ABSTRACT

Objective To explore the significance and value of the anti-nucleosome antibodies(AnuA)in the diagnosis of system-ic lupus erythematosus(SLE).Methods The serum AnuA was detected in 177 patients with SLE,138 patients with other rheumat-ic diseases and 56 healthy controls by Western blot.The clinical manifestations,autoantibodies and other test results were recorded in the SLE patients.The AnuA and other autoantibodies were analyzed.Results The positive rate of AnuA in the SLE group was significantly higher than that in the disease control group,AnuA was negative in the healthy control group;the sensitivity of AnuA in the SLE group was 48.6% and the specificity was 95.3%;the sensitivity of AnuA was significantly higher than that of the anti-ds-DNA antibodies and anti-Sm antibodies,the difference had statistical significance (P <0.05).The specificity of AnuA was higher than that of ANA and histone (P <0.05 ).The sensitivity of joint detection of AnuA,anti-ds-DNA and anti-Sm antibodies was 89.8%,which was significantly higher than that of a single index detection.The positive rate of AnuA in the active period of SLE was significantly higher than that of the non-active period,moreover higher than that of the ds-DNA antibodies (P <0.05).Conclu-sion AnuA might participate in the pathogenesis of SLE.The joint detection of autoantibodies including AnuA,etc.has importance significance in the diagnosis,condition judgment and curative efficacy evaluation of SLE.

ABSTRACT

Objectives To investigate the changes and signiifcance of serum tumor necrosis factor-α(TNF-α) and anti-ds-DNA antibody in systemic lupus erythematosus (SLE) and lupus nephritis (LN) in children. Methods Sixty-seven children with SLE among whom 52 children had LN were selected and 30 healthy children were selected as normal controls. The level of serum TNF-αwas measured by enzyme-linked immunosorbent assay, and the anti-ds-DNA antibody level was measured by radioimmunoassay. Results The serum levels of TNF-αand anti-ds-DNA antibody were signiifcantly higher in children with SLE or LN than those in healthy controls (P all<0.01). Furthermore, the serum levels of TNF-αand anti-ds-DNA antibody were signiifcantly higher in active phase than in stationary phase of SLE and LN (P all<0.01). The serum TNF-αlevel was positively correlated with SLE disease activity index score and serum anti-ds-DNA level in 67 SLE patients (P<0.05). Conclu-sions The serum levels of TNF-αand anti-ds-DNA antibody were increased in children with SLE and LN and were positively correlated with disease activity.

ABSTRACT

Anti-F(ab')_2 and anti-dsDNA-F(ab'):fragment(idiotype)Were prepared from digested normal human IgG and anti-dsDNA antibody with pepsin.Anti-F(ab')_2 antibody was determined by ELISA in 46 patients with SLE.A significant correlation between titers of anti-dsDNA-F(ab')_2 and anti-F(ab'):antibodies was observed.Results show high levels of anti-F(ab')_2 and low levels of anti-dsDNA antibodies in patients with remisson SLE activity.These findings suggest that the general IgG-F(ab')_2 antibodies and the anti-dsDNA-F(ab')_2 idiotypes in SLE sera may express some common structural epitopes.The measurement of anti-F(ab')_2 antibody levels may prove usefuladditional parameters assessing the clinical status.
