ABSTRACT

OBJECTIVE: To analyze active ingredients,targets,pathways and biological processes of xianglian pills by means of network pharmacology and explore its pharmacodynamics material base and mechanism of action.METHODS: Active ingredients and corresponding targets of xianglian pills were obtained by using TCMSP database and BATMAN-TCM analytical platform,the network of xianglian pills active ingredients-targets was constructed by using Cytoscape 3.5.1 software, using the STRING platform to build protein-protein interaction; GO bioprocess enrichment and clustering analysis of target genes were finished by using BINGO and MCODE plug-ins; KEGG signaling pathways of target genes were analyzed by means of DAVID database.RESULTS: Eleven active ingredients in xianglian pills act on 291 potential targets. these targets participate in eighteen major GO biological process adjustment sub-clusters of the inflammatory response, leukotriene metabolism, lipid metabolism, apoptosis, smooth muscle contraction and platelet activation etc. involve in thirty-three signal pathways of seven categories: inflammation, metabolism,signal transduction, cancer, secretion of digestive juices, ardiovascular system etc.CONCLUSION: The different therapeutic value, pharmacodynamic substances and molecular mechanism of Xianglian pills in the treatment of inflammation,diabetes,cancer and cardiovascular diseases have been preliminarily elucidated, which provides new clues for the development of its clinical application.

ABSTRACT

Objective: Based on traditional Chinese medicine (TCM) inheritance system and integrated pharmacology platform for data mining of ulcerative colitis prescription, The difference of molecular mechanism between high-frequency herb combination and potential new prescriptions were screened to explore the potential molecular targets and signal pathways. Methods: The prescriptions of ulcerative colitis were collected from “Chinese medicine prescription dictionary”. Association rules and complex system entropy clustering were used to determine the frequency of usage and association rules of each drug, and identify high-frequency herb combinations and new potential prescriptions. By using integration platform, network construction and target prediction, the disease-target network, herb-target network, GO and KEGG pathway of ulcerative colitis related preparations of high frequency as well as potential new pairs were analyzed. The potential molecular mechanism of ulcerative colitis related targets was compared in high-frequency and new prescriptions by analysis of functions and pathways. Results: From 257 prescriptions, the frequency of usage and the association rules of prescription herb were analyzed. A total of six core combinations and three new potential prescriptions were identified, and the disease-target relationship obtained, and molecular mechanism differences between the high-frequency herb combination and the new potential prescription were compared. Conclusion: Combination of TCM inheritance system and integrated pharmacology platform can help to explore the TCM formula and potential molecular mechanism of ulcerative colitis treatment from a macro and micro perspective, and provide the guidance for the further research of classic prescription and its derivatives.

ABSTRACT

Objective To investigate the effects of mesalazine combined with Xianglian pills on cytokines and immunocytes in ulcerative colitis. Methods A total of 86 patients with ulcerative colitis from our hospital were collected and randomly divided into experimental group and control group with 43 cases in each group.Patients in the control group were treated by mesalazine, patients in the experimental group were treated by mesalazine combined with Xianglian pills treatment.The cytokines and immunocytes levels and efficacy between two groups were compared.Results After treatment, the clinical effective rate of the control group 69.77% was lower than the experimental group 88.37%, with statistical significance ( P <0.05 );compared with the control group, the TNF-αlevel of the experimental group was lower than the control group, IL-2, IL-4 and IL-10 levels were higher than the control group;the serum TFF1 and TFF3 levels were higher than the control group;CD3 +, CD4 +/CD8 +and NK cells levels were higher than the control group, the level of CD8 +was lower than the control group, with statistical significance ( P<0.05 ) .Conclusion Mesalazine combined with Xianglian pills in the treatment of patients with ulcerative colitis could reduce the inflammatory reactions and improve the clinical efficacy, with lower adverse reactions.

ABSTRACT

Objective To establish a determining method for the content of berberine hydrochloride in Coptis chinensis Franch.and Xianglian pills.Method The sample was extracted and developed,the berberine hydrochloride was determined by thin layer chromatograph-fluorescence of spectrophotometry with Benzen-ethylaceate-isopropanol-methanol-ammonia water(6:3:1.5:1.5:0.5) as the developing system and Ex=365 nm,Em=409 nm.Result The calibration curve was linear in the range of 0.473~ 3.784 ?g,r=0.999 3.The recovery rate in Coptis chinensis Franch.was 99.03%,RSD=1.86%,and in Xianglian pills was 97.10%,RSD=1.09%.Conclusion The method is simple and accurate.
