ABSTRACT

Aim: The study investigated the anti-hyperglycemic and anti-hyperlipidemic potentials of methanol extracts of Piper guineense and Aframomum melegueta leaves with a view to utilizing the plants in the treatment and management of cardiovascular disorders. Methodology: Twenty-eight healthy albino rats were randomly divided into seven equal groups: Group I received normal saline (2 ml/kg bwt); Group II received a single dose of alloxan(150 mg/kg bwt) intraperitoneally; Group III received alloxan (150 mg/kg bwt) + glibenclamide (5 mg/kg bwt);Group IV received alloxan (150 mg/kg bwt) +PG (200 mg/kg bwt); Group V received alloxan (150 mg/kg bwt) + PG (400 mg/kg bwt); Group VI received alloxan (150 mg/kg bwt) + AM 200 (mg/kg bwt); Group VII received alloxan (150 mg/kg bwt) + AM (400 mg/kg bwt). The blood glucose level was determined before and after treatment with the extracts. The lipid: (total cholesterol (TC), triglycerides (TG), high density lipoprotein (HDL) and low density lipoprotein (LDL) were estimated using the Randox diagnostic kits. Results: The results revealed that alloxan was able to induce hyperglycemia at 150 mg/kg bwt and post-treatment with P. guineense and A. melegueta at 200 mg/kg and 400 mg/ kg bwt were able to significantly lower the blood glucose level which was quite apparent in AM treated groups. Also, the extracts at 200 mg/kg and 400 mg/kg were able to bring a significant (p < 0.05) reduction in TC, TG and LDL concentrations when compared to the alloxan treated group with the highest reduction in AM treated groups. Conclusion: These results revealed that the methanol extract of P. guineense and A. melegueta elicited anti-hyperglycemic and anti-hyperlipidemic potentials of the extracts with the highest effect observed in A. melegueta treated rats.

ABSTRACT Chemical profile analyses of artichoke (Cynara scolymus L., Asteraceae) edible parts (fleshy receptacle, inner bracts) as well as roots are compared with the commercially usable leaf extract using HPLC-DAD-ESI-MS via chlorogenicacid as a marker. Overall polyphenolic constituents demonstrated by means of LC/MS profiling. The nutritional values and inulin contents of different assessed parts were investigated. The present study was designed to determine the effect of artichoke: leaves, bracts, receptacles and roots alcoholic extracts against CCl4-induced acute hepatotoxicity and hyperlipidemia in rats by means of histopathological and biochemical parameters. Serum liver enzymes levels of aspartate amino transferase, alanine amino transferase, alkaline phosphatase and lipid peroxidase content (malondialdehyde MDA) were estimated. Blood glutathione, total cholesterol, triacylglycerides and high density lipid level were estimated in plasma. The ethanol extract of roots, leaves, bracts and receptacles were standardized to (0.82 ± 0.02, 1.6 ± 0.06, 2.02 ± 0.16 and 2.4 ± 0.27 mg chlorogenic acid/100 mg extract), respectively. The receptacle showed the highest content of polyphenols and exhibits the highest antioxidant activity. HPLC analysis of inulin in the receptacles of globe artichoke revealed high content of inulin (41.47 mg/g) dry extract. All artichoke parts contain comparable vitamins and minerals. Artichokes receptacles extract when taken in dose of (500 mg/kg/day) reduce the lesion caused by CCl4 alone more than groups receiving silymarin. Bracts and leaves extract exert nearly the same effect.

ABSTRACT

OBJECTIVE Diabetes mellitus is a chronic disease, for which there is no known cure except in a very specific situation. The available orthodox drugs used for its management are having disadvantages. This study investigated the anti-diabetic potential of aqueous and ethanol extracts of Strophanthus hispidus(SH)stem bark in Wistar rats.METHODS Glucose concentrations,serum α-amylase and lipid profile parameters of normal and diabetic rats were monitored for 12 weeks.Diabetes mellitus was induced by intraperitoneal injection of streptozotocin (55 mg·kg-1). 30 Male rats were randomly selected into six groups of five rats per group. Two groups served as normal and diabetic controls, respectively, and received distilled water for 12 weeks. Another two groups (normal rats) were treated orally with the aid of a gavage,250 mg·kg-1of aqueous and ethanol extracts of SH respectively for 12 weeks whereas the other two groups(diabetic rats)received the respective dose of aqueous and ethanol extracts for the same period. RESULTS The results revealed significant (P<0.05) progressive decrease in the fasting blood glucose concentration on the 2nd-12thweeks in normal rats, whereas the diabetic treated rats,showed reduction(P<0.05)in the fasting blood glucose concentration on the 4th-12thweeks.Total cholesterol,LDL and TG levels were lowered significantly(P<0.05)by the extracts in normal and diabetic treated rats, whereas HDL levels (17.6±0.50) mg·dL-1(aqueous) and (21.4±1.28) mg·dL-1(ethanol) were elevated in diabetic treated rats.The activity of serum α-amylase was also elevated by the extract in diabetic treated rats. CONCLUSION These findings indicated that SH showed hypoglycemic, anti-hyperlipidemic and anti-diabetic activities. These findings may be connected with the presence and amount of phytochemicals in the plant.

Abstract Securigera securidaca (L.) Degen & Döefl., Fabaceae, has been widely used in the Iranian, Indian and Egyptian folk medicine as antidiabetic and anti-hyperlipidemic remedy. Phenolic profiling of the ethanolic extract (90%) of the flowers of S. securidaca was performed via HPLC-DAD-MS/MS analysis in the positive and negative ion modes. The total polyphenols and flavonoids in the flowers were determined colorimetrically, and the quantification of their components was carried out using HPLC-UV. Total phenolics and flavonoids estimated as gallic acid and rutin equivalents were 82.39 ± 2.79 mg/g and 48.82 ± 1.95 mg/g of the dried powdered flowers, respectively. HPLC-DAD-MS/MS analysis of the extract allowed the identification of 39 flavonoids and eight phenolic acids. Quantitative analysis of some flavonoids and phenolics (mg/100 g powdered flowers) revealed the presence of isoquercetrin (3340 ± 2.1), hesperidin (32.09 ± 2.28), naringin (197.3 ± 30.16), luteolin (10.247 ± 0.594), chlorogenic acid (84.22 ± 2.08), catechin (3.94 ± 0.57) and protocatechuic acid (34.4 ± 0.15), in the extract. Moreover, the acute toxicity, hypoglycemic and hypolipidemic effects of the extract were investigated using alloxan induced diabetes in rats in a dose of 100, 200, and 400 mg/kg bwt. The ethanolic extract was safe up to a dose of 2000 mg/kg. All tested doses of the flower extract showed marked decrease in blood glucose level by 31.78%, 66.41% and 63.8% at 100, 200 and 400 mg/kg bwt, respectively, at p < 0.05. Regarding the anti-hyperlipidemic effect, a dose of 400 mg/kg of the flower extract showed the highest reduction in serum triacylglycerides and total cholesterol levels (68.46% and 51.50%, respectively at p < 0.05). The current study proved the folk use of the flowers of S. securidaca as anti-diabetic and anti-hyperlipidemic agent which could be attributed to its high phenolic content.

ABSTRACT

Tocotrienols are members of the vitamin E family essential for human nutrition. It has four isomeric forms i.e., α, β, γ, δ. Palm oil and rice bran oil represent two major nutritional sources of natural tocotrienols. Taken orally, tocotrienols are bioavailable to all vital organs. Apart from tocotrienols antioxidant, anti-inflammatory, antineurodegeneration, antimicrobial, anticancer properties it also has antihypercholesterolemic and antiangiogenic properties. During the last 7 years, tocotrienol research has gained substantial momentum. More than 75% of the entire PubMed literature on tocotrienols has been published on or after 2000. This represents major swing in the overall direction of tocotrienol research. The objective of this review is to highlight the potential significance of the tocotrienol in cardiovascular diseases, mainly anti hypercholesterolemic and anti hyperlipidemic properties along with its efficacy and safety.

ABSTRACT

Background: Glucova Active Tablet is a proprietary Ayurvedic formulation with ingredients reported for anti-hyperglycemic, anti-hyperlipidemic activity and antioxidant properties. Objective: Evaluation of anti-diabetic activity of Glucova Active Tablet on Type I and Type II diabetic model in rats. Materials and Methods: Experimental Type I diabetes was induced in 24 albino rats with intra-peritoneal injection of streptozotocin (50 mg/kg). Type II diabetes was induced in 18 albino rats by intra-peritoneal injection of streptozotocin (35 mg/kg) al ong with high fat diet. The rats were divided in 5 groups for Type I model and 4 groups for Type II model. Normal control group was kept common for both experimental models. Glucova Active Tablet (108 mg/kg) treatment was provided for 28 days twice daily orally. Fasting blood glucose level, serum lipid profi le and liver anti-oxidant parameters like superoxide dismutase and reduced glutathione was carried out in both experimental models. Pancreas histopathology was also done. Statistical analysis were done by 'analysis of variance' test followed by post hoc Tukey’s test, with signifi cant level of P < 0.05. Results and Discussion: Glucova Active Tablet showed signifi cant effect on fasting blood glucose level. It also showed signifi cant alteration in lipid profi le and antioxidant parameters. Histopathology study revealed restoration of beta cells in pancreas in Glucova Active Tablet treated group. Conclusion: Finding of this study concludes that Glucova Active Tablet has shown promising anti-diabetic activity in Type I and Type II diabetic rats. It was also found showing good anti-hyperlipidemic activity and anti-oxidant property.

ABSTRACT

DWP208 is a sodium succinate form of ZYM-201 which is a triterpenoid glycoside isolated from Sanguisorba officinalis, a medicinal plant prescribed for various diseases, such as duodenal ulcers and bleeding in East Asian counties. We demonstrated that this compound is able to normalize the altered lipid metabolism induced by hyperglycemia and a high fat diet. In this study, we determined whether hyperlipidemic conditions induced with chronically treated alcohol can also be restored by DWP208. Similar to our previous results, orally administered DWP208 (1 to 10 mg/kg) also ameliorated the hyperlipidemia that was induced by alcohol. This compound reversed the alcohol-induced hyperlipidemia including (i) up-regulated hyperlipidemic parameters such as low-density lipoprotein (LDL), very low-density lipoprotein (VLDL), atherosclerotic index (AI), triglyceride, and total cholesterol, and (ii) down-regulated hyperlipidemic parameters such as absolute body weight, superoxide dismutase (SOD) activity, and high-density lipoprotein (HDL) in serum and liver. According to our data, the ameliorative activity of DWP208 is due to its indirect anti-oxidative activity as a result of which lipid peroxide and hydroxyl radical levels were reduced and the activity of SOD was enhanced. Therefore, our data strongly suggest that DWP208 can be used as a remedy against alcohol-induced hyperlipidemia.

ABSTRACT

Ten caffeoylquinic acids (CQAs) were obtained from the anti-hyperlipidemic extract of fruits of Pandanus tectorius. All of the compounds were isolated and purified by various column chromatographies especially by preparative HPLC method. Their structures were determined under the aid of spectroscopic methods. All compounds were isolated from genus pandanus for the first time. The anti-hyperlipidemic activities of three compounds characterized with high content in the fruits were tested in HepG2 cells.The three CQAs all significantly reduced the intracellular content of TC and TG.

ABSTRACT

O presente trabalho verificou a utilização de plantas medicinais encontradas no Cerrado mato-grossense para o tratamento de hiperlipidemias e obesidade. Entrevistas com 180 pessoas acima de 50 anos foram realizadas em Nova Xavantina-MT. O questionário abordou nome popular, parte utilizada, forma de preparo e uso das espécies citadas, além de informações gerais sobre o uso de plantas. As dez plantas mais citadas foram coletadas, identificadas e estudadas por meio de uma revisão bibliográfica. A maioria dos entrevistados (95,6 por cento) declarou utilizar plantas medicinais regularmente, sendo que 71,5 por cento deles herdaram o conhecimento sobre plantas dos pais e avós e 94,20 por cento relataram aconselhar o uso aos mais jovens. Além disso, 93,6 por cento atestaram que as plantas são mais eficazes que os medicamentos de farmácia e 57 por cento consideraram forte o efeito das mesmas, ou seja, sempre resolvem o problema de saúde. Quanto aos efeitos adversos, 95,9 por cento disseram nunca ter sentido após o uso de plantas. Dos entrevistados, 56,7 por cento conheciam ou já haviam utilizado plantas medicinais no tratamento de hiperlipidemias e obesidade, sendo citadas 54 espécies medicinais diferentes pertencentes a 53 gêneros e 38 famílias, com destaque para Fabaceae (13 por cento). As dez plantas mais citadas foram: guatambu (Aspidosperma tomentosum Mart.), quina-do-cerrado (Strychnos pseudoquina St. Hil.), ipê-roxo [Tabebuia impetiginosa (Mart. Ex DC.) Standl], embaúba (Cecropia pachystachya Trec.), calunga (Simaba sp.), pata-de-vaca [Bauhinia rufa (Bong.) Steud.], mangaba (Hancornia speciosa Gomez), batata-de-tiú [Jatropha elliptica (Pohl.) Muell. Arg.], folha-de-carne (Casearia sylvestris Sw.) e manacá (Spiranthera odoratissima A. St.-Hil.). A folha foi a parte mais utilizada (46 por cento) e o preparo das plantas ocorre principalmente por meio de infusão citado por 36,5 por cento dos entrevistados. Apesar do uso popular destas plantas no combate as hiperlipidemias e obesidade, há necessidade de estudos fitoquímicos e farmacológicos que comprovem estas atividades, com vistas ao desenvolvimento de um fitoterápico.


This study checked the use of medicinal plants found in the Cerrado mato-grossense for the treatment of hyperlipidemia and obesity. Interviews with 180 people over 50 years were held in Nova Xavantina-MT. The questionnaire addressed popular name, part used, type of preparation and use of the species mentioned, beyond general information about the use of plants. The ten plants most cited were collected, identified and studied through a literature review. Most respondents (95.6 percent) said using medicinal plants regularly, while 71.5 percent of them inherited the knowledge of plants of parents and grandparents and 94.20 percent reported the use to advise youngsters. Furthermore, 93.6 percent attested that the plants are more effective than the drugs from pharmacy and 57 percent considered the strong effect of the same, i.e., they always solved health problems. Regarding adverse effects, 95.9 percent said they have never felt them after the use of plants. Of the respondents, 56.7 percent knew or had used medicinal plants in the treatment of hyperlipidemias and obesity, being cited 54 medicinal species belonging to 53 different genera and 38 families, with emphasis on Fabaceae (13 percent). The ten most cited were: guatambu (Aspidosperma tomentosum Mart.), quina-do-cerrado (Strychnos pseudoquina St. Hil.), ipê-roxo [Tabebuia impetiginosa (Mart. Ex DC.) Standl], embaúba (Cecropia pachystachya Trec.), calunga (Simaba sp.), pata-de-vaca [Bauhinia rufa (Bong.) Steud.], mangaba (Hancornia speciosa Gomez), batata-de-tiú [Jatropha elliptica (Pohl.) Muell. Arg.], folha-de-carne (Casearia sylvestris Sw.) and manacá (Spiranthera odoratissima A. St.-Hil.). The leaf was the most used (46 percent) and the preparation of the plants occurs mainly through the infusion cited by 36,5 percent of respondents. Despite the popular use of these plants for treatment of obesity and hyperlipidemia, further chemical and pharmacological studies are required to demonstrate these activities, in order to develop a phytotherapic product.
