lesions in mice ApoE-/-. Methods: Fifteenweek- old ApoE-/- mice were randomly divided into 4 groups according to the treatment giving by gavage during 56 days: control - vehicle (dimethyl sulfoxide); E200 - 200 mg/kg/day M. indica leaf extract; E400 - 400 mg/kg/day M. indica leaf extract, M40 - 40 mg/kg/day mangiferin. Administrations of vehicle, extracts and mangiferin were performed every day by gavage during 8 weeks. Blood parameters were measured using enzymatic kits and atherosclerotic lesions were evaluated by en face method. Results: The dired extract showed 17% of mangiferin. Total cholesterol, HDLc, LDLc and triglycerides blood levels, as well as the percentage of lipid deposition in the aortic arch and thoracic aorta were not significantly different between the groups (p> 0.05). Conclusion: The administration of M. indica leaf extract and mangiferin in ApoE-/- mice did not affect serum lipids and did not decreased pre-existing atherosclerotic lesions

ABSTRACT

Vismia genus is an important source of natural medicinal products, thus, information collected in this review is an attempt to cover the most recent developments in the ethnopharmacology, pharmacology and phytochemistry of this genus. Anthraquinones and other quinonoid derivates, terpenoids and volatile constituents have been reported as the major constituents isolated from different Vismia species. On the other hand, pharmacological studies carried out to date have revealed the variety of anti-plasmodium, antioxidant, antimicrobial and antifungical properties of extracts and pure isolated compounds of the different species tested. The information summarized in this paper intends to serve as a reference tool to practitioners in the fields of etnopharmacology and chemistry of natural products.


El género Vismia es una fuente importante de productos naturales medicinales, es por esto que la información reunida en la presente revisión cubre los estudios más recientes en la etnofarmacología, farmacología y fitoquímica de este género. Antraquinonas y otros derivados quinoides, terpenos y constituyentes volátiles han sido reportados como los compuestos mayormente aislados de las diferentes especies de Vismia. Por otro lado, los estudios farmacológicos realizados hasta los momentos muestran las diversas propiedades antiplasmodicas, antioxidantes, antimicrobianas y antifúngicas que presentan tanto los extractos como los compuestos puros aislados de las diferentes especies ensayadas. La información resumida en este documento intenta servir de material de apoyo para investigadores en los campos de la etnofarmacologia y la química de productos naturales.

ABSTRACT

Do extrato em diclorometano do tronco de Kielmeyera cuspidata Saddi, Clusiaceae, foram isoladas cinco xantonas, 1,6-diidroxi-3,5-dimetoxixantona, 2-hidroxi-1-metoxixantona, 2,3-metilenodioxixantona, 4-hidroxi-2,3-metilenodioxixantona e 3-hidroxi-2,4-dimetoxixantona e dois triterpenos, α-amirina e β-amirina. O extrato exibiu toxicidade para Artemia salina e atividade antibacteriana para Micrococcus luteus, Bacillus subitilis, Staphylococcus aureus e Streptococcus mutans, não sendo ativo para Escherichia coli, Pseudomonas aeruginosa e Salmonela choleraesuis.


From the dichloromethane extract of trunk of Kielmeyera cuspidata five xanthones were isolated, 1,6-dihydroxy-3,5-dimethoxyxanthone, 2-hydroxy-1-methoxyxanthone, 2,3-methylenedioxyxanthone, 4-hydroxy-2,3-methylenedioxyxanthone and 3-hydroxy-2,4-dimethoxyxanthone and two triterpenes α-amirin and β-amirin. The extract displayed toxicity against Artemia salina and exhibited antibacterial activity against Micrococcus luteus, Bacillus subitilis, Staphylococcus aureus and Streptococcus mutans and it was not active against Escherichia coli, Pseudomonas aeruginosa and Salmonela choleraesuis.

ABSTRACT

Leiothrix (Eriocaulaceae) é um gênero exclusivo da América do Sul, com 37 espécies restritas ao Brasil. Leiothrix flavescens (Bong.) Ruhland e Leiothrix curvifolia (Bong.) Ruhland, são espécies endêmicas em áreas dos Estados de Minas Gerais e Bahia. Existem na literatura poucos estudos químicos e biológicos realizados com espécies desse gênero. Neste trabalho, foi estudada a atividade antioxidante pelo ensaio TEAC (Trolox Equivalent Antioxidant Capacity) e de autoxidação, utilizando ácido linoléico, de xantonas isoladas dos extratos etanólicos de L. curvifolia e L. flavescens. As substâncias apresentaram moderada atividade antioxidante, quando comparadas com padrões de quercetina e de BHT (2, 6 - ditercbutil - 4 - metóxifenil).


Leiothrix (Eriocaulaceae) is an exclusively South American genus of which 37 species are restricted to Brazil. Leiothrix flavescens (Bong.) Ruhland and Leiothrix curvifolia (Bong.) Ruhland, are species endemic to areas of Minas Gerais and Bahia States. Chemical and biological investigations of this genus are scarse. The objective of this work was to investigate the antioxidant effect using TEAC and linoleic acid assays of the xanthones isolated from the ethanolic extract of these species. These substances showed a moderate antioxidant activity when compared to quercetin and to BHT.
