ABSTRACT

Chronic unpredicted mild stress(CUMS) combined with isolated feeding was used to induce depressed rat model. The anti-depressant effects of Zhizichi Decoction(ZZCD) and its solid fermented product(ZZC) were analyzed by behavioral test and comparison of pathological tissues of hippocampus and liver, metabolic characteristics of intestinal flora, and relative abundance of species. The results showed that ZZC could increase sucrose preference, shorten the immobility time in the forced swim test and tail suspension test(P<0.05), and repair damaged hippocampus and liver tissues, and the effect was superior to that of ZZCD. The results of Biolog ECO plates showed that the average well color development(AWCD) of intestinal flora in the model group significantly decreased and the metabolic levels of sugar and amino acids were reduced, while the AWCD of the treatment groups increased. The metabolic levels of the two carbon sources were improved in the ZZC group, while only sugar metabolic level was elevated in the ZZCD group. Metagenomic analysis of intestinal flora showed that the ratio of Firmicutes/Bacteroidetes was 3.87 in the control group, 21.77 in the model group, 5.91 in the ZZC group, and 18.48 in the ZZCD group. Lactobacillus increased by 3.28 times, and Prevotella and Bacteroidetes decreased by 75.59% and 76.39%, respectively in the model group as compared with that in the control group. Lactobacillus decreased by 31.13%, and Prevotella and Bacteroidetes increased by more than three times in the ZZC group as compared with that in the model group, while the corresponding changes in the ZZCD group were not significant. ZZC could improve depression-like beha-viors by regulating the structure of intestinal flora and metabolic functions and repairing damaged hippocampus and liver tissues in depressed rats, showing an anti-depressant effect superior to that of ZZCD. This study is expected to provide a basis for the development of new anti-depressant food products.

ABSTRACT

OBJECTIVE:To investigate t he effects of different compatibility ratio of Gardenia jasminoides to fermented soybean on the content of genistein and total flavonoids ,and to investigate the compatibility regularity of Zhizichi decoction. METHODS:The decoction method was used to prepare the mixed decoction with different compatibility ratio of G. jasminoides to fermented soybean (2∶1,1∶1,1∶2,1∶4,m/m,the same hereinafter ). UPLC-MS/MS method was used to determine the content of genistein in Zhizichi decoction with different compatibility ratio and corresponding fermented soybean single decoction. UV method was used to determine the content of total flavonoids in Zhizichi decoction with different compatibility ratio and corresponding gardenia single decoction and fermented soybean single decoction. RESULTS :The established method had good linearity , precision, repeatability, stability and accuracy. Compared with single decoction ,the content of genistein in the mixed decoction with different compatibility ratio of G. jasminoides to fermented soybean (2∶1,1∶1,1∶2,1∶4)was decreased to different extents , while the content of total flavonoids was increased to different extents. With the increase of fermented soybean ,the content of genistein in the decoction increased at first and then decreased. When the compatibility ratios of G. jasminoides to fermented soybean were 1 ∶ 1 and 1 ∶ 2,the content of genistein in the decoction was the highest (all 0.071 μg/mL). With the increase of fermented soybean ,the content of total flavonoids in the decoction did not change regularly ;when the ratio of G. jasminoides to fermented soybean was 1 ∶ 1,the content of total flavonoids in the decoction was the highest (1.861 μg/mL). CONCLUSIONS : When the compatibility ratio of G. jasminoides to fermented soybean was 1 ∶ 1,the content of flavonoids in the decoction is the highest.

ABSTRACT

OBJECTIVE: To explore the protective effect of Zhizichi Decoction extract against glutamic acid-induced neurotoxicity in PC 12 cells. METHODS: PC 12 cells were treated with glutamic acid to establish the cell model. Cell viability and membrane permeability were examined by MTT assay and LDH kit. The apoptosis rates were measured by flow cytometry using fluorescein labeling FITC-Annexin V/PI. The production of ROS was determined by flow cytometry. The levels of GSH and SOD in the cells were determined by kit respectively. RESULTS: Zhizichi Decoction extract significantly decreased the contents of LDH, enhanced the activities of SOD and GSH induced by glutamate. In addition, Zhizichi Decoction extract treatment decreased the intracellular ROS generation. CONCLUSION: Zhizichi Decoction extract can significantly ameliorate glutamic acid induced PC 12 cells injury. The mechanism may be partly through anti-oxidative stress.

ABSTRACT

Objective To optimize separation and purification technology of Zhizichi decoction by macroporous resina . Methods The content of total iridoid glycosides and total isoflavones were determined by ultraviolet spectrophotometry .Tak-ing the content of total iridoid glycosides and total isoflavones as indexes ,the effect of the concentration of sample solution , medicinal herbs on the amount of sample ,concentration and volume of eluent was investigated by single factor test .Results The optimum separation and purification technology was as following :the concentration of sample solution was 0 .1 g/ml ,the volume ratio of resin to material drug was 2∶1 ,the adsorption time was 2 h ,and the sample was firstly eluted with 1 BV wa-ter ,then 6 BV 20% ethanol and 60% ethanol ,and the eluent was collected .Conclusion This optimized separation and purifi-cation technology was reasonable and stable ,and it could be extended to large-scale production applications .

ABSTRACT

Objective To discuss the specific role of association rules in the compatibility of herbs and the relationship between herbs and symptoms by analyzing ancient analogous prescriptions Zhizichi Decoction with SQL Server software. Methods The prescriptions which contain Gardenia and Semen Sojae Preparatum were searched in the database of “Ancient Prescriptions for Epidemic Disease”, and they were imported to software with the standardized data of their symptoms and ingredients, then the proper association rules were found. The laws between parameter setting and rule generation were searched through parameter adjustment. Results Many laws of the drug compatibility and relationship between herbs and symptoms were obtained effectively in data mining by adjusting the parameters:The support setting was good for finding common used drug compatibility, and the confidence was good for finding specific drug compatibility and relationship between herbs and symptoms;the item set which had high frequency, sometimes made the rule’s importance low. Conclusion The laws of compatibility of herbs and the relationship between herbs and symptoms for data mining of analogous prescriptions are discovered by analyzing the support, confidence and importance of the association-rules and the clinical magnificence between symptoms and herbs.
